Cryptococcus neoformans virulence is enhanced after growth in the genetically malleable host Dictyostelium discoideum.

Cryptococcus neoformans is an encapsulated, environmental fungus that can cause life-threatening meningitis. Pathogenicity of C. neoformans for macrophages and vertebrate hosts may be a mechanism selected in evolution for protection against environmental predators. In this study, we investigated whether Dictyostelium discoideum could serve as an alternate host for C. neoformans. D. discoideum has a defined genetic system which provides significant advantages for the study of fungus-amoeba interactions. Our results show that D. discoideum is susceptible to infection with C. neoformans and that

Helicobacter pylori In Free-Living Amoebae: A Potential Environmental Reservoir for Persistence and Transmission.

Helicobacter pylori (H. pylori) is a highly adapted gastric pathogen responsible for one of the most prevalent chronic bacterial infections worldwide and a major cause of gastritis, peptic-ulcer disease, and gastric cancer. Traditionally considered predominantly a human-associated microorganism transmitted via oral-faecal routes, growing evidence challenges this paradigm by demonstrating its persistence outside the human host through interactions with free-living amoebae (FLA). This review critically examines current knowledge of the biological and ecological interplay between H. pylori and FL

Multi-target inhibitors of white spot syndrome virus envelope proteins (VP28, VP26, and VP24) to protect shrimp (Penaeus monodon): An integrated virtual screening, pharmacokinetic, and molecular dynamics study.

White Spot Syndrome Virus (WSSV) remains the most devastating pathogen in shrimp aquaculture worldwide, causing annual economic losses exceeding US$3 billion. To date, no effective prophylactic or therapeutic agents currently exist to mitigate the rapid, high-mortality outbreaks associated with WSSV. So, we employed an in silico framework, utilizing ClusPro 2.0, to map interactions between Penaeus monodon (host) receptors (Rab7, C-type lectin, GLUT-1, and β integrin) and the viral envelope proteins VP28, VP26, and VP24. The result of the protein-protein interactions guided to develop a multi-t

Oral fungal microbiome in gout: composition, cross-kingdom interactions, and effects of intervention.

BACKGROUND: Oral fungi, despite their low abundance, may influence microbial interactions, host responses, and disease progression; however, their role in gout remains poorly understood. MATERIALS AND METHODS: Between March and December 2024, patients with chronic gouty arthritis (CGA), acute gouty arthritis (AGA), and age- and sex-matched healthy controls (HC) were enrolled. Unstimulated saliva underwent ITS2 sequencing. Analyses included α/β-diversity, differential abundance, and genus-level co-occurrence networks. Cross-kingdom associations were assessed using partial Spearman correlation

Potential of brain mast cells for therapeutic application in the immune response to bacterial and viral infections.

A wide range of microorganisms can infect the central nervous system (CNS). The immune response of the CNS provides limited protection against microbes penetrating the blood-brain barrier. This results in a neurological deficit and sometimes leads to high morbidity and mortality rates despite advanced therapies. For the last two decades, different studies have expanded our understanding of the molecular basis of human neuroinfectious diseases, especially concerning the contributions of mast cell interactions with other central nervous system compartments. Brain mast cells are multifunctional c

Interaction of primary human endometrial cells with Neisseria gonorrhoeae expressing green fluorescent protein.

Infection of the endometrium by Neisseria gonorrhoeae is a pivotal stage in the development of pelvic inflammatory disease in women. An ex vivo model of cultures of primary human endometrial cells was developed to study gonococcal-host cell interactions. To facilitate these studies, gonococci were transformed with a hybrid shuttle vector containing the gfp gene from Aequoria victoria, encoding the green fluorescent protein (GFP), to produce intrinsically fluorescent bacteria. The model demonstrated that both pili and Opa proteins were important for both mediating gonococcal interactions with e

Tick-borne disease risk in a forest food web.

Changes to the community ecology of hosts for zoonotic pathogens, particularly rodents, are likely to influence the emergence and prevalence of zoonotic diseases worldwide. However, the complex interactions between abiotic factors, pathogens, vectors, hosts, and both food resources and predators of hosts are difficult to disentangle. Here we (1) use 19 yr of data from six large field plots in southeastern New York to compare the effects of hypothesized drivers of interannual variation in Lyme disease risk, including the abundance of acorns, rodents, and deer, as well as a series of climate var

Changes in the prevalence of three disease-causing pathogens in bird-borne blacklegged ticks: Borrelia burgdorferi sensu stricto, Anaplasma phagocytophilum, and Babesia microti.

Borrelia burgdorferi sensu stricto, Anaplasma phagocytophilum, and Babesia microti are common tick-borne pathogens of medical and veterinary concern in the United States and are transmitted by the blacklegged tick (Ixodes scapularis). The range expansion of ticks and their pathogens depends on the movements of vertebrate hosts, including birds. Flight grants birds high mobility - giving them the potential to rapidly expand the range of ticks and their pathogens, thus impacting human, wildlife, and livestock risk of exposure to tick-borne diseases. We examined the prevalence of B. burgdorferi s

Post-splenectomy antibiotic prophylaxis--unfinished story: to treat or not to treat?

Overwhelming infection in asplenic patients is a well documented occurrence in the literature. The introduction of immunization with polyvalent pneumococcal, Hemophilus Influenza and meningococcal vaccines significantly cut down the incidence of post-splenectomy sepsis and mortality. However, the issue of prophylactic antibiotic therapy in these patients remains inconclusive. There are contradictory reports bouncing between life-long treatment to no treatment. There are also more flexible approaches emerging from patient difficulties complying with a prolonged therapy. This debate calls for de

Exploring Bacteroidota strains from human stool and animal feces: phenotyping and functional diversity.

Bacteroidota, a diverse phylum of bacteria, includes classes whose members are increasingly recognized for their significant contributions to host health, particularly through their antimicrobial properties. This study investigates the functional diversity of 42 new Bacteroidia and Sphingobacteriia strains enriched and identified from diverse hosts, including mouse ceca and human stool samples. Using 16S rRNA gene sequencing, we phylogenetically characterized the strains of the genera Bacteroides, Phocaeicola, and Sphingobacterium and assessed their functional properties related to potential b

Localizing the Glycan Binding Sites of SARS-CoV-2 Receptor-Binding Domain.

Mapping the binding sites on viral proteins that interact with host glycans is essential for understanding host-pathogen interactions and guiding therapeutic design. While the SARS-CoV-2 spike (S) protein is known to bind host glycans through its N-terminal domain and receptor-binding domain (RBD), the specific glycan-interacting residues on the RBD remain undefined. Here, we introduce antibody masking-native mass spectrometry (AM-nMS), a spatially resolved assay for mapping glycan-binding sites by combining a panel of nanobodies targeting distinct regions within the RBD with center-of-mass mo

Assembly by disassembly of the gut microbiota in Riptortus pedestris and other stinkbugs.

The gut is one of the largest internal organs in animals and is central to nutrition. Its need to uptake food from the environment implies it is open at both ends, exposing its lumen to the outside world and to colonization by ingested microbial communities. As the primary site of host-associated microbiota, the gut has, over evolutionary time, fostered stable associations with specific microbes to which the host has delegated key biological functions. Insect gut microbiota is of interest both for its impact on this ecologically important animal group and as a simplified model for understandin

An integrated transcriptomics and proteomics analysis of the secretome of the helminth pathogen Fasciola hepatica: proteins associated with invasion and infection of the mammalian host.

To infect their mammalian hosts, Fasciola hepatica larvae must penetrate and traverse the intestinal wall of the duodenum, move through the peritoneum, and penetrate the liver. After migrating through and feeding on the liver, causing extensive tissue damage, the parasites move to their final niche in the bile ducts where they mature and produce eggs. Here we integrated a transcriptomics and proteomics approach to profile Fasciola secretory proteins that are involved in host-pathogen interactions and to correlate changes in their expression with the migration of the parasite. Prediction of F.

Insights into the pathogenic mechanisms associated with the SARS-CoV-2 spike protein.

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), the pathogenic agent responsible for the coronavirus disease 2019 (COVID-19) pandemic, uses the trimeric spike protein to gain entry into the host cell. Structural studies have revealed that the spike protein is comprised of the S1 and S2 subunits. The S1 subunit of the spike protein contains the receptor-binding domain (RBD), which binds to the human angiotensin-converting enzyme 2 (ACE2) receptor. The interaction between the RBD and ACE2 facilitates membrane fusion and host cell infection. The SARS-CoV-2 spike protein also contain

Enterohemorrhagic Escherichia coli O157:H7 Infection Inhibits Host Endoplasmic Reticulum Stress in Intestinal Epithelial Cells via the PERK Pathway.

Enterohemorrhagic Escherichia coli (EHEC) O157:H7 is a foodborne pathogen that causes a variety of diseases, ranging from self-limiting gastroenteritis to life-threatening extra-intestinal diseases such as hemolytic uremic syndrome. EspF, an effector protein secreted by the type III secretion system of EHEC, is primarily responsible for the development of inflammatory colitis. Our previous study revealed that EspF interacts with the host Annexin A6 (ANXA6) protein and targets the endoplasmic reticulum (ER). Given the critical effects of ER stress on the host responses of gastroenteritis, we ex

Mechanisms Underlying Pneumococcal Transmission and Factors Influencing Host-Pneumococcus Interaction: A Review.

Streptococcus pneumoniae (also called pneumococcus) is not only a commensal that frequently colonizes the human upper respiratory tract but also a pathogen that causes pneumonia, sepsis, and meningitis. The mechanism of pneumococcal infection has been extensively studied, but the process of transmission has not been fully elucidated because of the lack of tractable animal models. Novel animal models of transmission have enabled further progress in investigating pneumococcal transmission mechanisms including the processes such as pneumococcal shedding, survival in the external environment, and

Pathogenesis of infection with Bordetella pertussis in hamster tracheal organ culture.

Hamster tracheal organ culture was employed as a model for study of the pathogenesis of infection due to Bordetella pertussis. Infected tracheal explants were examined with light, immunofluorescence, and electrom microscopy. B. pertussis organisms preferentially attached to the ciliated cells, producing ciliostasis and marked destruction of the subcellular organelles followed by expulsion of these cells from the epithelial layer. Other nonciliated respiratory epithelial cells appeared to be unaffected. Metabolic studies on infected tracheal cultrues indicated that significant deficiencies in s

Fungi in the Gut Microbiota: Interactions, Homeostasis, and Host Physiology.

The mammalian gastrointestinal tract is a stage for dynamic inter-kingdom interactions among bacteria, fungi, viruses, and protozoa, which collectively shape the gut micro-ecology and influence host physiology. Despite being a modest fraction, the fungal community, also referred to as mycobiota, represents a critical component of the gut microbiota. Emerging evidence suggests that fungi act as early colonizers of the intestine, exerting a lasting influence on gut development. Meanwhile, the composition of the mycobiota is influenced by multiple factors, with diet, nutrition, drug use (e.g., an

Antibiotic Activity Altered by Competitive Interactions Between Two Coral Reef-Associated Bacteria.

Microbes produce natural products that mediate interactions with each other and with their environments, representing a potential source of antibiotics for human use. The biosynthesis of some antibiotics whose constitutive production otherwise remains low has been shown to be induced by competing microbes. Competition among macroorganism hosts may further influence the metabolic outputs of members of their microbiomes, especially near host surfaces where hosts and microbial symbionts come into close contact. At multiple field sites in Fiji, we collected matched samples of corals and algae that

Interactions of Muscovy duck reovirus, gut microbiota, and host innate immunity: Transcriptome and gut microbiota analysis.

It has been shown that Muscovy duck reovirus (MDRV) infection causes severe intestinal barrier damage and intestinal mucosal immune suppression. The health and balance of gut microbes is essential for the progression of intestinal infectious diseases. To investigate the interaction of MDRV, intestinal bacteria with host intestinal innate immunity, an MDRV contact-infection model was established in this study. High-throughput sequencing technology was used to sequence 16S rDNA and transcripts in ileal samples from experimental Muscovy ducklings. Our results suggest that intestinal opportunistic
