Development and Optimization of a Hollow Fiber-Based Countercurrent Dialysis Process for Continuous Manufacturing of Monoclonal Antibodies.

Countercurrent dialysis using hollow fiber modules offers several advantages over both inline diafiltration (ILDF) and hemodialyzer based countercurrent dialysis in terms of buffer requirements and compatibility with GMP manufacturing of biologics. In this work, we evaluate GMP-grade hollow fiber modules of varying lengths (20, 41.5, and 65 cm) and benchmark their performance against an ILDF unit as a reference process for continuous exchange of buffer in manufacturing of monoclonal antibodies. Hollow fiber modules outperformed the ILDF configuration in terms of buffer consumption and pressure

Serum metabolomics identifies ganglioside dysregulation and pyruvate dehydrogenase complex-related metabolic disturbances in pediatric drug-resistant epilepsy.

INTRODUCTION: Despite advances in epilepsy management, the exact causes of epilepsy remain unclear, and diagnostic biomarkers are still lacking. Besides, about one-third of cases involve drug-resistant epilepsy (DRE). METHODS: This study employed untargeted metabolomics to identify potential metabolic signatures in the sera of pediatric patients with epilepsy using liquid chromatography-high-resolution mass spectrometry. RESULTS: Our results revealed several disrupted energetic pathways and metabolites indicative of oxidative stress and mitochondrial dysfunction in both controlled epilepsy (

The H3K27me3-NR1D1-IFN-γ axis in trained macrophages induced by early respiratory virus infection drives non-T2 asthma in children.

Unlike that of T2 asthma, the pathophysiology of non-T2 asthma remains largely unexplored. However, increasing evidence supports a role for trained macrophages in asthma. In this study, we utilized a non-T2 asthma mouse model associated with early respiratory viral infection to identify the H3K27me3-NR1D1-IFN-γ axis in lung-trained macrophages, which drives non-T2 asthma by promoting Th1 differentiation. Early infection combined with allergen sensitization induced the formation of trained macrophages, which displayed hyperactive methionine cycling and elevated levels of S-adenosylmethionine (S

Transcriptome and volatile analyses identify key genes involved in ester biosynthesis in durian (Durio zibethinus) during fruit ripening.

Durian (Durio zibethinus), which is an important economic crop in Southeast Asia, is known for its distinct aroma combining sulfurous and fruity notes. Volatile esters are the main contributors to the sweet, fruity aroma of ripe durian fruit. Volatile ester contents in fruit are generally regulated by the activities of alcohol acyltransferase (AAT) and enzymes that metabolize amino acids, fatty acids, and carbohydrates into aroma volatiles. However, the molecular regulation of ester biosynthesis in durian remains unexplored. In this study, we aimed to identify key durian genes involved in este

Sex-specific dynamics of MASLD reveal early hepatic and extrahepatic metabolic deterioration in females despite long-term protection.

BACKGROUND: Metabolic dysfunction-associated steatotic liver disease (MASLD) is a chronic condition characterized by hepatic fat accumulation and systemic metabolic dysfunction. MASLD exhibits clear sex differences, yet the mechanisms underlying these disparities remain poorly defined. METHODS: To investigate the early temporal dynamics of MASLD, male and female mice were fed a choline-deficient, methionine-restricted (0.1%) high-fat (61%) diet (CDAHFD; characterized by impaired hepatic lipid export and enhanced lipotoxic stress) for two weeks, enabling assessment of initial metabolic respons

Dietary Glycerol Monolaurate Enhances Growth and Immune Function in Calves via Hepatic Immunometabolic Reprogramming.

Early-life nutrition is critical for the development and health of dairy calves, necessitating alternatives to in-feed antibiotics. This study investigated whether dietary glycerol monolaurate (GML) enhances growth performance, reduces diarrhea incidence, improves systemic antioxidant and immune status, and modulates hepatic immunometabolic function in calves. Twenty-four Holstein bull calves (7 ± 0.5 d of age) were randomly assigned by body weight and age to a control group or a GML-supplemented group, both fed milk replacer with starter feed provided throughout the 45-day trial. Calves in th

The monoacylglycerol acyltransferase 2 inhibitor VB-85387 reduces MASLD.

Metabolic dysfunction-associated steatotic liver disease (MASLD) arises from the accumulation of triglycerides within the liver. MASLD can advance to metabolic dysfunction-associated steatohepatitis (MASH), cirrhosis, and hepatocellular carcinoma. Monoacylglycerol acyltransferase 2 (MOGAT2) is essential for triglyceride synthesis and plays a significant role in regulating lipid metabolism. Here, we demonstrate the ability of a new human MOGAT2 inhibitor, VB-85387, to inhibit the development of MASLD/MASH and further define its effects on the key metabolic pathways that progress MASH developmen

Effects of complex enzyme preparation supplementation in a low-protein amino acid-balanced diet on production performance, serum biochemistry, nitrogen metabolism and nitrogen balance of mid-lactation dairy cows.

OBJECTIVE: High-protein diets for lactating dairy cows reduce nitrogen ( N ) use efficiency (NUE) and cause environmental nitrogen pollution. Low-protein (LP), amino acid (AA)-balanced diets are a promising alternative but carry the risk of impairing nutrient digestibility and cow productivity. Exogenous enzyme preparations may mitigate these limitations, but their specific effects and underlying mechanisms in this feeding scenario remain not fully elucidated. METHODS: Ninety mid-lactation Holstein dairy cows were assigned to 3 treatment groups in a randomized block design: (1) normal protein

Integrated transcriptomic and metabolomic analyses reveal coordinated molecular responses associated with nano-enabled arbuscular mycorrhizal symbiosis for salt stress tolerance in rice.

Salinity stress is a major environmental constraint limiting rice growth and yield. Seed nanopriming with calcium oxide nanomaterials (CaO NMs) in combination with arbuscular mycorrhizal fungus (AMF) has recently emerged as an effective strategy to enhance resilience, although the underlying responses remain unexplored. In the present study, rice seeds primed with CaO NMs (80 ppm) and inoculated with the AMF Claroideoglomus claroideum at transplanting were evaluated under 175 mM NaCl stress to assess their effects on plant performance and metabolic responses. The combined treatment significant

Nutrient Digestibility and Growth Performance of Juvenile Penaeus vannamei Fed Diets Containing the Filamentous Fungus, Paecilomyces variotii, Produced From Corn Ethanol Thin Stillage.

Fungal proteins are emerging ingredients having high potential for use in aquafeeds, yet digestibility and practical inclusion limits remain unclear for marine shrimp. This study evaluated a filamentous fungus (Paecilomyces variotii [PV]) produced from corn ethanol thin stillage as a replacement for salmon by-product meal (SLM) and poultry by-product meal (PBM) in diets for juvenile Penaeus vannamei. Five diets were formulated by replacing SLM and PBM with PV (0%-100% replacement; PV from 0% to 22.60% of the diet). After 78 days, shrimp performance was similar across treatments. Survival remai

Tied to Work: Topologically Knotted SPOUT Methyltransferases and UCH Deubiquitinases in Cellular Biochemistry.

Topologically knotted proteins are emerging as functional elements of the proteome rather than structural curiosities. Among the recurring protein knot types, the 31 trefoil, 52 Gordian and 71 septoil knots provide useful models for understanding how topology may reshape catalysis, substrate recognition, folding and mechanical behaviour. This review focuses on two systems in which these links can already be made with unusual clarity: the 31-knotted SPOUT methyltransferase superfamily and the 52-knotted ubiquitin C-terminal hydrolases (UCHs). In SPOUT methyltransferases, the knot defines a comp

Effects of rumen-protected choline supplementation on metabolic responses of dairy cows: a dose-response meta-analysis.

Several studies suggest that dietary supplementation with rumen-protected choline (RPC) during the transition period may support lactation performance in dairy cows, but the metabolic responses underlying these effects remain incompletely understood. We conducted a systematic review and dose-response meta-analysis following PRISMA guidelines to evaluate the effects of RPC supplementation on selected blood metabolites and liver-function indicators in dairy cows. A comprehensive literature search in Web of Science, Scopus, PubMed, and Google Scholar identified 18 articles and one dissertation, c

Phase 1 dose-escalation trial combining sulfasalazine and stereotactic radiosurgery in patients with recurrent glioblastoma.

Glioblastoma radioresistance is linked to glutathione, a key antioxidant protecting against oxidative stress. Sulfasalazine inhibits the xCT antiporter mediating cystine uptake required for glutathione synthesis in glioma, potentiating radiotherapy in preclinical studies. This trial evaluated safety and therapeutic potential of combining sulfasalazine with stereotactic radiosurgery (SRS) for recurrent glioblastoma. Adults with recurrences were enrolled in a 3 + 3 dose-escalation trial receiving 1.5, 3.0, 4.5, or 6.0 g sulfasalazine orally for 3 days before SRS. Primary endpoint was safety. Sec

Studies on functional differentiation of xpr1a and xpr1b genes in zebrafish.

Xenotropic and polytropic retrovirus receptor 1 (XPR1) is known to be involved in various biological processes, including phosphate homeostasis, cellular signaling, brain and vascular mineralization, whereas its specific contribution to bone development remains incompletely characterized. Due to genome duplication in teleosts, zebrafish Danio rerio possess two paralogous genes of XPR1 namely xpr1a and xpr1b, whose functional divergence remains unclear. The amino acid sequence similarity between zebrafish xpr1a and xpr1b was 83.26%. In situ hybridization demonstrated overlapping localization in

Polyporus umbellatus β-glucan ameliorates metabolic dysfunction-associated steatohepatitis by remodeling the gut microbiota-bile acid-FXR signaling axis.

ETHNOPHARMACOLOGICAL RELEVANCE: Polyporus umbellatus (Pers.) Fries has been used in traditional Chinese medicine for millennia to "promote water metabolism and eliminate dampness", a pathogenic concept central to metabolic dysfunction-associated steatohepatitis (MASH). Its polysaccharide extract is clinically applied in China for dampness-heat related liver disorders, but its specific bioactive components and mechanisms against MASH remain unknown. AIM OF THE STUDY: To evaluate the anti-MASH efficacy of the (1,3),(1,6)-β-D-glucan from P. umbellatus (PUP) and determine its mechanisms, focusing

BDNF Val66Met Variant as a Genetic Risk Factor for Major Depressive Disorder in Saudis.

OBJECTIVE: Major depressive disorder (MDD) is a prevalent and disabling psychiatric condition in Saudi Arabia, with genetic susceptibility remaining incompletely characterized. Reduced brain-derived neurotrophic factor (BDNF) activity has been implicated in MDD. The Val66Met polymorphism (rs6265), involving the substitution of valine (Val, G allele) with methionine (Met, A allele), impairs activity-dependent BDNF secretion. This study examined the frequency of Val66Met and its association with MDD in a Saudi cohort. MATERIALS AND METHODS: A case-control study was conducted, including 87 patie

Different metabolic pathways associated with total cortisol exposure and the cortisol time profile: a randomized crossover trial.

Excess cortisol exposure and disruption of its circadian pattern have both been linked to adverse health outcomes; however, whether distinct metabolic signatures differentiate total cortisol exposure from cortisol secretion dynamics remains unclear. This study aimed to identify metabolites and metabolic pathways uniquely associated with total cortisol exposure and/or variation of the cortisol time profile. In a randomized, 12-week, cross-over trial 18 adults with primary adrenal insufficiency (AI) received the same total daily dose of hydrocortisone (HC) administered as a once-daily (OD) dual-
