Effects of dietary Zanthoxylum bungeanum seed powder on growth performance, immune function, and cecal microbiota in broilers.

This study was conducted to evaluate the effects of Zanthoxylum bungeanum seed powder (ZBS) on growth performance, immune function, and cecal microbiota in 817 broilers. A total of 260 one-day-old 817 broilers were randomly assigned to 4 treatment groups (CON, ZBS1, ZBS2, and ZBS3) with 5 replicates per treatment and were fed diets containing 0, 1%, 2%, or 3% ZBS, respectively, for 63 d. The results showed that dietary ZBS had no significant effects on average daily feed intake, average daily gain, or gain-to-feed ratio (P > 0.05). However, body weight at 28 and 63 d and average daily gain dur

Testosterone plus lifestyle therapy improves skeletal muscle glycolysis in older men with obesity and hypogonadism.

OBJECTIVE: Weight loss in older men with obesity and hypogonadism accelerates musculoskeletal decline, yet the underlying metabolic mechanisms remain unclear. Testosterone replacement therapy (TRT), when added to lifestyle therapy (LT), mitigates this decline, but its metabolic basis has not been defined. We examined skeletal muscle metabolomic adaptations to LT with or without TRT, focusing on glycolysis, the pentose phosphate pathway (PPP), the tricarboxylic acid (TCA) cycle, and carnitine metabolism to identify dominant pathways of metabolic adaptation. DESIGN: Randomized, double-blind, pl

Integrated Multi-Omics Analysis Reveals Molecular Features Associated with Energy Metabolism Adaptations in Brooding Taihe Black-Bone Silky Fowls.

Broodiness is a natural behavior whereby female birds stop laying eggs to sit on and hatch them. This behavior is regulated through genetic, hormonal, and environmental factors. The Taihe Black-Boned Silky Fowl (TBSF), a Chinese traditional domestic breed, exhibits a strong brooding tendency; however, the molecular mechanisms underlying this trait remain unclear. In this study, we performed an integrated multi-omics analysis to characterize differences between two groups of TBSF hens: 8 individuals undergoing 30 days of active brooding (BR30) and 8 individuals in the normal laying egg stage (N

Exposure to known and emerging groundwater contaminants significantly alters poultry microbiome and metabolome.

UNLABELLED: The exposome encompasses all lifetime environmental exposures affecting health. Its complexity and high data dimensionality make it challenging to link specific exposure combinations to adverse health outcomes. Establishing relevant exposome criteria is key to addressing current knowledge gaps. This study evaluated contaminant levels in Wisconsin groundwater and their effects on host health. We focused on three co-occurring chemicals that were detected at concentrations exceeding groundwater standards: nitrate, atrazine, and imidacloprid, and the emerging contaminant, microplastics

Liver-originated selective autophagy of BTD by alectinib underlies concurrent hepatotoxicity and dermatotoxicity.

Alectinib serves as an indispensable treatment for ALK (ALK receptor tyrosine kinase)-positive non-small-cell lung cancer (NSCLC), yet its hepatotoxicity and dermatotoxicity pose significant clinical concerns due to poorly understood mechanisms. This study demonstrated that alectinib-induced dermatotoxicity was secondary to hepatotoxicity. Integrated multi-omics analysis revealed that alectinib triggered excessive macroautophagic/autophagic degradation of hepatic BTD (biotinidase), causing systemic biotin deficiency that drove both hepatocyte apoptosis and skin barrier dysfunction. Mechanistic

The impact of working onboard ships on individual metabolic profiles: Comparing pre-embarkation and post-embarkation testing among Filipino seafarers.

BACKGROUND: Filipino seafarers comprise nearly one-third of the global maritime workforce and face oc-cupational exposures that may elevate cardiometabolic risk. Prolonged deployments marked by irregular schedules, limited physical activity, and constrained dietary diversity can influence metabolic regulation. The objective was to examine pre- to post-embarkation changes in metabolic parameters among Filipino seafarers and identify demographic and occupational predictors of these changes. MATERIAL AND METHODS: A retrospective paired analysis of 402 male seafarers was conducted using pre- and

Metformin treatment impairs the adenine nucleotide translocator activity and energy metabolism in human clear cell renal carcinoma cells.

Although metformin (MET), the well-known antidiabetic drug, exhibits clear antineoplastic effects and is reported to target mitochondria, several issues are still open in this regard, thus limiting its utilization as an anticancer drug alone or in combination with other molecules. Here a functional investigation was carried out to reveal how MET impacted on mitochondrial functions and cell energy metabolism in human cultured clear cell renal carcinoma cells (ccRCCs), in which the anticancer effect of MET is already known. The in vitro effect of increasing MET concentrations on cell viability,

Conversion of CO2 into valuable products: engineering the PirC-PGAM switch in cyanobacteria to direct carbon flux into desired products.

BACKGROUND: In response to rising CO2 emissions driving global warming, there is an urgent need for a transition toward a sustainable bioeconomy. Photo-biotechnological processes based on oxygenic photosynthesis hold high potential for achieving CO2 neutrality and in this regard, cyanobacteria have emerged as promising biocatalysts. Rational metabolic engineering of cyanobacteria depends on a thorough understanding of regulatory mechanisms governing primary metabolism, because native metabolic flux through specific pathways and, consequently, the formation of target products can be limited. Metabolomic properties of the fluid from the surgical ligation after breast-conserving therapy and intraoperative radiotherapy.

Intraoperative radiotherapy with low-energy X-rays (IOXRT) is an increasingly utilized modality during breast conserving therapy (BCT). However, the molecular mechanisms by which it affects the postoperative microenvironment have not yet been fully elucidated. Surgical wound fluid (WF) has been shown to modulate cancer cell behavior; however, its metabolomic composition has not previously been characterized in the context of breast cancer. The objective of this study was to evaluate metabolic alterations in postoperative WF and to determine whether IOXRT induces different metabolic signatures

Sperm energy-restriction and recovery enhances equine sperm function and embryo development after ICSI.

Assisted reproductive technologies (ARTs) are extensively used in domestic animal reproduction; however, improving their efficiency and consistency remains a major challenge for achieving reliable reproductive outcomes. The equine industry faces unique challenges related to sperm capacitation, which limited the success of conventional in vitro fertilization (IVF) for many years. Consequently, intracytoplasmic sperm injection (ICSI) became the most widely used technique for commercial in vitro embryo production (IVP). Even though a successful IVF protocol was recently developed, substantial opp

Macrophage metabolic reprogramming in organ transplantation: mechanisms, transplant outcomes, and therapeutic implications.

Organ transplantation is the definitive treatment for end-stage organ failure, yet long-term graft survival remains substantially limited by ischemia-reperfusion injury (IRI), allograft rejection, and chronic graft dysfunction. Current immunosuppressive regimens have not fully exploited the metabolic plasticity of macrophages, which are central orchestrators of both innate and adaptive immune responses in transplanted organs. Macrophages display remarkable functional plasticity, classically defined by pro-inflammatory (M1)/anti-inflammatory (M2) polarization, and this dual capacity renders the

Orchestrating organotropism: miRNA-driven mechanisms of site-specific metastasis in triple-negative breast cancer (Review).

Triple-negative breast cancer (TNBC), characterized by its aggressive nature and poor prognosis, exhibits a pronounced propensity for organ-specific metastasis, which remains the primary cause of treatment failure and mortality. The present review synthesizes current knowledge on the pivotal role of microRNAs (miRNAs/miRs), particularly those shuttled via tumor-derived exosomes, in orchestrating the complex molecular mechanisms underlying TNBC metastasis to the brain, bone, liver and lungs. In addition, the review highlights how specific miRNAs function as master regulators of organotropic met

Dietary tryptophan supplementation prevents sepsis by enhancing macrophage bacterial defense through GPR37 activation.

BACKGROUND: Sepsis, a critical and life-threatening condition, is one of the most common causes of death among inpatients. Emerging evidence suggests that active metabolites derived from gut microbe-associated metabolism of dietary essential amino acid L-tryptophan (Trp) help the host in combating infectious diseases. This study aims to investigate the mechanisms through which these active metabolites regulate sepsis progression. METHODS: The effects of dietary Trp and indole-3-pyruvate (IPyA) on the mouse septic model were evaluated by 72-hour survival rate monitoring, organ pathological inj

Hederagenin suppresses inflammation-cancer transformation in chronic atrophic gastritis by modulating glycolysis through the mTOR/HIF-1α axis.

Chronic atrophic gastritis (CAG) is a critical precancerous condition with limited therapeutic options to halt its progression toward gastric cancer. Targeting dysregulated metabolism and inflammation-driven transformation represents a promising yet underexplored strategy. Herein, through systematic screening of Jianwei Xiaoyan Granule (JWXYG)-derived compounds via molecular docking and surface plasmon resonance (SPR), we identified hederagenin as a potent and novel mechanistic target of rapamycin (mTOR) binder (Kd = 1.30 μM). In a rat CAG model, hederagenin administration resulted in marked h
