Sorbitol dehydrogenase aids grain-fill and integrates carbohydrate metabolism with redox in the hypoxic maize endosperm.

Extreme low oxygen is typical of microenvironments inside developing grains, thus requiring metabolic adaptations to hypoxia. Potential contributions from sorbitol dehydrogenase (SDH) are tested here for maize (Zea mays) since the reversible reaction (fructose + NADH ↔ sorbitol + NAD+) can balance both redox state and metabolism during assimilate import into kernels. To do so, we developed and analyzed maize sdh1 mutants and over-expression lines. Dysfunction of Sdh1 decreased seed weight by 17%, impaired starch accumulation, and enhanced levels of glucose, fructose, and sucrose. Redox balance

Odorant-binding proteins and chemosensory proteins in insects: structural insights, functional plasticity, and prospects for targeted pest management.

Odorant-binding proteins (OBPs) and chemosensory proteins (CSPs) are integral to the chemical communication systems of insects, mediating the detection, transport, and discrimination of hydrophobic ligands such as pheromones, odorants, and xenobiotics. While OBPs are largely confined to olfactory tissues and exhibit high structural specificity-with six conserved cysteines forming three disulfide bridges-CSPs are structurally simpler, featuring four conserved cysteines and two disulfide bridges, yet display remarkable conformational plasticity and functional versatility. Recent evidence undersc

Programming Failure Mode Transitions in Polyurea-Reinforced 3D-Printed ABS and PA-GF Cellular Metamaterial Composites.

Additively manufactured cellular architectures frequently exhibit brittle failure under impact due to layer-induced stress concentrations. Through the programming of architectural and material design, specifically combining Fused Deposition Modeling (FDM) lattice topology with hyperelastic polyurea infiltration, this study achieves active control over the macroscopic transition from catastrophic structural fragmentation to stable progressive collapse. To evaluate this, auxetic and honeycomb specimens printed with ABS and glass-fiber-reinforced polyamide (PA-GF) were evaluated in unreinforced a

Ultra-high-field solid-state NMR of intact Saccharomyces cerevisiae cells at 1.2 GHz.

The increasing availability of ultra-high-field NMR spectrometers is opening new opportunities for the characterization of complex and structurally heterogeneous biological assemblies by solid-state NMR, including lipid membranes, cell walls, and even intact cells. Here, we investigate the use of a 1.2 GHz (28.2 T) spectrometer, the highest magnetic field currently commercially available, for the analysis of intact Saccharomyces cerevisiae cells. Budding yeast is an experimentally tractable eukaryotic model organism that is widely used in biotechnology and industrial fermentation. Characteriza

Immune-neural crosstalk in chronic low back pain: from intervertebral disc degeneration to pain sensitization.

Chronic low back pain (CLBP) is a leading cause of disability, yet radiographic degeneration correlates poorly with symptoms. This mini-review examines how immune-neural crosstalk may link intervertebral disc degeneration to pain initiation and persistence. Degenerated nucleus pulposus and annulus fibrosus cells develop inflammatory and catabolic phenotypes and release mediators that may promote neural and vascular ingrowth, sensitize nociceptors, and increase dorsal root ganglion neuron excitability. Sustained peripheral input may engage spinal glial pathways and contribute to central sensiti

A multi-targeted therapeutic strategy for triple-negative breast cancer: the tumor-targeting peptide p28 enhances chemotherapy, induces apoptosis, crosses the blood-brain barrier, and suppresses metastasis.

Triple-negative breast cancer (TNBC) is an aggressive and molecularly heterogeneous subtype of breast cancer characterized by limited targeted treatment options, frequent chemoresistance, and a strong propensity for metastasis, particularly to the central nervous system (CNS). These challenges highlight the need for multi-targeted therapeutic strategies. The tumor-targeting peptide p28, which exhibits antitumor activity and potential blood-brain barrier (BBB) penetration, represents a promising candidate for enhancing therapeutic efficacy in TNBC through distinct mechanisms of action. In this

Menstrual cycle patterns in primary care: a structured approach using primary ovarian insufficiency as a sentinel condition.

Primary Ovarian Insufficiency, a condition characterized by early loss of ovarian function, illustrates the consequences of failing to act on menstrual cycle changes as early clinical signals. Menstrual disturbance is often the first presenting feature, yet diagnosis is frequently delayed despite repeated clinical encounters. For primary care clinicians, a disrupted menstrual pattern in an adolescent or young adult should prompt consideration of whether Primary Ovarian Insufficiency could be present in an early stage. This Perspective presents a structured approach to interpreting menstrual cy

Family polyvictimization in households with parental health conditions: a cross-sectional study of disparities and risk factors.

BACKGROUND: Families with parents with health conditions often experience socioeconomic hardship, caregiving strain, and elevated exposure to family violence. However, differences in family polyvictimization (FPV) between these families and those without health conditions remain underexplored. This study examined disparities in FPV exposure and associated family-level risk factors. METHODS: A cross-sectional household survey was conducted with 1,400 Hong Kong families with children under 18. Measures included FPV (FPV Screen), social support (MSPSS), family hierarchy (PRP), health (SF-12v2, P

Grape pomace-derived pectin complex as a sole oleogelator: thermally induced structural reinforcement and application in pound cakes.

Oleogelation has emerged as a promising strategy for structuring unsaturated oils to address the conflict between food texture and health concerns. However, most oleogelators used in indirect methods rely on complicated pre-processing and lack natural sources. In this study, a pectin-protein-polyphenol complex derived from white grape pomace was investigated as a sole oleogelator to structure canola oil via an emulsion-templated approach. High oil contents of 94.9-97.9% (w/w) were achieved at pectin-to-oil ratios of 2:100-5:100 (w/v), with liquid oil effectively entrapped within the pectin com

Advances in ecotoxicological research using planarians: a comprehensive review.

Planarians, distinguished by their extraordinary regenerative capacity and ecological role as benthic predators, have emerged as a transformative model in ecotoxicology. This review provides a comprehensive synthesis of research on the effects of diverse classes of pollutants, including metals, pesticides, pharmaceuticals, personal care products, nanomaterials, and a dedicated category of "other pollutants," namely biotoxins and physical stressors, on these organisms. We introduce a novel conceptual framework of "regenerative toxicology," which focuses on how contaminants impair an organism's

Molecular and toxicogenetic safety profile of short-term nitrous oxide anesthesia in surgical patients.

BACKGROUND: Nitrous oxide (N2O) is a widely used anesthetic; however, its potential to compromise genomic stability by disrupting vitamin B12 and folate metabolism remains a critical concern in clinical molecular biology. Since N2O can oxidize cobalamin, it may theoretically impair the methionine synthase pathway, leading to DNA damage. Thus, this study aimed to evaluate the toxicogenetic impact-DNA damage and gene expression-and the metabolic profile of vitamins B9 and B12, homocysteine (HCY) and micronutrients/antioxidants in patients undergoing short-term N2O anesthesia. METHODS AND RESULT

Loss of epigenetic adaptation to a high-fat diet in alpha-synuclein transgenic mice.

INTRODUCTION: In humans, a high-fat diet and obesity are associated with a higher risk and accelerated progression of Parkinson's disease (PD). Similarly, in animal models a high-fat diet exacerbates PD-related phenotypes, including dopaminergic neurodegeneration, and alpha-synuclein aggregation. We previously demonstrated that transgenic mice overexpressing human, mutated A30P alpha-synuclein failed to transcriptionally adapt to metabolic stress which could be a potential explanation for the high-fat diet-dependent aggravation of PD pathology. However, the underlying epigenetic mechanisms tha

Obesity and Metabolic Dysfunction-Associated Steatotic Liver Disease (MASLD): A Literature Review on Pathophysiology and Treatment.

Metabolic dysfunction-associated steatotic liver disease (MASLD) represents a key hepatic manifestation of obesity and systemic metabolic dysfunction and is a leading cause of chronic liver disease worldwide. Obesity-driven mechanisms, including adipose tissue dysfunction, insulin resistance and increased free fatty acid flux to the liver, promote ectopic lipid accumulation and lipotoxic injury. These processes trigger mitochondrial and endoplasmic reticulum stress, inflammation, immune activation and disturbances in the gut-liver axis, driving disease progression from simple steatosis to meta

Genetic regulatory axis between AGR2 and ESR1 promotes breast cancer progression.

Anterior gradient protein 2 (AGR2), a member of the protein disulfide isomerase family, plays a critical role in endoplasmic reticulum proteostasis and has been implicated in breast cancer progression. However, the downstream regulatory programs and signaling pathways governed by AGR2 remain incompletely defined. Here, we employed CRISPR-Cas9-mediated knockout of AGR2 in breast cancer cells to systematically investigate the functional and transcriptional consequences of AGR2 loss. AGR2 depletion resulted in significant suppression of cell migration, invasion, and chemoresistance. Unbiased tran

Insights into Fe0, Fe2O3, and Fe3O4-Mediated reduction of antibiotic resistance genes and horizontal gene transfer via reactive oxygen species during composting.

Reactive oxygen species (ROS) generated by Fe-based additives mitigate antibiotic resistance during composting, yet the associated patterns remain unclear. The objective of this study was to examine the specific roles of Fe0, Fe2O3, and Fe3O4 in ROS-mediated reduction of antibiotic resistance genes (ARGs) and associated host bacteria during composting of chicken manure (CM) and wine grape pomace (WGP). The results showed that Fe0, Fe2O3, and Fe3O4 achieved ARG attenuation efficiencies of 81%, 92%, and 83%, respectively, representing a 52-63% increase over the control (29%). Notably, Segmented

Why glycemic control remains suboptimal in type 1 diabetes in the era of advanced technologies?

Type 1 diabetes (T1D) remains associated with suboptimal glycemic control in a substantial proportion of individuals despite major advances in insulin formulations and technological systems. This apparent paradox highlights that glucose control in T1D is not determined by technology alone, but rather emerges from the interaction of multiple biological, therapeutic, behavioral, and psychosocial factors. In this structured narrative review, we examine the main determinants that continue to limit optimal control in real-world practice. Pathophysiological barriers include residual or absolute insu

One-pot pyrene-functionalized UiO-66-NH2 enables pH-Gated speciation of serum copper: Exchangeable versus total copper.

Metal ion dysregulation, particularly involving copper, has been implicated in neuropsychiatric disorders including depression. Herein, we report a pyrene-functionalized UiO-66 platform (UiO-66-NH-PCA) prepared by a facile one-pot solvothermal strategy that avoids complicated pyrene-ligand presynthesis and tedious post-synthetic modification. Spectroscopic analyses supported amidation-consistent chemical anchoring of the pyrene unit onto UiO-66-NH2, affording a fluorescent MOF that retained its framework integrity under acidic conditions. UiO-66-NH-PCA acted as a turn-off fluorescence probe fo

Lysine Carboxymethyl Cysteinate (LCC) Protects the Epidermis from UVB-Induced Barrier Damage Through the Activation of Autophagy.

Lysine carboxymethyl cysteinate (LCC) has been identified as a glutathione (GSH) precursor for the use of cosmetic products, providing a defense against oxidative stress by elevating GSH levels, and mitigating UVB-induced pigmentation and barrier disruption. In this study, the protective efficacy of LCC on epidermal barrier integrity under UVB irradiation was systematically evaluated and its underlying mechanisms were investigated. Results from the UVB-exposed 3D living skin equivalent model (LSE) indicated that LCC effectively restored UVB-induced reductions in epidermal living cell thickness

Responsive nanoparticles modulating microglia lactate transport alleviate M1-type polarization and neuroinflammation for brain injury therapy.

Neuroinflammation mediated by microglia and excessive oxidative stress are key pathological processes driving the progression of brain injuries such as intracerebral hemorrhage (ICH) and traumatic brain injury (TBI). Modulating lactate transport has emerged as a promising approach to mitigate M1 microglia polarization and alleviate neuroinflammation. Liposomal nanoparticles provide a safe and efficient platform for drug delivery. Here, we developed reactive oxygen species (ROS) responsive MiRCM nanoparticles that co-deliver monocarboxylate transporter 1 (MCT1) inhibitor AR-C155858 and a ROS-sc
