Bacterialized tumor cells as vaccine.

Tumor vaccines represent a promising modality in cancer immunotherapy, but the limited immunogenicity of whole-tumor cell vaccines (WTCVs) restricts their clinical translation. Here, we developed a novel bacterialized tumor cell (BTC) vaccine by coating amino-functionalized bacterial lysates onto the surface of tumor cells, a process we define as "bacterialization". In vitro and in vivo experiments demonstrated that BTCs efficiently promote dendritic cell (DC) phagocytosis, maturation, and antigen cross-presentation, enhance DC migration to draining lymph nodes (DLNs), and remodel the DLN micr

Simvastatin as an immunomodulator and anti-myeloma agent - epidemiological, in vitro and murine model studies.

INTRODUCTION: Simvastatin (SIM), a member of the statin family, is commonly used to lower blood cholesterol levels, particularly low-density lipoprotein. Statins inhibit the action of hydroxy-methyl-glutaryl-coenzyme A reductase, an important enzyme in the synthesis of cholesterol and isoprenoids; they also have anti-inflammatory effects and can regulate membrane synthesis in cancer cells. Preliminary evidence suggests an association between SIM use and reduced cancer incidence. SIM's anti-cancer efficacy in vivo has been shown in studies on several solid tumors, but little is known about its

Alcohol Pre-Exposure in T. cruzi-Infected Mice Enhances IL-4 Production by CD8 T Cells and Modulates Innate Immunity Cytokine Response.

Trypanosoma cruzi infection, which causes Chagas' disease, induces an immune response in the host whose efficiency is important for the infection to persist or be eliminated. Alcohol consumption produces a great impact on the immune system, inducing alterations in the determination of T lymphocyte effector function, directing the profile of these cells to tolerance or inflammation. Our study aimed to evaluate, in C57BL/6 mice, the cytokine production in splenic leukocytes from T. cruzi infected and treated (EtOH) for 15 days and controls. Twenty-four mice were randomised into four groups, with

Antigen-specific TH17 cells offset the age-related decline in durable T cell immunity.

Older adults are susceptible to infections in part due to waning of immune memory. To uncover mechanisms of a long-lasting immune memory, we contrasted varicella zoster virus antigen-specific memory T cell responses in adults vaccinated at young (<20 years) or older age (>50 years) with a live-attenuated vaccine conferring durable protection only when given at young age or with an adjuvanted component vaccine eliciting long-lasting immunity in older adults. Unlike VZV-specific CD4+ T cells, CD8+ T cells exhibited profound age-sensitive changes including memory subset shifts, reduced T cell rec

HIV virion capturing liposomes for therapeutic vaccination.

HIV infection currently has no effective cures and requires lifelong antiretroviral treatments. Cures have failed due to HIV's immune evasion and rapid mutation rate. Here we present a first in class HIV therapeutic vaccine, termed nanotrap therapeutic vaccines (NTVs) that are designed to capture circulating HIV virions and facilitate internalization by local antigen presenting cells. NTVs are modified liposomes that display the CD4 mimetic molecule, CJF288, on their surfaces and have the TLR 7/8 agonist R848 loaded into their bilayer. We show that NTVs can 1) bind gp120 and capture pseudovira

Pre-treatment polyfunctionality percentage (PFA) of CD8+ T cells is associated with development of immune-related adverse events (irAEs) in patients receiving immune checkpoint inhibitors (ICIs).

INTRODUCTION: Immune checkpoint inhibitors (ICIs) have improved cancer survival, but immune-related adverse events (irAEs) occur frequently and can have devastating consequences. There are no validated methods to evaluate risk of irAEs prior to initiation of ICIs. MATERIALS AND METHODS: We conducted a pilot study evaluating the ability of blood-based, single-cell secretomic analysis to characterize irAEs. A total of 10 patients with thoracic malignancies who were scheduled to receive ICIs were enrolled. Each patient had a pre-ICI blood sample drawn as well as a sample at the time of irAE deve

Supermeres Containing the Discoidin Domain Receptor 1 Extracellular Domain Promote Collagen Alignment and Immune Exclusion in Microsatellite-Stable Colorectal Cancer.

UNLABELLED: Immune checkpoint blockade (ICB) is an effective treatment for microsatellite instability-high (MSI-H) colorectal cancers that are highly infiltrated by CD8+ T cells. Microsatellite-stable (MSS) colorectal cancers are unresponsive to ICB, at least in part, due to the paucity of intratumoral CD8+ T cells. We recently identified discoidin domain receptor 1 (DDR1) as one of four genes associated with CD8+ T-cell exclusion in MSS colorectal cancer. There are conflicting reports about the presence and role of the cleaved ectodomain (cECD) of DDR1 in mouse models of breast and pancreatic

Immune Activation Signatures Associated with Fatigue in Cancer Patients Undergoing Immune Checkpoint Inhibitor Therapy.

UNLABELLED: Fatigue is a common and often debilitating adverse event among patients receiving immune checkpoint inhibitor (ICI) therapy, yet its underlying immunologic mechanisms remain poorly defined. We prospectively collected clinical data and blood samples from patients with solid tumors receiving ICI therapy. Patients were prospectively surveyed at month 2, 4, or 6 on treatment to assess for the presence and severity of fatigue as compared with treatment baseline. We analyzed peripheral lymphocyte populations by cytometry by time of flight and peripheral levels of 39 cytokines with a Lumi

TIGIT blockade increases efficacy of PD-1 blockade combined with radiation therapy in triple-negative breast cancer model.

BACKGROUND AND PURPOSE: T-cell immunoreceptor with Ig and ITIM domains (TIGIT) suppresses functions of CD8+ T cells, and radiation therapy (RT) induces stimulation of regulatory T cells (Tregs), thereby limiting antitumor efficacy. This study aims to investigate the role of TIGIT in the immunosuppressive tumor environment and evaluate the potential of TIGIT blockade (αTIGIT) to enhance antitumor immune responses. METHODS: We analyzed public transcriptomic data to identify the expression patterns of TIGIT on T cells in breast cancer and its prognostic impact. In addition, a murine TNBC model w

Lymph node delivery of immunogenic dendritic cell exosomes via extended-tip microneedles for cancer prevention.

Cancer prevention remains a significant challenge due to the ubiquitous presence of potentially malignant cells in the body. This study presents a novel cancer vaccine platform aimed at training the immune system to prevent the development of solid tumors. We engineered a nanostimulator (nano-IS3) comprising survivin-encoding plasmid DNA, polyethylenimine, and interferon-β-expressing nanoprotoplasts to activate dendritic cells (DCs), producing immunogenic exosomes enriched with survivin-derived antigens and costimulatory molecules (IdE@S). These IdE@S were loaded into long-tipped, high-mechani

Tumour and normal tissue radiosensitivity: 2025 update.

The personalization of oncology and radiation treatments is becoming increasingly crucial in patient care. Over the past few decades, numerous assays have been developed to predict tumour response and assess the risk of radiation-induced toxicity. These tests have not yet been integrated into routine clinical practice, but recent advances in radiation oncology are paving the way for personalized strategies that consider both tumour recurrence risk and normal tissue reactions. Regarding tumour radiosensitivity prediction, no tests are currently in clinical use, although the radiosensitivity ind

CD8 + T cells may mediate the effect of gut microbiota on psoriasis: evidence from two-step mendelian randomization and bayesian weighting.

Emerging research indicates that gut microbiota and the associated immune responses are crucial in the development of chronic inflammatory skin diseases. This investigation employs Mendelian Randomization (MR) and Bayesian weighting to elucidate the causal links between gut microbiota, immune cells, and psoriasis, with a specific emphasis on CD8 + T cells. We leveraged summary statistics from genome-wide association studies (GWAS) related to gut microbiota, immune cells, and psoriasis. Single nucleotide polymorphisms (SNPs) were chosen as instrumental variables (IVs) to evaluate causal relatio

NR2F6 as a Disease Driver and Candidate Therapeutic Target in Experimental Cerebral Malaria.

Cerebral malaria (CM) is the severe progression of an infection with Plasmodium falciparum, causing detrimental damage to brain tissue and is the most frequent cause of Plasmodium falciparum mortality. The critical role of brain-infiltrating CD8+ T cells in the pathophysiology of CM having been revealed, our investigation focuses on the role of NR2F6, an established immune checkpoint, as a candidate driver of CM pathology. We employed an experimental mouse model of CM based on Plasmodium berghei ANKA (PbA) infection to compare the relative susceptibility of Nr2f6-knock-out and wild-type C57BL6

Peroxisome proliferator-activated receptor alpha is an essential factor in enhanced macrophage immune function induced by angiotensin-converting enzyme.

Increased expression of angiotensin-converting enzyme (ACE) by myeloid lineage cells strongly increases the immune activity of these cells, as observed in ACE10/10 mice, which exhibit a marked increase in antitumor and antibactericidal immunity. We report that peroxisome proliferator-activated receptor alpha (PPARα), a transcription factor that regulates genes critical for lipid metabolism, is a key molecule in the enhanced macrophage function induced by ACE. Here, we used a Cre-LoxP approach with LysM-Cre to create a modified ACE10/10 mouse line in which macrophages continue to generate abund

SPP1 + macrophages cause exhaustion of tumor-specific T cells in liver metastases.

Functional tumor-specific CD8+ T cells are essential for effective anti-tumor immune response and immune checkpoint inhibitor therapy. Here we show that, compared to other organ sites, primary, metastatic liver tumors in murine models contain a higher number of tumor-specific CD8+ T cells which are also dysfunctional. High-dimensional, multi-omic analysis of patient samples reveals a higher frequency of exhausted tumor-reactive CD8+ T cells and enriched interactions between these cells and SPP1+ macrophages in profibrotic, alpha-SMA rich regions specifically in the liver. β2-integrins as biomarkers in urothelial cancer.

β2-integrins are a family of adhesion proteins expressed in immune cells that play multiple roles in anti-tumor immunity. β2-integrins regulate tumor infiltration of anti-tumorigenic immune cells such as cytotoxic CD8 + T cells and NK cells. However, they also regulate the activity of myeloid cells, such as macrophages, which can have both anti- and pro-tumorigenic properties. The role of β2-integrins in urothelial cancer remains poorly understood. Here, we have investigated the role of different β2-integrins, and their cytoplasmic regulators, in urothelial cancer, by utilizing RNA expression

CD8+T cells and monocytes were associated with brain alterations in human immunodeficiency virus-infected individuals with cognitive impairment.

Cognitive impairment (CI) continues to be a concern for people living with HIV-1 (PLWH) in the era of antiretroviral therapy (ART), yet the underlying mechanisms remain unclear. We aimed to elucidate the structural and functional brain alterations and peripheral immune profile of PLWH with CI, as well as the correlation between them. PLWH were divided into CI (n = 30) and cognitive normal (CN, n = 59) groups based on the Montreal Cognitive Assessment, and underwent multi-modal magnetic resonance imaging. Mass cytometry was utilized to profile immune cells, while the liquid chip technique was e

IgA-dependent cell phagocytosis of HIV-infected cells elicits cross-presentation to CD8+T cells and immune memory in effector monocytes.

Mucosal IgA antibodies are the first defence against mucosal infections. Besides targeting specific antigens by their Fab-region, IgA also mediates antiviral functions via their Fc-domain, allowing infected cells destruction by antibody-dependent cellular phagocytosis (ADCP). Passive immunisation with anti-HIV-1 IgG protected Non-Human Primates in a CD8+ T cell-dependent manner, a process likely involving ADCP. Here, we unravel the consequences of ADCP of HIV-1-infected cells mediated by anti-HIV envelope IgA compared to IgG. We found that IgA-mediated ADCP, not IgG, drives viral antigen cross

CD8 T cell dynamics and immune cell trafficking in ZIKV infection: implications for neuroinflammation and therapy.

The 2015-2016 Zika virus (ZIKV) epidemic underscored the severe consequences of congenital Zika syndrome (CZS) and the broader challenges posed by neurotropic flaviviruses. As key mediators of cytotoxic immunity, CD8 T cells play a crucial and multifaceted role in ZIKV pathogenesis. While essential for controlling viral replication, their infiltration into the central nervous system (CNS)-an immune-privileged site-raises potential concerns regarding immunopathology. This review explores the dual roles of CD8 T cells during ZIKV infection, emphasizing both their antiviral functions and their po
