Exosomal Non-Coding RNAs: Emerging regulators of the tumor microenvironment in gastrointestinal and hepatobiliary cancers.

Extracellular vesicle (EV)-associated non-coding RNAs (ncRNAs) are increasingly recognized as mediators of intercellular communication within the tumor microenvironment (TME), yet their mechanistic and translational significance varies substantially across gastrointestinal and hepatobiliary cancers. This narrative review, informed by a structured literature search, integrates evidence on EV-associated microRNAs, long non-coding RNAs, and circular RNAs in esophageal, gastric, colorectal, pancreatic, hepatocellular carcinoma, and cholangiocarcinoma. Rather than organizing the literature by RNA c

Exosome RAB10 inhibits JAK1/STAT1 to hinder macrophage M1 polarization and promote tumor immune escape.

Exosomes are key mediators of communication between tumor cells and the tumor microenvironment(TME); however, the mechanisms underlying exosome-mediated crosstalk between tumor cells and macrophages remain largely unclear. This study investigated the effect of exosomal RAB10 on macrophage polarization and tumor growth. Mechanistically, RAB10 delivered by breast cancer cells binds to the interferon receptor IFNAR1 and inhibits JAK1/STAT1 pathway phosphorylation, thereby impeding M1 polarization and promoting M2 polarization. RAB10 expression was significantly upregulated in drug-resistant breas

Telocytes Twenty Years on: A Critical Reappraisal of Identity, Function, and Pathological Relevance.

Telocytes are stromal cells defined by extremely long, moniliform prolongations termed telopodes and have been described in most mammalian organs since their formal designation in 2010. During the past two decades, the field has expanded from ultrastructural organ-mapping to hypotheses concerning stem-cell niche regulation, extracellular-vesicle-mediated communication, fibrosis, inflammation, and cancer-associated stromal remodelling. This expansion, however, has also generated methodological heterogeneity, with frequent reliance on non-specific markers such as CD34, PDGFRα, vimentin, and c-ki

Molecular mechanisms of folliculogenesis and oogenesis.

Folliculogenesis is a complex, multi-stage process crucial for the establishment and maintenance of female fertility through the production of a developmentally competent oocyte. Folliculogenesis, including follicular formation, activation, growth and maturation, relies on a finely tuned spatiotemporal crosstalk between germ cells, somatic cells, and the hypothalamic-pituitary-ovarian axis. This work provides a comprehensive overview of the cellular dynamics and molecular mechanisms underlying each stage of follicular development. A particular emphasis is placed on the interaction of growth fa

GDF15: A Hijacked Metabo-Hormone Orchestrating Cachexia and Immunosuppression in Cancer.

Cancer is responsible for systemic burdens, most notably cachexia and immunosuppression, that extend far beyond local tumor growth and collectively dictate poor outcomes. While often studied separately, these debilitating syndromes are deeply interconnected. On the basis of emerging evidence of growth differentiation factor 15 (GDF15)'s dual actions in immunity and metabolism, we propose that the stress-responsive hormone GDF15 is hijacked by tumors and repurposed as a central metaboceptive hub that integrates diverse oncogenic stress signals to launch a coordinated, dual pathological cascade.

The dual role of exosomes in renal fibrosis and their potential for clinical translational applications.

Renal fibrosis is the core pathological process in the progression of chronic kidney disease to its end stage. There is to date no novel therapeutic strategies that are both safe and efficient in reversing renal fibrosis in humans. Exosomes, as key mediators of intercellular communication, play significant regulatory roles in renal fibrosis. Translating mechanistic studies on exosome-mediated promotion or inhibition of renal fibrosis into clinically applicable anti-fibrotic strategies remains a challenging issue in this field. In this review, we systematically elucidate the distinct dual role

Requirements-driven automation of a social home-cage access-control system for rat cognitive testing.

BACKGROUND: Understanding cognition is essential for advancing treatments for human disorders. Rodent studies remain important for investigating underlying mechanisms. Traditional testing often involves handling, isolation and experimenter variability, which can reduce data quality. NEW METHOD: We present the automated Social RatPad, a home-cage access-control system enabling socially housed rats to reach touchscreen-based RatPad chambers without manual separation. The chamber presents stimuli, records responses and delivers pellets; this study validates the access-control and logging layer c

Macrophage Metabolic Reprogramming in Rheumatoid Arthritis: Pathogenic Mechanisms and Therapeutic Implications.

Rheumatoid arthritis (RA) is a chronic autoimmune disease characterised by persistent synovitis, progressive cartilage destruction and bone erosion. Recent advances in single-cell and spatial omics, together with immunometabolic studies, have revealed marked state heterogeneity among synovial macrophages in RA. Their metabolic reprogramming appears to sustain pathogenic cellular states, drive aberrant intercellular communication and impair the resolution of inflammation. Rather than acting as an independent initiating factor, it more likely operates as a downstream amplifier of disease. In thi

A NETs-centric multi-omics framework prioritizes PGLYRP1 and MMP9 as subtype-associated thromboinflammatory biomarker candidates and putative therapeutic hypotheses in ischemic stroke.

Neutrophil extracellular traps are implicated in immunothrombosis and neuroinflammation in ischemic stroke, but blood-based markers that distinguish subtype-specific thromboinflammatory patterns remain limited. We applied an integrated multi-omics strategy combining multi-cohort peripheral-blood transcriptomics, machine-learning feature selection, Mendelian randomization, and single-cell RNA sequencing, followed by in silico perturbation analyses and compound prioritization with molecular docking, molecular dynamics simulation, and cellular thermal shift assays. Single Cell RNA Sequencing Reveals THBS1+CD14+ Monocyte Modulates Inflammatory Activation via NRLP3-Inflammasome in Congenital Heart Block.

Isolated congenital heart block (iCHB) is defined as atrioventricular block without structural cardiac defects, characterized by irreversible fibrosis of the cardiac conduction system. Maternal autoantibodies may elicit systemic exaggerated immune responses involving type I interferon (IFN) signalling cascade, yet peripheral circulating immunity in CHB pathogenesis remains poorly understood. To investigate this, we performed single-cell RNA sequencing (scRNA-seq), followed by differential expression gene (DEG) analysis, SCENIC analysis, pseudotime analysis and cell communication analysis, to c

Neuroinflammation and vascular pathology in Alzheimer's Disease: The NRF2/HO-1 axis as a modulator of microglial phenotypes.

Alzheimer's disease (AD) frequently co-occurs with vascular pathology, and this overlap is increasingly recognized as a major driver of cognitive decline in mixed dementia and vascular cognitive impairment and dementia (VCID). Disruption of the neurovascular unit (NVU) creates a perivascular microenvironment rich in inflammatory and oxidative cues that can instruct microglial state transitions. In this review, we argue that perivascular microglia represent a key "decision hub" at the neurovascular interface, where shifts from homeostatic surveillance toward disease-associated microglia (DAM) p

Targeting NCAM1/FGFR1 Signaling to Alleviate Sleep Disturbance-Induced Neuroinflammation in the Hypothalamus.

Sleep disturbance is a prevalent public health concern associated with cognitive deficits and emotional dysregulation, yet the mechanisms underlying SD-induced hypothalamic dysfunction remain incompletely understood. Here, we found that two weeks of SD induced marked hypothalamic neuroinflammation, accompanied by increased expression of the pro-inflammatory cytokines IL-1β, IL-6, and TNF-α, as well as the chemokine Ccl2, together with reduced neuronal markers, including decreased Nissl-body and NeuN+ cells. Single-nucleus RNA sequencing (snRNA-seq) suggested that inhibitory neurons exhibited p

Patient-Derived Organoid and Single-Cell Multiomics Reveal Evolution Driven by CEACAM6/KRT19-Mediated Heterogeneity.

Colorectal cancer (CRC) remains a leading cause of cancer mortality globally, with therapeutic efficacy hindered by tumor heterogeneity and drug resistance. While organoid technology offers unprecedented opportunities to model tumor complexity, systematic platforms for chemo-targeted combination screening and molecular mechanisms underlying chemoresistance remain underexplored in Chinese populations. Here, we established a biobank of 17 patient-derived CRC organoids (85% success rate) from treatment-naïve surgical specimens, preserving histopathological fidelity and molecular diversity of pare

CRABP2 promotes peritoneal metastasis in CRC through TGF-β/Smad-mediated EMT signaling and invadopodia formation.

Peritoneal metastasis (PM) in colorectal cancer (CRC) is typically associated with a lower likelihood of successful conversion therapy and a poorer prognosis compared to metastases at other sites. Invadopodia, cancer-specific protrusive structures, play a vital role in the metastatic cascade by degrading the basement membrane and surrounding stroma. However, the exact mechanism of invadopodia regulation in PM of CRC still remains unclear. In the research, we have found that CRABP2 as a PM-associated differentially expressed gene (DEG) that promoted cell protrusion formation in CRC. CRABP2 was

Bone marrow-derived extracellular vesicles from multiple myeloma patients promote adaptive immune dysfunction via HLA-G, PD-1, and PD-L1.

INTRODUCTION: Extracellular vesicles (EVs) are critical mediators of intercellular communication and contribute to cancer progression and immune regulation. METHODS: We characterized EVs isolated from bone marrow (BM) plasma harvested from healthy donors and patients affected by Multiple Myeloma (MM) by Nano Tracking Analysis and by flow cytometry. RESULTS: EVs from MM patients were significantly more abundant and enriched in CD138, supporting their partial origin from malignant plasma cells, with additional input from BM resident cells, including monocytes and NK cells. Extracellular Vesicles and Glaucoma: Opportunities and Challenges.

PURPOSE: Glaucoma is one of the leading causes of irreversible blindness, characterized by progressive visual field loss. Several risk factors are associated with developing the disease. However, the exact mechanisms or pathological pathways involved are still unknown. There is an urgent need to find the mechanisms and biomarkers for early detection and therapy to halt progression or cure the disease. Extracellular vesicles (EVs), specifically exosomes, have emerged as a crucial player in all aspects of glaucoma, including pathogenesis to therapeutic application with their cell-cell communicat

The ErbB2-Dock7 Signaling Axis Mediates Excessive Cell Morphogenesis Induced by Autism Spectrum Disorder- and Intellectual Disability-Associated Sema5A p.Arg676Cys.

Characterized by social communication deficits and the presence of restricted and repetitive behaviors, autism spectrum disorder (ASD) is a significant neurodevelopmental condition. Genetic studies have revealed a strong association between ASD and numerous mutations that alter the function of key proteins, either through activation or inactivation. These alterations are widely hypothesized to affect neuronal morphogenesis; however, a comprehensive understanding of the specific molecular cascades driving these cellular and symptomatic changes remains lacking. In this study, we report for the f

Proteoglycans in Breast Cancer: Friends and Foes.

Proteoglycans (PGs) are highly glycosylated proteins of great importance both structurally and for signalling in the extracellular matrix (ECM) as well as cell surfaces. In breast cancer (BC), they control the structure of tissue architecture, cellular communication pathways and tumour-stroma interactions, thus affecting adhesion, migration, angiogenesis, immune evasion, and metastasis. Their structural heterogeneity supports either subtype- or context-dependent functions. This review combines current studies of PGs in BC according to their classification into intracellular, cell-surface, peri
