Interleukin 2- and interferon-production and expression of interleukin 2 receptors by human mononuclear cells stimulated with mumps virus or phytohaemagglutinin.

Interleukin 2 (IL 2) production, expression of IL 2-receptors and interferon (IFN) production in vitro by human mononuclear cells stimulated with inactivated mumps-virus antigen and phytohaemagglutinin (PHA, a T lymphocyte mitogen) were characterized. The detectable level of the T-cell lymphokine IL 2 in culture supernatants peaked after 18-24 hours for PHA stimulation and after 48 hours for mumps stimulation. The expression of IL 2 receptors was maximal on day 3 of PHA stimulation and days 5-6 of mumps stimulation. The IFN-level in culture supernatants showed only small variations during the

Treatment of common and plantar viral warts with human lymphoblastoid interferon-alpha--pilot studies with intralesional, intramuscular and dermojet injections.

Pilot studies with human lymphoblastoid interferon-alpha(Wellferon), utilizing the intralesional, intramuscular and dermojet routes, have been performed in 39 patients with common and plantar viral warts resistant to conventional therapy. Clinical efficacy, local and systemic toxicity, and effects on haematological and clinical chemistry parameters has been measured. Our results indicate that the intralesional route is the most favourable as it combines an acceptable therapeutic ratio with a procedure that may be performed quickly and with no prior preparation of the lesion.

Genetics of resistance to the African trypanosomes. V. Qualitative and quantitative differences in interferon production among susceptible and resistant mouse strains.

The induction of interferon (IFN) was examined in different inbred mouse strains infected with Trypanosoma brucei rhodesiense. Relatively susceptible C3HeB/FeJ mice that do not exhibit variant-specific immunity or control parasitemia did not exhibit detectable IFN throughout the infection. Relatively resistant B10.BR mice that exhibit variant-specific immunity and control the first peak of parasitemia exhibited detectable IFN at two intervals. The appearance of IFN in B10.BR serum first coincided with the onset of the parasitemia 4 days after infection and then disappeared; this IFN peak was p

Effect of mouse lymphokines and cloned mouse interferon-gamma on the interaction of Rickettsia prowazekii with mouse macrophage-like RAW264.7 cells.

We studied the effects of crude mouse lymphokines and cloned mouse interferon-gamma on the interaction of Rickettsia prowazekii with mouse macrophage-like RAW264.7 cells. Treatment of RAW264.7 cells with lymphokines before infection, after infection, or both before and after infection with R. prowazekii led to killing of a substantial proportion of the RAW264.7 cells. Such cytotoxicity required both lymphokines and viable R. prowazekii and did not occur in mouse fibroblastic L929 cells. Untreated cultures of RAW264.7 cells supported good growth of the Breinl strain of R. prowazekii, but in lym

Antiviral treatment of chronic hepatitis B virus infection: pharmacokinetics and side effects of interferon and adenine arabinoside alone and in combination.

In an uncontrolled trial, 29 patients with chronic hepatitis B virus infection were treated with 93 courses of adenine arabinoside at doses ranging from 2.5 to 15 mg/kg per day. Most patients were treated concomitantly with human leukocyte interferon. Significant, but transient, neurotoxicity was seen with adenine arabinoside therapy in 44% of all courses. Manifestations of toxicity were mainly neurological and ranged from pain syndromes to tremors and, rarely, seizures. Suppression of numbers of lymphocytes was also noted. All effects were reversible with time. The extent of toxicity was depe

Defective immune interferon production and natural killer activity associated with poor neutrophil mobility and delayed umbilical cord separation.

Two infants with recurrent infections and a history of delay in separation of the umbilical cord (1 month and 17 days) had severely impaired neutrophil mobility. In addition very poor natural killer cell (NK) activity of blood lymphocytes against a leukaemia cell line (Molt 4F) was found. Incubation of lymphocytes with lymphoblastoid interferon increased NK activity in the one case tested. No immune (gamma) interferon production was detected in Raji cell and phytohaemagglutinin (PHA) stimulated cultures from the other case. Apart from an abnormal dose-response curve in thymidine uptake after P

Persistent Epstein-Barr virus infection in a child with hypergammaglobulinaemia and immunoblastic proliferation associated with a selective defect in immune interferon secretion.

A 5-year-old girl had a chronic disease characterised by fever, lymphoid hyperplasia, interstitial pneumonitis, thrombocytopenia, and polyclonal hypergammaglobulinaemia. Evidence for severe, persistent Epstein-Barr virus (E.B.V.) infection was found: titres of antibody to E.B.V. viral capsid antigen (IgM and IgG) and early antigen were extremely high, cells containing E.B.V.-associated nuclear antigen (E.B.N.A.) were found in lymph nodes and blood, and spontaneous permanent lymphoblastoid cell lines were established from both sources over a period of a year. After exacerbation of the polyclona

Tuberculosis as an unconventional interferonopathy.

Tuberculosis is caused by Mycobacterium tuberculosis, a bacterium that accounts for more human mortality than any other. Evidence is accumulating for the view that tuberculosis is an interferonopathy - a disease driven by type I interferons. However, how type I interferons exacerbate tuberculosis remains poorly understood. As an infection, tuberculosis is distinct from conventional interferonopathies, which are autoinflammatory diseases. Here I consider the hypothesis that type I interferons promote bacterial replication by impairing key antibacterial immune responses, including those orchestr

[Progress in vaccination against AIDS].

Two vaccination trials against AIDS viruses are reported. The first trial was a prophylactic vaccination carried out in the pig-tailed macaque (Macacca nemestrina) against simian immunodeficiency virus (SIV) variant PBj14. The immunogen was a Semliki forest virus (SFV) recombinant expressing SFV - PBj14 envelope protein. Vaccination did not prevent infection but protected the animals against the disease (a fulminant form of AIDS that kills the animal within 12-15 days). The second trial was a therapeutic vaccination carried out with HIV-I-infected hemophilic patients. The immunogen was a toxoï

Modulatory effects of interferon-γ and interleukin-13 on tumor necrosis factor-α-induced chemokine gene expression in canine keratinocytes.

Keratinocytes play a crucial role in the pathogenesis of canine atopic dermatitis (cAD). Tumor necrosis factor (TNF)-α, a proinflammatory cytokine, is highly expressed in the lesional skin of dogs with atopic dermatitis. Additionally, interferon (IFN)-γ, a T-helper (Th)1-type cytokine, and interleukin (IL)-13, a Th2-type cytokine, are also detected in the lesional skin during the acute and/or chronic phases of cAD. However, their effects on chemokine expression in canine keratinocytes remain unclear. This study investigated the effects of TNF-α on chemokine mRNA expression in canine keratinocy

Pathway-level transcriptional convergence between Gaucher disease and common variable immunodeficiency reveals concordant interferon and antigen-processing signatures.

BACKGROUND: Gaucher disease (GD), an inborn error of metabolism, and common variable immunodeficiency (CVID), an inborn error of immunity, have distinct etiologies but may exhibit convergent downstream transcriptional responses. We investigated transcriptional robustness and pathway-level convergence using a robustness-oriented transcriptomic framework. METHODS: Public datasets were analyzed independently for GD and CVID. Patient-level GD fibroblast profiles (GSE124283) underwent differential-expression analysis, leave-one-patient-out (LOPO) testing, expression-scale sensitivity analysis, and

Keratinocyte VISTA attenuates UV light-induced skin injury by suppressing cutaneous type I interferon (IFN-I) response.

Persistent production of type I interferons (IFN-Is) is a hallmark of cutaneous lupus erythematosus (CLE). Ultraviolet (UV) light stimulates IFN-I response in the skin and exacerbates CLE. Here, we identify V-type immunoglobulin domain-containing suppressor of T cell activation (VISTA) as a negative regulator of both basal and UV-induced IFN-I response in the skin and show that VISTA limits skin photosensitivity in an IFN-I-dependent manner, in part through Stimulator of Interferon Genes (STING). Furthermore, we demonstrate a novel role for VISTA in keratinocytes both at steady state and in re

Uncoupling Type I Interferon Benefits From Inflammatory Toxicity: Transformer-Prioritized Precision Agonists for Potent and Safer Cancer Immunotherapy.

Paclitaxel (PTX) chemotherapy is constrained by an "immunomodulatory paradox," where antitumor Type I Interferon (IFN-I) activation is coupled with detrimental pro-inflammatory cascades. To address this challenge, we developed Deep Learning for Innate Immunity Modulatory Potential (DLINP), a Transformer-based framework designed to identify precision immunomodulators that uncouple IFN-I induction from deleterious inflammatory signaling. Screening 123 million entities identified Co68-an organometallic PNP-pincer complex-as a dual-functional agent with superior potency to conventional taxanes. In

Bilateral otomastoiditis caused by Cryptococcus neoformans, a rare manifestation in patient with adult-onset immunodeficiency due to anti-interferon-γ autoantibodies.

Cryptococcus is an opportunistic fungal pathogen found in immunocompromised patient such as HIV or transplant recipients. Infection of central nervous system is the most common manifestation but pulmonary and skeletal involvement have been reported in non-HIV population. We report a case of cryptococcal otomastoiditis which was extremely rare and majority of cases were reported in individual without comorbidity. In this case, we also identified adult-onset immunodeficiency associated with interferon-gamma autoantibodies in this patient as an important underlying which predisposed cryptococcal

AI-Assisted Computed Structure Models for Pre-Ubiquitylation Complexes Assembled by Respiratory Syncytial Viral Suppressors of Cellular Interferon Response.

Multiple viruses suppress the antiviral defense system of the host for optimal growth and pathogenesis by co-opting the ubiquitin-mediated proteasomal system (UPS) that promotes the degradation of cellular substrates belonging to the interferon pathway. In the Orthopneumovirus genus, respiratory syncytial virus (RSV), a significant pathogen in human and other animals, employs a pair of viral nonstructural proteins (NS1, NS2) to assemble the UPS. The lack of experimental three-dimensional structures of the substrate proteins and the NS-assembled UPS has impeded progress in our understanding of

The Effect of Interferon Type I Adjuvant Therapy on the Lifespan and Complications of Glioma Patients Undergoing Chemotherapy: A Systematic Review and Meta-Analysis.

BACKGROUND: The malignant glioma, as the most common and aggressive primary brain and spinal cord neoplasm, has shown limited responsiveness to available treatments, including tumor dissection, radiation, and chemotherapy. Thus, interferon type I, as a supplemental therapy, is added to the main therapies to overcome neoplasm resistance and prolong the patients' lifespan. METHODS: To clarify the effects of interferon adjuvant therapy on the lifespan and complications of glioma patients, we conducted a systematic review and meta-analysis by searching valid databases, scanning, and screening ful
