We investigated the role of prostanoid-mediated pressor mechanisms in setting the level of blood pressure in renin-dependent and renin-independent models of hypertension in unanesthetized rats. Intravenous administration of a blocker of thromboxane A2/prostaglandin endoperoxide receptors, SQ29548 (2 mg/kg bolus injection plus 2 mg/kg/hr for 3 hours), reduced from 162 +/- 4 to 144 +/- 5 mm Hg (p less than 0.05) the blood pressure of rats with aortic coarctation-induced hypertension at 7-14 days after coarctation when plasma renin activity is greatly increased. In contrast, treatment with SQ2954

Bacterial sepsis often precedes the development of the adult respiratory distress syndrome (ARDS) and bacterial endotoxin (LPS) produces a syndrome similar to ARDS when infused into experimental animals. We determined in isolated, buffer-perfused rabbit lungs, free of plasma and circulating blood cells that LPS synergized with platelet activating factor (PAF) to injure the lung. In lungs perfused for 2 h with LPS-free buffer (less than 100 pg/ml), stimulation with 1, 10, or 100 nM PAF produced transient pulmonary hypertension and minimal edema. Lungs perfused for 2 h with buffer containing 100

Current treatments for acute myeloid leukemias (AMLs) cure fewer than 30 % of patients. This low efficacy is due, in part, to the inter-patient and intra-patient heterogeneity of AMLs; accordingly, all current AML treatment regimens involve drug combinations (polypharmacy). A recently-completed clinical trial in relapsed/refractory AML using a combination of two newer targeted antileukemics, the BCL-2 inhibitor venetoclax (VEN) plus the FLT3 inhibitor gilteritinib (GIL), yielded highly promising results for this two-drug polypharmacy combination. Polypharmacology - wherein a single drug molecu

We report a case of a 24-year-old liver transplant recipient who developed hepatic artery thrombosis and graft failure, which was complicated by subphrenic abscess and persistent Klebsiella pneumoniae carbapenemase (KPC)-producing K. pneumoniae bacteremia. Ceftazidime-avibactam treatment led to emergence of resistance, and alternative combination therapy failed due to persistent infection and toxicity. The infection resolved after initiation of meropenem-vaborbactam, which created a bridge to retransplantation. Treatment-emergent ceftazidime-avibactam resistance is increasingly recognized, sug

Functional imaging studies have revealed that certain brainstem areas are activated during migraine attacks. The neuropeptide calcitonin gene-related peptide (CGRP) is associated with activation of the trigeminovascular system and transmission of nociceptive information and plays a key role in migraine pathophysiology. Therefore, to elucidate the role of CGRP, it is critical to identify the regions within the brainstem that process CGRP signaling. In situ hybridization and immunofluorescence were performed to detect mRNA expression and define cellular localization of calcitonin receptor-like r

BACKGROUND: Although international guidelines primarily position the sofosbuvir/velpatasvir/voxilaprevir (SOF/VEL/VOX) combination as a salvage therapy for treatment-experienced patients, specific national reimbursement policies in Türkiye have uniquely positioned this regimen as a first-line treatment for treatment-naive individuals. Because international protocols largely restrict this combination to salvage settings, there remains limited real-world evidence regarding its efficacy and safety as a first-line therapy. This study aims to evaluate the real-world clinical performance of the SOF/

Through the application of TRAP (target-related affinity profiling), we identified a novel class of heteroaroylphenylureas that inhibit human CCL2-induced chemotaxis of monocytes/macrophages both in vitro and in vivo. This inhibition was concentration-dependent and selective with regard to other chemokines. The compounds, however, did not antagonize the binding of (125)I-labeled CCL2 to the CCR2 receptor nor did they block CCR2-mediated signal transduction responses such as calcium mobilization. Optimization of early leads for potency and pharmacokinetic parameters resulted in the identificati

Hematopoietic cell transplantation is a curative therapy for certain hematologic malignancies but is limited in use due to post-transplant complications such as graft-versus-host disease (GVHD), wherein donor cells mount an immune response against host tissues. GVHD is a major cause of post-transplant mortality, inspiring the development of prophylactic strategies to reduce GVHD severity. However, these involve administration of cytotoxic or immunosuppressive compounds into patients, increasing adverse side effects. To address this issue, we have developed a strategy to reduce GVHD through ex

BACKGROUND: Thromboxane A 2 (TXA 2 ), a metabolite of arachidonic acid, is well known for its role in vasoconstriction and platelet aggregation via activation of thromboxane prostanoid receptors (TPR). Although other prostanoid receptors have established roles in pain modulation, the contribution of TXA 2 -TPR signaling to nociceptive processing remains unclear. This study evaluated the analgesic effects of a TPR agonist in mouse models of pain.
METHODS: Four-week-old male ddY mice, an outbred mouse strain, received intrathecal (i.t.) administration of (Z)-7-((1S,2R,3R,4R)-3-((R,E)-3-hydroxy-

BACKGROUND: The pan-AKT inhibitor MK2206 has shown improved but limited efficacy in achieving pathologic complete response (pCR) in patients with breast cancer receiving standard neoadjuvant chemotherapy (NACT). This study aimed to develop non-invasive tumor heterogeneity index (THI) from dynamic-contrast-enhanced magnetic resonance imaging (DCE-MRI) that can better select AKT-inhibition responders than immunohistochemistry (IHC)-based receptors and explore associated tumor microenvironment (TME). PATIENTS AND METHODS: We included 987 patients who underwent surgery following neoadjuvant chemot

We evaluated whether concomitant 5-aminosalicylic acid (5-ASA) influences clinical remission in patients with ulcerative colitis (UC) receiving Janus kinase inhibitors (JAKi). In this retrospective, multicenter cohort study, UC patients receiving tofacitinib (n = 181), upadacitinib (n = 313), or filgotinib (n = 139) were included. Time to clinical remission was analyzed using interval-censored Cox models. Among the 633 patients treated with JAKi, 476 patients received 5-ASA, and 151 did not. Cumulative probability of remission at week 48 was similar with and without 5-ASA, at 81.3% and 77.0%,

Bioelectric signaling is well characterized in neurons and cardiomyocytes but remains largely unexplored in epithelia. Using multielectrode arrays, we demonstrate that localized laser injury to epithelial monolayers (primary human keratinocytes and MDCK cells) triggers voltage spikes in the range of 4 to 12 per min for over 60 min postinjury. These spikes exhibit depolarization, repolarization, and hyperpolarization phases lasting 1 to 2 s, a timescale three orders of magnitude slower than neuronal action potentials. Spike amplitudes and frequencies detected at 740 μm from the injury site (the

Spinal cord injury (SCI) is accompanied by a significant microglia-associated inflammatory response that is associated with secondary tissue damage and poorer functional outcomes. Serum and glucocorticoid-regulated kinase 1 (SGK1) has been implicated in the regulation of cell survival and neuronal excitability in various diseases. However, the role and cell-specific mechanism of SGK1 in SCI remain to be elucidated. In this study, we observed that SGK1 was predominantly expressed in microglia located at the lesion margin during the early phase of SCI in a mouse contusion model. Inhibition of SG
