Tissue flow acts as a guidance cue for immune cell polarization and directional migration.

Embryonic myeloid (EM) cells are the first immune-cell population to emerge during development and must disperse throughout the embryo to act as the first line of defence against infection. Although EM cells migrate directionally toward wounds, how they navigate through unwounded tissues during early colonization remains unclear. Here we show that EM cell dispersion in Xenopus embryos is driven, at least in part, by cell-on-cell migration, an important yet underappreciated phenomenon, guided by dynamic tissue flows. We have established a new ex vivo EM cell migration system that allows for liv

How temperature modulates the combined toxicity of aged polystyrene microplastics and copper in early zebrafish development.

Environmental factors, such as temperature, exert a critical influence on aquatic ecosystems. It is important to assess how climate change and pollutants affect contaminants released into water. In this study, the combined effects of environmentally relevant temperature variation, polystyrene microplastics (PSMPs), and copper were evaluated in zebrafish. Toxicity tests were conducted at 23, 26, and 29°C. The PSMPs were aged via the Fenton reaction, to simulate environmental oxidative conditions. Temperature influenced embryonic development, with hatching strongly delayed at 23 °C, while contam

TM4SF1 mediates YAP/TAZ regulation of coronary artery formation.

Although the role of YAP/TAZ in mediating VEGF-driven angiogenesis is well established, their downstream effectors in coronary artery development remain elusive. In this study, we identify TM4SF1 as a novel downstream target of YAP/TAZ in coronary endothelial cells. Conditional deletion of YAP/TAZ in the endocardial lineage resulted in embryonic lethality due to congenital heart defects, including ventricular noncompaction. Mutant embryos exhibited severe coronary vessel defects, including the absence of major arteries. Loss of YAP/TAZ significantly inhibited endothelial cell proliferation and

Ibogaine as a double-edged sword: Developmental cardiotoxicity and neurobehavioral alterations in zebrafish.

Ibogaine has attracted increasing attention as a potential treatment for substance use disorders; however, its clinical application remains limited by well-documented cardiotoxicity, and its safety during early development is poorly understood. Here, we investigated the developmental cardiotoxic and neurobehavioral effects of ibogaine using zebrafish embryos and larvae as a translational model. Ibogaine induced early-onset bradycardia at concentrations as low as 100 ng/mL and pericardial edema at concentrations of 500 ng/mL, indicating both functional and morphological cardiac impairment. Failure to Fuse Shut Eyelids, a Novel Unique Sign in Affected Fetus with Homozygous PPP1R13L Pathogenic Variant-A Case Report and Review of the Literature.

Prompted by a fetus with a homozygous PPP1R13L pathogenic variant and persistent open eyelids on sonography, suggesting fusion failure, this study marks the first prenatal human description linking PPP1R13L to eyelid closure, mirroring waved with open eyelids 2(woe2) and waved 3 (wa3) mouse models. Eyelid morphogenesis involves complex epidermal-dermal interactions. We reviewed the literature to understand embryonic/fetal development and identify molecular pathways crucial for proper eyelid morphology. Known signaling pathways in eyelid closure and reopening include NF-κB, p53, Wnt, TGF-β, BMP

MCM3 Safeguards Neural Progenitor Maintenance and Cortical Development Against Replication-Associated Stress.

Faithful genome duplication during neurogenesis relies on the licensing of surplus replication origins, and hypomorphic variants in MCM3 have been associated with microcephaly and related growth disorders. Using in utero electroporation in embryonic mouse cortex, we show that acute partial depletion of MCM3 disrupts cortical progenitor development. MCM3 knockdown reduced EdU incorporation and PCNA positivity among GFP-positive cells in the VZ, indicating decreased S-phase engagement or impaired cell-cycle progression. It also increased replication-stress-associated signals, including γ-H2AX an

Shifts in embryonic oxygen levels cue heterochrony in limb initiation.

Heterochrony, or the alteration of developmental timing, is an important mechanism of evolutionary change. Avian species display synchronized growth of the forelimbs and hindlimbs, while mammalian species show delayed hindlimb development. We find that mammalian limb heterochrony is evident from the start of limb bud formation and is associated with heterochronic expression of T-box transcription factors. This heterochronic shift is not due to changes in cis-regulatory sequences controlling T-box gene expression but, unexpectedly, is dependent upon the differential oxygen levels to which avian

H3K9me3 controls epidermis morphogenesis by regulating RNA Pol II dynamics at developmental promoters and enhancers.

Histone H3K9me3 silences repetitive elements and represses non-lineage genes during early development, but its role in organogenesis is understudied. Here, we show that H3K9me3 deposition is dynamic during epidermis morphogenesis and essential for lineage diversification. We ablate Suv39h1, Suv39h2, and Setdb1 histone methyltransferases, in the embryonic mouse epidermis, to induce H3K9me3 loss. This causes complete failure of keratinocyte differentiation, skin barrier formation, hair follicle development, and Merkel cell specification. Single-cell transcriptomics reveals aberrant cell fates wi

Single-cell landscape of goose thymus uncovers developmental and regulatory dynamics of T cell differentiation.

The thymus is vital for T cell development, yet avian thymocyte maturation remains poorly characterized. Here, we present a single-cell transcriptomic atlas of thymic development in Zhedong White geese across embryonic, gosling, juvenile, and adult stages. Morphological and histological analysis revealed a multilobulated, paired thymus with rapid post-hatch expansion and age-dependent involution. Single-cell RNA sequencing identified all major thymic cell types, with T cells predominant and γδ T cells unusually abundant compared to mammals. High-resolution profiling defined key T cell subsets

CRISPR and Beyond: Genome-Editing Strategies in Retinal Stem Cell Research.

Genome editing has emerged as a transformative approach for understanding and treating retinal degenerative diseases. Combining this technology with pluripotent stem cells provides an ideal platform for modeling human development and disease, and investigating emerging therapeutic strategies ultimately aimed towards in vivo correction. This approach enables both functional studies to understand retinal degeneration and the early development of targeted therapies for inherited disease. This review offers a comprehensive overview of genome-editing techniques and the ability to create new clinica

The extracellular protease AdamTS-B is a dosage-sensitive regulator of unicellular tracheal branch morphogenesis.

Epithelial tube morphogenesis requires coordinated interactions between epithelial cells and their extracellular environment, yet the extracellular mechanisms that regulate this process remain poorly understood. The Drosophila trachea provides an excellent model for investigating how extracellular remodeling contributes to epithelial tube morphogenesis. AdamTS-B, an extracellular ADAMTS protease, is expressed in the embryonic trachea from early to late stages of tracheal development. Loss of AdamTS-B caused mild defects in unicellular branch organization, whereas overexpression produced ectopi

Generation of germinal-vesicle oocytes from mouse embryonic stem cells under an ovarian soma-free condition.

In vitro oogenesis provides a platform to elucidate the mechanisms of oocyte development and advance reproductive medicine. The prevalent in vitro oogenesis model requires ovarian somatic cells (OSCs) to support oocyte development; yet, complex three-dimensional oocyte-OSC interactions pose difficulties in systems regulation and mechanistic understanding. Here, we present an OSC-free system of in vitro oogenesis: upon optimized provision of retinoic acid and bone morphogenetic protein on feeders, mouse primordial germ-cell-like cells induced from embryonic stem cells propagate robustly, and en

Emerging insights into lineage plasticity in pancreatic cancer initiation, progression, and therapy resistance.

Lineage plasticity, the ability of cells to switch from one specialized identity to another, is a fundamental cellular process in embryonic development and tissue regeneration. The process is often hijacked by tumor cells at various stages to facilitate cancer initiation, progression, metastasis, and therapy resistance. It is also recognized as a key contributor to intratumor heterogeneity (ITH). In pancreatic ductal adenocarcinoma (PDAC), lineage plasticity is central to acinar-to-ductal metaplasia (ADM) and its associated acinar-to-ductal reprogramming (ADR), epithelial-mesenchymal transitio

mir-276a Is Required for Muscle Development in Drosophila and Regulates the FGF Receptor Heartless During the Migration of Nascent Myotubes in the Testis.

MicroRNAs function as post-transcriptional regulators in gene expression and control a broad range of biological processes in metazoans. The formation of multinucleated muscles is essential for locomotion, growth, and muscle repair. microRNAs have also emerged as important regulators for muscle development and function. In order to identify new microRNAs required for muscle formation, we have performed a large microRNA overexpression screen. We screened for defects during embryonic and adult muscle formation. Here, we describe the identification of mir-276a as a regulator for muscle migration

Spatially organized cellular communities shape functional tissue architecture in the pancreas.

Organ function depends on the precise spatial organization of cells across multiple scales, from individual units to cellular communities that form local niches and, ultimately, higher-order structures. Although cell identities are increasingly well defined, the spatial arrangement and interactions among diverse cell types remain poorly understood. Here, we combine single-cell and spatial transcriptomics to map pancreatic cell populations across space and time, from embryonic development to adult homeostasis in mice. Using these maps, we resolve spatial heterogeneity among pancreatic cell type

Glycogen metabolism in mouse embryonic Sertoli cells sustains the germ line through the lactate shuttle.

Metabolites are key regulators of cell fate decisions, epigenetic remodeling, and lineage commitment. While genetic pathways governing testis differentiation are well studied, the role of metabolism remains poorly understood. In this study, we investigated a transient, male-specific accumulation of glycogen in supporting (Sertoli) cells of embryonic testis in mice. Although glycogen metabolism was dispensable for Sertoli cell differentiation, its disruption led to reduced lactate production and impaired PGCs ability to colonize the newly forming testis cords. Inhibiting lactate transport furth

Interleukin-34-dependent perivascular macrophages promote vascular function in the brain.

The development of most macrophages depends on the colony-stimulating factor 1 (CSF-1) receptor, which has two ligands: CSF-1 and interleukin-34 (IL-34). While IL-34 is required for the homeostasis of microglia, the parenchymal macrophages in the central nervous system (CNS), it is unclear whether brain border-associated macrophages (BAMs) also depend on this cytokine. Here, we demonstrated that the embryonic development of murine BAMs in the choroid plexus, leptomeninges, and perivascular spaces required CSF-1, while IL-34 was critical for their maintenance in adulthood. In the brain, Il34 wa

Functional regeneration strategies of hair follicles: advances and challenges.

Hair follicles are essential appendages of human skin that function in protection, sensation, thermoregulation and social interactions. The multicellular components, particularly the dermal papilla, matrix and bulge housing stem cells, enable cyclic hair growth postnatally. However, miniaturization and loss of hair follicles can occur in the context of ageing, trauma and various alopecia-related diseases. Conventional treatments involve the redistribution of existing follicles, which may not be viable in patients lacking follicular resources. Recent progress in the comprehension of morphogenes

Cardiac trabeculation in vertebrates: Convergent evolution or evolutionary adaptations associated with heart complexity?

One of the most important processes during early heart development is the formation of trabecular myocardium. Cardiac trabeculation is the process by which the ventricular chambers develop a complex sponge-like myocardium essential for optimal cardiac function to provide efficient oxygenation and nourishment to the developing embryo. Indeed, its importance is highlighted by the fact that defects in trabecular formation lead to embryonic lethality and congenital heart disease. In the last decades, our understanding of cardiac trabeculation in different vertebrate models has advanced significant

Fine-tuning of gene expression through the Mettl3-Mettl14-Dnmt1 axis controls ESC differentiation.

The marking of DNA, histones, and RNA is central to gene expression regulation in development and disease. Recent evidence links N6-methyladenosine (m6A), installed on RNA by the METTL3-METTL14 methyltransferase complex, to histone modifications, but the link between m6A and DNA methylation remains scarcely explored. This study shows that METTL3-METTL14 recruits the DNA methyltransferase DNMT1 to chromatin for gene-body methylation. We identify a set of genes whose expression is fine-tuned by both gene-body 5mC, which promotes transcription, and m6A, which destabilizes transcripts. We demonstr
