Cycle dependent LH/hCG receptor gene expression in porcine nongonadal reproductive tissues.

Uterine and other nongonadal reproductive tissues are a new target for LH and CG action in pig and many other mammalian species, including humans. Since rat and pig LH/CG receptor cDNAs have been cloned we have been prompted to look for the expression of this gene in porcine uteri and oviducts. Reverse transcription-polymerase chain reaction (RT-PCR) was carried out on total RNA isolated from endometrium, myometrium and oviduct. Ovarian and testicular RNA served as positive and skeletal muscle RNA as negative control. RT-PCR was designed to amplify part of the extracellular domain (exons II-IX

Molecular study of the 5 alpha-reductase type 2 gene in three European families with 5 alpha-reductase deficiency.

The molecular basis of 5 alpha-reductase (5 alpha R) deficiency was investigated in four patients from three European families. In the French family, the first patient was raised as a female, and gonadectomy was performed before puberty. The second sibling, also raised as female, differed in that gonadal removal was performed after the onset of pubertal masculinization. The other two patients, both from Polish families, developed masculinization of external genitalia during puberty. All patients developed a female sexual identity. In all cases, no known consanguinity or family history of 5 alp

The amplification refractory mutation system (ARMS): a rapid and direct prenatal diagnostic technique for beta-thalassaemia in Singapore.

beta-Thalassaemia major patients have chronic anaemia and since 3-4 per cent of Singaporeans carry the beta-gene, prenatal diagnosis is essential. We evaluated the amplification refractory mutation system (ARMS) technique as a routine test for prenatal diagnosis of beta-major. Six mutations along the beta-gene were studied--41-42 (-TCTT), IVSII #654 (C-T), 17 beta (A-T), -28 TATA (A-G), IVSI #5 (G-C), and IVSI #1 (G-T). Our results indicate that prenatal diagnosis using these mutations can be offered to 90 per cent (35/39) of our Chinese couples and 54.6 per cent (12/22) of our Malay couples a

The role of the sex-determining region Y gene in the etiology of 46,XX maleness.

The condition of 46,XX maleness is characterized by testicular development in subjects who have two X chromosomes but who lack a normal Y chromosome. Several etiologies have been proposed to explain 46,XX maleness: 1) translocation of the testis-determining factor from the Y to the X chromosome, 2) mutation in an autosomal or X chromosome gene which permits testicular determination in the absence of TDF, and 3) undetected mosaicism with a Y-bearing cell line. We evaluated 10 affected subjects who were ascertained for different reasons and who had several distinct phenotypes. Six subjects had i

Baboon T cell lymphomas expressing the B cell-associated surface proteins CD40 and Bgp95.

Papio hamadryas baboons in the Sukhumi colony develop enzootic outbreaks of malignant lymphomas with an incidence of about 1.5% per year among adults of the high-risk stock. We investigated the surface phenotypes of cells from normal and lymphomatous animals using antibodies against human lymphocyte antigens. We found that more than 80% of the lymphomas that developed during the last 3 years were characterized histologically to be of the peripheral T cell type. Generally, the lymphomatous cells also expressed high levels of MHC class II DR protein, CD18 (LFA-1 beta chain), and CD45RO. Surprisi

The role of the sex-determining region of the Y chromosome (SRY) in the etiology of 46,XX true hermaphroditism.

The syndrome of 46,XX true hermaphroditism is a clinical condition in which both ovarian and testicular tissue are found in one individual. Both Mullerian and Wolffian structures are usually present, and external genitalia are often ambiguous. Two alternative mechanisms have been proposed to explain the development of testicular tissue in these subjects: (1) translocation of chromosomal material encoding the testicular determination factor (TDF) from the Y to the X chromosome or to an autosome, or (2) an autosomal dominant mutation that permits testicular determination in the absence of TDF. Associations between mutations and a VNTR in the human phenylalanine hydroxylase gene.

The HindIII RFLP in the human phenylalanine hydroxylase (PAH) gene is caused by the presence of an AT-rich (70%) minisatellite region. This region contains various multiple of 30-bp tandem repeats and is located 3 kb downstream of the final exon of the gene. PCR-mediated amplification of this region from haplotyped PAH chromosomes indicates that the previously reported 4.0-kb HindIII allele contains three of these repeats, while the 4.4-kb HindIII allele contains 12 of these repeats. The 4.2-kb HindIII fragment can contain six, seven, eight, or nine copies of this repeat. These variations perm

Efficient duck hepatitis B virus production by an avian liver tumor cell line.

Duck hepatitis B virus (DHBV) is produced in small amounts following transfection of human hepatoma or hepatoblastoma cell lines with cloned viral DNA. In a search for better hosts for DHBV replication, two avian liver cell lines were investigated. One of these cell lines, LMH, produced 5 to 10 times more DNA replicative intermediates and 10 to 20 times more infectious DHBV than did either of the two human cell lines, HuH-7 and Hep G2. Utilization of cell lines in genetic analyses of virus replication is often dependent upon obtaining efficient complementation between cotransfected viral genom

Determinants of dihydroartemisinin-piperaquine treatment failure in Plasmodium falciparum malaria in Cambodia, Thailand, and Vietnam: a prospective clinical, pharmacological, and genetic study.

BACKGROUND: The emergence and spread of resistance in Plasmodium falciparum malaria to artemisinin combination therapies in the Greater Mekong subregion poses a major threat to malaria control and elimination. The current study is part of a multi-country, open-label, randomised clinical trial (TRACII, 2015-18) evaluating the efficacy, safety, and tolerability of triple artemisinin combination therapies. A very high rate of treatment failure after treatment with dihydroartemisinin-piperaquine was observed in Thailand, Cambodia, and Vietnam. The immediate public health importance of our findings

Mutational patterns in a large cohort of parathyroid carcinomas.

PURPOSE: A lack of targeted therapies make parathyroid carcinoma, a diagnostically challenging malignancy, difficult to treat. The rarity of this tumor type necessitates international collaboration to collect a sizable sample set for study. Prior studies have revealed the importance of driver mutations in the CDC73 gene and identified several putative drivers/aberrant pathways including PI3K/mTOR activation and CCND1 (cyclin D1) amplification. In this study, we sought to better understand the prevalence of putative oncogenic drivers in parathyroid carcinoma. METHODS: We subjected an expanded

Improvement the accuracy and reproducibility of telomere length measurement utilizing qPCR.

Telomere length serves as a well-established molecular biomarker for evaluating aging and age-associated diseases. Among various methods, quantitative PCR for telomere length detection is convenient, rapid, cost-effective, and capable of high-throughput analysis in large epidemiological cohorts. However, numerous studies have indicated that issues related to differences in DNA quality caused by DNA extraction process significantly affect the accuracy and reproducibility of qPCR-based telomere length quantification. Initially, we established a model of DNA integrity variation, by utilizing nucl

Graphene foam biosensors for molecular diagnostics of antimicrobial resistance and emerging pathogen threats.

Pandemic preparedness for the surge of antimicrobial resistance (AMR) and newly emerging pathogens creates an urgent need for rapid diagnostic tools which can be mass manufactured and ideally used at the point of care. Rapid adaptation to new diagnostic targets is therefore a key requirement. To meet this need, we developed a sensitive electrochemical biosensor based on disposable graphene foam (GF) electrodes functionalised with peptide nucleic acid (PNA) probes, along with a rapid and generic functionalisation strategy. Pyrene-modified PNA probes were co-immobilised with pyrene-butanol on GF

Reexamining Novel Risk Factors for Community-Acquired Clostridioides difficile: A Multimodal Approach.

BACKGROUND: Geospatial surveys have identified livestock operations as potential sources for community-acquired Clostridioides difficile infection. Confirmatory studies integrating individual-level risk factors, geospatial data, and environmental surveys are limited. METHODS: We conducted 2 parallel studies: (1) a case-control assessment of C difficile risk associated with livestock and (2) a microbiologic survey of surface waters and wastewater influent for C difficile. For aim 1, adult residents of North Carolina tested for C difficile through the Duke University Health System from March 20

Viral Heterogeneity in an Avian Reovirus Isolate Despite Plaque Purification and Differences in Segment-Wise Abundance Between Different Amplification and Purification Steps.

Avian reovirus (ARV) remains a significant concern in the poultry industry. Three rounds of plaque purification are routinely employed to obtain clonal populations of a viral isolate for experimental purposes. However, the effectiveness of this approach in achieving viral homogeneity has usually been taken for granted, especially in older publications, when there was no means of checking the purity. This study aimed to determine the purity of a plaque-purified ARV isolate (strain AL) using conventional PCR, Sanger sequencing, and whole-genome sequencing (WGS). WGS analysis of multiple samples

Development of a multipurpose diagnostic marker using PCR, real-time PCR, and LAMP assays for rapid detection of Rhizoctonia solani in rice plants and soil.

Rhizoctonia solani AG1-IA is a major soil-borne pathogen responsible for sheath blight in rice, causing significant yield losses globally. Early, rapid, and accurate detection is essential for effective disease management, as traditional methods are time-consuming and less sensitive. In this study, we developed and validated a highly specific molecular marker, BrD3 targeting the bromodomain-containing Major Facilitator Superfamily (MFS) gene. This marker was successfully employed in PCR, quantitative PCR (qPCR), and Loop-mediated Isothermal Amplification (LAMP) assays for the detection of the

Review of Mechanisms of Photodynamic and Photothermal therapy by Water-Soluble Conjugate Polymers and Their Applications in Drug and Gene Delivery.

Water-soluble conjugated polymers (WSCPs) represent an important class of functional macromolecules that combine the electronic advantages of π-conjugated polymer backbones with aqueous processability, biocompatibility, and tunable biointerfacial behavior. Unlike conventional small-molecule photosensitizers or inorganic nanomaterials, WSCPs offer polymeric design flexibility through controlled backbone engineering, side-chain functionalization, amphiphilicity, charge density, molecular weight, and nanoparticle self-assembly. These structural parameters govern their optical absorption, fluoresc

Dual dynamics of persistence and recruitment characterise the ACPA-expressing B cell response in rheumatoid arthritis.

OBJECTIVES: Rheumatoid arthritis, a prototypic autoimmune disease, is characterised by the presence of anticitrullinated protein antibodies (ACPAs). Unlike responses to recall antigens, ACPA-expressing B cells display a continuous phenotype of recent activation, even in patients with sustained clinical remission, indicating that these cells are continuously activated over long periods without 'dying out' or acquiring a resting or anergic phenotype. To elucidate whether this persistent activation reflects ongoing stimulation of the same B cell clones or recruitment of new cells, we longitudinal

Integrated transcriptomic and functional analyses identify PRR7 as a therapeutic target that associated with Wnt/β-catenin signaling and docetaxel resistance in prostate cancer.

BACKGROUND: Docetaxel resistance represents a major therapeutic challenge in advanced prostate cancer. Despite extensive studies on drug efflux, apoptosis, and metabolic adaptation, the specific cellular states and molecular determinants that sustain resistance remain poorly understood. METHODS: To delineate the cellular basis of chemoresistance, we integrated bulk RNA sequencing of docetaxel-resistant prostate cancer models with single-cell transcriptomic data from patient tumors. Resistance-associated gene signatures were projected onto a single-cell atlas to identify docetaxel-resistant as

Optimizing the hybridization chain reaction-fluorescence in situ hybridization (HCR-FISH) protocol for Pleurodeles waltl.

Advances in transcriptomic technologies have transformed the study of complex biological processes, including tissue regeneration, through high-resolution characterization of gene expression programs. In regenerative vertebrate models such as the Iberian ribbed newt (Pleurodeles waltl), these approaches provide insight into the molecular mechanisms underlying retina and lens regeneration. However, single-cell RNA sequencing studies lack spatial resolution, meaning the ability to validate gene expression patterns within ocular tissues is essential and requires optimization.
