Motor Timing in Tourette Syndrome: The Effect of Movement Lateralization and Bimanual Coordination.

The study of motor timing informs on how temporal information integrates with motor acts. Cortico-basal ganglia and cortico-cerebellar circuits control this integration, whereas transcallosal interhemispheric connectivity modulates finely timed lateralized or bimanual actions. Motor timing abilities are under-explored in Tourette syndrome (TS). We adopted a synchronization-continuation task to investigate motor timing in sequential movements in TS patients. We studied 14 adult TS patients and 19 age-matched healthy volunteers. They were asked to tap in synchrony with a metronome cue (SYNC) and

Reversing Cocaine-Induced Plasticity with Zeta Inhibitory Peptide.

Cocaine-induced plasticity persists during abstinence and is thought to underlie cue-evoked craving. Reversing this plasticity could provide an opportunity for therapeutic intervention. Converging evidence suggest that zeta inhibitory peptide (ZIP) eliminates memories for experience-dependent behaviors, including conditioned drug associations. However, the effect of ZIP on reward seeking and drug-induced plasticity is unknown. The current study examined the effect of ZIP administration in the nucleus accumbens on reinstatement (RI) of cocaine seeking, a rodent model of relapse. We demonstrate

Neural Dynamics of Variable Grasp-Movement Preparation in the Macaque Frontoparietal Network.

Our voluntary grasping actions lie on a continuum between immediate action and waiting for the right moment, depending on the context. Therefore, studying grasping requires an investigation into how preparation time affects this process. Two macaque monkeys (Macaca mulatta; one male, one female) performed a grasping task with a short instruction followed by an immediate or delayed go cue (0-1300 ms) while we recorded in parallel from neurons in the grasp preparation relevant area F5 that is part of the ventral premotor cortex, and the anterior intraparietal area (AIP). Initial population dynam

Oxytocin receptor signaling in the dorsal lateral septum is essential for social transmission of food preference in mice.

Social learning is a fundamental adaptive strategy that critically shapes decision-making and feeding behaviors. Through the social transmission of food preference (STFP), animals acquire information about food safety via social interactions, forming a food-odor memory that overrides innate preferences and guides subsequent food preferences. However, the neural mechanisms through which social cues durably influence feeding preferences remain unclear. Here, we identify the lateral septum (LS) as a key node in STFP. We show that LS neuronal activity increases following social learning and reliab

Limonene Modulates Event-Related Potentials in a 2-Back Working Memory Task.

INTRODUCTION: Olfactory inputs can influence visual cognition. However, it remains unclear whether such effects are driven by the chemical properties of odorants or by mood-related associations, especially under conditions of high cognitive demand. This study examined whether limonene, a key citrus odorous compound, modulates cognitive processing during a visual 2-back task under different levels of perceptual load. METHODS: We developed a 2-back working memory task using color lightness discrimination. The task included both low-load (orange) and high-load (gray) conditions. Participants wor

The right posterior parietal cortex mediates spatial reorienting of attentional choice bias.

Attention facilitates behavior by enhancing perceptual sensitivity (sensory processing) and choice bias (decisional weighting) for attended information. Whether distinct neural substrates mediate these distinct components of attention remains unknown. We investigate the causal role of key nodes of the right posterior parietal cortex (rPPC) in the forebrain attention network in sensitivity versus bias control. Two groups of participants performed a cued attention task while we applied either inhibitory, repetitive transcranial magnetic stimulation (n = 28) or 40 Hz transcranial alternating curr

Is P3 amplitude associated with greater gaze distraction effect in schizotypy?

A recently proposed "Hyperfocusing hypothesis" suggests that schizotypy is associated with a more narrow but more intense way of allocating attention. The current study aims to test a vital prediction of this hypothesis in a social context, that schizotypy may be related to greater difficulty overcoming the distracting effects of gaze. This could cause a longer time to respond to targets that are invalidly cued by gaze. The current study tested this prediction in a modified Posner cueing paradigm by using P3 as an indicator for attentional resources. Seventy-four young healthy individuals with

Increased risky choice during forced abstinence from fentanyl on the cued rat gambling task.

RATIONALE: The use of illicit opioids has arguably never been more risky; street drug potency can be dangerously high, is often unknown to the consumer, and results in multiple daily fatalities worldwide. Furthermore, substance use disorder (SUD) is associated with increased maladaptive, risky decisions in laboratory-based gambling tasks. Animal studies can help determine whether this decision-making deficit is a cause or consequence of drug use. However, most experiments have only assessed psychostimulant drugs. OBJECTIVES: To assess differences in decision-making strategies both before, dur

Cocaine diminishes consolidation of cued fear memory in female rats through interactions with ventral hippocampal D2 receptors.

In addition to cocaine's addictive properties, cocaine use may lead to heightened risk-taking behavior. The disruptive effects of cocaine on aversive memory formation may underlie this behavior. The present study investigated the effects of cocaine on fear memory using a cued fear conditioning paradigm in female Sprague Dawley rats, and further determined the role of D2 receptors in modulating the effect of cocaine on cued fear expression. Animals received six evenly spaced shocks preceded by a tone. The following day, rats were returned to the fear chamber where tones, but no shocks, were del

Context-drug-associations and reinstatement of drug-seeking behavior in male rats: Adolescent and adult time-dependent effects.

RATIONALE: Drug use during adolescence results in a life-long risk to develop substance-use disorders. Adolescent rats are sensitive to different drug-associated cues, compared to adults; however, the contribution of adolescent-formed context-drug-associations to elicit relapse-like behavior is underexplored. OBJECTIVES: The present study compared the effect of adolescent vs adult-formed context-drug associations to elicit time-dependent increases in cocaine-seeking behavior. This objective was accomplished using an abbreviated (ABRV) operant cocaine self-administration (Coc-SA), Extinction (

Neural evidence that disengaging memory retrieval is modulated by stimulus valence and rumination.

To remember information from our personal past we need to be in a cognitive state where we treat stimuli as cues for memory retrieval. In this study we considered whether participants could exert control and disengage from a memory state when it was no longer required for the task at hand. In particular, we examined whether this ability was affected by the valence of the stimuli and participant's rumination scores. After a study phase participants completed test blocks where the task switched every two trials between a memory task (retrieving information from the study phase) and a perceptual

Mapping sign-tracking and goal-tracking onto human behaviors.

As evidenced through classic Pavlovian learning mechanisms, environmental cues can become incentivized and influence behavior. These stimulus-outcome associations are relevant in everyday life but may be particularly important for the development of impulse control disorders including addiction. Rodent studies have elucidated specific learning profiles termed 'sign-tracking' and 'goal-tracking' which map onto individual differences in impulsivity and other behaviors associated with impulse control disorders' etiology, course, and relapse. Whereas goal-trackers are biased toward the outcome, si

Assaying Fear Memory Discrimination and Generalization: Methods and Concepts.

Generalization describes the transfer of conditioned responding to stimuli that perceptually differ from the original conditioned stimulus. One arena in which discriminant and generalized responding is of particular relevance is when stimuli signal the potential for harm. Aversive (fear) conditioning is a leading behavioral model for studying associative learning and memory processes related to threatening stimuli. This article describes a step-by-step protocol for studying discrimination and generalization using cued fear conditioning in rodents. Alternate conditioning paradigms, including co

Unaware of the amount consumed: Systematic error in estimating food- and drink intake.

BACKGROUND: Our current food environment promotes overconsumption due to the overrepresentation of foods that have a high calorie density and can be easily consumed. These food characteristics lead to limited oro-sensory exposure, which may lead to overconsumption due to insufficient perception of the amount consumed. Better perception of the amount eaten and thus a better ability to estimate intake may help control actual food intake through prolonged inter-meal interval and smaller meal sizes. OBJECTIVE: The aim of this study was to determine whether food form, flavor and portion size influ

Human frequency following responses to iterated rippled noise with positive and negative gain: Differential sensitivity to waveform envelope and temporal fine-structure.

The perceived pitch of iterated rippled noise (IRN) with negative gain (IRNn) is an octave lower than that of IRN with positive gain (IRNp). IRNp and IRNn have identical waveform envelopes (ENV), but differing stimulus waveform fine structure (TFS), which likely accounts for this perceived pitch difference. Here, we examine whether differences in the temporal pattern of phase-locked activity reflected in the human brainstem Frequency Following Response (FFR) elicited by IRNp and IRNn can account for the differences in perceived pitch for the two stimuli. FFRs using a single onset polarity were

Interpretation training to target repetitive negative thinking in generalized anxiety disorder and depression.

OBJECTIVE: Repetitive negative thinking (RNT) for example, worry in generalized anxiety disorder (GAD) and rumination in depression, is often targeted during psychological treatments. To test the hypothesis that negative interpretation bias contributes to worry and rumination, we assessed the effects of inducing more positive interpretations in reducing RNT. METHOD: Volunteers diagnosed with GAD (66) or depression (65) were randomly allocated to one of two versions of cognitive bias modification for interpretation (CBM-I), either with or without RNT priming prior to training, or a control con

Attentional impairments in Huntington's disease: A specific deficit for the executive conflict.

OBJECTIVE: Huntington's disease (HD) is characterized by motor and cognitive impairments including memory, executive, and attentional functions. However, because earlier studies relied on multidetermined attentional tasks, uncertainty still abounds regarding the differential deficit across attentional subcomponents. Likewise, the evolution of these deficits during the successive stages of HD remains unclear. The present study simultaneously explored 3 distinct networks of attention (alerting, orienting, executive conflict) in preclinical and clinical HD. METHOD: Thirty-eight HD patients (18 p

Imitation inhibition in children with Tourette syndrome.

OBJECTIVE: Echopraxia, that is, the open and automatic imitation of other peoples' actions, is common in patients with Gilles de la Tourette syndrome, autism spectrum disorder, and also those with frontal lobe lesions. While systematic reaction time tasks have confirmed increased automatic imitation in the latter two groups, adult patients with Tourette syndrome appear to compensate for automatic imitation tendencies by an overall slowing in response times. However, whether children with Tourette syndrome are already able to inhibit automatic imitation tendencies has not been investigated. ME

Disrupted Default Mode Network and Basal Craving in Male Heroin-Dependent Individuals: A Resting-State fMRI Study.

BACKGROUND: Craving is associated with a high probability of relapse. However, the relationship between functional connectivity in the default mode network (DMN) during resting state and basal craving of heroin-dependent individuals remains unknown. METHODS: Data used in the present study were collected between August 10, 2009, and June 28, 2011. Twenty-four male heroin-dependent individuals based on DSM-IV criteria and 20 male healthy control subjects participated in a study of resting-state functional magnetic resonance imaging. The Hyper-Modular Associative Mind: A Computational Analysis of Associative Responses of Persons with Asperger Syndrome.

Rigidity of thought is considered a main characteristic of persons with Asperger syndrome (AS). This rigidity may explain the poor comprehension of unusual semantic relations, frequently exhibited by persons with AS. Research indicates that such deficiency is related to altered mental lexicon organization, but has never been directly examined. The present study used computational network science tools to compare the mental lexicon structure of persons with AS and matched controls. Persons with AS and matched controls generated free associations, and network tools were used to extract and compa
