[Catecholamines and their metabolites in children with Asperger and Kanner syndromes].

Children with Asperger and Kanner syndromes in the stable state demonstrate similar decrease in plasma norepinephrine. In the aggravated state, these changes become more expressed and are characterized by a decrease in plasma tyrosine, norepinephrine, normetanephrine and by an increase in dopamine and homovanylic acid and a decrease in excretion of norepinephrine and an increase in excretion of homovanylic acid, epinephrine and MHPG. Only in children with Kanner syndrome in the aggravated state plasma MHPG increases, excretion of tyrosine decreases and excretion of normetanephrine increases. T

Psilocybin induces schizophrenia-like psychosis in humans via a serotonin-2 agonist action.

Psilocybin, an indoleamine hallucinogen, produces a psychosis-like syndrome in humans that resembles first episodes of schizophrenia. In healthy human volunteers, the psychotomimetic effects of psilocybin were blocked dose-dependently by the serotonin-2A antagonist ketanserin or the atypical antipsychotic risperidone, but were increased by the dopamine antagonist and typical antipsychotic haloperidol. These data are consistent with animal studies and provide the first evidence in humans that psilocybin-induced psychosis is due to serotonin-2A receptor activation, independently of dopamine stim

[Treatment of orthostatic hypotension with metoclopramide].

A 38 year-old woman, with orthostatic hypotension secondary to autonomic dysfunction accompanied by sympathetic hyperactivity and excess of dopamine, was treated with an antagonistic dopaminergic drug, metoclopramide, 30 mg/day, with good tolerance. Support intervention as well as the use of various drugs (beta-blockers, caffeine, fludrocortisone, haloperidol) showed no effect. The answer to metoclopramide both subjective and objective were remarkable. The way in which metoclopramide acts has not yet been fully clarified. Being a widely used drug, well tolerated, its use must be considered in

[Primary dexamethasone-suppressible hyperaldosteronism and hyperprolactinemia].

This report describes a clinical case with rare association: dexamethasone-suppressible hyperaldosteronism (DSH) and hyperprolactinemia. Previous studies reported that endogenous dopaminergic activity would be increased in both primary aldosteronism and in hyperprolactinemic syndromes. By manipulating dopamine tone with metoclopramide, bromocriptine, ACTH and amineptine, the aldosterone and prolactin responsiveness was evaluated. Our results showed a lack of dopamine inhibiting aldosterone secretion, despite dopaminergic tone is known to be increased in hyperprolactinemia. Thus it seems unlike

Relationship between urinary excretion of homovanillic acid and norepinephrine metabolites in normal subjects and patients with orthostatic hypotension.

Patients with neurogenic orthostatic hypotension due to multiple system atrophy (MSA) or pure autonomic failure (PAF) excrete lower amounts of homovanillic acid (HVA) than do normal subjects. There is a highly significant correlation between the rates of excretion of HVA and norepinephrine metabolites. The regression line relating excretion of the dopamine and norepinephrine metabolites suggests that about one third of dopamine formed in noradrenergic neurons is converted to norepinephrine and the remainder metabolized, mainly to HVA. About one fourth of urinary HVA appears to be derived from

In vivo electrochemical evidence for an enkephalinergic modulation underlying stereotyped behavior: reversibility by naloxone.

The effect of the enkephalin pentapeptide analog (WY 42,896) on amphetamine-induced stereotypy was studied in male, albino rats. WY 42,896 significantly inhibited amphetamine-induced stereotypy. The inhibition of the head-bobbing and sniffing components was significantly reversed by naloxone. Disinhibition of the rearing components by naloxone reached borderline significance. In vivo electrochemical measurements in rat caudate showed that WY 42,896 inhibited both basal and amphetamine-induced dopamine release. This inhibition was prevented by naloxone. These data show an enkephalinergic-dopami

Urinary excretion of catecholamines and their metabolites in patients with renovascular hypertension.

Urinary excretion of catecholamines, vanillylmandelic acid, and methoxycatecholamines was studied in 50 patients with renovascular hypertension. The urinary excretion of noradrenaline was increased in patients with elevated peripheral plasma renin activity and in those with elevated renal vein renin ratio in comparison with the patients with normal plasma renin activity. The urinary excretion of dopamine was diminished and vanillylmandelic acid increased in the entire group of patients with renovascular hypertension. In the investigated group, following sodium restriction, the mean urinary exc

Abnormal central monoamine metabolism in humans with "true hypersomnia" and "sub-wakefulness".

A case of Kleine-Levin syndrome with true hypersomnia and a case of sub-wakefulness are described. In both patients lumbar cerebrospinal fluid homovanillic acid, 5-hydroxyindoleacetic acid, 3-methoxy-4-hydroxyphenylethylene glycol levels have been assayed during episodes of hypersomnia and normal sleep-waking cycles. Besides an increased 5-hydroxytryptamine turnover, mainly an increased dopamine turnover has been detected in both kinds of hypersomnia, and this finding was more remarkable in the case with sub-wakefulness. The probable role of dopamine in abnormalities in the sleep-waking cycle

Biochemical aids in the diagnosis of Parkinson's disease.

Degeneration of dopamine (DA) containing neurons in the basal ganglia of Parkinsonian patients causes a reduction in cerebrospinal fluid (CSF) concentrations of the principal metabolite of dopamine, homovanillic acid (HVA). Not only are steady state HVA levels depressed, but there is substantial diminution in the rate of HVA accumulation in CSF during the oral administration of probenecid, a drug which inhibits HVA transport from the spinal fluid compartment. Two methods for the measurement of HVA in human CSF, employing fluorometry or gas liquid chromatography-mass spectroscopy, are described

Shifts in the neurobiological mechanisms motivating cocaine use with the development of an addiction-like phenotype in male rats.

RATIONALE: The development of addiction is accompanied by a shift in the mechanisms motivating cocaine use from nucleus accumbens (NAc) dopamine D1 receptor (D1R) signaling to glutamate AMPA-kainate receptor (AMPA-R) signaling. OBJECTIVE: Here, we determined whether similar shifts occur for NAc-D2R signaling and following systemic manipulation of D1R, D2R, and AMPA-R signaling. METHODS: Male rats were given short-access (20 infusions/day) or extended-access to cocaine (24 h/day, 96 infusions/day, 10 days). Motivation for cocaine was assessed following 14 days of abstinence using a progressiv

Sexual dysfunction and hyperprolactinemia in schizophrenia before and after six weeks of D2/3 receptor blockade - An exploratory study.

Sexual side-effects along with antipsychotic treatment may be linked to hyperprolactinemia and dopamine D2 receptor blockade. High prevalence of sexual dysfunction in un-medicated patients challenges the notion of sexual dysfunction as merely a side-effect of antipsychotic medication. Sexual dysfunction was assessed in fifty-six initially antipsychotic-naïve patients with schizophrenia using the UKU (Udvalget for Kliniske Undersøgelser) questionnaire. Serum-prolactin was obtained before and after six weeks of D2/3 receptor blockade with amisulpride. At baseline 68% of patients reported one or

Dorsal striatum D1-expressing neurons are involved with sensorimotor gating on prepulse inhibition test.

Prepulse inhibition (PPI) is a behavioral test in which the startle reflex response to a high-intensity stimulus (pulse) is inhibited by the prior presentation of a weak stimulus (prepulse). The classic neural circuitry that mediates startle response is localized in the brainstem; however, recent studies point to the contribution of structures involved in higher cognitive functions in regulating the sensorimotor gating, particularly forebrain regions innervated by dopaminergic nuclei. The aim of the present study was to verify the role of dorsal striatum (DS) and dopaminergic transmitting medi

Acute systemic blockade of D2 receptors does not accelerate the extinction of cocaine-associated place preference.

Facilitation of extinction can be used as a therapeutic tool in treatment of both post-traumatic stress disorder and drug addiction. The present study examined whether the blockade of D2 receptors before each extinction trial would accelerate the extinction of cocaine-induced place preference. Male Wistar rats were initially conditioned and tested for a cocaine-associated place-preference (20 mg/kg). On the following day after the initial test, the animals were submitted to extinction training. This training consisted of daily sessions in which the subjects were alternatively confined during 3

Adverse effects in the treatment of Parkinson's disease.

Side effects to antiparkinsonian drugs constitute an important component of the daily management of patients with Parkinson's disease. Treatment with levodopa frequently leads to motor fluctuations and dyskinesias. Hallucinosis, nausea and vomiting, drowsiness, orthostatic hypotension and peripheral edema can be managed by dose reduction, medication substitution and specific counteractive measures. Anticholinergics frequently cause cognitive or autonomic symptoms while ergot-derived dopamine agonists carry unique, albeit rare, risks of fibrotic, vasoconstrictive and dermatological side effects

Agreement between noninvasive hemoglobin monitoring (SpHb) and laboratory hemoglobin in patients undergoing free-flap reconstruction for oral squamous cell carcinoma.

BACKGROUND: Surgery for head and neck squamous cell carcinoma (HNSCC) often involves extensive resection with free-flap reconstruction, leading to significant blood loss. Accurate intraoperative hemoglobin (Hb) monitoring is crucial to guide transfusion. Non-invasive Hb monitoring (SpHb) offers continuous Hb measurement; however, its reliability in major oncological surgeries remains uncertain. Thus, this study evaluated the agreement between SpHb and point-of-care testing-measured Hb (POCT-Hb). METHODS: In this retrospective study, patients undergoing oral squamous cell carcinoma (SCC) resec

Sequential delivery of sinigrin and dabigatran by an in situ self-stabilizing dynamic hydrogel attenuates intervertebral disc degeneration.

Intervertebral disc degeneration (IDD) is characterized by an imbalance between nucleus pulposus catabolism and anabolism, driven by metabolic dysfunction of nucleus pulposus cells (NPCs) and a chronic inflammatory microenvironment. Effective treatments for IDD are lacking. Here, we report an injectable hydrogel that achieves reactive oxygen species (ROS)-triggered self-stabilization within the degenerative microenvironment for adaptive intradiscal therapy. The CAD hydrogel is constructed from chitosan-phenylboronic acid (CS-PBA), aldehyde-β-cyclodextrin (A-β-CD), and hyaluronic acid-dopamine

Glucagon-like Peptide-1 Boosts Plumbagin's Neuroprotection Against Rotenone-Induced Motor Deficits.

INTRODUCTION: Parkinson's disease (PD), a condition that involves neural degeneration, develops due to dopaminergic neuronal death in the substantia nigra pars compacta, resulting in reduced striatal dopamine levels. This shortage causes problems with movement and thinking. The neurodefensive response of GLP-1 is especially important in PD. Assessments have demonstrated the neuroprotective advantages of activating GLP-1 receptors in distinct models of PD, resulting in enhancements in motor as well as non-motor behaviour. These characteristics suggest that GLP-1 signalling could be a promising

First Clinical Report on the Treatment of Parkinson's Disease with Fetal Midbrain Precursor Cells.

BACKGROUND: Because human fetal ventral mesencephalic tissue grafts provide promising results in ameliorating Parkinson's disease-implicated motor dysfunctions, human fetal midbrain-derived dopamine neuronal precursor cells are considered good candidates for cell-based therapy for Parkinson's disease in that large quantities of cells can be supplied through a good manufacturing practice-compliant system. OBJECTIVE: We conducted a prospective, phase I/IIa, dose-escalation, open-label "first-in-human" clinical trial with fetal neural precursor cells to assess their safety and therapeutic effica

Histone H3 dopaminylation in nucleus accumbens, but not medial prefrontal cortex, contributes to cocaine-seeking following prolonged abstinence.

Enduring patterns of epigenomic and transcriptional plasticity within the mesolimbic dopamine system contribute importantly to persistent behavioral adaptations that characterize substance use disorders (SUD). While drug addiction has long been thought of as a disorder of dopamine (DA) neurotransmission, therapeutic interventions targeting receptor mediated DA-signaling have not yet resulted in efficacious treatments. Our laboratory recently identified a non-canonical, neurotransmission-independent signaling moiety for DA in brain, termed dopaminylation, whereby DA itself acts as a donor sourc
