The Mechanism of G Protein-Coupled Receptor Regulation of Ferroptosis in Hepatic Ischemia-Reperfusion Injury.

Hepatic ischemia-reperfusion injury (HIRI) is a significant clinical challenge in the field of liver surgery and transplantation, and its pathological mechanisms are complex. In recent years, ferroptosis, a novel form of iron-dependent programmed cell death, plays a central role in this injury process. G protein-coupled receptors (GPCRs), as the largest family of membrane receptors in the body, regulate cellular stress and death through extensive signaling networks. This review elucidates the specific molecular mechanisms by which GPCRs regulate ferroptosis in HIRI by affecting key pathways su

How Lorentz Boosts Reshape Relaxation Spectra.

In relativity, relaxation processes are often assumed to undergo time dilation under Lorentz boosts. We show that this intuition fails generically. Because of relativity of simultaneity, Lorentz boosts can split a single relaxation mode into a continuum of excitations, with a width set by the maximal signal propagation speed. Focusing on linearized relativistic (kinetic or rheological) theories with an Onsager-type symmetry, we derive rigorous bounds on relaxation spectra in arbitrary inertial frames, expressed solely in terms of rest-frame spectral data at zero wave number. As a consequence,

Potential Roles of Prostaglandins PGD2, PGJ2, and PGE2 in the Pathogenesis of Systemic Lupus Erythematosus.

Prostaglandins (PGs) are eicosanoid compounds with various hormone-like effects within the human body. Prostaglandin D2 (PGD2), prostaglandin J2 (PGJ2, a metabolite of PGD2), and prostaglandin E2 (PGE2) are common prostaglandins. They are produced from arachidonic acid by cyclooxygenase isoenzymes (COX-1 and COX-2) and PG synthases and are involved in many biological metabolisms, including the inflammatory response. Recent studies show PGs are mediators for both proinflammatory and anti-inflammatory processes. In systemic lupus erythematosus (SLE), inflammation caused by autoimmunity damages t

Muscle-to-CNS signaling in physiological homeostasis, aging, and disease.

The central nervous system (CNS) orchestrates homeostatic responses and organismal behaviors by integrating cues from the whole body. Like other peripheral tissues, skeletal muscle can signal to the brain, and this occurs via muscle-secreted signaling factors (myokines/myometabolites). In this review article, we examine exercise-induced myokines and myometabolites that improve cognitive capacity and impede neurodegeneration and, conversely, detrimental myokines secreted by diseased muscles that negatively impact brain function. Cellular processes modulated by myokines in the CNS include proteo

The Heart's Electromagnetic Field in Emotions, Empathy and Human Connection: Biosensor-Derived Insights into Heart-Brain Axis Mechanisms and a Basis for Novel BioMagnetoTherapies.

The heart's electromagnetic field (HEMF) represents the strongest magnetic signal in the human body and has been increasingly associated with processes related to the Heart-Brain Axis (HBA). The present review summarizes its biophysical basis along with current and emerging biosensing technologies. It examines hypotheses regarding interpersonal interactions and interactions with external fields, including geomagnetic activity, and reviews evidence linking the HEMF to autonomic activity and emotional states. It provides an overview of magnetic field-based therapeutics, introduced here as our ow

Organ symmetry establishment during gynoecium development.

Symmetry is a key factor in the morphological diversity and reproductive success of angiosperms (flowering plants). The gynoecium, the female reproductive organ of the flower, displays remarkable variation in symmetry types, ranging from bilateral to radial, from its base (ovary) to its apex (style). Proper tissue growth and differentiation occur along the body axes to form three-dimensional structures and establish symmetric forms within the organ. In this review, we summarise the latest understanding on cellular, molecular and genetic mechanisms governing pivotal symmetry changes during gyno

Inter-Organ Crosstalk and Regulatory Mechanisms of Skeletal Muscle-Derived Extracellular Vesicles in Systemic Metabolic Homeostasis.

Extracellular vesicles (EVs), including exosomes, play a pivotal role in intercellular communication by facilitating the transfer of bioactive molecules between cells. These vesicles, which encompass a variety of subtypes such as exosomes, microvesicles and apoptotic bodies, carry functional proteins, mRNAs, miRNAs and other molecular cargo that influence various physiological processes. In particular, skeletal muscle-derived EVs have recently emerged as a novel category of myokines, contributing to muscle homeostasis through paracrine signalling and exerting systemic endocrine effects on meta

The conundrum of esophagogastric junction outflow obstruction: Answers to key clinical questions.

Esophagogastric junction outflow obstruction (EGJOO) is a heterogeneous disorder characterized by abnormal esophagogastric junction residual pressures in the setting of intact esophageal body peristalsis. This manometric finding is most clinically relevant in patients with dysphagia and chest pain, and may arise from various mechanisms, including structural processes, catheter positioning artifact, and medications. Rarely, manometric EGJOO is a consequence of abnormal relaxation of the lower esophageal sphincter (LES) akin to achalasia. Thus, it is critical in the evaluation of EGJOO to determ

Tribocorrosion of Metallic Biomaterials under Simulated Inflammation Conditions: Current Status and Future Prospects.

Metallic biomaterials are widely employed in orthopedic, dental, and other clinical applications. Tribocorrosion is the combined effect of mechanical wear and corrosion and hence has become one of the crucial limitations of metallic biomaterials. The tribocorrosion of metallic biomaterials can release metal ions and wear debris into bodily fluids, leading to inflammatory reactions in the human body. Inflammatory mediators from the immune cells, for example, reactive oxygen species and acids, can further influence the tribocorrosion process and accelerate the failure of the implanted prosthesis

[Surgical Strategy for Pseudoarthrosis and Delayed Nerve Palsy after Osteoporotic Vertebral Fracture].

Osteoporotic vertebral fractures are among the most common fractures associated with osteoporosis. Approximately 80% of cases heal by fusion in the natural process without the need for surgery, and if we include the so-called "incidental vertebral body fractures," which are discovered by chance during radiography, the frequency of prolongation of back pain due to pseudoarthrosis or delayed-onset neurological deficits is low. However, the prognosis of osteoporotic vertebral fractures is affected by the number of fractured vertebrae. In patients who become bedridden because of prolonged pain due

The carotid body oxygen sensor.

Carotid body (CB) chemoreceptor glomus cells sense hypoxia through the inhibition of plasmalemmal K+ channels, which leads to the opening of Ca2+ channels, Ca2+ influx, and neurotransmitter release. The mechanism of O2 sensing and the regulation of membrane ion channels by O2 have remained undefined and a subject of debate. Here, we summarize the molecular pathway that underlies acute O2 sensing in the CB. This process does not rely on a single-molecule O2 sensor expressed in glomus cells but rather on HIF2α-dependent genetically specialized mitochondria that can detect changes in O2 tension,

Biochemical and medical aspects of vitamin B1 research.

Vitamin B1 is an indispensable food factor for the human and animal body. In animals, vitamin B1 is found in the form of thiamine and its phosphate esters - thiamine mono-, di- and triphosphate, as well as an adenylated derivative - adenosine thiamine triphosphate. At present, the only vitamin B1 form with biochemical functions being elucidated is thiamine diphosphate, which serves as a coenzyme for several important enzymes involved in carbohydrate, amino acid, fatty acid and energy metabolism. Here we review the latest developments in the field of vitamin B1 research in animals. Transport, m

The use of imaging tests in paediatric button battery ingestion complications.

The ingestion of foreign bodies is a common issue among children, especially those under the age of three. Most pass through the gastrointestinal tract without any complications. However, button batteries are associated with a higher risk of complications, particularly in infants or when batteries are larger, due to the risk of oesophageal impaction, which can cause severe injuries. The initial clinical presentation is ambiguous, delaying and complicating diagnosis. There are times when complications are only suspected after symptoms become severe. Radiography, oesophagography, computerised to

Th9-derived IL-9 in autoimmune diseases: An update.

Autoimmune diseases (AIDs), such as rheumatoid arthritis, multiple sclerosis, and systemic lupus erythematosus, are caused by abnormal immune response that targets the body's own tissues. T-helper cell 9 (Th9) is a recently discovered subpopulation of T cells that primarily participates in inflammatory responses and immune regulation by secreting interleukin-9 (IL-9). IL-9 is a multifunctional cytokine that regulates the function of T and B cells, and also affects the activation and response of mast cells, macrophages, and other cells. Studies have shown that Th9 cells significantly affect the

An accelerated Parkinson's disease monkey model using AAV-α-synuclein plus poly(ADP-ribose).

The etiology of Parkinson's disease (PD) remains elusive, and the limited availability of suitable animal models hampers research on pathogenesis and drug development. We report the development of a cynomolgus monkey model of PD that combines adeno-associated virus (AAV)-mediated overexpression of α-synuclein into the substantia nigra with an injection of poly(ADP-ribose) (PAR) into the striatum. Our results show that pathological processes were accelerated, including dopaminergic neuron degeneration, Lewy body aggregation, and hallmarks of inflammation in microglia and astrocytes. The injured axon: intrinsic mechanisms driving axonal regeneration.

Injury to the central nervous system (CNS) often results in permanent neurological impairments because axons fail to regenerate and re-establish lost synaptic contacts. By contrast, peripheral neurons can activate a pro-regenerative program and regenerate following a nerve lesion. This relies on an intricate intracellular communication system between the severed axon and the cell body. Locally activated signaling molecules are retrogradely transported to the soma to promote the epigenetic and transcriptional changes required for the injured neuron to regain growth competence. These signaling e

The Role of Intestinal Microbiota and Diet as Modulating Factors in the Course of Alzheimer's and Parkinson's Diseases.

Many researchers propose manipulating microbiota to prevent and treat related diseases. The brain-gut axis is an object that remains the target of modern research, and it is not without reason that many researchers enrich it with microbiota and diet in its name. Numerous connections and mutual correlations have become the basis for seeking answers to many questions related to pathology as well as human physiology. Disorders of this homeostasis as well as dysbiosis itself accompany neurodegenerative diseases such as Alzheimer's and Parkinson's. Heavily dependent on external factors, modulation

Induction of Meiosis from Human Pluripotent Stem Cells.

An in vitro model of human meiosis would accelerate research into this important reproductive process and development of therapies for infertility. We have developed a method to induce meiosis starting from male or female human pluripotent stem cells. We demonstrate that DNMT1 inhibition, retinoid signaling activation, and overexpression of regulatory factors (anti-apoptotic BCL2, and pro-meiotic HOXB5, BOLL, or MEIOC) rapidly activates meiosis, with leptonema beginning at 6 days, zygonema at 9 days, and pachynema at 12 days. Handling the sugar rush: the role of the renal proximal tubule.

Blood glucose homeostasis is critical to ensure the proper functioning of the human body. Through the processes of filtration, reabsorption, secretion, and metabolism, much of this task falls to the kidneys. With a rise in glucose and other added sugars, there is an increased burden on this organ, mainly the proximal tubule, which is responsible for all glucose reabsorption. In this review, we focus on the current physiological and cell biological functions of the renal proximal tubule as it works to reabsorb and metabolize glucose and fructose. We also highlight the physiological adaptations
