New local ganirelix sustained release therapy for uterine leiomyoma. Evaluation in a preclinical organ model.

Currently, there is a limited number of treatment options available for patients with symptomatic leiomyomas, and surgical removal is by far the most frequent procedure. Previous studies found that GnRH agonists and antagonists acting through GnRH receptors led to cell death and decreased extracellular synthesis in cultured leiomyoma cells. In this study, we encapsulated the GnRH antagonist ganirelix in PLGA microspheres contained in an alginate scaffold that also supports a leiomyoma ex vivo tissue explant. Microspheres maintained ganirelix concentration stably during six days of culture, ind

The Proteome of Preretinal Tissue in Proliferative Vitreoretinopathy.

BACKGROUND AND OBJECTIVE: Proliferative vitreoretinopathy (PVR) is the leading cause of retinal detachment repair failure. However, the molecular pathogenesis remains incompletely understood. Determining the proteome of PVR will help to identify novel therapeutic targets. MATERIALS AND METHODS: Preretinal tissue samples, delaminated during surgery from six PVR cases and one idiopathic epiretinal membrane (ERM) were analyzed by mass spectrometry. Tandem mass spectra were extracted using the UniProt database, generating a list of 896 proteins, which were subjected to pathway set and fold-change

Polarized Exosome Release from the Retinal Pigmented Epithelium.

The retinal pigmented epithelium (RPE) forms the outer blood-retinal barrier and provides nutrients and recycling of visual pigment to the photoreceptors, among many other functions. The RPE is also a key site of pathophysiological changes in age-related macular degeneration, making it an important focus of study in both visual health and disease. Exosomes are nanometer-sized vesicles that are released by cells in a controlled fashion and mediate a range of extra- and intercellular activities. Some key exosome actions include cell-cell communication, immune modulation, extracellular matrix tur

Interaction of Bacterial Exotoxins with Neutrophil Extracellular Traps: Impact for the Infected Host.

Since their discovery in 2004, neutrophil extracellular traps (NETs) have been characterized as a fundamental host innate immune defense against various pathogens. Released in response to infectious and pro-inflammatory stimuli, NETs can immobilize invading pathogens within a fibrous matrix consisting of DNA, histones, and antimicrobial peptides. Conversely, excessive or dysregulated NET release may hold a variety of detrimental consequences for the host. A fine balance between NET formation and elimination is necessary to sustain a protective effect during infectious challenge. In recent year

Heparan sulfate saccharides modify focal adhesions: implication in mucopolysaccharidosis neuropathophysiology.

Mucopolysaccharidoses type III (MPSIII, Sanfilippo syndrome) are genetic diseases due to deficient heparan sulfate (HS) saccharide digestion by lysosomal exoglycanases. Progressive accumulation of undigested saccharides causes early-onset behavioural and cognitive symptoms. The precise role of these saccharides in the pathophysiological cascade is still unclear. We showed that exposure of wild-type neural cells to exogenous soluble HS fragments of at least eight saccharides activated integrin-based focal adhesions (FAs), which attach cells to the extracellular matrix. FAs were constitutively a

Macrophage matrix metalloproteinase-12 dampens inflammation and neutrophil influx in arthritis.

Resolution of inflammation reduces pathological tissue destruction and restores tissue homeostasis. Here, we used a proteomic protease substrate discovery approach, terminal amine isotopic labeling of substrates (TAILS), to analyze the role of the macrophage-specific matrix metalloproteinase-12 (MMP12) in inflammation. In murine peritonitis, MMP12 inactivates antithrombin and activates prothrombin, prolonging the activated partial thromboplastin time. Furthermore, MMP12 inactivates complement C3 to reduce complement activation and inactivates the chemoattractant anaphylatoxins C3a and C5a, whe

A pharmacoproteomic study confirms the synergistic effect of chondroitin sulfate and glucosamine.

Osteoarthritis (OA) is the most common age-related rheumatic disease. Chondrocytes play a primary role in mediating cartilage destruction and extracellular matrix (ECM) breakdown, which are main features of the OA joint. Quantitative proteomics technologies are demonstrating a very interesting power for studying the molecular effects of some drugs currently used to treat OA patients, such as chondroitin sulfate (CS) and glucosamine (GlcN). In this work, we employed the iTRAQ (isobaric tags for relative and absolute quantitation) technique to assess the effect of CS and GlcN, both alone and in

Neural adhesion molecule L1 as a member of the immunoglobulin superfamily with binding domains similar to fibronectin.

Diverse glycoproteins of cell surfaces and extracellular matrices operationally termed 'adhesion molecules' are important in the specification of cell interactions during development, maintenance and regeneration of the nervous system. These adhesion molecules have distinct functions involving different cells at different developmental stages, but may cooperate when expressed together. Families of adhesion molecules which share common carbohydrate domains do exist, despite the structural and functional diversity of these glycoproteins. These include the Ca2+-independent neural adhesion molecul

Decoding intervertebral disc degeneration pathomechanics: from mechanisms to therapeutic horizons.

BACKGROUND: Intervertebral disc degeneration (IVDD) is widely recognized as a key factor in various lumbar spine disorders. Biomechanical factors play a crucial role in maintaining homeostasis and influencing the degeneration process of the intervertebral disc (IVD). The nucleus pulposus (NP), located at the core of the disc, is responsible for distributing mechanical stresses from spinal movements, making it a primary site for degeneration. AIM OF REVIEW: This review designs to elucidate the key mechanisms of pathological mechanics in the nucleus pulposus and provide insights for biomechanic

Continuous aerobic granular sludge activated by a contracted final settler and associated hydraulic measures.

A contracted & baffled final settler, with a low H (height)/D (diameter) ratio and a high hydraulic selective pressure (HSP) imposed by elevating up-flow velocity by influent or effluent, was applied to promote in-situ granulation in a continuous biological nutrient removal (BNR) process. Under observational HSPobs = 0.9-3.2 m3/(m2·h), baffles could retain potential granules, and wash out flocs via 4-5 times higher up-flow velocity through baffle gaps. Continuous granulation was efficient within 44 d (HSPobs = 0.9-1.9 m3/m2·h): granulation increased to 42%, and SVI30 decreased to 52 mL/g from

Mechanical Stiffening Promotes Growth, Invasion-Associated Phenotypes, and Reduced Selumetinib Sensitivity in 3D Plexiform Neurofibroma Cultures.

Plexiform neurofibromas (pNF1s) are benign peripheral nerve sheath tumors caused by NF1 loss, leading to dysregulated RAS/mitogen-activated protein kinase (MAPK) signaling. While the mitogen-activated protein kinase kinase (MEK) inhibitors, selumetinib and mirdametinib, can reduce tumor volume, surgical resection remains the primary treatment for immediate debulking and symptom relief. Complete removal is often limited by tumor infiltration along nerve plexuses, and residual tumors may undergo postsurgical tissue remodeling, producing localized regions of stiffened extracellular matrix (ECM).

Antibody subclass deficiency accelerates tumorigenesis in genetically engineered mouse models of pancreatic cancer.

Antibody production by B cells has emerged as an important factor in regulating antitumor immunity with both suppressive and promotive roles in cancer. However, the specific effect of antibody deficiency during development of pancreatic ductal adenocarcinoma (PDAC) has not been explored. To address this question, we crossed the well-established KPC mouse model to mice lacking all circulating immunoglobulin (Ig) due to genetic ablation of both Ig secretion and Ig class switching (KPC-μSAID mice). KPC-μSAID mice exhibited a two-fold acceleration in tumor formation, a two-fold reduction in median

Anterior cruciate ligament overuse increases collagen fibril compliance: collagen unraveling and elevated glycosaminoglycan activity in a murine model.

Data increasingly indicates that many anterior cruciate ligament (ACL) injuries are likely due to tissue damage accumulation and the gradual decrease in ACL strength and stiffness over time. Previous ex vivo experiments found molecular-level ACL damage (collagen unraveling) to be associated with increased tissue compliance at the collagen fibril-level. However, it remains unclear if this biomechanical effect is present in vivo, or what effect other modulators of tissue mechanics (e.g., glycosaminoglycans (GAGs)) have in fatigued-damaged ACL matrices. Herein, we investigated the in vivo relatio

4-Methylumbelliferone Restores Age-Related Changes in Perineuronal Nets, Memory, and Neuroinflammation.

Aging-associated cognitive decline is closely linked to dysregulated glial activity and chronic neuroinflammation, which together constrain neuronal plasticity. Astrocytes and microglia are key regulators of the brain extracellular matrix (ECM), including perineuronal nets (PNNs), whose accumulation during aging contributes to reduced synaptic flexibility. Targeting ECM-glia interactions therefore represents a promising strategy to modulate neuroinflammatory and plasticity-related processes. Here, we investigated the effects of long-term oral administration of 4-methylumbelliferone (4-MU), an

Effects of adhesion pattern of 2D substrate on cell morphology and migration.

Focal adhesions play critical roles in a variety of cellular behaviors and physiological processes, including cell migration, proliferation, wound healing, and tumor invasion. Although focal adhesions are recognized as key protein signaling and mechanosensory hubs that mediate interactions between the cell and the extracellular matrix (ECM), the mechanisms by which cells sense and respond to ECM geometry at the subcellular level, and how these cues are translated into cell-scale behaviors, remain unclear. In this study, we develop a computational cell model to investigate the effects of adhesi

Molecular mechanisms of skeletal muscle fibrosis and potential targeted therapeutic strategies.

Skeletal muscle fibrosis is a pathological process characterized by excessive deposition of extracellular matrix (ECM). It commonly occurs in various diseases such as muscular dystrophy, aging, cancer cachexia, and muscle injury. This condition leads to destruction of muscle structure, loss of function, and impaired regeneration, significantly affecting patients' quality of life. This review systematically summarizes the molecular mechanisms underlying skeletal muscle fibrosis. Key signaling pathways include transforming growth factor-beta (TGF-β)/Smad, yes-associated protein/transcriptional c

Decoding collagen cues: the interplay of integrins and discoidin domain receptors in health and disease.

The extracellular matrix (ECM) provides critical biochemical and biophysical cues that regulate cell behavior in health and disease. Collagens dominate in abundance and structural importance, shaping tissue-specific ECM signatures that guide cellular behavior. Two major and distinct transmembrane receptor families, integrins and discoidin domain receptors (DDRs), serve as primary sensors for collagens, yet they employ fundamentally distinct binding mechanisms and signaling kinetics. While both can activate shared downstream pathways, their functional interplay remains complex and context-depen

CD31+ Cell Enrichment Enhances Therapeutic Effects of Stromal Vascular Fraction in Experimental Primary Osteoarthritis: A Preclinical Study in the Dunkin Hartley Guinea Pig Model.

Despite the growing burden of osteoarthritis (OA), effective disease-modifying treatments remain limited. Hyaluronic acid, platelet-rich plasma, and hydrogels are in clinical use, but their efficacy beyond placebo is debated. Cell-based therapies are continuously investigated for their disease-modifying potential. Stromal vascular fraction (SVF), a minimally processed and clinically accessible cell product, has shown therapeutic potential, but remains poorly characterized in respect to its pro-regenerative cell types and molecular signaling. To determine whether CD31+ cell enrichment enhances

We continuously recorded oxygen-consumption rate (OCR) in nasal chondrocyte (NC), MSC, and NC-MSC co-culture pellets cultured for 27 days under normoxia, and integrated OCR trajectories with time-resolved gene expression, matrix histology, glycosaminoglycan (GAG)/DNA, and unconfined compression mechanics across defined NC:MSC ratios (monocul
