Description and phylogenetic position of a new Serpentirhabdias (Nematoda: Rhabdiasidae) from Hydrodynastes gigas (Serpentes: Colubroidea) in Pantanal, Brazil.

Serpentirhabdias Tkach, Kuzmin and Snyder, 2014 comprises about 23 species of lung-dwelling nematodes parasites of snakes distributed worldwide. Among them, eight species were found in Brazil. We describe and examine the phylogenetic position of Serpentirhabdias kuzmini n. sp., a parasite of Hydrodynastes gigas from the Pantanal, Brazil. The new species is characterized by a triangular oral opening, absence of a buccal capsule, presence of onchia in the oesophastome, excretory glands longer than the esophagus, and a gradually tapering tail. Moreover, comparative analysis of partial sequences o

Forensic analysis of formalin-fixed non-paraffin-embedded tissues: a comparative study of STR and mtDNA profiling.

Formalin fixation is a common practice for preserving forensic samples in developing countries like India, but it can adversely affect DNA quality and hinder downstream molecular analyses. This study investigated the effectiveness of mitochondrial DNA (mtDNA) typing in formalin-fixed, non-paraffin-embedded tissues when conventional Short Tandem Repeat (STR) profiling fails due to DNA degradation. Tissue samples from 30 deceased individuals were fixed in 10% unbuffered formalin and divided into four groups based on fixation duration (1-4 weeks). DNA was extracted, quantified, and subjected to S

The phylogenetic position of the unusual diplostomoidean genus Schwartzitrema Vigueras, 1941 (Digenea, Diplostomoidea) and description of 2 new species from Neotropics.

Schwartzitrema Vigueras, 1941, is a small, rarely reported genus of diplostomoideans that parasitize avian definitive hosts, predominantly anhingas, on several continents. No DNA sequences were previously available for any Schwartzitrema spp. In the present study, we describe adults of 2 new Schwartzitrema spp. from anhingas in Brazil. We also provide description of metacercaria of one of the new species from Argentina. Schwartzitrema macrotesticulata n. sp. lacks a clear separation between its testes, appearing almost as a contiguous, irregularly shaped, lobular mass. A highly characteristic

Human fascioliasis endemic areas in Argentina: multigene characterisation of the lymnaeid vectors and climatic-environmental assessment of the transmission pattern.

BACKGROUND: In South America, fascioliasis stands out due to the human endemic areas in many countries. In Argentina, human endemic areas have recently been detected. Lymnaeid vectors were studied in two human endemic localities of Catamarca province: Locality A beside Taton and Rio Grande villages; Locality B close to Recreo town. METHODS: Lymnaeids were characterised by the complete sequences of rDNA ITS-2 and ITS-1 and fragments of the mtDNA 16S and cox1. Shell morphometry was studied with the aid of a computer image analysis system. Climate analyses were made by nearest neighbour interpol

Complete genomic characterization of a pathogenic A.II strain of Francisella tularensis subspecies tularensis.

Francisella tularensis is the causative agent of tularemia, which is a highly lethal disease from nature and potentially from a biological weapon. This species contains four recognized subspecies including the North American endemic F. tularensis subsp. tularensis (type A), whose genetic diversity is correlated with its geographic distribution including a major population subdivision referred to as A.I and A.II. The biological significance of the A.I - A.II genetic differentiation is unknown, though there are suggestive ecological and epidemiological correlations. In order to understand the di

T2T-CHM13 reference genome reduces mapping bias and enhances alignment accuracy at disease-associated variants.

The T2T-CHM13v2.0 reference genome added previously uncharacterized genomic sequences and improved the accuracy of repetitive stretches compared to former human genome assemblies. By comprehensive allelic variation analysis and read mapping statistics from sequencing reads aligned to hg38 and T2T-CHM13 assemblies in samples encompassing different sequencing designs, we observed that T2T-CHM13v2.0 assembly significantly reduces the reference mapping bias (RMB) and increases read mapping precision at clinically relevant sites, including BRCA1 pathogenic variants. We report the presence of sequen

Diversity of prophage DNA regions of Streptococcus agalactiae clonal lineages from adults and neonates with invasive infectious disease.

The phylogenetic position and prophage DNA content of the genomes of 142 S. agalactiae (group-B streptococcus, GBS) isolates responsible for bacteremia and meningitis in adults and neonates were studied and compared. The distribution of the invasive isolates between the various serotypes, sequence types (STs) and clonal complexes (CCs) differed significantly between adult and neonatal isolates. Use of the neighbor-net algorithm with the PHI test revealed evidence for recombination in the population studied (PHI, P = 2.01 × 10(-6)), and the recombination-mutation ratio (R/M) was 6:7. Mutation analysis of the human homologue of Drosophila patched and the xeroderma pigmentosum complementation group A genes in squamous cell carcinomas of the skin.

The human homologue of Drosophila patched (PTCH), located at chromosome 9q22.3, was recently identified as a candidate tumor suppressor gene for familial and sporadic basal cell carcinomas. Squamous cell carcinomas (SCCs) of the skin display allelic loss in this chromosomal region, which, in addition to the PTCH gene, contains the DNA repair gene xeroderma pigmentosum complementation group A (XPA). Patients with xeroderma pigmentosum are predisposed to non-melanoma skin tumors because of deficient excision repair of ultraviolet-induced DNA damage. Mutation analysis by single-strand conformatio

Chromosome-level genome assembly of Oncomelania hupensis: the intermediate snail host of Schistosoma japonicum.

BACKGROUND: Schistosoma japonicum is a parasitic flatworm that causes human schistosomiasis, which is a significant cause of morbidity in China, the Philippines and Indonesia. Oncomelania hupensis (Gastropoda: Pomatiopsidae) is the unique intermediate host of S. japonicum. A complete genome sequence of O. hupensis will enable the fundamental understanding of snail biology as well as its co-evolution with the S. japonicum parasite. Assembling a high-quality reference genome of O. hupehensis will provide data for further research on the snail biology and controlling the spread of S. japonicum.

Familial nonsyndromic craniosynostosis with specific deformity of the cranium.

An otherwise healthy, developmentally normal 3-week-old male infant presented with complex multisuture craniosynostosis involving the metopic suture and bilateral coronal sutures with frontal prominence and hypotelorism. Frontal craniectomy and bilateral frontoorbital advancement remodeling were performed at the age of 5 months. The postoperative course was uneventful. The child's development was normal up to 8 months after the operation. His father and grandfather had similar specific deformities of the cranium, but no anomaly of the extremities was found, and conversation suggested that thei

Caudal dysplasia sequence with penile enlargement: case report and a potential pathogenic hypothesis.

The clinical spectrum of caudal dysplasia sequence (CDS) is noted for its diversity. The origin of CDS remains unknown, though poorly controlled gestational diabetes has been implicated in some cases. Here we describe the case of a newborn with CDS associated with penile enlargement (PE). The main anomalies included anal atresia, agenesis of the kidneys and of the sacrococcygeal vertebrae, dysgenesis of lumbar vertebrae, and bilateral cryptorchidism. Penile enlargement (7 cm), a rather unusual finding, has so far not been reported in association with CDS. Chromosomal analysis failed, and the n

Multi-Perspective Natural Vector: A Novel Method for Viral Sequence Feature Extraction.

The rapid expansion of biological data in recent decades has highlighted the need for efficient methods in sequence analysis. Traditional pairwise alignment approaches are both time-consuming and memory-intensive. Alignment-free (AF) methods such as natural vector (NV) and k-mer operate on a one-dimensional framework, interpreting DNA primarily as a linear string of nucleotides. To achieve a more comprehensive interpretation of molecular structure, this study incorporates the three-dimensional architectural features of DNA and introduces a novel AF method named Multi-perspective natural vector

TET2 mutations in Ph-negative myeloproliferative neoplasms: identification of three novel mutations and relationship with clinical and laboratory findings.

High-throughput DNA sequence analysis was used to screen for TET2 mutations in peripheral blood derived DNA from 97 patients with BCR-ABL-negative myeloproliferative neoplasms (MPNs). Overall six mutations in the coding region of the gene were identified in 7 patients with an overall mutational frequency of 7.2%. In polycythemia vera patients (n = 25) 2 mutations were identified (8%), and in those with essential thrombocythemia (n = 55) 2 mutations (3.6%); in those with unclassifiable MPN (n = 8) 3 mutations (37.5%). No primary myelofibrosis patients (n = 6) harboured TET2 mutations. Three unr

Sensitive detection of the c-KIT c.1430G>T mutation by mutant-specific polymerase chain reaction in feline mast cell tumours.

Here, we describe the establishment of mutant-specific polymerase chain reaction (PCR) for detection of a c-KIT c.1430G>T mutation in feline mast cell tumours. Several mutations in feline c-KIT have been identified, with the c.1430G>T mutation accounting for a significant portion of feline mast cell tumour mutations. The c.1430G>T mutation in c-KIT exon 9 was detected in 15.7% (11 of 70) of samples by mutant-specific PCR but in only 7.1% (5 of 70) by PCR-restriction fragment length polymorphism (RFLP) in the genomic DNA isolated from 70 formalin-fixed paraffin-embedded sections or cells collec

Secretory expression of human ScFv against keratin in Pichia pastoris and its effects on cultured keratinocytes.

We have got the humanized antibody with high affinity and specificity against keratin by phage antibody library technology. To improve protein yields and get high affinity and specific anti-keratin antibody, we chose to express it in Pichia pastoris. Anti-keratin ScFv gene from plasmid p3MH/ScFv was subcloned into vector pPIC9K. After confirmed by DNA sequence analysis, the recombinant plasmid pPIC9K/ScFv was transduced into the genome of GS115 P. pastoris. Mut(s) multiple insert transformants were screened by G418 and induced by 5 mL/L methanol to express soluble ScFv. After 6 days of methano
