[Anatomical and neurochemical bases accounting for the frequent association between headaches and sleep: the hypnic headache paradigm].

INTRODUCTION: The relationship between headache and sleep is complex and runs in two directions. Headache may be the consequence of a (primary or secondary) sleep disorder or its cause (in chronic tension-type headache and/or chronic migraine with or without painkiller abuse). It can also be related to sleep in an intrinsic way, as in the case of hypnic headache (which only appears during sleep) or other primary headaches, such as migraine and certain trigeminal-autonomic cephalgias (which can appear during the waking state or during sleep); this type of headache occurs mostly during REM sleep

Seasonal variations of gonadotropins and prolactin in the laboratory rat. Role of maternal pineal gland.

Sexual and gender minority individuals experience higher rates of adverse childhood experiences (ACEs) than heterosexual individuals. Yet, the general ACE framework ignores unique ACEs faced by SGM individuals (i.e., cisheterosexism during childhood). The Sexual and Gender Minority ACE (SGM-ACE; Schnarrs et al., 2022, 2023) framework fills this gap by accounting for ACEs that are unique for SGM individuals resulting from systems of oppression (i.e., cisheterosexism during childhood). However, past studies employing this framework were conducted in the U.S. context, limiting the understanding o

Calmodulin-regulated adenylyl cyclases: cross-talk and plasticity in the central nervous system.

Gene disruption studies have shown that the Ca(2+)-stimulated adenylyl cyclases, AC1 and AC8, are critical for some forms of synaptic plasticity, including long-term potentiation as well as long-term memory formation (LTM). It is hypothesized that these enzymes are required for LTM to support the increased expression of a family of genes regulated through the cAMP/Ca(2+) response element-binding protein/cAMP response element transcriptional pathway. In contrast to AC1 and AC8, AC3 is a Ca(2+)-inhibited adenylyl cyclase that plays an essential role in olfactory signal transduction. Coupling of

Properties of clock-controlled and constitutive N-acetyltransferases from chick pineal cells.

The pineal gland synthesizes its hormone melatonin (O-methyl-N-acetylserotonin) from serotonin. Acetyl-CoA: serotonin N-acetyltransferase (SNAT), the enzyme that catalyzes the committed step in this biosynthesis, is largely restricted to the pineal gland and is regulated by adrenergic and circadian mechanisms. Another enzyme, acetyl-CoA: arylamine N-acetyltransferase (ANAT), having an apparently similar activity, is also present in the pineal. This enzyme, however, is not rhythmically regulated. SNAT activity of cultured chick pineal cells was obtained without ANAT after ammonium sulfate preci

Long term beneficial effects of weak electromagnetic fields in multiple sclerosis.

A 39 year-old severely disabled woman with a 19 year history of chronic relapsing-remitting multiple sclerosis (MS) began to experience improvement in symptoms within 24 hours after she received experimental treatment with picotesla electromagnetic fields (EMFs). Pattern reversal visual evoked potential (VEP) study obtained three weeks after the initiation of the first magnetic treatment showed a return to normal of the P100 latencies in each eye. The patient continued to receive 1-2 EMFs treatments per week and during the following 32 months she made a dramatic recovery with resolution of dip

Bone turnover marker responses to sleep restriction and weekend recovery sleep.

BACKGROUND: Prior data demonstrated three weeks of sleep restriction and concurrent circadian disruption uncoupled bone turnover markers (BTMs), indicating decreased bone formation and no change or increased bone resorption. The effect of insufficient sleep with or without ad libitum weekend recovery sleep on BTMs is unknown. METHODS: BTMs were measured in stored serum from 20 healthy adults randomized to one of three study groups consisting of a control group (N = 3 men; 9 h/night) or one of two nocturnal sleep restriction groups in an inpatient laboratory environment. One Sleep Restriction

The Relationship Between Shift Work and Men's Health.

BACKGROUND: More than 21 million Americans and nearly 20% of the U.S. workforce are shift workers. Non-standard shift work, defined as work that falls outside of 6 am-6 pm, can lead to poor diet, exercise, and sleep habits that lead to decreased productivity, increased workplace accidents, and a variety of negative health outcomes. AIM: To investigate the associations between shift work exposure and chronic medical conditions such as metabolic syndromes, cardiovascular disease, gastrointestinal disturbances, and depression as well as urologic complications including hypogonadism, male inferti

Neuroendocrine Stress Response in Acute Stroke: Physiological and Clinical Perspectives.

Acute ischemic stroke constitutes a significant systemic stress event that initiates a complex cascade of neuroendocrine and autonomic responses. Central to this process is activation of the hypothalamic-pituitary-adrenal (HPA) axis and the sympathetic nervous system, reflecting the brain's integrated response to acute cerebral injury. These changes are accompanied by widespread alterations in physiological regulation, including endocrine signaling, autonomic balance, circadian organization, and cardiovascular control. Understanding the nature and coordination of these responses is important f

Human milk oligosaccharide 2'-fucosyllactose attenuates oxidative stress-induced intestinal barrier injury in mice via regulating microbial tryptophan metabolism.

The present study aimed to investigate the protective effects of 2'-fucosyllactose (2'-FL) on diquat-induced intestinal oxidative injury and the underlying involvement of the gut microbiota and microbial tryptophan metabolism in this process. Thirty-six mice were assigned to CON, diquat, and 2'-FL + diquat groups. The results indicated that the diquat injection significantly impaired the intestinal barrier, suppressed the antioxidant capacities in serum and the jejunum, downregulated the mRNA expression of genes related to redox balance, and elevated proinflammatory cytokines. All these advers

Irregular sleep-wake rhythm disorder: From the pathophysiologic perspective to the treatment.

Irregular sleep-wake rhythm disorder (ISWRD) is an intrinsic circadian rhythm disorder caused by loss of the brain's circadian regulation, through changes of the input and/or output to the suprachiasmatic nucleus (SCN), or of the SCN itself. Although there are limited prevalence data for this rare disease, ISWRD is associated with neurodegenerative disorders, including the Alzheimer disease (AD) and the Parkinson disease (PD), which will become increasingly prevalent in an aging population. It additionally presents in childhood developmental disorders, psychiatric disorders, and traumatic brai

Successful deslorelin implant treatment in a neutered female Pomeranian dog with Alopecia X.

The Pomeranian dog is recognized as one of the predisposed breeds for alopecia X, a noninflammatory hair cycle arrest disorder. Although it is considered a cosmetic issue unrelated to systemic illness, having a pet with alopecia can be distressing for owners. Sex steroid imbalance seems to influence the development of alopecia X, but the exact pathogenesis is still unknown. Although some treatment alternatives are currently available for alopecia X, results can vary, as well as costs, risks, and owner adherence. GnRH analogs have been used extensively in the modulation of sex hormone synthesis

Tryptophan Stability and Palatability in the Food Formulation: A Review.

Tryptophan, an essential amino acid, plays a vital role in the synthesis of critical compounds like serotonin, melatonin, and niacin, which impact mood, sleep, and metabolic processes. It holds promise for improving the well-being of individuals with mood issues or sleep disorders through dietary enrichment. However, incorporating tryptophan into food products presents challenges related to stability, bitterness, and susceptibility to oxidative degradation. These issues can reduce consumer acceptability and effectiveness and may lead to the formation of harmful byproducts. This review comprehe

Oxidative Imbalance in Endometriosis-Related Infertility-The Therapeutic Role of Antioxidants.

Endometriosis in half of affected women is closely related to problems with fertility. Endometriosis-associated infertility is caused by a wide range of abnormalities affecting the female reproductive tract, from oocyte quality impairment to disturbances in the eutopic endometrium or mechanical abnormalities resulting from disease progression. Since supportive antioxidant therapies, in addition to surgical treatment or assisted reproductive techniques (ARTs), have overall been proven to be effective tools in endometriosis management, the objective of our review was to analyze the role of antio

Effect of 60 days of head down tilt bed rest on amplitude and phase of rhythms in physiology and sleep in men.

Twenty-four-hour rhythms in physiology and behaviour are shaped by circadian clocks, environmental rhythms, and feedback of behavioural rhythms onto physiology. In space, 24 h signals such as those associated with the light-dark cycle and changes in posture, are weaker, potentially reducing the robustness of rhythms. Head down tilt (HDT) bed rest is commonly used to simulate effects of microgravity but how HDT affects rhythms in physiology has not been extensively investigated. Here we report effects of -6° HDT during a 90-day protocol on 24 h rhythmicity in 20 men. During HDT, amplitude of li

Intra-myocardial hemorrhage and cardiac microvascular injury in ischemia/reperfusion. A systematic review of current evidences.

The in-hospital mortality rate in acute myocardial infarction (AMI) remains high despite the undoubted achievements in treatment of this disease achieved in the last 40 years. The dangerous complications of AMI remain cardiac microvascular injury (CMI) and intramyocardial hemorrhage (IMH). IMH is a widespread pathology that occurs in 42 - 57% of patients with ST-segment elevation myocardial infarction and percutaneous coronary intervention. IMH is associated with larger infarct size and contractile dysfunction. IMH is accompanied by inflammation. The appearance of IMH is depending on the durat

Correlation between gastroesophageal reflux disease lung volumes and exacerbation of bronchial asthma: Italian pilot observational retrospective study GERDAS.

Reflux asthma is an entity characterized by typical symptoms and, in some cases, is "silent"; it is more dangerous when associated with obesity and sleep apnea syndrome. Numerous studies demonstrate its high prevalence in the general population, particularly in the pediatric population, where, despite medical specialists' treatment, asthma symptoms remain poorly controlled with a high risk of acute exacerbations. This clinical study aims to show how the addition of a particular type of alginate (Deflux Plus sachets) containing hyaluronic acid and melatonin at low doses administered over a prol

Effects of Antioxidant Intake on Fetal Development and Maternal/Neonatal Health during Pregnancy.

During pregnancy, cycles of hypoxia and oxidative stress play a key role in the proper development of the fetus. Hypoxia during the first weeks is crucial for placental development, while the increase in oxygen due to the influx of maternal blood stimulates endothelial growth and angiogenesis. However, an imbalance in the number of oxidative molecules due to endogenous or exogenous factors can overwhelm defense systems and lead to excessive production of reactive oxygen species (ROS). Many pregnancy complications, generated by systemic inflammation and placental vasoconstriction, such as preec

Exploring the Tryptophan Metabolic Pathways in Migraine-Related Mechanisms.

Migraine is a complex neurovascular disorder, which causes intense socioeconomic problems worldwide. The pathophysiology of disease is enigmatic; accordingly, therapy is not sufficient. In recent years, migraine research focused on tryptophan, which is metabolized via two main pathways, the serotonin and kynurenine pathways, both of which produce neuroactive molecules that influence pain processing and stress response by disturbing neural and brain hypersensitivity and by interacting with molecules that control vascular and inflammatory actions. Serotonin has a role in trigeminal pain processi
