HSP72 alleviates heat stress-induced oxidative damage in boar Sertoli cells through modulating the AGE-RAGE/NOX4/NF-κB axis.

Heat stress is widely recognized to exert detrimental effects on boar semen quality. This occurs primarily through oxidative stress, which perturbs spermatogenesis by damaging the structural integrity and functional capacity of the involved cells. Heat shock protein 72 (HSP72) plays crucial roles in maintaining redox balance against oxidative stress. A clearer understanding of the functional roles and mechanistic actions of HSP72 in boar Sertoli cells under heat stress, however, requires more definitive evidence. Herein, we mainly investigated the regulatory and functional roles of HSP72 in al

Sperm Retrieval in Men With AZFc Microdeletions Is Independent of Age but Associated With Intermediate Follicle-Stimulating Hormone Levels.

INTRODUCTION: Y-chromosome microdeletions, particularly in the azoospermia factor c (AZFc) region, are a common genetic cause of male infertility. This study evaluates sperm retrieval rates (SRRs) and testicular histology across age groups in men with isolated AZFc deletions. METHODS: We identified men with isolated complete AZFc microdeletions who underwent microdissection testicular sperm extraction from 2000 to 2024. Genitourinary pathology reports categorized histology as Sertoli cell-only, tubular atrophy, maturation arrest, or hypospermatogenesis. χ2 tests compared histology and SRR. Mu

SARS-CoV-2 infection reduces the number of spermatogonial stem cells and dysregulates the transcriptional landscape of the human testis.

SARS-CoV-2 coronavirus emerged in 2019, leading to the Coronavirus disease 2019 (COVID-19). Expression of viral entry factors such as ACE2 and TMPRSS2 is higher in the testis, particularly in Sertoli cells, Leydig cells, and spermatogonia. To understand COVID-19's impact on testicular cell populations and gene expression, we analyzed testicular tissue samples from 28 COVID-19 patients and compared them with 23 non-diseased controls. COVID-19 samples showed increased immune cell infiltration, thrombosis, and reduced numbers of testicular cells. There was a significant decrease in Sertoli cells

Creatine monohydrate modulates testicular morphology and SOD1/NFκB-p65 immunoexpression in streptozotocin-induced diabetic rats.

Diabetes mellitus (DM) is a chronic metabolic disorder characterized by persistent hyperglycemia that may impair testicular function, including spermatogenesis and steroidogenesis. Creatine, an endogenously synthesized nitrogenous compound and one of the main reservoirs in the testes, is widely used, particularly among men. However, evidence regarding its effects on male reproductive health, especially under DM, remains limited. This study evaluated the impact of creatine supplementation on testicular morphology in streptozotocin-induced diabetic rats. Forty-eight adult Wistar rats were random

Furosemide induces dose-dependent testicular toxicity via disrupting blood-testis barrier, inflammation, and apoptosis: A comprehensive biochemical, hormonal, and histopathological study.

Furosemide (FSD) is a widely prescribed loop diuretic; however, its potential reproductive toxicity and its underlying mechanism have not been explored yet. The current study assessed the dose-dependent effects of FSD on testicular function in Sprague Dawley rats. Thirty-two male Sprague Dawley rats were apportioned into four groups i.e., control, FSD (10 mg/kg), FSD (20 mg/kg), and FSD (30 mg/kg) treated group. FSD exposure significantly downregulated the expression of blood-testis barrier genes (CLDN11, OCLN, TJP1, F11R, CDH2, GJA1), as well as upregulated pro-inflammatory mediators (NF-κB,

Swimming in Plastamination: Polylactic Acid (PLA) Nanoplastics Affect Bull Sperm Functions.

Plastic contamination (plastamination) is one of the main challenges of the 21st century. Microplastics (MPs, 5 mm-1 µm) and nanoplastics (NPs, <1 µm) are mainly produced from the environmental degradation of plastic waste and enter the food chain, bypass biological barriers, and bioaccumulate in tissues where they exert toxic/inflammatory effects. In the male reproductive system, MP/NPs can cross the blood-testis barrier, impair spermatogenesis, and affect semen quality parameters. While biodegradable polymers like polylactic acid (PLA) may represent an ecofriendly alternative to carbon fossi

Lactational exposure to polystyrene nanoplastics induces oxidative stress, testicular histological alterations, and impaired sperm quality in rat offspring.

The widespread presence of nanoplastics in the environment has raised concerns regarding their potential adverse effects on reproductive health, particularly during the early developmental stages. Among these contaminants, polystyrene nanoplastics (PSNPs) can cross biological barriers and accumulate in tissues; however, the reproductive consequences of lactational exposure remain insufficiently understood. This study investigated the effects of maternal exposure to PSNPs during lactation on testicular development, oxidative stress, spermatogenesis, and sperm quality in male rat offspring on po

Histological effect of melatonin and its agonist on testes in light-exposure and pinealectomized rats.

Melatonin (MEL), beyond its role as a pineal hormone, supports male reproduction by regulating circadian rhythms and exerting antioxidant and anti-apoptotic actions in the testes. Disruption of pineal function by pinealectomy and/or continuous light exposure is negatively correlated with spermatogenic activity and is linked to testicular degeneration. This study assessed testicular changes in MEL-deficient models and the capacity of MEL and the MEL-receptor agonist ramelteon (RAM) to ameliorate injury. Adult male albino rats underwent continuous light exposure and/or pinealectomy, then were al

TSGA10 as a Model of a Thermal Metabolic Regulator: Implications for Cancer Biology.

TSGA10, a multifunctional protein critical for mitochondrial coupling and metabolic regulation, plays a paradoxical role in cancer progression and carcinogenesis. Here, we outline a potential mechanism by which TSGA10 mediates metabolism in oncogenesis and thermal modulation. Initially identified in spermatogenesis, TSGA10 interacts with mitochondrial Complex III: it directly binds cytochrome c1 (CytC1). In our model, TSGA10 optimizes electron transport to minimize reactive oxygen species (ROS) and heat production while enhancing Adenosine Triphosphate (ATP) synthesis. In cancer, TSGA10's expr

A recurrent loss-of-function variant in DRC1 causes non-syndromic severe asthenozoospermia with favorable intracytoplasmic sperm injection and pregnancy outcomes.

BACKGROUND: Asthenozoospermia, characterized by reduced sperm motility, is a common cause of male infertility. Multiple morphological abnormalities of the sperm flagella (MMAF) represent a severe and genetically heterogeneous form of asthenozoospermia. Over 50 genes have been associated, but approximately half of MMAF cases remain unexplained. DRC1, a gene involved in the nexin-dynein regulatory complex (N-DRC), has been linked to MMAF and primary ciliary dyskinesia (PCD), often with significant variability in clinical presentation. OBJECTIVES: His study aimed to identify novel pathogenic DRC

HSPA2 emerges as a key biomarker: Insights from global lysine acetylproteomic profiling in idiopathic male infertility.

Spermatozoa are highly specialized cells, and any alterations in their protein profiles may affect their function and fertilizing ability. In spermatozoa, which are transcriptionally and translationally inactive, molecular chaperones, particularly heat shock proteins, play crucial roles in maintaining redox balance and preserving protein integrity. Post-translational modifications, particularly lysine acetylation, influence chaperone function and are lately being recognized in the pathophysiology of male infertility. To assess the impact of lysine acetylation on sperm chaperone proteins in idi

Seminal Plasma Exosomal miRNA Profiling Reveals hsa-miR-7-5p as a Key Regulator of Sperm Motility in Asthenozoospermia.

BACKGROUND: Asthenozoospermia is a significant contributing factor to male infertility. Accumulating evidence indicates that impaired sperm motility is closely linked to dysregulated microRNA expression during spermatogenesis. Seminal plasma exosomes are enriched with diverse microRNAs, which play pivotal roles in modulating sperm motility, the acrosome reaction, capacitation, and fertilization. OBJECTIVE: To investigate the association between exosomal microRNA profiles and sperm motility by comparing microRNA expression patterns in seminal plasma exosomes from asthenozoospermic patients and

A systematic review and meta-analysis of the effects of diabetes on human and murine epididymis.

BACKGROUND: Diabetes mellitus has increased significantly over the past decades. This disease affects the reproductive competence of diabetic men by disrupting spermatogenesis, fertility potential, penile erection, and ejaculation. However, hyperglycemic conditions' effects on the epididymis remain elusive despite its importance for sperm maturation. OBJECTIVE: We aimed to determine the effects of diabetes on the epididymis, using qualitative (systematic review) and quantitative (meta-analysis) approaches, to address the question: Can diabetes disrupt epididymal structure and function?
