Functional characterization of bicarbonate transporters from the cyanobacterial SbtA2 family and subsequent expression in tobacco.

Cyanobacteria rely on bicarbonate (HCO3-) as the primary inorganic carbon (Ci) source for photosynthesis in aquatic environments. To use this Ci source, cyanobacteria employ CO2-concentrating mechanisms that elevate cytoplasmic HCO3- via plasma membrane transporters, enhancing carboxylation by carboxysomal Rubisco. The sodium-dependent SbtA1 transporter family is well characterized in freshwater cyanobacteria, but the related SbtA2 family, prevalent in marine α-cyanobacteria, remains uncharacterized. Here, we report functional characterization of SbtA2 homologues from marine Synechococcus spp.

Reversible Bond Dynamics Enable Crystallinity-Healed COF Membranes for Selective Ion Transport.

Covalent organic frameworks (COFs) offer ordered, nanometer-scale channels with programmable chemistry and topology, making them promising membrane materials for selective ion transport. However, fabricating robust COF membranes that preserve high crystallinity remains a key challenge. Here we directly address this challenge by decoupling crystallization from membrane formation. COF membranes are first made by interfacial polymerization and subsequently healed under acid-catalyzed hydrothermal conditions, which activate reversible COF linkage bond exchange and framework self-correction. Using

Phosphate Acquisition in Protozoan Parasites: Transport Systems, and Extracellular Phosphate-Releasing Enzymatic Activities.

Inorganic phosphate (Pi) is an essential nutrient required for energy metabolism, macromolecule biosynthesis, and signal transduction. Protozoan parasites are exposed to pronounced fluctuations in nutrient availability throughout their life cycles and therefore rely on adaptive strategies to secure phosphate from diverse host and environmental niches. This review summarizes current knowledge on phosphate acquisition mechanisms in protozoan parasites, with emphasis on membrane transport systems and extracellular phosphate-releasing enzymatic activities. Phosphate transport systems energized by

Manipulating Fluorinated Nanochannels in Porous Aromatic Frameworks for Ultra-Stable Sodium-Ion Transport.

Quasi-solid-state sodium metal batteries suffer from sluggish Na+ transport and unstable interphases due to strong ion pairing and unfavorable solvation structures. Herein, fluorinated nanochannels are constructed by grafting flexible perfluoroalkyl chains into porous aromatic frameworks (PAF-1-F), generating a weakly coordinating environment that promotes ion-pair dissociation and fluorine-polarization-assisted Na+ transport. The resulting PAF-1-F quasi-solid-state electrolyte exhibits an impressive ionic conductivity of 2.79 × 10-4 S cm-1, a high Na+ transference number of 0.562, and a wide

Early inotropic support in pediatric shock: evidence and challenges in prehospital setting and interfacility transport.

BACKGROUND & AIM: Pediatric shock requires rapid recognition and timely intervention. In the prehospital and interfacility transport setting, the challenge is further compounded by limited vascular access, limited monitoring capabilities, and the need for rapid decision-making in dynamic, resource-constrained environments. The aim of this review is to summarize the available literature on the use of vasoactive agents in pediatric shock, provide guidance for evidence-based decision-making, and identify key gaps for future research. METHODS: A literature search was conducted using electronic da

Radial illumination enables the concentration, dispersion, lateral transport, and sorting of photocatalytic TiO2 microspheres.

The precise manipulation of a large number of colloidal particles in lateral (xy) dimensions is important for various applications. We introduce a simple way of concentrating, dispersing, moving, and sorting a large group of isotropic, photocatalytic TiO2 microspheres in 2D by moving the optical condenser of a microscope up and down. This operation varies the vertical (z) position of the beam waist of the illumination light and causes the light beam to be radially oblique, and the nonuniform photocatalysis on the surface of TiO2 microspheres causes them to move away from light via diffusiophor

Asymmetrically Wetted Trilayer-Structured Wound Dressing with Unidirectional Moisture Transport and Hemostatic Function.

An effective wound dressing should be hemostatic and able to absorb excess exudates while protecting the wound from infection and inflammation. However, conventional dressings with a single structure and property exist with non-negligible limitations in exudate management owing to their limited draining ability. Herein, a self-pumping Janus hemostatic dressing with unidirectional moisture transport (Janus@BG5) was reported by an electrospun trilayered fibrous matrix, in which the inner hydrophobic polylactic acid (PLA) nanofiber array, the interlayer PLA and silk fibroin (SF) composite fibrous

Electrothermal Regulation of Ultrafast Nanoconfined Water Transport in Conductive Carbon Membranes for Molecular Sieving.

Manipulation of nanoconfined water dynamics by heat and electricity is attractive for advancing membrane separation processes, yet integrating ångström-scale molecular sieving with robust electrothermal performance remains challenging. Herein, we demonstrate that a conductive carbon membrane enables ultraselective water/organic molecular separation under electrothermal regulation. Compared with the thermally driven pervaporation process, the electrically driven Joule heating enhanced the permeation flux and separation factor by 48% and 270%, respectively. The improved separation performance wa

pH-Responsive Hydroxypropyl Cellulose-Based Membranes for Controlled Mass Transport and Drug Release.

The swelling-regulated transport properties of modified and cross-linked HPC-based hydrogel formulations containing NaCMC and citric acid were studied as stimuli-responsive polymeric membranes under various conditions, including deionized water. Physiological conditions were simulated by evaluating various pH conditions (1.2, 6.8, and 7.4). The pseudo-second-order kinetic model best described the swelling process, suggesting that both solvent uptake capacity and polymer network relaxation contribute to the extent of swelling. The swelling behavior of the hydrogel formulations was significantly

Hit-to-Lead Optimization of Energy-Coupling Factor (ECF) Transporter Inhibitors as Novel Antibiotic.

Multiparameter optimization of a previously identified class of inhibitors of the energy-coupling factor (ECF) transporters enabled the confirmation of in vivo efficacy. ECFs are a class of transmembrane proteins that play a vital role in the active translocation of essential nutrients across cell membranes and are therefore important in the fight against antimicrobial resistance. Aiming to improve the drug-like properties of our inhibitory class, we performed a focused structure-activity relationship study around the Eastern part of our starting molecule 3 by exploiting click chemistry. Cilia-independent gas-liquid transport, a third mechanism mediating airway mucus clearance.

Airway mucus clearance from the lungs occurs by 2 widely recognized mechanisms: cilia-mediated clearance and high-velocity airflow-mediated cough clearance. However, a potentially important third mechanism of mucus clearance, referred to as cilia-independent gas-liquid transport (GLT), was proposed based on in vitro model systems to occur during normal tidal breathing but has largely been overlooked. We conducted in vitro and in vivo studies to investigate the role of tidal breathing airflow rates in mucus clearance. An in vitro airway culture bead-tracking model demonstrated airflow-dependent

Metabolic and transport engineering enable efficient de novo biosynthesis of patchoulol in Saccharomyces cerevisiae.

Patchoulol is a valuable sesquiterpene for fragrance and cosmetics, but plant extraction is slow and resource-intensive. Here, we combined enzyme, pathway and transporter engineering to build a yeast chassis for high-titer patchoulol production. Semi-rational engineering of patchoulol synthase (PTS) increased catalytic efficiency; together with multi-copy PTS expression, the shake-flask titer rose from 0.40 ± 0.05 mg/L (PT01) to 258.72 ± 27.35 mg/L. Strengthening glycolysis and the mevalonate pathway and down-regulating ERG9 redirected flux to FPP. Guided by docking and transcript analysis, we

Ileal Bile Acid Transporter Inhibitors in Cholestasis: Potential for More Than Just Paediatrics?

Ileal bile acid transporter inhibitors (IBATi) are a new, attractive therapeutic mechanism to alter the enterohepatic circulation through depletion of the bile acid pool by blocking bile acid reuptake in the ileum leading to improvements in pruritus and liver function in cholestatic liver diseases. These drugs may also have an impact on immunity, the gut microbiome, and motility. IBATi are approved in Japan for the treatment of idiopathic chronic constipation. There are two IBATi, maralixibat and odevixibat, that have been extensively investigated in clinical trials and are FDA approved for ch

The dromedary camel HPA axis: Baseline ultrastructure and functional correlates of cortisol following transport.

BACKGROUND: The hypothalamic-pituitary-adrenal (HPA) axis is central to vertebrate adaptation, yet its structural organization remains poorly characterized in large desert-adapted mammals at the subcellular level. The dromedary camel (Camelus dromedarius) is a species of considerable comparative interest due to its tolerance of extreme environmental conditions, but foundational integrated neuroendocrine anatomy is lacking. Furthermore, whether subcellular structural variation within this axis correlates with functional HPA output has not been investigated enough. METHODS: This study provides

Scale-dependent transport in deep eutectic solvents: nanoscale dynamics of cholinium ions in reline.

Transport in deep eutectic solvents (DESs) exhibits a pronounced dependence on length scale, with macroscopic hydrodynamic descriptions failing to capture the complex dynamics that emerge at the molecular level. This study seeks to elucidate the molecular origins of these transport anomalies by examining the nanoscale diffusion of cholinium ions within reline using quasielastic neutron scattering (QENS). Our findings reveal that nanoscale transport within DESs arises from a combination of two distinct dynamical processes: rapid localized motion and slower long-range translational diffusion. Th

circ_0043837 attenuates macrophage mitochondrial damage through XPO1-associated HuR transport in atherosclerosis.

Macrophage mitochondrial damage is implicated in the pathogenesis of atherosclerosis. Circular RNAs (circRNAs) regulate mitochondrial energy metabolism and contribute to the development of various diseases, but the mechanism in atherosclerosis regulation remains to be elucidated. This study aimed to investigate the underlying mechanisms by which circRNAs modulate mitochondrial function in the progression of atherosclerosis. We established a macrophage-derived foam cell model to define the role of circ_0043837 in mitochondrial dysfunction during atherosclerosis. Foam cells showed loss of mitoch

Linking Laboratory and Field Observations of Carbon Dioxide Transport at the Nevada National Security Site.

Subsurface gas transport is critical for containment performance following underground nuclear detonations or high-explosive tests. Such gases can provide fingerprinting needed to identify whether detected seismic signals have a nuclear or chemical source, provided that their subsurface transport can be accurately interpreted. In this study, we examine carbon dioxide (CO2) diffusion through dry core samples taken from representative lithologies at the Nevada National Security Site (NNSS) and compare laboratory-measured transport with field observations after a subsurface high-explosive experim

Water chemistry controlled enhancement of electrokinetic transport and reactant delivery in low-permeability zones.

Electrokinetic (EK) transport and remediation represent a promising approach for delivering charged reactants into low-permeability zones (LPZs) in the subsurface, where contaminants often accumulate, and conventional hydraulic-based in situ remediation is limited by preferential flow through surrounding permeable regions. EK transport in physically heterogeneous porous media is governed by the coupled action of pressure-driven flow, electromigration, and electroosmosis, the latter of which depends substantially on pore water ionic composition and ionic strength and can significantly influence

The Caenorhabditis elegans mitochondrial electron transport chain: its role in adaptation, longevity, and biotechnology.

In Caenorhabditis elegans (C. elegans) the mitochondrial electron transport chain (ETC) exhibits remarkable functional plasticity. This review summarizes the composition, regulation, and adaptive roles of complexes I-V. Depending on oxygen availability, the ETC uses either ubiquinone (UQ) or rhodoquinone (RQ), an ancestral strategy for hypoxia or high hydrogen sulfide (H2S) conditions. Mild ETC impairments can extend lifespan through redox signaling, mitohormesis, and activation of the mitochondrial unfolded protein response. These processes likely represent conserved mechanisms of bioenergeti

Tolfenpyrad Modulates Antibiotic Activity on Francisella novicida and Synergizes with Electron Transport Chain Inhibitors.

Francisella tularensis is a highly virulent intracellular pathogen for which treatment options remain limited and vulnerable to resistance development. Tolfenpyrad, a pesticide with selective antibacterial activity against Francisella species, has emerged as a promising compound, yet its mechanism of action remains poorly defined. Here, we investigated how tolfenpyrad modulates the activity of established antibiotics and probed its effects on bacterial energy metabolism in Francisella. In this study, we used combinatorial drug experiments to identify both synergistic and antagonistic drug-drug
