Optic chiasmal neuritis: clinical features, aetiologies, MRI patterns and prognosis in a real-world cohort.

BACKGROUND: Optic chiasmal neuritis (OCN) is considered atypical for multiple sclerosis (MS) and more frequently associated with neuromyelitis optica spectrum disorders (NMOSD). Yet the true aetiological spectrum of OCN in the antibody era remains poorly defined. METHODS: We conducted a retrospective single-centre study of consecutive patients with MRI-confirmed chiasmal enhancement between 2017 and 2025. Non-inflammatory, non-demyelinating aetiologies and previous optic neuritis were excluded. Clinical features, ophthalmological outcomes, MRI findings, and aetiologies were analysed. RESULTS

EHBMT, a method for visualizing tumor evolution, identifies a surge in gastric cancer with hybrid epithelial-mesenchymal phenotypes due to an inflammatory microenvironment.

A method for analyzing tumor evolution based on bulk RNA-sequencing data has not been reported yet. The epithelial-mesenchymal transition (EMT) is an evolutionarily conserved cellular program with high heterogeneity and plasticity. In this study, we proposed an EMT heterogeneity-based molecular typing (EHBMT) method to visualize cancer evolution and guide personalized medicine. Multiplex immunohistochemical assay and single-cell analysis were performed to confirm the feasibility of this method. EHBMT divided gastric (cancer) tissues into an epithelial phenotype cluster (EPC), hybrid epithelial

Dissecting the Causal Role of Early Inferior Frontal Activation in Reading.

Cognitive models of reading assume that speech production occurs after visual and phonological processing of written words. This traditional view is at odds with more recent magnetoencephalography studies showing that the left posterior inferior frontal cortex (pIFC) classically associated with spoken production responds to print at 100-150 ms after word-onset, almost simultaneously with posterior brain regions for visual and phonological processing. Yet the theoretical significance of this fast neural response remains open to date. We used transcranial magnetic stimulation (TMS) to investigat

A detailed review of genetically encodable RFPs and far-RFPs and their applications in advanced super-resolution imaging techniques.

The red fluorescent proteins (RFPs) and far-red fluorescent proteins (far-RFPs) that are encoded genetically can emit fluorescence within the spectral ranges of 580-680 nm when exposed to the light of appropriate wavelengths. Unlike many RFPs derived from coral species, numerous far-RFPs are optimized synthetic constructs engineered from different orange or red-emitting progenitors. Various categories have been established for the available RFPs and far-red fluorescent proteins based on their photo-chemical profile, fluorescence mechanism, and switching kinetics. Fluorescent probes (FPs) deriv

Combined evidence from artificial neural networks and human brain-lesion models reveals that language modulates vision in human perception.

Comparing information structures in between deep neural networks (DNNs) and the human brain has become a key method for exploring their similarities and differences. Recent research has shown better alignment of vision-language DNN models, such as contrastive language-image pretraining (CLIP), with the activity of the human ventral occipitotemporal cortex (VOTC) than earlier vision models, supporting the idea that language modulates human visual perception. However, interpreting the results from such comparisons is inherently limited owing to the 'black box' nature of DNNs. Here we combine mod

Dynamic in vivo imaging of microplastic-induced hepatic injury via enzyme-activated far-red fluorescent probe.

Microplastic pollution, particularly from polystyrene (PS), poses increasing risks to human health due to its persistence in the environment, tendency to bioaccumulate, and ability to induce oxidative stress and hepatic injury. However, tools for dynamic imaging that elucidate the mechanistic link between microplastic exposure and liver toxicity remain lacking. Herein, we report a far-red fluorescent probe (emission maximum at 674 nm), DDAO-CT, derived from the hydroxyl-substituted red-emitting fluorophore DDAO (9,9-dimethylacridin-2(9H)-one derivative), designed for in vivo visualization of c

Shining light on photoreceptors: A minireview on the development and clinical applications of optoretinography.

Retinal diseases often result in photoreceptor dysfunction and cell death, leading to progressive vision impairment and eventual blindness. Clinical management can benefit from assessing photoreceptor mediated visual function, such as for progression monitoring. Meanwhile, therapeutic development requires accurate and reliable vision end points. The emerging optoretinography (ORG) technologies promise noninvasive, objective, and highly sensitive markers of photoreceptor function. ORG is an umbrella term that covers several imaging modalities that use intrinsic optical signal, i.e., without dye

Shift in excitation-inhibition balance underlies perceptual learning of temporal discrimination.

Temporal perceptual learning (TPL) constitutes a unique and profound demonstration of neural plasticity within the brain. Our understanding for the neurometabolic changes associated with TPL on the other hand has been limited in part by the use of traditional fMRI approaches. Since plasticity in the visual cortex has been shown to underlie perceptual learning of visual information, we tested the hypothesis that TPL of an auditory interval involves a similar change in plasticity of the auditory pathway and if so, whether these changes take place in a lower-order sensory-specific brain area such

CCR3 Expression in Relation to Delayed Microbleeds in a Rat Model of Large Vessel Occlusion.

UNLABELLED: Thirty percent of ischemic stroke patients develop vascular cognitive impairment and dementia (VCID) within 1 year of stroke onset. The expression of C-C motif chemokine receptor 3 (CCR3) is associated with endothelial dysfunction and memory impairment. CCR3 has been reported to increase after experimental stroke and in human stroke patients. Using an in vivo model of stroke, our study aims to link CCR3 expression with endothelial dysfunction in this rodent stroke model. METHODS: 5-hour transient Middle Cerebral Artery Occlusion (5t-MCAO) or sham surgery was performed on rats and

Visual evoked potential in generalized joint hypermobility: A case-control study.

INTRODUCTION: Generalized joint hypermobility (GJH) can be the result of several hereditary connective tissue disorders, especially Ehlers-Danlos syndrome. Cerebrovascular manifestations are among the most common complications in this disorder, and understanding their extent can help better diagnosis and prevention of hazardous events. We investigated visual evoked potential (VEP) changes in patients with GJH and compared them with healthy individuals. METHODS: Our case-control study included 90 patients who fulfilled the Beighton score (B score) for joint hypermobility and other 90 healthy p

Dementia Ideal Care: Ecosystem Map of Best Practices and Care Pathways Enhanced by Technology and Community.

BACKGROUND: Globally, much work has been done by nonprofit, private, and academic groups to develop best practices for the care of people living with dementia (PLWD), including Alzheimer's disease. However, these best practices reside in disparate repositories and tend to focus on one phase of the patient journey or one relevant group. OBJECTIVE: To fill this gap, we developed a Dementia Ideal Care Map that everyone in the dementia ecosystem can use as an actionable tool for awareness, policy development, funding, research, training, service delivery, and technology design. The intended audie

Re-WRAP (Remyelination for women at risk of axonal loss and progression): A phase II randomized placebo-controlled delayed-start trial of bazedoxifene for myelin repair in multiple sclerosis.

INTRODUCTION: Multiple sclerosis (MS) is a major cause of disability in young and middle-aged people, and myelin repair therapies are needed to slow or potentially reverse this damage. Bazedoxifene (BZA) is a selective estrogen receptor modulator identified in a novel high-throughput unbiased screen for its remyelinating potential, and its remyelinating effects were demonstrated in pre-clinical models. METHODS: This is a single-center, double blind, randomized, controlled, delayed-start Phase 2 clinical trial (NCT04002934) investigating the remyelinating effects of BZA relative to placebo. Fe

Investigation of the efficacy and safety of retinal inactivation as a treatment for amblyopia in cats.

INTRODUCTION: Deprivation of normal vision early in postnatal development elicits modifications of neural circuitry within the primary visual pathway that can cause a severe and intractable vision impairment (amblyopia). In cats, amblyopia is often modeled with monocular deprivation (MD), a procedure that involves temporarily closing the lids of one eye. Following long-term MD, brief inactivation of the dominant eye's retina can promote recovery from the anatomical and physiological effects of MD. In consideration of retinal inactivation as a viable treatment for amblyopia it is imperative to

Utilizing multimodal imaging to visualize potential mechanism for sudden death in epilepsy.

Sudden death in epilepsy or SUDEP is a fatal condition that accounts for more than 4000 deaths each year. Limited clinical and preclinical data on sudden death suggest critical contributions from autonomic, cardiac, and respiratory pathways. A potential mechanism for such sudden and severe cardiorespiratory dysregulation may be linked to acid reflux-induced laryngospasm. Here, we expand on our previous investigations and utilize a novel multimodal approach to provide visual evidence of acid reflux-initiated cardiorespiratory distress and subsequent sudden death in seizing rats. We used systemi

A Unified Rule for Binocular Contrast Summation Applies to Normal Vision and Common Eye Diseases.

PURPOSE: Binocular summation refers to better visual performance with two eyes than with one eye. Little is known about the mechanism underlying binocular contrast summation in patients with common eye diseases who often exhibit binocularly asymmetric vision loss and structural changes along the visual pathway. Here we asked whether the mechanism of binocular contrast summation remains preserved in eye disease. METHODS: This study included 1035 subjects with normal ocular health, cataract, age-related macular degeneration, glaucoma, and retinitis pigmentosa. Monocular and binocular contrast s
