Type I unconventional protein secretion of the SARS-CoV-2 nucleocapsid protein promotes inflammatory cytokine release.

SARS-CoV-2 infection remains a global health threat, yet its pathogenic mechanisms are incompletely understood. Here, we show that viral nucleocapsid protein (NP) is detectable in the serum of patients with SARS-CoV-2 infection independently of viral RNA. Using virus-like particle (VLP) systems and in vitro infection models, we demonstrate that NP is secreted in a vesicle-free form via type I unconventional protein secretion (UPS) pathway. This process is regulated by NP phosphorylation and oligomerization, coordinated by viral structural proteins, and dependent on membrane components includin

Ephedrae Herba as a potential source of SARS-CoV-2 RNA-dependent RNA polymerase inhibitory activity.

INTRODUCTION: Maoto () is a traditional Japanese Kampo formula composed of four crude drugs-Ephedrae Herba, Armeniacae Semen, Cinnamomi Cortex, and Glycyrrhizae Radix-and has been used to treat febrile viral illnesses. Among these, Ephedrae Herba has been associated with antiviral activity in previous studies. However, little is known about the effects of Ephedrae Herba on viral infection, spike (S) protein-mediated syncytium formation (cell-cell fusion), and viral enzymes, or the specific metabolites underlying these activities. In this study, we focused on Ephedrae Herba and comparatively ev

Circadian immunometabolic states impart a temporal response to SARS-CoV-2 spike proteins in mammalian macrophages.

Circadian rhythms, driven by 24-h molecular oscillators, or "clocks", widely tune physiology to the daily rhythms of light and dark to enhance organismal fitness. In mammals, the cellular immune response is tightly regulated by these rhythms such that immunometabolic output is coordinated across the day, consolidating macrophage physiology into temporally distinct phases that determine the macrophage response to stimuli. Importantly, key proteins in the macrophage response to viral infection have been found to be under circadian control, and time of day of adjuvant application is known to affe

Geospatial disparities in post-pandemic SARS-CoV-2 vaccination: Evidence from Chile beyond the national success.

While Chile achieved one of the world's highest and most equitable SARS-CoV-2 vaccination coverages during the pandemic, less is known about the post-pandemic stage. This study examines within-country geospatial disparities in COVID-19 vaccination uptake and their socio-demographic determinants in two urban areas of Chile. A population-based survey conducted in May 2024 collected individual data on vaccination status and socio-demographic characteristics. These were georeferenced and linked to census information at the household block level. Spatial analyses explored clustering and correlation

Emergence of neutralizing RBD antibodies following Omicron infection with limited activity against ancestral SARS-CoV-2.

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) evolution reduces the efficacy of prophylactic vaccines and monoclonal antibody (mAb) therapies. To evaluate potency and breadth, receptor binding domain (RBD)-targeting neutralizing mAbs were identified from patient B cells using an Omicron KP3.1.1 Spike (S) protein bait. MAbs exhibiting the greatest neutralization potency (1332D4 and 1332E5) and binding breadth (1324A10 and 1316C10) were evaluated in greater detail. Cryo-electron microscopy (Cryo-EM) studies revealed 1332D4 and 1332E5 target RBD residue segments 439-446 and 498-506

Targeted FcRn to deliver epitope-optimized RBD confers broad-spectrum mucosal protection against SARS-CoV-2.

Current respiratory vaccines face two major obstacles: limited breadth of protection and insufficient induction of mucosal immunity. Here, we present a neonatal Fc receptor (FcRn)-targeted mucosal delivery strategy integrating computational antigen design to address both challenges. Using severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) as a model virus, we employed Epigraph to generate T cell epitope-optimized consensus sequences for the receptor-binding domain (RBD) and incorporated them into a modified human IgG1 Fc framework engineered for enhanced FcRn binding, yielding a sing

Perovskite-gold shell composite nanoprobe for trimodal lateral flow immunoassay detection of SARS-CoV-2 antibodies.

A CsPbBr3@SiO2-Aushell nanoprobe enables trimodal (colorimetric/fluorescent/photothermal) lateral flow immunochromatographic assay for SARS-CoV-2 IgG detection. This multi-signal approach offers environmental adaptability and internal cross-verification, with the photothermal mode achieving the highest sensitivity (LOD: 0.4 ng mL-1). The work presents a robust strategy for sensitive point-of-care testing.

Increased anti-SARS-CoV-2 antibody titers in plasma-derived intravenous immunoglobulin collected in Southern China.

Although the COVID-19 pandemic is no longer considered an emergency, the ongoing spread of the virus poses a persistent threat to public health. COVID-19 poses a challenge to the system due to the risk of saturation and the associated economic burden. To assess changes in the anti-SARS-CoV-2 antibody profile, we analyzed nine batches of intravenous immunoglobulin (IVIG) prepared from plasma samples collected between November 2020 and June 2023. Antibody changes against SARS-CoV-2 spike, nucleocapsid, and membrane proteins, and neutralizing antibodies were measured. Antibody titers against wild

Inequities and global declines in SARS-CoV-2 genomic data availability hinder response to emerging variants.

Genomic epidemiology has transformed the way public health scientists detect, monitor, and respond to infectious disease threats such as SARS-CoV-2 on a global scale. Early in the COVID-19 pandemic, vast inequities in whole-genome sequence data availability between high- and low-income countries were highlighted, but the persistence of these disparities five years into a global pandemic has not been quantified. Also, while it is generally known that genomic surveillance of SARS-CoV-2 largely declined following the end of the COVID-19 public health emergency, this has not been formally measured

Delivery of monoclonal antibodies using mRNA lipid nanoparticles confers protection against SARS-CoV-2 and influenza.

Monoclonal antibodies (mAbs) are an emerging class of therapeutics for the prevention and treatment of viral infections. Recent advances in mRNA/lipid nanoparticle (LNP) technology provide a promising new modality for the production of mAbs in vivo, potentially bypassing the need for recombinant manufacturing of mAb proteins. In this study, we compared traditional infusion of protein-based neutralizing mAbs targeting severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) or influenza viruses to mRNA/LNP-based production of mAbs in treated mice. High serum concentrations of mAbs were achi

Fusion Strategy of DNA-Encoded Libraries Drives Discovery of Allosteric Inhibitors of SARS-CoV‑2 RdRp.

Allosteric regulation is a central mechanism for modulating biological functions and offers an attractive strategy in drug discovery, particularly for targets considered challenging or "undruggable." However, the discovery of allosteric inhibitors is hindered by poorly defined binding sites and the lack of effective screening approaches. Here, we present a dual DNA-encoded library (DEL) screening strategy that integrates reversible DEL and covalent DEL (CoDEL) technologies to identify novel allosteric inhibitors of the SARS-CoV-2 RNA-dependent RNA polymerase (RdRp). Using this approach, we dis

Computational Evaluation of Multitarget Capabilities of Phenylethanoid Glycosides Against SARS-CoV-2's 3CLpro and PLpro.

Background: The development of multitarget drugs capable of simultaneously inhibiting SARS-CoV-2 proteases-3CLpro and PLpro-may enhance therapeutic efficacy against COVID-19. Given the historical use of Traditional Chinese Medicine (TCM) in the management of respiratory diseases, phenylethanoid glycosides (PGs) represent an attractive and chemically diverse natural product scaffold for the discovery of antiviral agents. Objectives: This study aimed to identify promising candidates within this class capable of simultaneously inhibiting both target proteases. Methods: The PG structures described

Fuzzy Support Vector Regression for QSAR Modeling of 9,10-Dihydrophenanthrene Derivatives Against SARS-CoV-2 3CLpro.

INTRODUCTION/OBJECTIVE: Quantitative Structure-Activity Relationship (QSAR) modeling is a critical computational strategy in drug discovery; however, data heterogeneity and experimental noise frequently compromise model reliability. In this study, QSAR models were developed to predict the inhibitory activity of 9,10-dihydrophenanthrene derivatives against the SARS-CoV-2 3CLpro enzyme using a novel fuzzy Support Vector Regression (FSVR) framework. The proposed approach incorporates a fuzzy membership function to weight activity data according to their reliability and employs both the full molec

SARS-CoV-2 3CLpro mutations T21I and E166A confer differential resistance to simnotrelvir, bofutrelvir, and ensitrelvir.

UNLABELLED: Inhibiting the catalytic activity of 3CLpro is a mainstream strategy to block coronavirus replication. However, the appearance of SARS-CoV-2 3CLpro resistance to protease inhibitors raises concerns for effective therapies. In this work, we first investigated the resistance profile of simnotrelvir, an approved anti-SARS-CoV-2 drug that targets 3CLpro. We found that the T21I/E166A mutations in 3CLpro equally emerged when SARS-CoV-2 was passaged in the HEK293T-hACE2 cells with increasing concentrations of simnotrelvir. The SARS-CoV-2 isolate carrying 3CLproT21I/E166A (SARS2-T21I/E166A

Pediatric post-acute sequelae of SARS-CoV-2 infection in Taiwan: Insights from the DISCOVER cohort.

The post-acute sequelae of SARS-CoV-2 infection (PASC), or long COVID, represent a multifaceted challenge in pediatric populations, characterized by symptoms persisting beyond the acute phase. In Taiwan, where early public health measures initially contained the pandemic, the 2022 Omicron surge prompted focused investigation into pediatric PASC, highlighting the critical need for longitudinal data in this specific demographic. To address this challenge, the Diagnosis and Support for COVID Children to Enhance Recovery (DISCOVER) study was established as a prospective, multidisciplinary cohort.

Engineering antigenic breadth against SARS-CoV-2 by pairing divergent RBDs within a single mRNA immunogen.

Vaccines capable of eliciting broadly neutralizing antibodies (bnAbs) are a major goal for pandemic preparedness. A challenge across vaccine fields is how to deliberately recruit the rare B cell clones that recognize conserved epitopes shared across diverse viral variants. BnAbs have been known to frequently emerge through extensive somatic hypermutation during affinity maturation, here we describe an alternative mechanism for bnAb selection. We designed an mRNA vaccine in which two antigenically distinct (severe acute respiratory syndrome coronavirus 2) SARS-CoV-2 variant's (Omicron and Delta

Exclusion by design: enrollment criteria limit older adult participation in SARS-CoV-2 vaccine clinical trials.

BACKGROUND AND OBJECTIVES: Older adults (≥65 years) face a disproportionate burden of COVID-19 mortality yet remain underrepresented in clinical trials despite this group accounting for 76% of COVID-19 deaths. Hundreds of SARS-CoV-2 vaccine clinical trials have been registered on ClinicalTrials.gov; however, the extent of older adult representation remains unclear. Our objective was to evaluate the representation of older adults in SARS-CoV-2 vaccine clinical trials registered on ClinicalTrials.gov and identify enrollment criteria that disproportionately limit older adult participation. Effect of SARS-CoV-2 infection on pregnant outcomes after embryo transfer: a multicenter retrospective study.

RESEARCH QUESTION: With the nationwide relaxation of controls for the COVID-19 epidemic from December 2022 in China, fertility and in vitro fertilization (IVF) centers reported increasing numbers of infected patients in a short time. However, there is a lack of information regarding the effects of SARS-CoV-2 infection on pregnancy outcomes after IVF–embryo transfer (ET)/intracytoplasmic sperm injection (ICSI)–ET/frozen–thawed embryo transfer (FET), especially the live birth rate. DESIGN: This multicenter retrospective study was conducted at three public IVF centers in Jiangsu province of China

Longitudinal SARS-CoV-2 seroprevalence among learners, parents, and teachers in a South African school community.

INTRODUCTION: Longitudinal assessments of SARS-CoV-2 seroprevalence among a triad of parents, teachers and children in low-middle income countries are scarce. CoKiDSS assessed changes in seroprevalence among asymptomatic primary school learners, their parents, and teachers in a semi-rural South African school setting. METHODS: SARS-CoV-2 seroprevalence was estimated at baseline (May-August 2023) and follow-up (October 2023) using a point-of-care COVID-19 IgG/IgM test. Results were adjusted for test sensitivity and specificity. Between-group differences and within-participant serostatus change
