The involvement of the cholinergic system in Alzheimer disease.

This chapter recounts how memory mechanisms were related to the CNS cholinergic system. It also examines how memory loss in aging individuals was later linked to cholinergic deficiencies and how extensive pharmacologic studies in primates led to the "Cholinergic Hypothesis" of geriatric memory loss. Building upon this, a series of influential publications revealed a loss of cholinergic markers in the cerebral cortex of postmortem Alzheimer disease brain samples. These studies were reinforced by histologic evidence of the degeneration of "magnocellular" neurons in the nucleus basalis of Meynert

Presynaptic Changes in Mouse Rod Photoreceptors During Early Retinitis Pigmentosa.

PURPOSE: Homeostatic plasticity is crucial for maintaining stable neural activity by adjusting strength and intrinsic properties of synapses. This mechanism is vital for normal nervous system function and plays a role in various neurological conditions, including retinal degenerations. Sensitive night vision has been shown in P23H/Gnat2-/- retinitis pigmentosa (RP) mice, which lack cone phototransduction and rely solely on rods, even after losing more than half of their rod photoreceptors. While homeostatic plasticity has been proposed as a potential explanation, the underlying molecular mecha

Strengthening Medial Olivocochlear Feedback Reduces the Developmental Impact of Early Noise Exposure.

The early onset of peripheral deafness significantly alters the proper development of the auditory system. Likewise, exposure to loud noise during early development produces a similar disruptive effect. Before hearing onset in altricial mammals, cochlear inner hair cells exhibit spontaneous electrical activity that drives auditory circuit development. This activity is modulated by medial olivocochlear (MOC) efferent feedback through α9α10 nicotinic cholinergic receptors in inner hair cells. In adults, these receptors are restricted to outer hair cells, where they mediate MOC feedback to regula

Genetic and pharmacologic enhancement of SUMO2 conjugation prevents and reverses cognitive impairment and synaptotoxicity in a preclinical model of Alzheimer's disease.

INTRODUCTION: Amyloid beta oligomers (Aβos) are toxic to synapses and key to the progression of Alzheimer's disease (AD) and amyloid pathology, representing a target for therapeutic strategies. METHODS: Amyloid and small ubiquitin modifier 2 (SUMO2) transgenics were analyzed by electrophysiology and behavioral testing. A recombinant analogue of SUMO2, SBT02, was generated and assessed for brain penetration and the ability to mitigate amyloid pathology. RESULTS: Elevated SUMO2 expression prevents cognitive and synaptic impairment in a mouse model of AD amyloid pathology. Systemic administrati

Cell-type-specific plasticity in synaptic, intrinsic, and sound response properties of deep-layer auditory cortical neurons after noise trauma.

Peripheral trauma, such as noise-induced hearing loss (NIHL), triggers compensatory plasticity in the auditory cortex (ACtx) to maintain auditory function. While cortical plasticity in superficial cortical layers has been relatively well studied, the plasticity mechanisms governing deep-layer excitatory projection neurons remain less understood. Here, we investigated the plasticity of layer (L)5 extratelencephalic (ETs) and L6 corticothalamic neurons (CTs) following NIHL. Using a combination of in vitro slice electrophysiology, optogenetics, and in vivo two-photon imaging in a mouse model of N

Ia excitatory postsynaptic potentials are potentiated in slow-type motoneurons after a 5-week treadmill endurance training in rats.

The electrophysiological properties of motoneurons (MNs) are modified in response to motor activity; however, it remains unknown whether adaptations also involve peripheral input from muscle spindles, which are a potent source of excitatory connections to MNs. The aim of this study was to investigate whether endurance training evoked adaptive changes in Ia afferent synaptic transmission to MNs. Male Wistar rats (n = 15) underwent a 5-week treadmill running course. An electrophysiological experiment was performed on each rat under general anesthesia one day after the final training session.

Central amygdala astrocyte plasticity underlies GABAergic dysregulation in ethanol dependence.

Dependence is a hallmark of alcohol use disorder characterized by excessive alcohol intake and withdrawal symptoms. The central nucleus of the amygdala (CeA) is a key brain structure underlying the synaptic and behavioral consequences of ethanol dependence. While accumulating evidence suggests that astrocytes regulate synaptic transmission and behavior, there is a limited understanding of the role astrocytes play in ethanol dependence. The present study used a combination of viral labeling, super resolution confocal microscopy, 3D image analysis, and slice electrophysiology to determine the ef

Incubation of oxycodone craving is associated with CP-AMPAR upregulation in D1 and D2 receptor-expressing medium spiny neurons in nucleus accumbens core and shell.

A major problem in treating opioid use disorder is persistence of craving after protracted abstinence. This has been modeled in rodents using the incubation of craving model, in which cue-induced drug seeking increases over the first weeks of abstinence from drug self-administration and then remains high for an extended period. Incubation has been reported for several opioids, including oxycodone, but little is known about underlying synaptic plasticity. In contrast, it is well established that incubation of cocaine and methamphetamine craving depends on strengthening of glutamate synapses in

High-dose subacute and acute thymoquinone treatments alleviate LPS-induced depressive-like behavior in mice by reducing inflammation via the IDO/KYN pathway.

The present study aimed to evaluate the antidepressant-like activity of thymoquinone (TQ) in lipopolysaccharide (LPS)-induced depressive-like behavior by inhibiting indoleamine 2,3 dioxygenase (IDO). IDO degrades tryptophan (TRP) along the kynurenine (KYN) pathway. TQ, the main bioactive compound found in black seed (Nigella sativa) oil, possesses numerous biological activities, including anti-oxidants and anti-inflammation properties. Subacute and acute TQ treatments were administered to mice at doses of 5 and 20 mg/kg. Depression-like behavior was assessed using the forced swim test (FST), T

Recovery of retinal terminal fields after traumatic brain injury: evidence of collateral sprouting and sexual dimorphism.

The central nervous system is characterized by its limited regenerative potential, yet striking examples of functional recovery after injury in animal models and humans highlight its capacity for repair. Little is known about repair of pathways/circuits after traumatic brain injury (TBI), which results in disruption of connectivity. Here we utilize a mouse model of diffuse traumatic axonal injury (Impact-acceleration TBI) in order to explore, for the first time, the evolution of structural and functional changes in the terminal fields of the injured visual system. Spike-Based Neuromorphic Model of Spasticity for Generation of Affected Neural Activity.

Spasticity is a common motor symptom that disrupt muscle contraction and hence movements. Proper management of spasticity requires identification of its origins and reasoning of the therapeutic plans. Challenges arise because spasticity might originate from elevated activity in both the cortical and sub-cortical pathways. No existing models (animal or computational) could cover all possibilities leading to spasticity, especially the peripheral causes such as hyperreflexia. To bridge this gap, this work develops a novel computational, spike-based neuromorphic model of spasticity, named NEUSPA.

Non-invasive testing to diagnose the underlying auditory pathology of hearing impairment.

Of the three major classifications of inner ear pathology, sensory, neural and metabolic, it is relatively easy to distinguish a sensory pathology from a neural or inner hair cell pathology using current clinical tools. Deficiencies in both distortion product otoacoustic emissions (DPOAE) and auditory brainstem responses (ABR) suggest a sensory pathology involving outer hair cells. Normal DPOAEs with deficiencies in ABRs suggest a neural pathology, involving inner hair cells, auditory nerve synapses or neural deficiencies. However, there are currently no non-invasive methods to distinguish a m

Landscapes of gut microbiome and metabolic signatures in vitiligo patients with co-morbid emotional distress.

BACKGROUND: Vitiligo is a depigmentation disorder frequently associated with emotional distress; however, the precise mechanisms underlying this co-morbidity remain unclear. OBJECTIVE: This study aims to investigate whether gut dysbiosis and gut metabolites contributes to emotional distress in patients with vitiligo. METHODS: Depression and anxiety were assessed using the Patient Health Questionnaire-9 and Generalized Anxiety Disorder-7, respectively. Totally enrolled 28 vitiligo patients were diagnosed with depression or anxiety (VWD), 44 without such conditions (VTD), and 37 healthy contro

Olivocerebellar circuit development as a substrate for dystonia pathogenesis.

Despite the progress in identifying genetic and molecular contributors to dystonia, the synaptic and circuit mechanisms that disrupt motor maturation and serve as the substrate for disease pathogenesis are only beginning to emerge. In this chapter, we highlight evidence implicating the olivocerebellar node as a key site of pathogenesis in the dystonia network. As an example of aberrant olivocerebellar circuit activity leading to dystonia, here we examine a mouse model in which excitatory neurotransmission is silenced at inferior olive to cerebellum synapses upon Vglut2 mediated conditional com

5-HTTLPR does not moderate the effect of attention bias modification for depression: a randomized sham-controlled trial.

The 5-HTTLPR polymorphism in the serotonin transporter gene SLC6A4 has previously been dubbed a plasticity marker. Within the 5-HTTLPR polymorphism, a SNP (rs25531) in the L-allele in the promoter region, affects the transcription efficacy of the SCL6A4, leading to functionally important differences related to serotonin transporter availability in the synapses. 5-HTTLPR has been implicated in magnitude of negative attentional bias, a causal risk factor for depression, and the modifiability of attentional biases both in positive and negative directions. Hence, this genotype may moderate the out

Downmodulation of Potassium Conductances Induces Epileptic Seizures in Cortical Network Models Via Multiple Synergistic Factors.

Voltage-gated potassium conductances g K play a critical role not only in normal neural function, but also in many neurological disorders and related therapeutic interventions. In particular, in an important animal model of epileptic seizures, 4-aminopyridine (4-AP) administration is thought to induce seizures by reducing g K in cortex and other brain areas. Interestingly, 4-AP has also been useful in the treatment of neurological disorders such as multiple sclerosis and spinal cord injury, where it is thought to improve action potential propagation in axonal fibers. Here, we examined g K down

Complex N-glycosylation of mGluR6 is required for trans-synaptic interaction with ELFN adhesion proteins.

Synaptic transmission from retinal photoreceptors to downstream ON-type bipolar cells (BCs) depends on the postsynaptic metabotropic glutamate receptor mGluR6, located at the BC dendritic tips. Glutamate binding to mGluR6 initiates G-protein signaling that ultimately leads to BC depolarization in response to light. The mGluR6 receptor also engages in trans-synaptic interactions with presynaptic ELFN adhesion proteins. The roles of post-translational modifications in mGluR6 trafficking and function are unknown. Treatment with glycosidase enzymes PNGase F and Endo H demonstrated that both endoge
