Tissue-specific induction of SOCS gene expression by PRL.

The mechanisms whereby tissue sensitivity to PRL is controlled are not well understood. Here we report that expression of mRNA and protein for members of the SOCS/CIS/JAB family of cytokine signaling inhibitors is increased by PRL administration in ovary and adrenal gland of the lactating rat deprived of circulating PRL and pups for 24 h but not in mammary gland. Moreover, suckling increases SOCS mRNA in the ovary but not in the mammary gland of pup-deprived rats. Deprivation of PRL and pups for 48 h allows the mammary gland to induce SOCS genes in response to PRL administration, and this is a

Effect of the G-Protein-Coupled Receptor T2R14 on Proliferation and Cell Population Growth in Oral Cancer Cells.

Oral cancer is a leading cause of cancer-related deaths and significantly affects the quality of life of patients. However, many of its mechanisms remain unclear, and its treatment needs improvement. The G-protein-coupled receptor taste receptor type 2 member 14 (T2R14 or TAS2R14) is expressed in various cancer types. However, few studies have investigated its roles in oral cancer, and its effects on oral cancer cell proliferation and growth are unknown. This study aimed to examine T2R14's impact on proliferation and cell population growth (CPG) of oral cancer cells. TAS2R14 gene knockout was

Precisely Engineered Block Copolymers for Efficient PTEN mRNA Therapy of Non-Small Cell Lung Cancer.

Restoring the tumor suppressor PTEN (phosphatase and tensin homolog) offers a promising therapeutic strategy for nonsmall cell lung cancer (NSCLC). In particular, mRNA-based therapy provides a compelling alternative to small molecules and DNA-based gene therapy, combining high specificity with an excellent safety profile. However, its clinical translation hinges on the development of safe and efficient delivery systems. In this study, we present a rationally designed diblock copolymer that mediates highly effective PTEN mRNA delivery both in vitro and in vivo. The copolymer consists of a steal

miR-150-5p at the Interface of Cancer Biology and Immune Regulation: Dual Roles, Mechanisms, and Therapeutic Opportunities.

MicroRNAs (miRNAs) are small, non-coding RNAs that regulate gene expression post-transcriptionally and play crucial roles in cancer biology and immune function. Among them, miR-150-5p has emerged as a key regulator with complex, context-dependent roles in both tumorigenesis and immune cell differentiation. This review provides a comprehensive synthesis of current knowledge on miR-150-5p, highlighting its dual function as a tumor suppressor or oncogene depending on cancer type and cellular context. We examine its involvement in hematologic malignancies and solid tumors, detailing the molecular

Silencing the Signal: The Metastasis Suppressor NDRG1 Disrupts Small Extracellular Vesicle-Mediated Crosstalk in Pancreatic Cancer.

Pancreatic cancer (PaC) remains one of the deadliest cancers, with 5-year survival rates of 13%. A major driver of its aggressiveness is the tumour microenvironment (TME), which fuels tumour growth, metastasis, and therapeutic resistance through dynamic, bi-directional communication between cancer cells, fibroblasts, and immune cells. Emerging evidence highlights extracellular vesicles (EVs) as key mediators of oncogenic cross-talk within the PaC TME. This study demonstrates for the first time that the overexpression of metastasis suppressor N-myc downstream regulated gene 1 (NDRG1) significan

G-quadruplex homeostasis is a determinant of PARP inhibitor toxicity in BRCA2-deficient cells.

Inhibition of PARPs is a key strategy to treat tumours with defects in homologous recombination (HR), including those with mutations in the tumour suppressor gene BRCA2. PARP inhibitors generate replication stress, creating a dependence on HR to repair the resulting DNA damage. However, the DNA lesions generated upon PARP inhibition that impede replication fork progression and trigger a requirement for BRCA2 in cell survival are poorly defined. Here, we demonstrate that elevated levels of G-quadruplex (G4) DNA structures is a determinant of genome instability and PARP inhibitor toxicity, while

Role of microRNAs in cisplatin response of cancer stem cells.

Cisplatin (CDDP)-based chemotherapy remains a cornerstone of cancer treatment, but its efficacy is often limited by the development of chemo resistance that is mainly associated with cancer stem cells (CSCs). MicroRNAs (miRNAs) have emerged as pivotal post-transcriptional regulators of gene expression, critically influencing key cellular processes, including drug response. This review aims to provide a comprehensive analysis of the current evidence elucidating the role of specific miRNAs in governing the CDDP response of CSCs. We conducted a systematic assessment of all available scientific li

Immunohistochemical expression of E-cadherin and Bcl-2 in oral squamous cell carcinoma.

BACKGROUND: The most frequent malignancy of the oral cavity is oral squamous cell carcinoma (OSCC), which accounts for up to 80-90% of all malignant neoplasms of the oral cavity. It results from the gradual accumulation of diverse genetic alterations. Alcohol use, tobacco use, betel quid chewing, human papillomavirus (HPV) and poor nutrition are significant risk factors for oral squamous cell carcinoma (OSCC). The cancer suppressor gene E-cadherin establishes a cutoff for Wnt-catenin signaling. Cell-cell adhesion is lost as a result of the Wnt signaling pathway being amplified when E-Cadherin

Fine-tuning neuron number: Lzts as a key regulator of neurogenesis and cell proliferation in Ciona.

How progenitor mitotic timing is coupled to the number of neurons in a nervous system remains poorly understood, in part because most model systems have large, variable progenitor pools that obscure this relationship. The non-vertebrate chordate Ciona robusta offers a tractable alternative: its larval nervous system arises from an invariant lineage, including four Bipolar Tail Neurons (BTNs) that are reproducibly generated from the caudal neural plate border. Here we investigate the developmental function of the single Ciona ortholog belonging to the leucine zipper tumor suppressor (Lzts) gene

A-to-I RNA Editing Endows miR-3664-5p with Carcinogenicity in Breast Cancer Through Modulating LONP2 Mediated Glycolysis.

Breast cancer has become the main cause threatening women's health. RNA editing is one of the most important mechanisms for modifying genetic information. This study explained the relationship between A-to-I RNA edited miR-3664-5p and malignant phenotype of breast cancer. The results showed that high editing level of miR-3664-5p in breast cancer, which was related to tumor stage, recurrence and prognosis of breast cancer patients. ADAR perturbation experiments demonstrated that ADAR1 is the key enzyme regulating miR-3664-5p editing in breast cancer. A series of in vitro assays revealed that mi

EZH2 PROTACs outperform catalytic inhibitors in prostate cancer by targeting a methylation-independent function of PRC2.

Enhancer of Zeste Homolog 2 (EZH2) is the enzymatic subunit of the Polycomb Repressive Complex 2 (PRC2). It catalyzes H3K27 methylation for epigenetic silencing of tumor suppressors and critically drives prostate cancer (PCa) progression. However, inhibitors of EZH2 catalytic function (EZH2i), such as EPZ-6438, showed limited efficacy in PCa. Here, we designed and developed a series of VHL-based proteolysis-targeting chimera (PROTAC) degraders of EZH2 using EPZ-6438 as a ligand and identified PROTAC-6272 as a lead compound. PROTAC-6272 effectively degraded EZH2 and other PRC2 subunits across d

Therapeutic approaches for nonsense mutations in CFTR.

This manuscript reviews recent developments in suppressing nonsense mutations in the CFTR gene. Nonsense mutations lead to premature termination codons (PTCs)-UAG, UGA, or UAA- that cause the production of truncated, non-functional proteins and result in the degradation of transcripts by the nonsense-mediated decay (NMD) pathway, and in some cases exon skipping. Readthrough approaches facilitate incorporation of amino acids at nonsense codons, enabling the synthesis of a functional full-length protein. We focus on three readthrough strategies that may lead to therapeutics. First, high-throughp

Belzutifan in Von Hippel-Lindau Syndrome and Renal Cell Carcinoma: Mechanisms, Clinical Evidence, and Future Directions.

Clear cell renal carcinoma (ccRCC) and von Hippel-Lindau (VHL)-associated tumors arise due to dysregulation of the VHL/HIF pathway resulting from loss or inactivation of the VHL tumor suppressor gene, leading to constitutive activation of hypoxia-inducible factors (HIFs). Although immune checkpoint inhibitors and VEGF-targeted therapies have improved outcomes, these treatment modalities do not directly inhibit HIF-dependent transcription factors and are limited by resistance, cumulative adverse effects, and incomplete pathway suppression. With the introduction of belzutifan, a selective HIF-2α

The expanding landscape of Deltex: signaling crosstalk and clinical implications.

The deltex (DTX) gene family encodes RING-type E3 ubiquitin ligases that are central to the regulation of the Notch signaling pathway, a mechanism governing cell fate, differentiation, proliferation, and apoptosis during development and adult tissue homeostasis. In Drosophila melanogaster, the Deltex (Dx) fine-tunes Notch signaling by modulating receptor endocytosis and degradation, acting as either an enhancer or a suppressor depending on the context. Insights from flies have revealed the evolutionary conservation of the Dx-Notch interaction across species, including humans. However, recent s

Targeted NMR signal enhancement of RNA by site-directed bis-nitroxide labeling.

MicroRNAs regulate gene expression through sequence-specific interactions with target messenger RNAs (mRNAs), and their misregulation is a hallmark of cancer. MicroRNA-34a (miR-34a), a key modulator of the tumor suppressor p53, binds the mRNA encoding sirtuin 1 (mSirt1) and adopts multiple conformational states that influence repression efficiency. While such dynamics have been characterized in vitro, extending these studies to cellular environments is hampered by weak signals and substantial background inherent to nucleic acid NMR. To overcome this limitation, we developed a site-directed spi

Quantifying immune memory in colorectal cancer using fractional-order differential equations.

Traditional mathematical models of tumor-immune dynamics employ integer-order ordinary differential equations that assume Markovian behavior, wherein the system's instantaneous rate of change depends only on its current state. However, biological processes such as T cell exhaustion and myeloid-derived suppressor cell (MDSC) accumulation exhibit memory effects, where past immunosuppressive exposures create persistent functional states through epigenetic modifications and microenvironmental feedback loops. We develop a fractional-order differential equation framework to quantify these non-Markov

Suppression of Calcium Influx Reduces Coxsackievirus B3 Lethality With Elevated Interferon Beta Responses in Mice.

The disease course of severe enterovirus (EV) infection with hepatic necrosis is usually fulminant and fatal. Coxsackievirus (CVB3) is the emerging and important serotype of newborn EV inducing hepatic necrosis. Calcium (Ca2+) regulates host immunity, type I interferon (IFN-I), and viral infections. The interaction of Ca2+, IFN-I, and CVB3-induced hepatitis remains to be investigated. To address these issues, we used the Ca2+ blocker manidipine, which reduces Ca2+ influx into cells and is used in the clinic, the human hepatoma cell line (HuH7) for in vitro studies, and a murine infection model

Immune checkpoint inhibitors in medulloblastoma: current updates in preclinical and clinical developments.

Medulloblastoma (MB) is the most common pediatric embryonal tumor of the central nervous system. MB grows rapidly in the cerebellum and causes devastating disease in young children. The presence of high-risk molecular or histopathological features is associated with frequent recurrence, resistance to therapy, and poor prognosis, with few experiencing long-term survival. The current standard of care for MB includes surgical resection, radiation, and chemotherapy; however, there remains a need for more specific and effective treatment strategies, especially in recurrent disease. Promising develo

The SDR1-OsDSK2a-EUI1 module orchestrates plant height and multi-stress resilience in rice.

Optimal plant height is critical for yield performance and lodging resistance in rice (Oryza sativa L.). However, genetic resources available for optimizing yield and lodging resistance in breeding programs are currently limited. Here, we report the identification of semi-dwarf rice 1 (SDR1), a semi-dwarf gene encoding a RING-H2 type E3 ubiquitin ligase preferentially expressed in stem internodes. Mechanistically, SDR1 directly targets the key gibberellin (GA)-deactivating enzyme ELONGATED UPPERMOST INTERNODE 1 (EUI1) for degradation via the ubiquitin-proteasome system (UPS), thereby fine-tuni

Neo-Cysteine Molecular Glues for Targeting Mutated SMAD4 Protein.

Neo-protein-protein interactions (neoPPIs), directed by genetic mutation-encoded neo-amino acid residues, represent a promising class of precision medicine targets. Small molecules can mimic genetic mutational effects, creating neo-surfaces and acting as molecular glues to mediate neoPPIs and reprogram biological circuitry. This convergence of genomic alterations and chemical interventions highlights a strategy for targeting disease-associated mutations using neo-amino acid residue-directed molecular glues. Among these, neo-cysteine at the protein-protein interaction (PPI) interface represents
