Construction of a Golgi apparatus-localization polarity-responsive fluorescent probe with large Stokes shift to screen the small-molecule drugs for the inhibition of ferroptosis.

Ferroptosis is a distinctive cell death form and has close connections with many diseases, and screening of ferroptosis-associated drug has great importance to delve into creative approaches against the currently intractable diseases. Herein, we developed a method for the ferroptosis-related small-molecule drugs screening by a GA-targeting polarity-responsive fluorescent probe (GP-1). The probe GP-1 showed large Stokes shifts in solvents (up to 194 nm), and displayed desirable sensitivity and selectivity to polarity. GP-1 was primarily distributed in Golgi apparatus (GA) rather than the other

TM6SF2 acts at ER membrane contact sites to enable RAB7-dependent endolysosomal VLDL secretion and maintain Golgi structure.

Although a loss-of-function variant of TM6SF2 is a highly penetrant risk factor for steatotic liver disease, including progression to cirrhosis and cancer, its normal function is largely unknown. Here we show that endoplasmic reticulum (ER)-localized TM6SF2 forms membrane contact sites (MCSs) with late endosomes/lysosomes, where it activates the small GTPase RAB7 to enable an endolysosomal route for selective secretion of lipid-rich very low-density lipoprotein (VLDL) particles. In accord, TM6SF2 and RAB7 deficiency similarly cause endolysosomal VLDL retention and accumulation of enlarged late

TRPs-NCX2 Axis Jointly Preserves Pancreatic Beta Cell Susceptibility/Sensitivity to Elevated Glucose.

Background: For appropriate glucose sensing, pancreatic beta-cells essentially establish a constant mitochondria-directed endoplasmic reticulum (ER) Ca2+ leak via phosphorylated presenilin-1 (PS1). In the current work, we investigated how cells maintain the ER Ca2+ content during this continuous Ca2+ leak. Methods: Ca2+ imaging for ER, cytosolic and subplasmalemmal Ca2+, high resolution respirometry, and ELISA for insulin secretion were used. Results: A novel ER Ca2+ refilling mechanism driven by reverse-mode NCX2, which is fueled by local Na+ influxes through TRPC3 and TRPV4 channels that are

Mitochondrial Dynamics in Diabetes Mellitus and Its Complications: Mechanisms, Pathological Roles, and Therapeutic Implications.

Diabetes mellitus (DM) is a progressive metabolic disorder in which mitochondrial dysfunction is increasingly recognized as a central contributor to metabolic deterioration and diabetic complications. However, whether dysregulated mitochondrial dynamics represents a primary pathogenic driver, an adaptive response, or a consequence of metabolic stress remains incompletely understood. This review summarizes current advances in the molecular mechanisms governing mitochondrial fission, fusion, and mitophagy, with particular emphasis on their coordinated roles in mitochondrial quality control rathe

Parkin Deficiency Impairs ER-Mitochondria Associations and calcium homeostasis via IP3R-Grp75-VDAC1 Complex.

Disruption of mitochondria-associated endoplasmic reticulum membranes (MAMs) and calcium homeostasis has been implicated in the pathogenesis of Parkinson's disease (PD). Parkin, a PD-associated E3 ubiquitin ligase, has been shown to regulate MAM integrity and calcium dynamics. However, the mechanisms of Parkin recruitment and its substrate specificity have not been well understood. This investigation has demonstrated that loss of Parkin enhances ER-mitochondria associations and leads to excessive calcium flux in MAM, resulting in abnormal mitochondrial permeability transition pore (mPTP) openi

Understanding the unfolded protein response in glioblastoma.

Glioblastoma is the most aggressive and lethal malignancy of the central nervous system and exhibits a complex interplay with endoplasmic reticulum (ER) stress (ERS). The multifunctional ER is the primary site of the unfolded protein response (UPR), which is activated under conditions of ERS to enhance protein-folding capacity and promote cell survival. However, persistent ERS can trigger apoptosis. Despite extensive research over the past decades, the mechanistic insights into the impact of ERS and the UPR on glioblastoma pathogenesis and progression remain unclear. The UPR operates through t

Lipid Transporter TMEM24/C2CD2L Plays Essential Roles in Retinal Photoreceptor Survival by Regulating Calcium Signaling and Synaptic Transmission.

Retinal neurotransmission is fundamental to vision, relying on the precise calcium-dependent release of glutamate at photoreceptor ribbon synapses. Disruptions in this pathway underlie multiple visual disorders in humans. TMEM24/C2CD2L, an SMP domain-containing phospholipid transporter enriched at endoplasmic reticulum (ER)-plasma membrane (PM) contact sites, regulates phosphoinositide homeostasis and calcium signaling. However, its role in the retina remains unknown. Here, we show that C2CD2L is predominantly expressed in retinal photoreceptors. Using a rod-specific conditional knockout mouse

Curcumin Attenuates Glyphosate-Induced Mammary Toxicity via Suppression of ER Stress and the TNFα/MAPK/STAT3 Axis.

Curcumin, a natural polyphenol with well-established antioxidant and anti-inflammatory properties, was evaluated for its protective effects against glyphosate (GLY)-induced mammary toxicity. Pregnant mice received GLY (50 or 250 mg/kg/day) with or without curcumin (150 mg/kg/day) by oral gavage; GLY dose-dependently disrupted alveolar architecture, caused marked inflammatory infiltration, and downregulated tight-junction proteins. In parallel, goat mammary epithelial cells (GMECs) were treated with GLY (1 or 7 mM) and curcumin (1 µM) to dissect acute cellular stress pathways. These complementa

Axin1 stabilizes S-opsin and maintains cone photoreceptor survival by inhibiting GSK3β activity.

To ensure high phototransduction efficiency in the retina, the precise subcellular localization of signaling molecules must be tightly orchestrated by scaffold proteins. Aberrant localization of these scaffold proteins not only disrupts the transition of photoelectrical signals but also triggers endoplasmic reticulum (ER) stress, which leads to photoreceptor apoptosis. However, it is unknown how these proteins are localized to specific subcellular compartments of photoreceptors or how protein mislocalization is coupled with apoptotic signaling. Herein, we observed a specific spatiotemporal exp

Vincristine enhances doxorubicin cardiotoxicity in human iPSC-derived cardiomyocytes: Partial attenuation by SGLT2 inhibition.

BACKGROUND: Doxorubicin is a well-known cardiotoxic chemotherapeutic agent. Whether its combination with vincristine exacerbates cardiotoxicity and the underlying mechanisms remains unclear. METHODS: Human induced pluripotent stem cell derived cardiomyocytes (iPSC-CMs) were treated with doxorubicin, vincristine, or both. Cellular apoptosis, signaling, and mitochondrial function were assessed. Sodium-glucose cotransporter 2 (SGLT2) inhibitors and pharmacological modulators were used to test protective effects, and lymphoma cells were used to evaluate potential impacts on chemotherapeutic effic

Reticulocalbin1-mediated regulation of calcium homeostasis in naïve T lymphoncytes.

Reticulocalbin1 (RCN1), a calcium-binding protein localized in the endoplasmic reticulum (ER), is implicated in cancer progression, but its role in the immune system remains poorly understood. To clarify the function of RCN1, we generated a RCN1-deficient (Rcn1-/-) mice using CRISPR-Cas9 system. Immunological characterization by flow cytometry revealed that while T cell populations in the spleen were unaffected, the proportion of CD8 single-positive (SP) thymocytes was significantly reduced in Rcn1-/- mice. In contrast, in vitro stimulation of splenic CD8+ T cells revealed no significant diffe

Molecular insights into protein disulfide isomerase antagonism by punicalagin.

Punicalagin, an ellagic acid polyphenol from pomegranate, has been proposed as an antagonist of protein disulfide isomerase (PDI) and endoplasmic reticulum resident protein 57 (ERp57), thiol oxidoreductases that regulate protein folding and extracellular thrombotic signaling. Here, biochemical oxidase and reductase assays on PDI show that punicalagin inhibits both activities with micromolar potency, thereby extending earlier work that described only disulfide reductase inhibition. In parallel, thiol labeling of catalytic cysteines revealed no change in the redox state, supporting a noncovalent

Characterization of Novel Decenoic Acid Derivatives Exhibiting Cytotoxic Activity Against T98G Glioma Cells Under a Serum- and Glucose-deprived Condition.

The proliferative capacity and drug resistance of cancer cells are thought to be related to adaptations to the low nutrient and hypoxic conditions known as the tumor microenvironment. This adaptation is suggested to involve the unfolded protein response (UPR) triggered by the endoplasmic reticulum (ER) stress. Therefore, the ER stress induced under these conditions is considered one of the attractive targets for antitumor drug development. In this study, we designed and synthesized decenoic acid derivatives and found two compounds, 13-Chloro-6-(4-fluorophenoxy)tridec-6-en-8-yn-1-ol (Cpd6) and

Parkinson's disease-associated PLA2G6 protects IP3R1 protein to control ER-mitochondria tethering and Ca2+ transfer.

Mutations in the phospholipase A2 group VI (PLA2G6) gene have been linked to autosomal recessive Parkinson's disease (PD), yet the molecular mechanisms remain poorly understood. This study provides the in vitro and in vivo evidence, specifically in dopaminergic neurons derived from patients with PD, that PLA2G6 loss-of-function disrupts the mitochondria-associated endoplasmic reticulum (ER) membrane (MAM), a critical regulator of Ca2+ transfer and energy homeostasis. This study demonstrates that the PLA2G6 protein localizes to the MAM and physically associates with the IP3R1-GRP75-VDAC1 comple

FAM134B-mediated ER-phagy degrades APP and suppresses Alzheimer's disease pathology.

Endoplasmic reticulum autophagy (ER-phagy) is a selective autophagy pathway in which receptor proteins target ER membranes and proteins for degradation, yet its role in Alzheimer's disease (AD) remains unclear. Here, we identify FAM134B/RETREG1 as a specific ER-phagy receptor mediating amyloid precursor protein (APP) degradation. FAM134B directly interacts with ER-localized wild-type and familial mutant APP via their C-terminal domains and recruits LC3 through its LC3-interacting region (LIR) to promote APP delivery to phagophores for lysosomal degradation. In AD, epigenetic silencing at the F

TOMM40 suppression promotes neuronal cholesterol imbalance and molecular and behavioral phenotypes of Alzheimer's disease.

INTRODUCTION: While the apolipoprotein E (APOE) ε4 allele is a major risk factor for Alzheimer's disease (AD), the role of translocase of outer mitochondrial membrane 40 (TOMM40)-an adjacent gene involved in mitochondrial protein import-is not known. METHODS: Human brain tissue, human induced pluripotent stem cell-derived neurons (iNeurons), and mice were used for study of gene expression, cholesterol metabolism, mitochondrial function, and animal cognition. RESULTS: Human brain transcriptomics showed reduced TOMM40 expression that correlated with cholesterol regulatory gene expression, amyl

A pan-vertebrate signaling motif controls the molecular function of intracellular AQP12.

Unorthodox AQP12-type channels are poorly understood intracellular aquaporins localized in the endoplasmic reticulum and zymogen granules (ZGs) of pancreatic acinar cells. Despite connections to major diseases, their biophysical properties and intracellular trafficking regulation remain largely unknown. Here, we show that heterologously expressed plant and metazoan AQP12-related channels specifically localize to the intracellular yolk platelet (YP) membrane of frog oocytes, a feature that is recapitulated in vivo for invertebrate and vertebrate orthologs. Using native YP membranes, we show tha

PRRSV Nsp2 promotes inflammatory responses by restricting ER-to-Golgi translocation of STING through HMGB1.

STING serves as a central adaptor in innate immunity, with its trafficking from the endoplasmic reticulum (ER) to the Golgi apparatus determining its signaling outcomes. Retention of STING within the ER is associated with enhanced inflammatory responses, whereas its localization to the Golgi apparatus facilitates the production of type I interferon. In this study, we demonstrate that porcine reproductive and respiratory syndrome virus (PRRSV), an economically significant swine pathogen characterized by causing interstitial pneumonia, promotes the retention of STING within the ER, thereby enhan

Polysaccharides targeting organelle network dysfunction in Alzheimer's disease: A review.

Alzheimer's disease (AD) is driven by an interrelated pathological network, which to some extent explains the limited efficacy of single-target therapies and constitutes a significant challenge in current clinical treatment. Emerging evidence indicates that dysfunction and crosstalk among mitochondria, the endoplasmic reticulum, and lysosome-autophagy form a core organelle network that amplifies oxidative stress and exacerbates proteostatic imbalance, neuroinflammation, and synaptic failure. In this review, we summarize natural and engineered polysaccharides as potential multitarget modulators

GPX8+ cancer-associated fibroblast-derived lactate contributes to lenvatinib resistance by facilitating BRPF1 expression through histone H3 lysine 18 lactylation in hepatocellular carcinoma.

Cancer-associated fibroblasts (CAFs) are necessary constituents of the tumor microenvironment, significantly promoting cancer cell proliferation, invasion, and therapeutic resistance through the secretion of various factors. This study elucidates a novel metabolic-epigenetic mechanism by which glutathione peroxidase 8-positive (GPX8⁺) CAFs confer lenvatinib resistance in hepatocellular carcinoma (HCC). We demonstrate that GPX8 overexpression in CAFs activates the PI3K/AKT/mTOR signaling pathway by suppressing endoplasmic reticulum stress, driving glycolytic reprogramming and lactate production
