Genomic analysis of Enterobacter cloacae complex from Southern Thailand reveals insights into multidrug resistance genotypes and genetic diversity.

In this study, we conducted a comprehensive investigation into the Enterobacter cloacae complex (ECC), a group of notorious pathogens responsible for various hospital-acquired infections. We aimed to gain critical insights into antimicrobial resistance profiles and genomic diversity among 17 ECC isolates, which were previously collected as part of a short-term surveillance effort for 6 months in 2019. We identified two novel sequence types (ST-1936 in E. bugandensis PSU30 and ST-1937 in E. roggenkampii PSU45) among the 14 distinct STs identified in our ECC isolates. Furthermore, our expanded i

Genomic comparative analysis of Enterobacter asburiae harbouring a conjugative blaIMI-6-plasmid isolated from a public garden in Switzerland.

Ambler class A IMI enzymes are minor carbapenemases primarily associated with the Enterobacter cloacae complex. Screening soil surfaces of a well-visited public garden revealed an IMI-6-producing Enterobacter asburiae ST657 near a trash can. The strain harboured a 163-kb conjugative IncFII(Yp) plasmid containing blaIMI-6 and putative virulence genes. WGS comparative analysis with other clinical and non-clinical E. asburiae ST657 showed that it was genetically related to strains from a patient in France (62 ΔSNPs) and from retail salad in Switzerland (79 ΔSNPs). This finding suggests possible t

Study on the heteroresistance of carbapenem-resistant Klebsiella pneumoniae to Tigecycline and Polymyxin B and antibacterial activity of combined drugs in vitro.

BACKGROUND: Antimicrobial resistance is a global concern. Tigecycline (TGC) and polymyxin B (PMB) constitute some of the last treatment options remaining for carbapenem-resistant Klebsiella pneumoniae (CRKP). However, heteroresistance to these two drugs has rarely been reported. In this study, we investigated the prevalence of heteroresistance to TGC and PMB among clinically isolated CRKP strains, and assessed the synergistic activity of TGC plus PMB, TGC plus imipenem (IPM), and PMB plus IPM against heteroresistant CRKP. METHODS: A total of 90 nonduplicated CRKP clinical isolates were collec

Edwardsiella ictaluri type III secretion system effector EseG modulates cytoskeletal dynamics and immune response in macrophages.

Edwardsiella ictaluri is a gram-negative enteric pathogen responsible for enteric septicemia of catfish. One of the critical virulence factors identified in E. ictaluri is its type III secretion system (T3SS). In this study, we report that the T3SS effector protein EseG requires the small chaperone protein EscB for translocation. EseG shows partial homology to two Salmonella T3SS effectors, SseG and SseF, as well as to the Edwardsiella piscicida effector EseG, all of which also require chaperones for translocation. Functionally, EseG interacts with and inactivates Ras homolog family member A (

Water source, latrine type, and rainfall are associated with detection of non-optimal and enteric bacteria in the vaginal microbiome: a prospective observational cohort study nested within a cluster randomized controlled trial.

BACKGROUND: Less than one-third of sub-Saharan Africans have access to improved water sources. In US, Indian, and African studies, Bacterial vaginosis (BV) is increased among women with poor water, sanitation, and hygiene (WASH). We examined water source, sanitation (latrine type), and rainfall in relation to the vaginal microbiome (VMB). METHODS: In a cluster randomized controlled trial of menstrual cups and cash transfer, we measured the impact of cups on VMB via 16S rRNA gene amplicon sequencing in a subset of 436 adolescent girls. We analyzed how self-reported water source and latrine typ

A combined therapy of meropenem-ZnO nanoparticles efficiently eliminates carbapenem-resistant Klebsiella pneumoniae biofilms, with reduced nephrotoxicity (in vitro).

In response to the World Health Organization's research agenda of antimicrobial resistance in human health, this study appraised the antibacterial and antibiofilm synergistic activity of meropenem and ZnO nanoparticles (ZnO-NPs) combination against carbapenem-resistant Klebsiella pneumoniae (CRKP). The minimum inhibitory concentration (MIC) of meropenem in combination was found to be ~1/12 of its MIC alone. The results of microtiter dilution assay showed that the combination was more efficient in reducing the biofilm biomass than meropenem alone or ZnO-NPs alone. The scanning-electron-microsco

Diet-driven differential response of Akkermansia muciniphila modulates pathogen susceptibility.

The erosion of the colonic mucus layer by a dietary fiber-deprived gut microbiota results in heightened susceptibility to an attaching and effacing pathogen, Citrobacter rodentium. Nevertheless, the questions of whether and how specific mucolytic bacteria aid in the increased pathogen susceptibility remain unexplored. Here, we leverage a functionally characterized, 14-member synthetic human microbiota in gnotobiotic mice to deduce which bacteria and functions are responsible for the pathogen susceptibility. Using strain dropouts of mucolytic bacteria from the community, we show that Akkermansi

Isolation of OXA-48-like carbapenemase-producing Escherichia coli susceptible to piperacillin/tazobactam in a Japanese patient without a history of travel abroad.

Oxacillinase (OXA)-48-like β-lactamases are the most common carbapenemases in Enterobacterales in certain regions of the world and are being introduced on a regular basis into regions of non-endemicity. Japan has been characterized by low rates of carbapenemase-producing Enterobacterales, and among them, OXA-48-like carbapenemase-producing isolates are extremely rare. Here we describe a Japanese medical worker, without a history of travel abroad, who was diagnosed as having a community-acquired urinary tract infection, and whose urine sample was found to be positive for OXA-48-like carbapenema

A teleost interleukin-16 is implicated in peripheral blood leukocytes recruitment and anti-bacterial immunity.

Interleukin-16 (IL-16), as a lymphocyte chemoattractant cytokine, plays a crucial role in regulating cellular activities and anti-pathogen immunity. In teleost, the information about the antibacterial effect of IL-16 is scarce. In our study, we examined the immune functions of an IL-16 homologue (CsIL-16) from tongue sole Cynoglossus semilaevis. The CsIL-16 precursor (proCsIL-16) is comprised of 1181 amino acid residues, sharing 21.1%-67.3% identities with IL-16 precursor from invertebrate and vertebrate. The C-terminal proCsIL-16 containing two PDZ domains was designated as mature CsIL-16 whi
