Association of physical activity, sedentary behavior, and sleep duration with major depressive disorder: a cross-sectional and Mendelian randomization study.

BACKGROUND: Major depressive disorder (MDD) is a common mental illness that carries a serious public health and economic burden. Although physical activity (PA), sedentary behavior (SB), and sleep duration (SD) have been shown to be associated with MDD, the causal relationship is unclear. METHODS: This study analyzed data from 22,474 participants in NHANES from 2007 to 2018. PA, SB, and SD were assessed by questionnaire, and MDD was defined by the PHQ-9 questionnaire. Associations were assessed using logistic regression modeling and controlling for demographic and health-related covariates. A

Psychological, social factors, and smoking behavior mediated the effects of cannabis use on personality disorders: A Mendelian randomization study.

BACKGROUND: Rapid changes in attitudes, legality, and patterns of cannabis use (CU) underscore the importance of understanding its impact on mental health. Although links between CU and personality disorders (PDs) are documented, their causality remains uncertain. METHODS: Employing Genome-Wide Association Studies (GWAS) data, this study investigated the causal relationship between cannabis use disorder (CUD) and lifetime cannabis use (LCU) with 9 types of PD risk through Mendelian randomization (MR) analysis. The primary method was the inverse variance weighted (IVW) method, supplemented by

Investigating whether smoking and alcohol behaviours influence risk of type 2 diabetes using a Mendelian randomisation study.

Previous studies suggest that smoking and higher alcohol consumption are associated with greater type 2 diabetes (T2D) risk. However, studies examining whether this reflects causal relationships are limited and often do not consider continuous glycaemic traits. We conducted both two-sample and one-sample Mendelian randomisation (MR), using publicly available GWAS data and UK Biobank data, respectively, to examine the potential causal effects of lifetime smoking index (LSI) and alcoholic drinks per week (DPW) on T2D and continuous traits (fasting glucose, fasting insulin and glycated haemoglobi

New evidence for the protective effect of gut microbiota regulation of ferroptosis-related proteins against osteoporosis.

Osteoporosis (OP), characterized by bone degradation and increased fracture susceptibility, constitutes a significant global health burden. Recent findings implicate gut microbiota and ferroptosis in the regulation of bone metabolism; however, causal evidence for the gut microbiota's influence on OP specifically via ferroptosis regulation remains to be established. This study employed two-sample Mendelian randomization (MR) using genome-wide association study (GWAS) summary statistics to investigate these causal relationships and delineate mediating pathways. We assessed causal links between gu

Exploring the mediating role of potential therapeutic genes in the pathogenesis of hypopituitarism through the metabolites from a genomic perspective.

Hypopituitarism is a severe endocrine disorder characterized by a partial or complete hormone deficiency in the anterior or posterior pituitary gland. Current treatment relies on hormone replacement therapy, which is unable to mimic normal physiological circadian rhythm precisely, and long-term hormone replacement therapy can result in a variety of adverse effects. This study aimed to identify potential drug targets and clarify the mechanisms underlying hypopituitarism. To identify potential therapeutic targets for hypopituitarism, summary statistics from expression quantitative trait loci (eQ

Type 1 diabetes mellitus and non-alcoholic fatty liver disease: a two-sample Mendelian randomization study.

BACKGROUND: NAFLD (Nonalcoholic fatty liver disease) is becoming an increasingly common cause of chronic liver disease. Metabolic dysfunction, overweight/obesity, and diabetes are thought to be closely associated with increased NAFLD risk. However, few studies have focused on the mechanisms of NAFLD occurrence in T1DM. METHODS: We conducted a two-sample Mendelian randomization (MR) analysis to assess the causal association between T1DM and NAFLD with/without complications, such as coma, renal complications, ketoacidosis, neurological complications, and ophthalmic complications. The association between thyroid and breast cancers: a bidirectional mendelian randomization study.

BACKGROUND: Thyroid and breast cancers are the two most frequent types of endocrine-related tumors among women worldwide, and their incidence is still on the rise. Observational studies have shown a relationship between thyroid and breast cancers. Nevertheless, many confounders predispose the results to interference. Accordingly, we performed a two-sample Mendelian randomization (MR) study to investigate the causal association between thyroid and breast cancers. METHODS: We acquired breast cancer data from the UK Biobank (13,879 breast cancer cases and 198,523 controls) and the Breast Cancer

Pathogenic Variant Frequencies in Hereditary Haemorrhagic Telangiectasia Support Clinical Evidence of Protection from Myocardial Infarction.

Hereditary haemorrhagic telangiectasia (HHT) is a vascular dysplasia inherited as an autosomal dominant trait, due to a single heterozygous loss-of-function variant, usually in ACVRL1 (encoding activin receptor-like kinase 1 [ALK1]), ENG (encoding endoglin [CD105]), or SMAD4. In a consecutive single-centre series of 37 positive clinical genetic tests performed in 2021-2023, a skewed distribution pattern was noted, with 30 of 32 variants reported only once, but ACVRL1 c.1231C>T (p.Arg411Trp) identified as the disease-causal gene in five different HHT families. In the same centre's non-overlappi

Thyroid function, pernicious anemia and erythropoiesis: a two-sample Mendelian randomization study.

Autoimmune thyroid disease (AITD) and pernicious anemia (PA) often coexist, but the directionality is unknown. In a two-sample Mendelian randomization (MR) analysis, using summary statistics from large genome-wide association studies (GWASs) in Europeans (N = 49 269-755 406), we examined the genetic associations between thyroid function, PA and markers of erythropoiesis. We performed inverse variance weighted random-effects MR, several sensitivity MR analyses, and bidirectional MR and MR Steiger for directionality. AITD and PA were associated bidirectionally (P ≤ 8 × 10-6). Neither euthyroid t

Vitamin D Levels and Risk of Juvenile Idiopathic Arthritis: A Mendelian Randomization Study.

OBJECTIVES: Observational studies report mixed findings regarding the association between vitamin D and juvenile idiopathic arthritis (JIA) incidence or activity; however, such studies are susceptible to considerable bias. Because low vitamin D levels are common within the general population and easily corrected, there is potential public health benefit in identifying a causal association between vitamin D insufficiency and JIA incidence. To limit bias due to confounding and reverse causation, we examined the causal effect of the major circulating form of vitamin D, 25-hydroxy vitamin D (25-[O

Relationship between preparation of cells for therapy and cell quality using artificial neural network analysis.

OBJECTIVE: The successful preparation of cells for therapy depends on the characterization of causal factors affecting cell quality. Ultra scale-down methods are used to characterize cells in terms of their response to process engineering causal factors of hydrodynamic shear stress and time. This response is in turn characterized in terms of causal factors relating to variations as may naturally occur during cell preparation, i.e., passage number, generation number, time of the final passage stage and hold time in formulation medium. METHODS: To investigate the influence of all of these causa

[The plague].

The word plague evokes associations with old times, but plague actually does still occur all over the world. The survival chances are at the moment much better (antibiotics) than formerly, when half of the patients died; now this figure is 15%. The causal organism is Yersinia pestis, a bacterium in fleas on rodents. This bug is a plausible candidate for attacks by bioterrosists. So, in this time, there is good reason to pay attention to it, as well as to to smallpox and anthrax and their causal organisms.
