Sleep restriction reprograms systemic immunity through NK and T cell crosstalk.

Insufficient sleep is a recognized risk factor for inflammatory diseases, yet how sleep loss functionally perturbs immune regulation remains unclear. Here, we apply a multi-omics framework combining two-sample Mendelian randomization, sleep-restriction intervention microarray, and PBMC single-cell profiling to dissect immune consequences of sleep loss. Mendelian randomization associated genetically proxied sleeplessness/insomnia symptoms with alterations in 162 plasma proteins and increased risk across 94 clinical diagnosis traits. Gene expression microarray after sleep restriction confirmed i

Dexmedetomidine inhibits microglia activation, neuroinflammation, and glycolysis in postoperative cognitive dysfunction by promoting YTHDF2-mediated PKM2 mRNA degradation.

Postoperative cognitive dysfunction (POCD) is a common complication following surgery, particularly in elderly patients, and is closely linked to neuroinflammation driven by microglial activation. While dexmedetomidine (Dex) has shown neuroprotective potential in clinical settings, the molecular mechanisms underlying its beneficial effects—particularly in relation to RNA epigenetic regulation and metabolic reprogramming in microglia—remain poorly understood. Here, we uncover a novel mechanism by which Dex alleviates POCD through modulation of N6-methyladenosine (m6A)–dependent post-transcripti

Senescent-Like Myofibers Contribute to Anti-Regenerative Cytokine Signaling in Duchenne Muscular Dystrophy.

Duchenne muscular dystrophy (DMD) is a genetic muscular disease characterized by progressive muscle degeneration. p16 is expressed in skeletal muscles and induces cellular senescence in a rat model of DMD, whereas its ablation enhances muscle regeneration. However, the mechanism underlying this phenomenon remains unclear. This study aimed to elucidate the mechanism for p16-induced DMD exacerbation. RNA-seq analysis revealed p16-dependent upregulation of cytokine gene expression in DMD rat skeletal muscles, which also altered the systemic blood cytokine profile. Furthermore, the effect of an al

A NeuroD1 AAV-Based Gene Therapy for Functional Brain Repair in Alzheimer's Disease-Like Non-Human Primate Model.

There is a pressing demand for neuroregenerative treatment for Alzheimer's disease (AD). Recently, a NeuroD1-mediated neuroregeneration strategy has been proposed, yet its efficacy remains untested in non-human primate (NHP) AD models closely reflecting human pathology. This study evaluates the therapeutic potential of NeuroD1 AAV-based gene therapy in an NHP AD model with hippocampal hTau overexpression, utilizing immunostaining, fluorescence/confocal imaging, MRI and FDG PET scans, Simoa CSF biomarker analysis, behavioral tests, and bulk RNA sequencing. NeuroD1 AAV-based gene therapy prevent

High altitude-mediated immune remodeling accelerates aging.

High altitude-associated pathophysiological processes may potentially accelerate aging trajectory, while evidence remains limited. We present immune landscape characterization in human populations residing at 3656-meter (Lhasa) and 5070-meter (Tuiwacun) elevations on the Qinghai-Tibet Plateau, complemented by multiorgan single-cell RNA sequencing and spatially enhanced resolution omics sequencing (Stereo-seq) of mice under simulated 5000-meter hypoxic conditions. Comparative analysis revealed significantly elevated neutrophil proportions in high-altitude population (HAP) cohorts relative to lo

A Dual-Fusogenic Virus-like Vector Enables Direct Cytosolic Delivery of mRNA Vaccines to Dendritic Cells.

Messenger RNA (mRNA) vaccines hold significant potential for disease prevention and treatment; however, their effectiveness is limited by the inefficiency of current carriers in targeting dendritic cells (DCs) and facilitating endosomal escape to deliver mRNA into the cytosol. In this study, we develop a dual-fusogenic virus-like particle (VLP) cofunctionalized with a DC-targeting fusogen (DC-F) and an endosomal fusogen (E-F), termed DC/E-FVLP. We show that mRNA-loaded DC/E-FVLPmRNA selectively targets DCs and promotes fusion with both the plasma and endosomal membranes, achieving a 28.2% mRNA

ERH elicits cell lineage restriction in mammalian preimplantation development and differentiation from pluripotency via H3K9me3-heterochromatin.

Enhancer of Rudimentary Homolog (ERH) is an evolutionarily conserved protein originally characterized as promoting fission yeast heterochromatin and recently shown to maintain H3K9me3 heterochromatin in human fibroblasts. Here, we find that ERH depletion in fibroblasts reverts the somatic cell H3K9me3 landscape of broad megabase size domains to an embryonic stem cell (ESC) state composed of mainly H3K9me3 peaks and enables activation of naïve and pluripotency genes and transposable elements during induced pluripotent stem cell (iPSC) reprogramming. Concordantly, we find that ERH represses toti

Targeting lipid metabolism-associated lncHMGCS1 suppressed cell proliferation and restored intracellular lipid homeostasis by regulating ACSL3 in gastric cancer.

Previous work has indicated that lipid metabolic reprogramming plays a critical role in gastric cancer (GC) development, yet the underlying molecular mechanisms remain largely undefined. In the present study, we identified a novel lipid metabolism-associated long non-coding RNA, 3-Hydroxy-3-Methylglutaryl-CoA Synthase 1 (lncHMGCS1), which is aberrantly upregulated in GC tissues and cell lines. Functional assays demonstrated that lncHMGCS1 overexpression could promote intracellular lipid droplet accumulation, elevate the triglyceride, total cholesterol, and low-density lipoprotein cholesterol l

NSUN2 suppresses ferroptosis via the KLF4/GPX4 axis to preserve stem cells and tissues.

RNA 5-methylcytosine (m5C) methylation has emerged as an important epitranscriptomic regulator of gene expression, yet its role in stem cell maintenance remains not well understood. Here we identify a previously unrecognized function of the m5C methyltransferase NSUN2 in protecting stem cells from ferroptosis and promoting tissue regeneration. NSUN2 deletion reduced GPX4 expression, decreased intracellular GSH levels, increased Fe2+ accumulation, and enhanced lipid peroxidation in stem cells, leading to ferroptotic cell death, loss of tissue stem cells and impaired tissue regeneration across m

Woven dECM yarn scaffolds enable volumetric muscle loss regeneration via immunomodulation.

Volumetric muscle loss (VML) represents a significant unmet clinical need in current medical practice. Decellularized extracellular matrix (dECM) offers a promising clinical approach. However, its inherent architecture impedes adaptive topological guidance for rapid cellular infiltration, spatial organization, and coordinated immune response, limiting functional restoration. Here, we report a dECM yarn scaffold (YS) fabricated by 3D weaving of rotary-cut yarns, achieving precise structural control, full interconnectivity and high porosity. In murine VML models, YS significantly improved vascul

Spatially resolved multiomics of human meningeal development reveal lineage and disease dynamics.

The meninges are increasingly recognized as key stromal and immune niches supporting cranial development. Here we constructed a paired single-nuclei RNA and Assay for Transposase-Accessible Chromatin atlas of human meningeal development from 6 to 21 postconception weeks across cranial regions. By integrating spatial transcriptomics, we resolved cell types across meningeal layers. Our multiomic atlas defines gene regulatory networks underlying blood-brain barrier endothelial identity during development. Within immune compartments, the data support a yolk sac origin for meningeal myeloid lineage

Engineered internal architecture of core-shell lipid nanoparticles promotes efficient mRNA endosomal release.

Messenger RNA (mRNA) therapeutics rely on lipid nanoparticles (LNPs) for delivery, yet inefficient endosomal escape remains a major bottleneck, with only a small fraction of internalized cargo reaching the cytoplasm. Conventional LNPs encapsulate mRNA in amorphous lipid cores, where partial charge neutralization and lack of structural order limit protonation-driven membrane disruption. Here, we present an architectural strategy that engineers LNP internal structure using ionizable lipid-coated gold nanoparticles (IC-AuNPs) as rigid, pH-responsive cores. The Au cores template the formation of r

PPP4C: a potential molecular marker and therapeutic target in thyroid cancer and triple-negative breast cancer.

The protein phosphatase PPP4C has been implicated in oncogenic processes, but its role in thyroid carcinoma (THCA) and triple-negative breast cancer (TNBC) remains unclear. Here, we combined bioinformatic analyses with in vitro experiments to investigate the clinical and functional significance of PPP4C in these malignancies. PPP4C was significantly upregulated in both THCA and TNBC compared with normal tissues, and higher expression was associated with adverse clinicopathological features and poorer overall survival. Functional assays showed that PPP4C knockdown suppressed proliferation, migr

Transcriptome Analysis Reveals Key Drought-Stress-Responsive Genes in Two Bermudagrass Genotypes.

Drought inhibits grass development and survival. However, molecular-based studies on drought tolerance mechanisms in bermudagrass (Cynodon dactylon) remain scarce. Therefore, a drought-resistant bermudagrass (Tianshui) and a drought-sensitive (Zhengzhou) genotype were selected and subjected to 28 days of 50% drought stress. Leaves were sampled for RNA sequencing. Under drought stress, 2410 differentially expressed genes (DEGs) were discovered in which 1214 were upregulated (tolerant vs. sensitive) and 1196 downregulated. Kyoto Encyclopedia of Genes and Genomes (KEGG) indicated that these speci

Stage-resolved gene drivers of pancreatic ductal adenocarcinoma progression and therapeutic vulnerabilities.

Pancreatic ductal adenocarcinoma shows early dissemination, stromal remodeling, and therapy resistance, but how tumor programs co-evolve with immune and stromal changes across stages remains unclear. We built a stage-resolved transcriptomic atlas using bulk RNA sequencing of FFPE samples from 443 untreated tumors spanning resectable, locally advanced, primary metastatic, and liver metastatic disease. Integrative modeling identified ten transcriptional programs with monotonic dynamics during progression. Early stages were enriched for epithelial differentiation and immune-stimulatory signals. A

TDP2 drives immune evasion and metastatic progression in prostate cancer.

Immune evasion and epithelial-mesenchymal transition (EMT) are critical mechanisms driving tumor progression and therapy resistance in prostate cancer. In this study, we explored the role of TDP2 in modulating the tumor microenvironment (TME) through single-cell RNA sequencing and pathway enrichment analysis. Our results revealed that epithelial cells with high TDP2 expression extensively interact with myeloid cells, macrophages, and fibroblasts, thereby shaping immune responses and facilitating tumor progression. Specifically, TDP2 overexpression suppressed M1 macrophage polarization and dend

From Molecular Recognition to Behavioral Control: Dual-Target Aphid Repellent Discovery of Novel Ionone Derivatives.

Odorant-binding proteins (OBPs) are promising targets for insect behavior regulators. We established an original target-to-behavior strategy for developing dual-target aphid repellents. Structure-based virtual screening initially identified β-ionone as a lead compound, from which 24 novel ionone derivatives were designed and synthesized targeting Acyrthosiphon pisum OBP3 and OBP7. Most ionone derivatives exhibited enhanced binding affinities to both ApisOBP3 (5.14-20.70 μM) and ApisOBP7 (3.64-28.68 μM). Molecular dynamics simulations and alanine site-directed mutagenesis revealed that mutation

Double-stranded RNA and ROS scavenging nanoplatform for modulating skin inflammation.

Double-stranded RNA (dsRNA) and reactive oxygen species (ROS) contribute to the exacerbation of skin inflammatory diseases such as psoriasis and atopic dermatitis. While antioxidant strategies have been extensively explored, approaches to neutralize pathogenic dsRNA remain limited. Here, we show that Mo90Ce10, a cerium-doped polyoxometalate, concurrently targets dsRNA and ROS to mitigate cutaneous inflammation. In vitro, Mo90Ce10 scavenges ROS and suppresses type I interferon responses by directly binding dsRNA through its oxygen-rich, hydrogen bond-forming surface. Its structural template, {M

Ascites circRNA ASCOR Drives Platinum Resistance of High-Grade Serous Ovarian Cancer by Facilitating RPA1 Nuclear Translocation.

High-grade serous ovarian carcinoma (HGSOC) remains the most lethal gynecologic malignancy, with platinum resistance posing the major therapeutic barrier. Malignant ascites, a hallmark of advanced HGSOC, correlates with chemoresistance and poor prognosis, yet the contribution of ascites circRNAs in this process remains obscure. Here, we identify ASCOR, a circRNA upregulated in ascites small extracellular vesicles (sEVs) from platinum-resistant HGSOC patients, predicts poor survival. ASCOR promotes platinum resistance in vitro and in vivo by enhancing cell viability, reducing apoptosis, and all

DDX21 Enhances Radiosensitivity in Head and Neck Squamous Cell Carcinoma by Suppressing MK2-Mediated DNA Damage Response.

Radioresistance remains a significant challenge in the radiotherapy (RT) of head and neck squamous cell carcinoma (HNSCC). However, the biological factors that govern sensitivity to this therapy are not well-understood. The DEAD-box family is known for its role in genome stability, and inextricably linked to the radiotherapy resistance of tumors. This study found the role of the RNA helicase DDX21 in regulating radiosensitivity through extensive data mining. High DDX21 expression predicted improved survival after postoperative radiotherapy. Overexpression of DDX21 increased radiosensitivity in
