Circadian regulator PER1 inhibits osteoclastogenesis by activating inflammatory genes.

Disruption of circadian rhythms predisposes shift workers to many chronic conditions, including osteoporosis. However, the effects of disrupted circadian rhythms on bone remodeling remain largely unknown. Here, we show that one of the core circadian regulators PER1 inhibits osteoclastogenesis by upregulating genes involved in inflammation. Conditional knockout (KO) of Per1 in osteoclasts and related cells resulted in decreased bone mass in the femurs of mice, along with increased osteoclasts and decreased osteoblasts. Osteoclastogenesis was also promoted by Per1 depletion in vitro with 16 down

Single-cell transcriptome profiling reveals the heterogeneity of ossification of the posterior longitudinal ligament (OPLL) and its immune microenvironment.

BACKGROUND: Ossification of the posterior longitudinal ligament (OPLL) is a heterotopic ossification process of the posterior longitudinal ligament (PLL) that can compress the spinal cord and nerve roots, yet its cellular heterogeneity and pathogenesis remain unclear. METHODS: We performed single-cell RNA sequencing (scRNA-seq) with integrative computational analyses and histological validation on 4,683 cells from surgically resected cervical OPLL lesions obtained from human patients. RESULTS: We identified 15 major cell subsets, including chondrocyte-lineage populations-progenitor/prolifera

TNF-α blockade with adalimumab restores bone metabolism in rheumatoid arthritis by regulating cytokines and inflammatory cell infiltration.

OBJECTIVE: To investigate the mechanisms underlying the bone-sparing effects of the anti-TNF antibody adalimumab in rheumatoid arthritis (RA) by examining synovial tissue and its direct effect on osteoblasts and osteoclasts. METHODS: Paired synovial biopsies from 12 patients with RA were obtained before and 8 weeks after initiation of adalimumab. Immune-cell infiltrates (CD163+ macrophages, CD3+ lymphocytes) and expression of osteoprotegerin (OPG), receptor activator of nuclear factor κB (RANK), and its ligand (RANKL) were assessed by immunohistochemistry. In vitro, the effect of adalimumab o

[Exercise improves osteoporosis through bone-fat crosstalk].

Osteoporosis (OP) is a prevalent systemic metabolic bone disease characterized by reduced bone mass and compromised bone microstructure. Recent studies have demonstrated that the occurrence of OP is closely related to the imbalance of bone and lipid metabolism. Bone and lipid can communicate and influence each other by secreting bone-derived factors or adipokines. This kind of crosstalk is crucial for maintaining the equilibrium between bone and fat. Meanwhile, multiple signaling pathways such as Wnt/β-catenin, bone morphogenetic protein (BMP), mitogen-activated protein kinase (MAPK), peroxiso

Research Progress of Multiple Molecular Signal Pathways in the Pathogenesis of Steroid-induced Osteonecrosis of the Femoral Head.

Steroid-induced osteonecrosis of the femoral head (SONFH) is a terrible side effect of glucocorticoid therapy that causes problems with microvascular integrity, lipid metabolism, and a range of programmed cell death pathways. Recent studies have shown that autophagy can have different effects on osteoblasts and endothelial cells depending on the situation. These effects are mediated by the AMPK/mTOR, PINK1/Parkin, SIRT1/FoxO1, and PI3K/Akt/mTOR pathways. At the same time, apoptosis is caused by lowering Wnt/β-catenin, raising STAT1/caspase-3, and throwing off the balance of the OPG/RANKL/RANK

Ferric carboxymaltose increases fracture risk in patients and reduces bone formation in mice with iron deficiency anemia.

Modern IV iron formulations allow treatment of iron deficiency anemia (IDA) with 1 or 2 infusions. Ferric carboxymaltose (FCM) is a widely used IV iron, which causes hypophosphatemia in most of the patients. Osteomalacia and fractures are increasingly recognized after repeated infusions of FCM. It is unknown why ferric derisomaltose (FDI) rarely causes hypophosphatemia. In this study, we compare the effects of FCM and FDI on fracture risk and investigate potential underlying mechanisms explaining the different effects on bone and mineral metabolism. For this aim, fracture rate and osteomalacia

Isolation and characterization of mesenchymal stem cells from nasal polyps and olfactory mucosa: a comparative analysis of proliferation and multi-lineage differentiation capacity.

BACKGROUND: Nasal polyps (NPs) and olfactory mucosa (OM) are emerging as potential sources of mesenchymal stem cells (MSCs), which hold promise for regenerative medicine due to their multipotent differentiation capabilities. This study aimed to isolate MSCs from nasal polyps (NP-MSCs) and evaluate their differentiation potentials in comparison with healthy olfactory mucosa (OM-MSCs). METHODS: 16 human samples from NP and OM tissues were studied and MSCs were isolated enzymatically. Flow-cytometry analysis was done to characterize the isolated cells; then, tri-lineage differentiation capacity o

The growth plate: Zonal architecture, plasticity, and endocrine control of linear growth.

The growth plate (physis) is a highly specialized cartilaginous organ that drives longitudinal bone growth and ultimately determines adult stature. Its zonal architecture, including the resting, proliferative, hypertrophic, and calcification zones, integrates stem-like progenitor activity, clonal chondrocyte expansion, matrix remodeling, vascular invasion, and replacement by bone. Here, we review how the structural organization of the growth plate emerges from the interplay among extracellular matrix composition, sulfation pathways, and canonical paracrine signaling pathways, including Ihh-PTH

The effects of FOXC2-gene-manipulated human periodontal ligament stem cells on bone regeneration of craniofacial bone defect.

The progress and development of bone tissue engineering technology has brought new hope for the repair of oral and maxillofacial bone defects. As one of the ideal seed cells, the induction of human periodontal ligament stem cells (hPDLSCs) into osteoblasts has become a focus in current research. Forkhead box C2 (FOXC2) may be a good candidate gene for bone regeneration. Therefore, the aim of this study was to verify the role of FOXC2 in hPDLSCs osteogenesis and genetically-manipulate seed cells based on FOXC2 gene for enhancing their bone regeneration potentials. The hPDLSCs models with down-r

Activation of GLP-1 receptors enhances the osteogenic differentiation process of STRO-1-positive BMSCs.

BACKGROUND: Osteoporosis is a prevalent metabolic bone disorder, especially in individuals with diabetes. Recent studies suggest that GLP-1 plays a positive role in regulating bone metabolism. However, the impact of GLP-1R on bone marrow-derived mesenchymal stem cells, particularly STRO-1-positive BMSCs, remains unclear. This study aims to investigate the impact of GLP-1R activation on the osteogenic differentiation potential of STRO-1-positive BMSCs. METHODS AND RESULTS: BMSCs were isolated from the femurs and tibias of Sprague-Dawley rats, characterized by flow cytometry and immunofluoresce

DEC1 promotes breast cancer bone metastasis through transcriptional activation of CXCR4.

Bone metastasis is the primary cause of mortality in breast cancer (BC) patients. The present study elucidates the functional role of the differentiated embryonic chondrocyte-expressed gene 1 (DEC1) in promoting BC-related bone metastasis. Analysis of patient-derived samples and public databases revealed a significant upregulation of DEC1 and CXCR4 in breast tumors compared with adjacent normal tissues, with elevated levels correlating with increased metastatic potential, suggesting their synergistic involvement in BC progression. Intracardiac injection experiments demonstrated that Dec1-WT 4T

Astaxanthin Prevents Glucocorticoid-Induced Femoral Head Osteonecrosis by Targeting Ferroptosis through the JAK2/STAT3 Signaling Pathway.

Glucocorticoid (GC) is extensively used in clinical practice, and the osteonecrosis of the femoral head caused by them is a common issue in orthopedic surgery, yet the underlying mechanisms remain unclear. Astaxanthin (AST), a potent natural antioxidant, has an unexplored impact on GC-induced osteonecrosis of the femoral head (GIONFH). This study explores the effects and mechanisms of AST in counteracting dexamethasone (Dex)-induced ferroptosis and GIONFH. We developed a rat model of GIONFH using intraperitoneal Dex injections and conducted in vitro analysis by culturing osteoblasts (OBs) with

Dendrobine promotes bone formation via the canonical Wnt/β-catenin signaling pathway and prevents postmenopausal osteoporosis.

BACKGROUND: In early postmenopausal women with sex steroid deficiency, an imbalance between bone resorption and bone formation causes an accelerated decline in bone mineral density due to a longer lifespan of osteoclasts and a shorter lifespan of osteoblasts. Given this context, dendrobine, as a primary bioactive alkaloid isolated from Dendrobium nobile Lindl., has potential efficacy on osteoporosis due to the traditional use of Dendrobium nobile Lindl. in musculoskeletal disease. OBJECTIVE: To investigate the anti-osteoporotic efficacy of dendrobine in increasing osteoblastogenesis and to el

The gut-bone axis in osteoporosis: a multifaceted interaction with implications for bone health.

With the accelerated aging of the population, degenerative orthopedic diseases, particularly osteoporosis, have become a major public health challenge, threatening bone health and affecting the quality of life. Existing anti-osteoporosis regimens remain rather unitary or poorly adhered, which also limits the maintenance of bone health to some extent. Given the increasingly elucidated prominence of gut-related factors in osteoblasts/osteoclasts and bone formation/metabolism/maintenance, focusing on intestinal microecology and then targeting the distal bone tissue via the gut-bone axis have been

Osteocytes: master orchestrators of skeletal homeostasis, remodeling, and osteoporosis pathogenesis.

The skeleton functions as an endocrine organ. Osteocytes maintenance of skeletal strength and energy balance by sensing mechanical stress and communicating with surrounding cells. They are currently considered key regulators of bone remodeling, mineral metabolism, and systemic homeostasis. Osteocytes originate from osteoblasts and are embedded in the lacunar-tubular network. They express proteins such as DMP1, sclerostin, and FGF23, and influence Wnt signaling, the RANKL/OPG axis, and phosphate metabolism. We review the latest studies in the field of osteocyte biology, focusing on their mechan

Effects of microplastics on the bones: a comprehensive review.

A growing role for the skeleton in human health and longevity emerged recently. However, knowledge regarding the effects of new risk factors, linked to modern life, for bone diseases is not documented satisfactorily. Micro- and nanoplastics (MPs) are environmental and health concerns due to their widespread detection in the human surrounding environment. This narrative review aims to critically summarize current knowledge on the effects of MPs on bone and bone marrow and to explore potential links to human bone diseases, such as osteoporosis. Worryingly, MPs were recently detected in human bon

Vascular environment-responsive DNA nanoswitch controls the positive feedback system for spatiotemporal coupling of angiogenesis and osteogenesis.

Vascularization-ossification coupling is pivotal for bone defect repair. Neovascularization guides the migration of osteogenic precursor cells, and osteoblasts in the perivascular area support vascular regeneration through the secretion of cytokines. Delivery systems are being engineered to orchestrate the coupling of vascularization and ossification precisely. However, most delivery systems lack sensitivity to the inherent repair environment, hampering timely bone formation. This study developed a vascular environment-responsive biomaterial: a true bone ceramic/small intestine submucosa (TBC/

Periodontal ligament tissues support neutrophil differentiation and maturation processes.

INTRODUCTION: Periodontal ligament is the soft connective tissue joining the roots of teeth with alveolar bone. The periodontal ligament presents significant cellular heterogeneity, including fibroblasts, endothelial cells, cementoblasts, osteoblasts, osteoclasts, and immune cells such as macrophages and neutrophils. These cells have crucial roles for periodontium homeostasis and function. However, certain cell types, such as neutrophils, remain poorly characterized in this tissue, despite their natural abundance and relevance in processes and diseases affecting the periodontal ligament. Does crocin create new hope for the treatment of oral problems? A focus on periodontitis.

According to the World Health Organization (WHO) reports, oral health has an indispensable role in the maintenance of human public health. However, oral problems, especially periodontitis, are known as bad players in this issue. Periodontitis, as the most prevalent oral disease, is a type of chronic illness mediated by bacterial pathogens and immune system reactions, which is linked with the destruction of tooth-protecting tissues, such as alveolar bone and periodontal ligament. Periodontitis has a high prevalence (over 40% in the United States) and can be associated with other systemic ailmen
