IgG antibody and delayed-type hypersensitivity responses in nude mice grafted with thymic epithelium.

This study compared the ability of foetal thymic epithelium depleted of lymphocytes and dendritic cells, by low temperature or deoxyguanosine (dGuo) treatment in organ culture, to reconstitute T-cell function in nude mice. It is shown that renal capsule grafts of either type could promote the development of functional T lymphocytes in the periphery, as judged by in vivo assays. Both syngeneic and allogeneic thymic epithelium endowed nude mice with the capacity to mount IgG antibody and delayed-type hypersensitivity (DTH) responses to the T-dependent antigen ovalbumin (OVA). Functional reconsti

Androgen induction of beta-glucuronidase translational yield in submaxillary gland of B6.N mice.

Androgens induce the synthesis of murine beta-glucuronidase (GUS) 10-fold in the submaxillary gland of B6.PAC-Gusn mice without a concomitant increase in GUS mRNA levels. Since the rate of GUS synthesis per mRNA molecule, or translational yield, is a function of both the efficiency with which the message is translated and the fraction of newly made polypeptides that are incorporated into GUS tetramers, we conclude that androgen induces at least one of these components in the submaxillary gland of B6.N mice. Genetic variation in the submaxillary gland induction response was tested for using six

Molecular characterization of beta-globin gene mutations in patients with beta-thalassaemia intermedia in south China.

We have studied the spectrum of mutations producting beta-thalassaemia intermedia in South China. The methods of mutation detection include oligonucleotide analysis, polymerase chain reaction amplification of the beta-globin gene and direct genomic sequencing. The mutations have been identified in 22 beta-globin genes from the patients in 11 unrelated families. Seven different mutations have been identified and the A to G substitution in the TATA box of the beta-globin gene accounts for 42% of these mutant beta-globin genes. Most patients have a beta(+) thalassaemia and one copy of the TATA bo

HLA-linked control of predisposition to lepromatous leprosy.

In a study of the relation between HLA and lepromatous leprosy, HLA haplotype segregation was analyzed in 28 families with multiple cases of different types of leprosy. The inheritance of HLA-DR2, HLA-DR3, and HLA-MT1, which had previously been shown to be associated with susceptibility to leprosy or with a leprosy type, was analyzed separately. Segregation occurred in a significantly nonrandom fashion in both polar tuberculoid leprosy and lepromatous leprosy. This finding indicated HLA-encoded control of a predisposition to both of these forms of the disease. In both cases the segregation obs

Linear IgA dermatosis: a study of ten adult patients.

Ten adult patients with homogeneous-linear deposition of IgA along the basement membrane zone have been studied. The direct immunofluorescence IgA pattern was stable, and there was no deposition of IgG or IgM. The clinical presentations were heterogeneous and resembled dermatitis herpetiformis (DH) (3 cases) or bullous pemphigoid (5 cases). Two patients had widespread gyrate blistering lesions of acute onset. Pruritus was constantly present. The course of the disease was chronic except for one patient who had a spontaneous remission after 5 years. The histology was indistinguishable from "clas

Prolongation of renal allograft survival in DLA tissue-typed beagles after third-party blood transfusions and immunosuppressive treatment.

Significant prolongation of survival of nonrelated DLA-mismatched renal allografts has been obtained in beagle recipients receiving three blood transfusions from nonrelated donors prior to kidney transplantation and immunosuppression after transplantation. Nontransfused DLA-identical or DLA 1 haplotype-different littermates of the transfused dogs were used as controls. Lymphocytotoxic antibodies were formed after the blood transfusions. A quantitative immune reactivity score correlated with graft survival. Low scores prior to transplantation were found in five transfused dogs that did not reje

Recombination, mutation, or constitutive expression at a Gm locus and familial hypergammaglobulinemia.

In a hypercholesterolemic Lebanese family, an uncommon Gm haplotype carrying an unexpected C gamma 1 gene was inherited by only one of 10 siblings. A new recombination during the maternal or paternal meiosis could explain its formation. According to this hypothesis, our data would be informative for the linkage relationship between the gamma-cistrons and the alpha 2-cistron. The latter might be located near the N-terminal side of the gamma-cistron linkage group, and the sequence of genes would be alpha 2, gamma 4, gamma 3, and gamma 1. A mutation could also effect the change from G1m(17) (codo

Multi-environment genome-wide association study reveals genetic loci associated with drought tolerance during wheat germination.

This study used multi-index evaluation and GWAS to identify QTLs and candidate genes for wheat germination drought tolerance, notably a 5D pleiotropic hotspot associated with multiple traits, offering breeding targets. Drought tolerance during wheat germination is critical for uniform seedling establishment in arid and semi-arid regions, ultimately affecting final yield. However, the genetic basis of drought tolerance at this stage remains less understood compared to later growth phases. In this study, a natural population of 389 wheat varieties was evaluated under three independent annual env

Genomic Insights into Winter Wheat Breeding for Severely Cold Climates.

Wheat is one of the world's most important crops, cultivated across diverse ecogeographic zones on more than ~245 million hectares annually. Classified by vernalization requirement into spring, facultative, or winter types, the latter typically achieves higher yields due to its extended growing season, reaching ~18 t ha-1 and 9-10 t ha-1 as a national average for Western European countries such as Germany, France, and England, compared with the global average of barely above 3 t ha-1. Despite this potential, winter wheat is largely confined to regions with relatively mild winters, while vast t

Case Study: Genetic and In Silico Analysis of Familial Pancreatitis.

BACKGROUND/OBJECTIVES: Chronic pancreatitis (CP) is a progressive inflammatory condition of the pancreas that leads to irreversible changes in pancreatic structure. The pancreatic α and β cells secrete hormones such as insulin and glucagon into the bloodstream. The pancreatic acinar cells secrete digestive enzymes that break down macromolecules. When these digestive enzymes do not function properly, maldigestion, malabsorption, and malnutrition may result. Presented here is a case study of an individual newly diagnosed with chronic pancreatitis, along with a genetic analysis of his son and an

Ara h 2105-124-Specific TH2A Cells Drive Peanut Allergy in DRB1*15:01 Individuals: A Detailed Epitope Analysis.

BACKGROUND: The IgE-mediated CD4 T-cell response to peanut (Arachis hypogaea) is heterogeneous, yet TH2 cells remain central drivers of pathology. This study aimed to dissect this complexity at the epitope level by focusing on the HLA-DRB1*15:01-DRB5*01:01 haplotype. Specifically, we examined how distinct epitope-specific T-cell subsets shape the immunological landscape of peanut allergy in peanut-allergic (PA) versus non-peanut-allergic (NPA) individuals. METHODS: Using in vitro and ex vivo MHC-II tetramer approaches, the phenotype, frequency, function, and transcriptome of CD4 T-cell respon

MTNR1B variants increase gestational diabetes mellitus risk in young Chinese pregnant women.

The aim of this study is to explore the relationship between the MTNR1B gene variants rs1387153 and rs10830963 and the risk of gestational diabetes mellitus (GDM). Additionally, the study sought to investigate gene-environment interactions, assess the cumulative genetic risk through the application of Genetic Risk Scores (GRSs), and establish a predictive model for GDM. A case-control study was conducted with 500 GDM patients and 502 controls. MTNR1B gene variants were genotyped using SNPscan™. Associations between clinical data, genetic models, haplotype and GDM risk or blood glucose levels w

Serum xanthine oxidoreductase and oxidative stress are associated with bladder cancer: a case-control study from Jordan.

Xanthine oxidoreductase (XOR) is an oxidant enzyme that exists mainly in two distinct forms: the dehydrogenase form [xanthine dehydrogenase (XDH)] and the oxidized form [xanthine oxidase (XO)]. XO might contribute to tumorigenesis through direct metabolic activation of carcinogens and indirect generation of free radicals. Oxidative stress is one of the leading causes of bladder cancer (BC). Smoking and genetic susceptibility are also linked to oxidative stress and BC. This study investigated the association between XO serum levels and XOR genetic polymorphisms with BC. A case-control study was

Co-segregation of the c.489+3A>G variant with p.Cys1400Ter pathogenic CFTR mutation in Cyprus: prevalence and clinical implications.

BACKGROUND: The high variety of mutations found in the Cystic Fibrosis Transmembrane Regulator (CFTR) gene is responsible for the clinical heterogeneity observed in people with Cystic Fibrosis (CF) and the atypical manifestations in CFTR-related disorders (CFTR-RD). The intronic c.489+3A>G (c.621+3A>G) variant has been reported to have questionable pathogenicity, although its alleged severity was probably due to its co-segregation in cis with another undetected mutation, as previously reported from countries in the Mediterranean region. In the island of Cyprus, several rare CFTR variants have

Genetic structure and geographical distribution of Bithynia siamensis sensu lato from Khong and Mounlapamok districts, Champasak Province, Laos.

BACKGROUND: Bithynia spp., a key intermediate host of Opisthorchis viverrini, is widely distributed in the lower Mekong sub-region, where opisthorchiasis remains a major public health concern. Understanding the genetic diversity and population structure of these snails is crucial for disease control. Bithynia siamensis sensu lato has been classified into three genetic lineages (I-III) based on cytochrome c oxidase subunit 1 (cox1) and 16S ribosomal RNA (16S rRNA) sequence analysis. This study focuses on Champasak Province, Laos, a highly endemic area of opisthorchiasis with limited genetic dat

Genetic variations in the antioxidant genes and their role in modulating susceptibility towards chronic obstructive pulmonary disease in the North Indian population.

BACKGROUND: Chronic Obstructive Pulmonary Disease (COPD) is a persistent inflammatory lung condition characterized by an obstruction in removing oxygen from the lungs. Oxidant and antioxidant imbalance have long been hallmarks of COPD development, where the amount of antioxidants produced is less than that of oxidants. Here, polymorphism in the antioxidant enzymes like Catalase, Superoxide dismutase and Glutathione peroxidase plays an essential role in regulating the levels of oxidants. METHODS: 1000 subjects, including 500 COPD cases and 500 controls, have been recruited and genotyped to ass

Genetic Foundation of Prostaglandin Metabolism Influences Patent Ductus Arteriosus Closure in Extremely Low Birth Weight Infants.

OBJECTIVE: Prostaglandins (PGs) play a major role in maintaining patency of the ductal arteriosus (DA). Pulmonary 15-hydroxyprostaglandin dehydrogenase (PGDH), which is ecoded by the hydroxyprostaglandin dehydrogenase (HPGD) gene, is the primary enzyme responsible for PG breakdown. Animal studies have shown HPGD-knockout mice have significantly higher prostaglandin E2 levels and no ductal remodeling. Functional variants of the HPGD gene that alter PG breakdown have not been studied in preterm infants with patent ductus arteriosus (PDA). STUDY DESIGN: This was an observational cohort study inc
