Cardiac mitochondrial dysfunction and DNA depletion in children with hypertrophic cardiomyopathy.

Abnormalities in specific mitochondrial respiratory enzymes and DNA (mtDNA) have been reported in cardiomyopathy. In this study, we report 4 cases of severe hypertrophic cardiomyopathy (HCM) in which specific cardiac mitochondrial enzyme activity defects were found, including complex I (n = 2), complex III (n = 2), complex IV (n = 2) and complex V (n = 1). Other abnormalities were also noted including a marked depletion of mtDNA (n = 1) and decreased content of subunit 2 of cytochrome c oxidase (n = 1). None of the mtDNA point mutations and common deletions previously found in association with

[Progressive external ophthalmoplegia with extensive mitochondrial DNA deletion].

We report extensive deletion of muscle mitochondrial DNA and ophthalmological findings in a patient with progressive external ophthalmoplegia. The patient was a twenty-nine-year-old female who had suffered from slowly progressive external ophthalmoplegia for ten years. No abnormal finding was observed in the funduscopic examination, but fluorescein angiography showed mottled hyperfluorescence. The final rod threshold for the dark adaptation testing was slightly elevated. The flash electroretinogram showed a slightly attenuated b wave. Ragged-red fibers were observed in muscle biopsies. A bioch

Distinct Myogenic Stages Recapitulate Transcriptomic Networks in COPD Cachexia.

BACKGROUND: Cachexia is an extrapulmonary manifestation of Chronic Obstructive Pulmonary Disease (COPD) characterized by weight loss and muscle wasting. Transcriptomic profiling of vastus lateralis biopsies enables profiling of COPD-cachexia relevant dysregulation. As obtaining muscle biopsies is invasive and yields limited tissue, human muscle derived cultures (HMDC) may enable mechanistic research into cachexia. However, questions remain regarding the extent to which HMDC recapitulate transcriptomic signatures of bulk skeletal muscle in COPD-cachexia. To address this gap, we tested whether C

Oxidative stress-related biomarkers in thyroid eye disease: evidence from bioinformatics analysis and experimental validation.

BACKGROUND: Oxidative stress is a key contributor to the pathogenesis of the autoimmune condition thyroid eye disease (TED). However, its precise molecular mechanisms and reliable biomarkers remain unclear. Bioinformatics enables the identification of differentially expressed genes through transcriptomic analysis. However, distinguishing truly relevant findings from false discoveries remains challenging. Immunohistochemistry helps address this limitation by validating protein expression levels, revealing local immune responses, and linking microscopic tissue changes to clinical manifestations.

Identification and immune characteristics of N6-methyladenosine related ferroptosis-related genes in intervertebral disc degeneration.

The damage of ferroptosis is related to the pathogenesis of intervertebral disc degeneration (IDD). N6-methyladenosine (m6A) modification accounts for more than 80% of RNA modifications in eukaryotic cells. However, the key role of m6A related ferroptosis-related genes (FRGs) in IDD remain explorable. Firstly, batch correction between datasets was performed. Weighted gene co-expression network analysis (WGCNA) was then conducted to acquire the most relevant module genes. Differentially expressed genes (DEGs) between the IDD and Normal Group were obtained by differential analysis subsequently.

Gene Expression Analysis of Mitochondria-Associated Membrane (MAM)-Related Genes in ER Stress and Alzheimer's Disease.

Alzheimer's Disease (AD) is a devastating neurodegenerative disease, strongly linked to cellular stress originating from the accumulation of the Amyloid-beta (Aβ) peptide and phosphorylated tau protein. Endoplasmic Reticulum (ER) stress and Unfolded Protein Response (UPR) are reported as early events in the AD pathology. Mitochondria-Associated Membrane (MAM) is a proteinaceous tethering between the ER and mitochondria that plays a role in regulating ER stress and related responses. A high level of ER-mitochondria tethering, thereby mitochondrial Calcium (Ca2+) overload and cell death, has bee

Spatial gene expression and functional network abnormalities in multiple sclerosis: exploring biological influence on brain functional reorganization.

In multiple sclerosis (MS), functional network abnormalities arise as structural damage accumulates. However their biological basis and spatial distribution remain unclear. This study investigated the associations between MS-related functional network abnormalities and physiological gene expression using the Allen Human Brain Atlas (AHBA). Five-hundred fifty-eight MS patients and 214 healthy controls (HC) underwent neurological assessment and 3 T MRI; 491 patients also completed a neuropsychological evaluation. Resting-state functional MRI was used to generate degree centrality maps to identif

Disease duration impacts intestinal gene expression profiles in Crohn's disease but not in ulcerative colitis.

BACKGROUND: Disease duration is associated with lower treatment response and accrual of bowel damage in Crohn's disease (CD), but not in ulcerative colitis (UC). We aimed to understand intestinal transcriptomic changes associated with disease duration in CD and UC. METHODS: We analyzed intestinal tissue RNA sequencing data from two independent prospective cohorts of CD and UC patients, the Mount Sinai Crohn's and Colitis Registry (MSCCR; nCD = 498, nUC = 421), and the Study of a Prospective Adult Research Cohort with Inflammatory Bowel Disease (SPARC IBD; nCD = 777, nUC = 440). Uremic toxin receptor NR1H3 contributes to hyperlipidemia- and chronic kidney disease-accelerated vascular inflammation, which is partially suppressed by novel YBX2 anti-ROS pathway.

Hyperlipidemia and chronic kidney disease (CKD) are well-established risk factors for cardiovascular disease and act synergistically to promote vascular inflammation and disease progression. However, the mechanisms underlying this synergetic effect remain largely unknown. Using a mouse model combining hyperlipidemia (via high-fat diet feeding, HFD) with 5/6 nephrectomy-induced CKD, we made the following significant findings: 1) HFD + CKD upregulated 1179 genes in mouse aortas and induced prominent reactive oxygen species (ROS), far more than either HFD or CKD alone. 2) HFD + CKD upregulated 86

Exploration and Validation of Ferroptosis-Associated Genes in ADAR1 Deletion-Induced NAFLD through RNA-seq Analysis.

BACKGROUND: Ferroptosis, characterized by excessive iron ions and lipid peroxides accumulation, contributes to Nonalcoholic Fatty Liver Disease (NAFLD) development. The role of ADAR1, crucial for lipid metabolism and immune regulation, in ferroptosis-related NAFLD remains unexplored. METHODS: In this study, we analyzed the expression of ADAR1 in NAFLD patients using the GSE66676 database. Subsequently, We investigated the effects of ADAR1 knockdown on mitochondrial membrane potential (MMP), Fe2+ levels, oxidation products, and ferroptosis in NAFLD cells through in vitro and in vivo experiment

Short-term oral pre-exposure prophylaxis against HIV-1 modulates the transcriptome of foreskin tissue in young men in Africa.

Whilst short-term oral pre-exposure prophylaxis (PrEP) with antiretroviral drugs in men who have sex with men has shown protection against HIV-1 infection, the impact of this regimen on the in vivo foreskin transcriptome is unknown. We collected foreskin tissue after voluntary medical male circumcision from 144 young men (72 from Uganda and 72 from South Africa) randomized to one to two doses of either oral tenofovir (TFV) disoproxil fumarate (FTC-TDF) or tenofovir alafenamide (FTC-TAF) or no drug (untreated controls). This novel approach allowed us to examine the impact of short-term oral PrE

Phenotypic variability in a Spanish family with MNGIE.

Clinical, biochemical, and genetic features of a Spanish family with mitochondrial neurogastrointestinal encephalomyopathy are reported. The proband presented with severe gastrointestinal dysmotility and the affected sister had extraocular muscle weakness. In both affected individuals, biochemical defects of thymidine phosphorylase and a pathogenic G-to-A transition mutation at nucleotide 435 in the thymidine phosphorylase gene were identified. The first thymidine phosphorylase mutation identified in Spain showed phenotypic variability at onset.

Targeting NLRP3 Inflammasome Signaling After Myocardial Infarction: Mechanistic Rationale, Regulatory Networks, and Translational Perspectives.

Myocardial infarction (MI) remains a major cause of death and heart failure despite advances in reperfusion and secondary prevention. Sterile inflammation triggered by ischemic myocardial injury is increasingly recognized as a determinant of infarct expansion, adverse ventricular remodeling, and subsequent clinical outcomes. The NOD-like receptor protein 3 (NLRP3) inflammasome integrates danger-associated molecular patterns, mitochondrial dysfunction, ionic disturbance, metabolic stress, and innate immune signaling. Following activation, caspase-1 processes interleukin-1β and interleukin-18 an

Broad-spectrum antifungal activity of C12/C14 alkyl triphenylphosphonium salts (TPP-C12 and TPP-C14) against clinically relevant pathogens.

INTRODUCTION: Human fungal infections affect billions of people and result in more than 2 million deaths every year, however, they have historically been neglected as a cause of infectious disease-related deaths worldwide. Fungal drug resistance has become an increasingly serious problem with the wide use of antifungal drugs and the adaptive evolution of fungi. Resistance to all commonly used antifungal drugs has been reported, and the development of non-traditional antifungal drugs is urgently needed. METHODS: Minimal inhibitory concentrations (MICs) of clinical pathogenic fungi were assesse
