Systems biology framework for the rational design of operational conditions for in vitro/in vivo translation of tissue models.

Preclinical models are used extensively to study diseases and therapies. In vitro monoculture or microphysiological system (MPS) platforms incorporating multiple different human cell types can emulate diseased tissues, but determining experimental conditions (e.g., media supplements) that provide the most effective translatability to humans (in vivo) is a major challenge. Using metabolic dysfunction-associated steatotic liver disease (MASLD) as a case study, we developed a machine learning framework [called LIV2TRANS (Latent In Vitro to In Vivo Translation)] that first maps MPS onto in vivo da

Foxp3 drives context-dependent epigenetic programs that define regulatory T cell molecular identity and function.

Regulatory T cells (Treg cells) express the master regulator, Foxp3, and display distinctive epigenetic landscapes ensuring Treg cell-specific gene expression and stable suppressive functions, yet Foxp3's contribution to this epigenetic identity remains unclear. Leveraging Foxp3-transduced conventional T cells as a gain-of-function probe in mice, we identified a previously unrecognized subset that acquires endogenous Foxp3 expression, Treg cell-like transcriptomic and chromatin features, and suppressive functions exclusively in vivo. These Foxp3-driven features were conserved in Treg cells but

Pathological Mechanisms of Tendinopathy Pain: From Mechanical Overload to Neuroimmune-Vascular Crosstalk.

Characterized by localized pain and functional impairment, tendinopathy remains a significant challenge in sports medicine. Traditional models of structural degeneration and inflammation inadequately explain the clinical discordance between tissue damage and pain severity. This review explores the paradigm shift in understanding tendinopathic pain, elucidating the transition from a simple mechanical stress model to a complex neuroplastic disorder. We narratively review the multilayered pathophysiological mechanisms underlying chronic tendinopathic pain. Specifically, we outline abnormal mechan

USMSIT/NMUSIT Fundamental Guide on Diagnostic and Interventional Hip Ultrasonography.

Hip pain arises from a spectrum of intra-articular, periarticular, and neurogenic conditions. Ultrasound offers real-time, radiation-free visualization of the hip joint, adjacent soft tissues, and peripheral nerves - making it an essential tool in contemporary musculoskeletal evaluation. Guided by the EURO-MUSCULUS/USPRM framework, this guideline proposes a standardized, anatomy-based protocol for both diagnostic and interventional hip ultrasonography. Developed under the educational mission of the Ultrasound and Musculoskeletal/Neuromuscular Sonography Initiative in Taiwan - through its USMSI

PALM-Scan enables single-molecule site-specific detection of protein S-palmitoylation with a nanopore.

Protein S-palmitoylation is a dynamic and reversible post-translational modification (PTM) that governs diverse cellular processes, yet its study remains constrained by the indirect and ensemble-averaging nature of classical biochemical assays. Here, we report a nanopore-based platform, PALM-Scan (palmitoylation analysis via label-free monitoring and signature capture on nanopore) that directly transduces physicochemical signatures of S-palmitoylated peptides into digital electrical signals. Using an engineered Mycobacterium smegmatis porin A (M2-MspA) nanopore, PALM-Scan enables sequence-inde

Human iPSC-derived neural progenitor gene therapy improves outcomes in a neuropathic lysosomal disease.

Mucopolysaccharidosis type IIIA (MPS IIIA) is a neuropathic lysosomal storage disorder caused by SGSH mutations leading to heparan sulfate accumulation in the brain. This drives progressive cognitive decline, neuroinflammation, and behavioral abnormalities. No effective disease-modifying therapy exists, as current approaches fail to deliver sufficient enzyme to the brain or halt neurodegeneration. Here, we report a neural stem cell gene therapy that achieves high enzyme delivery and restores cognitive function, likely through integration of healthy neural cells across multiple brain regions. Blast-Induced Auditory Injury: From Peripheral Pathology to Central Neurodegeneration.

INTRODUCTION: Blast wave exposure represents a primary cause of complex auditory and neurological injuries. The ear acts as a sensitive pressure transducer, making it exceptionally vulnerable to the intense mechanical forces of a blast. This review provides an overview of the multidimensional damage to the auditory system following blast events. MATERIALS AND METHODS: We conducted a systematic literature search across PubMed, Scopus, Web of Science, and Google Scholar (January 2000-January 2025) using comprehensive search terms. After screening 462 unique records, 36 original research studies

Invasive Pulmonary Aspergillosis in a Young Adult With Hyperimmunoglobulin E Syndrome and Hypogammaglobulinemia Following Rituximab Therapy.

Hyperimmunoglobulin E syndrome (HIES) is a rare primary immunodeficiency marked by elevated IgE levels, recurrent skin and pulmonary infections, and immune dysregulation. While typically diagnosed in childhood, adult presentations can occur, often complicated by structural lung disease, opportunistic infections, and malignancies. We report a 37-year-old female with signal transducer and activator of transcription 3 (STAT3)-deficient HIES, diffuse large B-cell lymphoma (DLBCL) in remission, rituximab-induced hypogammaglobulinemia, and recurrent infections, who presented with acute-on-chronic dy

Adenoviral-mediated gene transfer into the scala media of the mouse cochlea in vivo.

Gene therapy may provide a way to restore inner ear function to deaf and dizzy patients. The mouse is a crucial model system for functional genomics because of the numerous genetic models for hearing loss and inner ear dysfunction. Using an advance generation, E1-/E3-/E2b- (preterminal protein-/polymerase-) Type 5 Adenovirus, we investigated several routes of virus microinjection to determine which were most reproducible in targeting the endolymphatic fluid compartment of the cochlea. We found that when adenovirus is injected via the round window, transduced cells are found only adjacent to th

Mutant p53 sensitizes KRAS-driven lung adenocarcinoma to immunotherapy by repressing SPP1.

The efficacy of immune checkpoint blockade (ICB) in lung adenocarcinoma (LUAD) is limited by primary or acquired resistance, especially in genetically defined subsets. We investigated the impact of KRAS/TP53 co-mutation on ICB response. Our analysis reveals that KRAS/TP53 co-mutation is associated with favorable ICB outcomes, a phenotype orchestrated by secreted phosphoprotein 1 (SPP1). Mechanistically, mutant p53 and KRAS exert opposing effects on SPP1. Mutant p53 synergizes with transcription factor FOXA1 to repress SPP1 transcription, whereas KRAS activates the mitogen-activated protein kin

Genetically engineered K562 cells augment NK cell cytotoxicity against acute myeloid leukemia and reduce dependency on IL-15.

Acute myeloid leukemia (AML) is an aggressive malignancy with limited treatment options. Enhancing natural killer (NK) cell functionality through artificial antigen-presenting cells (aAPCs) represents a promising immunotherapeutic strategy. This study evaluates the potential of genetically modified K562 cells, expressing CD137L and CD86, to enhance NK cell-mediated cytotoxicity against AML cell lines (HL-60, KG-1, and THP-1). Lentiviral transduction was used to generate aAPCs, confirmed by PCR, RT-PCR, and flow cytometry. Cord NK cells and the NK-92 cell line were co-cultured with aAPCs, and t

A zinc transporter drives glioblastoma progression via extracellular vesicles-reprogrammed microglial plasticity.

Glioblastoma (GBM) is the most aggressive form of brain cancer, with limited therapeutic options. While microglia contribute to GBM progression, the mechanisms by which they foster a protumorigenic immune environment remain poorly understood. We identify the zinc transporter Zrt- And Irt-Like Protein 4 (ZIP4) as a pivotal regulator of the GBM immune landscape. In orthotopic mouse models, ZIP4 drives tumor growth and behavioral changes. Mechanistically, ZIP4 modulates microglial plasticity through tumor-derived extracellular vesicles carrying triggering receptor expressed on myeloid cells-1 (TR

Immune-driven gene expression loss following intramuscular AAV delivery to non-human primates is only transient.

Recombinant adeno-associated virus (rAAV) vectors stand out as highly promising for in vivo gene transfer, particularly in targeting the skeletal muscle for treating muscular genetic diseases or secreting therapeutic factors. Despite the simplicity and efficacy of the established intramuscular (IM) route, it has been often associated with an immune-induced rapid loss of transgene expression, in particular in large animal models, and generally considered irreversible as a consequence of a cytotoxic elimination of transduced cells. Here, we report in a non-human primate model that transgene expr

Forces on the Incisor Teeth During Odontoplasty of the Cheek Teeth in Sedated Horses.

Mouth specula with incisor bite plates shift the pressure from all teeth exclusively to the incisors in the opening phase which leads to increased forces on dental, osseous, and soft tissue structures of the horse's head. The potentially pathological character of these forces is described clinically by single reports of maxilla and mandible fractures occurring during the use of opened mouth specula. This study describes these forces on incisor teeth in horses during odontoplasty of cheek teeth under sedation. Measurements were documented using a modified "Günther" mouth speculum fitted with a

Human bocavirus NP1 antagonizes host type I interferon response through repressing the nuclear transport of STAT1.

Human bocavirus 1 (HBoV1), first identified in human nasopharyngeal specimens in 2005, is a relatively recent member of the respiratory virus family and is primarily associated with respiratory tract infections. Despite this, the mechanisms underlying HBoV1 pathogenesis remain poorly understood, primarily due to the lack of suitable cell lines and animal models, as well as the complicating factor of co-infections with other pathogens. In this study, we demonstrate that the non-structural protein 1 (NP1) of HBoV1 antagonizes the type I interferon (IFN-I) signaling pathway. NP1 interacts with th

Combined effects of 40 Hz ultrasound and light stimulation on neural oscillations and cognition.

Single-modality 40 Hz ultrasound stimulation or 40 Hz light stimulation have demonstrated the efficacy in improving cognitive function in Alzheimer's disease (AD) mice. However, the therapeutic effect of combined 40 Hz ultrasound-light stimulation on AD-related cognitive deficits remain unexplored. To address this gap, we developed a 40 Hz dual-modality stimulation system comprising a dual-targeted holographic ultrasound transducer that enables precise, safe stimulation of the bilateral hippocampal CA1 regions, and a 40 Hz visual stimulation. In wild-type mice, acute 40 Hz ultrasound-light sti

DNA-Damaging Agents Induce PD-L1 Expression through the Src-STAT1-IRF1 Pathway.

Immune checkpoint inhibitors (ICIs) have been approved for the treatment of various cancers; however, their clinical efficacy remains limited in many patients due to resistance mechanisms. One of the mechanisms underlying this resistance is the downregulated expression of programmed cell death-ligand 1 (PD-L1) caused by mutations that impair Janus kinase 1/2 (JAK1/2) function. Therefore, finding an alternative JAK-independent pathway to enhance PD-L1 expression would be highly valuable. In the present study, we found that the chemotherapeutic agents, SN-38 and cisplatin, upregulated the transc

Nanocarrier imaging at single-cell resolution across entire mouse bodies with deep learning.

Efficient and accurate nanocarrier development for targeted drug delivery is hindered by a lack of methods to analyze its cell-level biodistribution across whole organisms. Here we present Single Cell Precision Nanocarrier Identification (SCP-Nano), an integrated experimental and deep learning pipeline to comprehensively quantify the targeting of nanocarriers throughout the whole mouse body at single-cell resolution. SCP-Nano reveals the tissue distribution patterns of lipid nanoparticles (LNPs) after different injection routes at doses as low as 0.0005 mg kg-1-far below the detection limits o

[Acupuncture effects and neuro-immune regulation: a breakthrough in "treating diseases at the root"].

The holistic philosophy of traditional Chinese medicine (TCM) embodied in acupuncture therapy has gained novel insights within the neuroimmune regulatory framework. Its mechanistic framework encompasses 3 pivotal dimensions: 1) Acupuncture stimulation at acupoints transduces mechanical signals into neuroelectrical and biochemical signals, initiating localized immunomodulatory microenvironment remodeling; 2) The autonomic nervous system orchestrates systemic and organ-specific immune homeostasis through vagus nerve-mediated anti-inflammatory effects and sympathetic nerve-mediated bidirectional

Pulmonary artery pressure via endobronchial ultrasound: A feasibility and safety study.

OBJECTIVES: Pulmonary hypertension significantly influences perioperative outcomes in thoracic surgery. Right heart catheterization, the standard method to assess pulmonary artery pressure, is invasive and time-consuming. We aimed to assess the feasibility and safety of direct pulmonary artery pressure measurement through endobronchial ultrasound-guided puncture of the right pulmonary artery. METHODS: Twenty patients underwent ultrasound-guided puncture of the right pulmonary artery using a 22-gauge needle under real-time ultrasound guidance. Systolic, diastolic, and mean pulmonary pressures
