Computational discovery and validation of 4'-O-methylochnaflavone as a novel HSP90AB1 inhibitor for hepatocellular carcinoma treatment.

Hepatocellular carcinoma (HCC) is a global health issue with limited therapeutic efficacy. Heat shock proteins (HSPs) could enhance adaptation to the oxygen-deprived tumor microenvironment (TME), leading to chemotherapy resistance. Regulating HSP to improve the immune microenvironment may enhance tumor treatment efficacy. However, pharmacological agents targeting HSPs to reverse the immunosuppressive TME remain scarce. In this study, we integrated bioinformatic analysis, virtual screening, molecular docking, molecular dynamics (MD) simulations, and experimental validation to identify therapeut

Multi-Halogenated Indoles as Antimicrobial and Antivirulence Agents Against Drug-Resistant Staphylococcus aureus.

The emergence of multidrug-resistant Staphylococcus aureus requires the development of novel agents that can target both planktonic cells and persistent biofilms. In this study, a library of multi-halogenated indoles was evaluated for antibacterial, antibiofilm and antivirulence activities against S. aureus including methicillin-resistant strains. Two lead compounds, 6-bromo-4-iodoindole and 4-bromo-6-chloroindole, exhibited potent bactericidal activity (MIC = 20-30 μg/mL) comparable to the antibiotic gentamicin, effectively inhibited biofilm formation and persister formation and suppressed ke

Prevalence of temporomandibular disorder in irritable bowel syndrome (IBS) patients: a systematic review.

BACKGROUND: Temporomandibular disorders (TMD) refer to a collection of pathological conditions that impact the stomatognathic system, often associated with psychiatric comorbidities. Interestingly, previous studies have reported a higher prevalence of TMD in individuals affected by irritable bowel syndrome (IBS), a condition commonly linked to stress-induced psychosomatic factors. The aim of this systematic review is to clarify the prevalence of TMD in IBS-diagnosed patients. METHODS: A systematic search was conducted in PubMed, Scopus, Web of Science and Cochrane Library databases up to May

Oral anticoagulant monotherapy in patients with chronic coronary disease: An updated meta-analysis.

BACKGROUND: The optimal antithrombotic strategy for patients with chronic coronary disease requiring long-term anticoagulation remains uncertain. While dual therapy with oral anticoagulants (OACs) and antiplatelet agents is common, it significantly increases bleeding risk. This meta-analysis was conducted to compare the efficacy and safety of OAC monotherapy against combination therapy. METHODS: We searched PubMed, Embase, ClinicalTrials.gov, and Cochrane library through August 2025 to identify randomized controlled trials (RCTs) comparing OAC monotherapy to dual therapy (OAC plus a single an

Genes driving three-dimensional growth of immortalized cells and cancer.

Unchecked growth in three-dimensions (3D) in culture is a key feature of immortalized cells on the path to malignant transformation and hence a potential target phenotype for prevention. Also, expression of genes driving this process, but not that of 2D growth, would likely be more specific to cancer development and their inhibition would be less toxic to normal cells, many of which can grow in 2D but rarely in 3D culture. To define such genes, we compared CRISPR depletion screens performed in HBLAK, a spontaneously immortalized, non-tumorigenic human urothelial cell line, grown in 2D to those

Embryonic Zebrafish Irritant-evoked Hyperlocomotion (EZIH) as a high-throughput behavioral model for nociception.

Behavioral models have served a key role in understanding nociception, the sensory system by which animals detect noxious stimuli in their environment. Developing zebrafish (Danio rerio) are a powerful study organism for examining nociceptive pathways, given the vast array of genetic, developmental, and neuroscience tools available for these animals. However, at present there are few widely-adopted behavioral models for nociception in developing zebrafish. This study examines the locomotor response of hatching-stage zebrafish embryos to dilute solutions of the noxious chemical and TRPA1 agonis
