Hemorrhagic disease in dogs infected with an unclassified intraendothelial piroplasm in southern Brazil.

A hemorrhagic disease affecting dogs in Brazil, referred to popularly as "nambiuvú" (bloody ears) and believed to be transmitted by ticks, has been observed in animals infected with an organism described originally in 1910 as a piroplasm, and known locally as Rangelia vitalii. In this series of 10 cases, the disease was characterized by anaemia, jaundice, fever, spleno- and lymphadenomegaly, hemorrhage in the gastrointestinal tract, and persistent bleeding from the nose, oral cavity and tips, margins and outer surface of the pinnae. The ixodid ticks Rhipicephalus sanguineus and Amblyomma aureo

Integrative single-cell and multi-omics analysis of ZBTB21-mediated serine metabolism in colorectal cancer: from metabolic reprogramming to immune microenvironment modulation.

BACKGROUND: Colorectal cancer (CRC) exhibits pronounced biological diversity, a feature increasingly attributed to alterations in cellular metabolic reprograms. Serine metabolism supports nucleotide synthesis, redox balance, and epigenetic regulation via one-carbon metabolism, yet its role in shaping the tumor cellular interactions and immune landscape at single-cell level remains unclear. METHODS: Single-cell transcriptomic profiles (GSE284449) were jointly analyzed with bulk expression datasets (GSE271719), together with Mendelian randomization (MR)-based inference, to dissect serine metabo

The GLP-1-Mitochondria Axis in Metabolic Aging.

Metabolic aging underlies a cluster of chronic conditions-type 2 diabetes, cardiovascular disease, sarcopenia, and neurodegeneration-that account for a substantial share of global morbidity and mortality. A common feature is progressive mitochondrial dysfunction: impaired bioenergetics, disrupted quality control, and loss of metabolic resilience. Reduced mitochondrial DNA copy number in peripheral blood leukocytes is associated with cardiometabolic disease and mortality, but pre-analytical variability, dependence on blood-cell composition, and uncertain relationship to tissue-level function me

Salvianolic acid B enhances adipose-derived stem cells (ADSCs)-mediated perforator flap survival through antioxidative and anti-apoptotic mechanisms.

Skin flap transplantation is often complicated by ischemia-reperfusion injury, leading to tissue necrosis. Salvianolic acid B (Sa B), a key component of Salvia miltiorrhiza, possesses known antioxidative and anti-apoptotic properties. This study investigated whether Sa B preconditioning could enhance the regenerative capacity of adipose-derived stem cells (ADSCs) before transplantation for improving flap survival. We assessed the effects of Sa B on ADSCs viability, migration, differentiation, and resilience to oxidative stress in vitro under normal and oxygen-glucose deprivation (OGD) conditio

Spinal Implant-Associated Infection in Type 2 and Type 1 Diabetes: Phenotype-Specific Inflammatory Features and Therapeutic Response to Semaglutide.

BACKGROUND: Diabetes mellitus (DM) is a major risk factor for postoperative infection and wound complications in spine surgery, yet distinctions between Type 2 (T2DM) and Type 1 (T1DM) pathophysiology are rarely addressed. This study compares infectious burden, wound healing, and immune response among a murine model of spinal implant-associated infection of T2DM, T1DM, and nondiabetic control mice before and after metabolic intervention with the GLP-1 receptor agonist (GLP-1RA), semaglutide. METHODS: Male C57BL/6J mice were rendered diabetic by streptozotocin induction (T1DM) or a high-fat, h

Ingestible Hydrogel-Encapsulated Optogenetic Yeast Controlled by Wearable Electronics Enables Programmable IL-10 Delivery for Inflammatory Bowel Disease Therapy.

Engineered yeast-living drugs offer significant potential for oral protein therapy but remain limited by poor intestinal retention and lack of precise spatiotemporal control over therapeutic delivery. Here, we developed an ingestible hydrogel-encapsulated optogenetic yeast system controlled by a wearable electronic device for programmable interleukin-10 (IL-10) delivery in inflammatory bowel disease (IBD). We engineered Pichia pastoris with an EL222-based blue-light-inducible circuit achieving rapid, stringent IL-10 secretion (50% saturated concentration within just 1 h upon induction). The ye

Efficient eukaryotic expression and potent antiviral activity of a long-acting recombinant feline interferon-ω2-Fc fusion protein against major feline viruses.

Feline interferon-ω2 (FeIFN-ω2) holds potential as a therapeutic agent against feline viral infections. However, its clinical application is limited by rapid clearance and suboptimal antiviral effectiveness. Thus, in this study, an Fc-fused construct, FeIFN-ω2-Fc, was engineered to improve antiviral potency and pharmacokinetic properties both in vitro and in vivo. Three recombinant constructs-native FeIFN-ω2, FeIFN-ω2-Dimer, and FeIFN-ω2-Fc-were expressed in Chinese Hamster Ovary cells. All showed strong antiviral activity (106.74-108.42 IU/mg) and effectively activated downstream interferon s

Avian HEV triggered egg decline: Viral-host, immune-neuroendocrine mechanisms in layers.

Avian hepatitis E virus (aHEV), a member of the Hepeviridae family, poses substantial threats to poultry health and productivity, particularly through its detrimental impact on egg production in laying hens. This review elucidates the multifaceted mechanisms underlying aHEV-induced egg production decline in laying hens, focusing on direct viral-host interactions, immune dysregulation, and neuroendocrine disruption. aHEV exhibits strong tropism for reproductive organs, with active replication in ovarian tissues causing follicular apoptosis, structural damage, and hormonal imbalance. Liposomal irinotecan (Onivyde): Exemplifying the benefits of nanotherapeutic drugs.

Irinotecan is a topoisomerase inhibitor, widely used in treatment of malignancies including pancreatic ductal adenocarcinoma (PDAC) as part of the FOLFIRINOX regimen prescribed as a first-line treatment in several countries. However, irinotecan has not been successfully introduced as a second-line treatment for pancreatic cancer and few randomized clinical studies have evaluated its added value. Efficacy of liposomal irinotecan (nal-IRI) combined with 5-fluorouracil and leucovorin (5-FU/LV) was reported in the phase III NAPOLI-1 trial in metastatic PDAC following failure of gemcitabine-based t

Strontium enhances BMP-2 mediated bone regeneration in a femoral murine bone defect model.

The application of strontium is one option for the clinical treatment of osteoporosis-a disease characterized by reduced bone density and quality-in order to reduce the risk of vertebral and nonvertebral fractures. Unlike other drugs used in osteoporosis therapy, strontium shows a dual effect on bone metabolism by attenuating cellular resorption and simultaneously enhancing new bone tissue formation. Current concerns regarding the systemic application of highly dosed strontium ranelate led to the development of strontium-modified scaffolds based on mineralized collagen (MCM) capable to release

Identification and validation of a dysregulated TME-related gene signature for predicting prognosis, and immunological properties in bladder cancer.

BACKGROUND: During tumor growth, tumor cells interact with their tumor microenvironment (TME) resulting in the development of heterogeneous tumors that promote tumor occurrence and progression. Recently, there has been extensive attention on TME as a possible therapeutic target for cancers. However, an accurate TME-related prediction model is urgently needed to aid in the assessment of patients' prognoses and therapeutic value, and to assist in clinical decision-making. As such, this study aimed to develop and validate a new prognostic model based on TME-associated genes for BC patients. METH

Folate metabolism in tumor-associated macrophages drives immunosuppressive function to promote tumor growth.

Tumor-associated macrophages (TAMs) promote tumor growth, inhibit effector lymphocytes, and induce resistance to immune checkpoint inhibitors (ICI). Therapies designed to deplete TAMs have had little success in the clinic, and strategies aimed to repolarize immunosuppressive TAMs to pro-inflammatory states have struggled with systemic toxicity. We have identified that the combination of low folate in the tumor microenvironment and TAM dependency on the high affinity folate receptor beta (FRβ) for folate uptake results in a unique metabolic dependency of TAMs necessary for their immunosuppressi

Gene-guided repurposing identifies dihydroergotamine as a candidate inhibitor of the BCL2-SIVA1 axis in advanced gastric cancer in vitro.

BACKGROUND: Advanced gastric cancer is frequently refractory to therapy because of multidrug resistance and apoptotic escape. BCL2 and SIVA1 are core apoptosis regulators, and their direct interaction suggests a potentially druggable BCL2–SIVA1 signaling axis that remains insufficiently explored in advanced gastric cancer. METHODS: We integrated bulk transcriptomics, single-cell RNA sequencing, spatial transcriptomics, and immune infiltration analyses from GEO/TCGA to characterize the BCL2–SIVA1 axis and applied a bioinformatics-guided drug-repurposing workflow (2D-QSAR machine learning, molec

Bone marrow mesenchymal stem cell-derived exosomes alleviate DSS-induced inflammatory bowel disease in mice through inhibiting intestinal epithelial cell pyroptosis via delivery of TSG-6.

BACKGROUND: Inflammatory bowel disease (IBD), characterized by chronic intestinal inflammation and epithelial barrier dysfunction, remains a therapeutic challenge due to the limitations of current treatments, including drug resistance and invasive surgical risks. Emerging evidence implicates intestinal epithelial cells (IECs) pyroptosis as a key contributor to IBD progression. Bone marrow mesenchymal stem cell exosomes (BMSCs-Exo) exhibit anti-inflammatory and tissue-reparative potential, yet the role of tumor necrosis factor-stimulated gene 6 (TSG-6), a critical anti-inflammatory mediator in

Essential roles of B cell subsets in the progression of MASLD and HCC.

BACKGROUND & AIMS: Hepatocellular carcinoma (HCC) is the third leading cause of cancer-related death. Metabolic dysfunction-associated steatotic liver disease (MASLD) is a significant cause of HCC. Current treatment options for HCC are very limited. Recent evidence highlights B cells as key drivers in MASLD progression toward HCC. However, it remains unclear whether multiple B cell populations or a distinct B cell subset regulates inflammatory responses during liver disease progression. The scope of this study was to define protumorigenic B cell subsets in MASLD and HCC. METHODS: Multicolor f

Shared PRAME Epitopes are T-Cell Targets in NUT Carcinoma.

BACKGROUND: NUT carcinoma is a rare but highly lethal solid tumor without an effective standard of care. NUT carcinoma is caused by bromodomain-containing NUTM1 fusion oncogenes, most commonly BRD4::NUTM1 . BRD4::NUTM1 recruits p300 to acetylate H3K27 forming expansive stretches of hyperacetylated chromatin called "megadomains" with the overexpression of corresponding oncogenes, including MYC . We hypothesized that transcriptional dysregulation caused by BRD4::NUTM1 would lead to the generation of cancer-specific antigens that could be therapeutically actionable. METHODS: We integrated genomi

Advancements in understanding neuropsychiatric lupus: deciphering immune heterogeneity with single-cell resolution.

Neuropsychiatric Systemic Lupus Erythematosus (NPSLE) represents one of the most severe and enigmatic manifestations of SLE, contributing significantly to disease morbidity and mortality. Its clinical presentation is profoundly heterogeneous, encompassing 19 distinct syndromes that pose considerable diagnostic and therapeutic challenges. While historical estimates of its prevalence varied widely, contemporary prospective studies suggest that approximately 56% of SLE patients experience a neuropsychiatric event, with 30-50% of these events being directly attributable to SLE-related pathogenic m

Osteogenesis-Related Long Noncoding RNA GAS5 as a Novel Biomarker for Osteonecrosis of Femoral Head.

Background: The lack of effective biomarkers makes it difficult to achieve early diagnosis and intervention for osteonecrosis of the femoral head (ONFH). Hence, we aimed to identify novel long noncoding RNA (lncRNA) biomarkers for ONFH. Methods: High-throughput RNA sequencing was performed to detect lncRNA and mRNA expression levels in subchondral bone samples from three patients with ONFH and three patients with femoral neck fractures. Integrated bioinformatics analyses were conducted to identify lncRNAs associated with ONFH development and their potential functions and signaling pathways. A
