Retrospective Review of Rush Allergen-Specific Immunotherapy Safety in Cats With Atopic Skin Syndrome.

BACKGROUND: Rush immunotherapy (RIT) accelerates the induction phase of allergen-specific immunotherapy treatment (ASIT) by administering incrementally increasing allergen doses over a single day. This approach reduces the time required to maintenance dosing. Safety information in cats with atopic skin syndrome (FASS) remains limited. HYPOTHESIS/OBJECTIVES: The objective of the study was to evaluate the safety of RIT in cats with hypersensitivity disease compatible with FASS and to describe the frequency and nature of adverse events. ANIMALS: Forty-seven client-owned cats that underwent RIT.

Exploring the impact of IL-2 cytokine family on skin barrier function: pathophysiology and therapeutic strategies.

This review examines the regulatory roles of the IL-2 cytokine family in maintaining or disrupting skin barrier immunity, focusing on mechanisms that impair or restore barrier function and their therapeutic exploitation. A narrative review of PubMed and Google Scholar literature up to November 2025 was conducted. The IL-2 cytokine family, which includes IL-2, IL-4, IL-7, IL-9, IL-15, and IL-21, shapes cutaneous immunity through effects on keratinocyte differentiation, T-cell activation, barrier protein expression, and inflammatory signaling. Among these cytokines, IL-4 occupies a pivotal posit

From Bald to Bold: Reversal of Alopecia Totalis in an Adolescent Using Dupilumab Monotherapy.

INTRODUCTION: Alopecia areata (AA) is an autoimmune condition marked by non-scarring, patchy hair loss of the scalp which progresses in <10% of cases to alopecia totalis (AT), which is marked by complete hair loss from the scalp, eyebrows, and eyelashes. AA is prevalent in pediatric patients and is associated with atopic dermatitis (AD). The interleukin (IL)-4 and IL-13 antagonist dupilumab is approved for use in both pediatric and adult patients with AD and asthma. However, in some cases, dupilumab has shown promising results in regrowing hair in patients with concurrent AA. CASE PRESENTA

A spatial transit-retention axis reveals adaptive immune organisation in psoriatic disease.

Spatial organisation of immune cells reflects a balance between tissue anchoring and migratory compatibility, yet how this balance is structured within inflammatory skin disease remains poorly understood. Spatial transcriptomic analyses defined transit-retention immune organisation across lesional and non-lesional skin in atopic dermatitis (AD) and psoriasis (PsO). A unified transit-retention axis captured contextual immune organisation within leukocyte-rich tissue microenvironments. Non-lesional skin in both diseases exhibited retention-dominant organisation. In contrast, psoriatic lesions sh

[Gut microbiota and allergic diseases].

Incidence of allergic diseases continues to increase, particularly among children, and has become an important public health concern. As a key link connecting environmental exposure, immune development, and susceptibility to allergic diseases, the gut microbiota has emerged as an important focus of related research. The gut microbiota plays a crucial role in immune system maturation during early life, establishment of immune tolerance, and maintenance of mucosal barrier integrity. Gut dysbiosis may contribute to the occurrence and progression of allergic diseases through multiple mechanisms, i

Dupilumab-associated lymphoproliferative disorders: a comprehensive review on clinicohistopathologic features and underlying mechanisms.

Dupilumab, a monoclonal antibody targeting interleukin 4 receptor, has shown significant efficacy in treating atopic dermatitis (AD). However, emerging case reports indicate that it may unmask or cause cutaneous T cell lymphoma (CTCL). This review analyzes 29 studies involving 124 patients who developed lymphoproliferative disorders after dupilumab, including 13 cases of lymphoproliferative reactions not meeting lymphoma criteria. The median time from dupilumab initiation to biopsy-confirmed lymphoproliferative disorders was 5 months, with 39.05% of cases in advanced stages. Histopathological

Oral JAK inhibitors for pediatric inflammatory skin disease.

PURPOSE OF REVIEW: JAK inhibitors have emerged as a promising new therapy for inflammatory skin disease, including in pediatric populations. However, due to their boxed warnings and lack of long-term safety and efficacy data, their specific role in the treatment ladder for various conditions remains to be determined. RECENT FINDINGS: JAK inhibitors are currently approved for pediatric atopic dermatitis and alopecia areata with evidence of both efficacy and safety. They are currently being investigated for use in children with vitiligo, psoriasis, juvenile dermatomyositis, and hidradenitis sup

Clinical characteristics and management of dupilumab-associated ocular surface disease in Japan.

Dupilumab-associated ocular surface disease (DAOSD) is one of the most common adverse events associated with dupilumab (an anti-interleukin-4-receptor-alpha monoclonal antibody) during the treatment of patients with atopic dermatitis (AD). However, it rarely occurs in patients with bronchial asthma or chronic rhinosinusitis with nasal polyps. Adequate understanding of DAOSD is important for proper diagnosis and appropriate ophthalmic intervention. The aim of this review was to summarize and discuss the clinical characteristics and management of DAOSD in Japan. The pathogenesis of DAOSD can be

The neuroscience of itch in relation to transdiagnostic psychological approaches.

The experience of itch and its associated chronic conditions (i.e., atopic dermatitis) form a significant burden of disease. Knowledge of how the brain processes itch, that might occur uniquely for chronic itch populations, could be used to guide more effective psychotherapeutic interventions for these groups. To build the evidence base for such approaches, we conducted a series of coordinates-based fMRI analyses, to identify the shared neural mechanisms for itch across the published literature. Upon so doing, we identified a core "itch network" that spans the Basal Ganglia/Thalamus, Claustrum

Neonatal skin health and associated dermatological conditions.

PURPOSE OF REVIEW: This review describes recent developments in neonatal skincare management and situates these findings within the preexisting literature on neonatal dermatology. RECENT FINDINGS: The studies included in this review expand research methods evaluating skincare management to different contexts across the world. Several studies explore the roles of emollient therapy, disinfection, and skin-to-skin contact on improving neonates' long-term health outcomes. Recent findings also assess the impact of neonatal interventions on atopic dermatitis risk later in life as well as epidemiolo

The association between atopic eczema and lymphopenia: Results from a UK cohort study with replication in US survey data.

BACKGROUND: Lymphocyte skin homing in atopic eczema (AE) may induce lymphopenia. OBJECTIVE: To determine if AE is associated with lymphopenia. METHODS: We used UK primary care electronic health records (Clinical Practice Research Datalink GOLD) for a matched cohort study in adults (18 years+) (1997-2015) with at least one recorded lymphocyte count. We matched people with AE to up to five people without. We used multivariable logistic regression to estimate the association between AE and lymphopenia (two low lymphocyte counts within 3 months) and linear mixed effects regression to estimate th

CD1a-mediated immunity from a molecular perspective.

Human CD1a is a non-polymorphic glycoprotein that presents lipid antigens to T cells. The most obvious role of CD1a is associated with its expression on Langerhans cells in epidermis, where it is involved in responses to pathogens. Antigen-specific T cells are believed to co-recognise CD1a presenting bacterial antigens such as species of lipopeptides from Mycobacterium tuberculosis. Further, human skin contains large amount of endogenous lipids that can activate distinct subsets of CD1a-restricted autoreactive T cells, mostly belonging to the αβ lineage, which are abundant in human blood and s

The Mind Body Connection in Dermatologic Conditions: A Literature Review.

Psychodermatology pertains to the relationship between the skin and brain. This review aims to summarize the evidence of the mind body connection in four psychophysiological conditions: rosacea, atopic dermatitis (AD), acne vulgaris (AV), and psoriasis. A literature search was conducted using several English language databases. All four conditions share similar psychiatric co-morbidities, including but not limited to anxiety, depression, and suicidality. In rosacea, the upregulation of transient receptor potential vanilloid type 1, Toll like receptor 2, and Th17 cells releases downstream produ

Individual Article: Efficacy of Over-The-Counter Cream in Reducing Eczema Signs.

BACKGROUND: Eczema, or atopic dermatitis (AD), is a chronic relapsing skin disease associated with unpredictable flares of erythema, rash, and pruritus. AD arises from a combination of immune system dysregulation and abnormal skin barrier function. Skin barrier support with proper skincare regimens have a central role in management. METHODS: This was a multi-center, 12-week in-use study of a skincare regimen in children and adults with mild-to-moderate eczema (6-16) on the Patient-Oriented Eczema Measure (POEM), and ≥2 flares within 3 months prior to screening. The regimen included Itch Re

Elevated serum IgE levels are not associated with poor treatment outcome in psoriasis vulgaris.

Psoriasis is a chronic inflammatory disorder which is associated with impaired skin barrier function. In this context, it was shown that serum IgE level was elevated in significant proportion of psoriasis patients. However, whether serum IgE levels are associated with treatment outcomes of psoriasis has not been understood. We retrospectively analyzed psoriasis patients who visited our clinics through electromedical records. Patients with history of atopic dermatitis were excluded. Total of 483 patients clinically or pathologically diagnosed with psoriasis vulgaris were included for analyses.

Bitter taste receptor T2R38 is expressed on skin-infiltrating lymphocytes and regulates lymphocyte migration.

Bitter taste receptors (T2Rs) are G protein-coupled receptors involved in the perception of bitter taste on the tongue. In humans, T2Rs have been found in several sites outside the oral cavity. Although T2R38 has been reported to be expressed on peripheral lymphocytes, it is poorly understood whether T2R38 plays immunological roles in inflammatory skin diseases such as atopic dermatitis (AD). Then, we first confirmed that T2R38 gene expression was higher in lesional skin of AD subjects than healthy controls. Furthermore, skin T2R38 expression levels were correlated with serum thymus and activa

Hyper-IgE and Carcinoma in CADINS Disease.

BACKGROUND: Atopic dermatitis (AD) affects up to 25% of children and 10% of adults in Western countries. When severe or recurrent infections and exceedingly elevated serum IgE levels occur in AD patients, an inborn error of immunity (IEI) may be suspected. The International Union of Immunological Societies classification lists variants in different genes responsible for so-called Hyper-IgE syndromes. Diagnosing an underlying IEI may influence treatment strategies. METHODS: Clinical and diagnostic workup of family members are presented including a detailed immunological description and histolo
