Preparation and in vitro anti-tumor properties of toad venom extract-loaded solid lipid nanoparticles.

In this study, we prepared solid lipid nanoparticles (TV-SLNs) loaded with toad venom extract and investigated their anti-tumor effects in vitro in HeLa and SKOV-3 cells. TV-SLNs were prepared using a cold homogenization technique, and the formulation was optimized by central composite design and response surface methods. The anti-tumor activities of TV-SLNs were evaluated by analyzing cell division and cell cycle distribution by using the MTT assay and flow cytometry. After incubation with TV-SLNs, the growth of both HeLa and SKOV-3 cells was inhibited significantly. The percentage of HeLa ce

One-step detection of circulating tumor cells in ovarian cancer using enhanced fluorescent silica nanoparticles.

Ovarian cancer is the fifth-leading cause of cancer-related deaths among women as a result of late diagnosis. For survival rates to improve, more sensitive and specific methods for earlier detection of ovarian cancer are needed. This study presents the development of rapid and specific one-step circulating tumor cell (CTC) detection using flow cytometry in a whole-blood sample with fluorescent silica nanoparticles. We prepared magnetic nanoparticle (MNP)-SiO2(rhodamine B isothiocyanate [RITC]) (MNP-SiO2[RITC] incorporating organic dyes [RITC, ëmax(ex/em) = 543/580 nm]) in the silica shell. We

Self-assembling modified β-cyclodextrin nanoparticles as neuronal siRNA delivery vectors: focus on Huntington's disease.

Huntington's disease (HD) is a rare autosomal dominant neurodegenerative disease caused by the expression of a toxic Huntingtin (HTT) protein. The use of short interfering RNAs (siRNAs) to silence the mutant protein is one of the most promising therapeutic strategies under investigation. The biggest caveat to siRNA-based approaches is the lack of efficient and nontoxic delivery vectors for siRNA delivery to the central nervous system. In this study, we investigated the potential of modified amphiphilic β-cyclodextrins (CDs), oligosaccharide-based molecules, as novel siRNA neuronal carriers. We

Nano-based antileishmanial agents: a toxicological study on nanoparticles for future treatment of cutaneous leishmaniasis.

Cutaneous leishmaniasis (CL) is endemic in the tropical and subtropical countries. Antileishmanial drugs that are traditionally used for treatment of CL are mainly toxic, ineffective for some parasite isolates, and mostly expensive. Previous studies showed that some metal and metal oxide nanoparticles have antimicrobial activity. Moreover, the use of nanoparticles together with ultra violet (UV) and infra red (IR) light increases toxic effects of nanoparticles by generation of reactive oxygen species (ROSs) and heat, respectively. There is little information on antileishmanial activity of nano

Proper design of silica nanoparticles combines high brightness, lack of cytotoxicity and efficient cell endocytosis.

Silica-based luminescent nanoparticles (SiNPs) show promising prospects in nanomedicine in light of their chemical properties and versatility. In this study, we have characterized silica core-PEG shell SiNPs derivatized with PEG moieties (NP-PEG), with external amino- (NP-PEG-amino) or carboxy-groups (NP-PEG-carbo), both in cell cultures as well as in animal models. By using different techniques, we could demonstrate that these SiNPs were safe and did not exhibit appreciable cytotoxicity in different relevant cell models, of normal or cancer cell types, growing either in suspension (JVM-2 leuk

Zinc oxide nanoparticles inhibit Ca2+-ATPase expression in human lens epithelial cells under UVB irradiation.

Epidemiological and experimental studies have revealed that lens epithelial cells exposed to ultraviolet B (UVB) light could be induced apoptosis, and lens epithelial cell apoptosis can initiate cataractogenesis. Posterior capsular opacification (PCO), the most frequent complication after cataract surgery, is induced by the proliferation, differentiation, migration of lens epithelial cells. Thus, inhibiting the proliferation of lens epithelial cells could reduce the occurrence of PCO. It is reported that zinc oxide (ZnO) nanoparticles have great potential for the application of biomedical fiel

Bifunctional polydopamine@Fe3O4 core-shell nanoparticles for electrochemical determination of lead(II) and cadmium(II).

The present paper has focused on the potential application of the bifunctional polydopamine@Fe3O4 core-shell nanoparticles for development of a simple, stable and highly selective electrochemical method for metal ions monitoring in real samples. The electrochemical method is based on electrochemical preconcentration/reduction of metal ions onto a polydopamine@Fe3O4 modified magnetic glassy carbon electrode at -1.1 V (versus SCE) in 0.1 M pH 5.0 acetate solution containing Pb(2+) and Cd(2+) during 160 s, followed by subsequent anodic stripping. The proposed method has been demonstrated highly s

Impact of dietary cadmium sulphide nanoparticles on Danio rerio zebrafish at very low contamination pressure.

To address the impact of cadmium sulphide nanoparticles (CdSNPs) of two different sizes (8 and 50 nm), Danio rerio zebrafish were dietary exposed to very low doses: 100 or 40 ng CdSNPs/day/g body weight for 36 or 60 days, respectively. The results obtained using RAPD-PCR genotoxicity test showed genomic alteration since the number of hybridisation sites of the RAPD probes was significantly modified after CdSNPs exposure. In addition, selected stress response genes were either repressed or upregulated in tissues of CdSNPs-exposed fish. Mitochondrial dysfunction was also caused by the presence o

Prussian blue nanoparticles as peroxidase mimetics for sensitive colorimetric detection of hydrogen peroxide and glucose.

Prussian blue nanoparticles (PB NPs) exhibits an intrinsic peroxidase-like catalytic activity towards the hydrogen peroxide (H2O2)-mediated oxidation of classical peroxidase substrate 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt to produce a colored product. The catalysis follows Michaelis-Menen kinetics and shows strong affinity for H2O2. Using PB NPs as a peroxidase mimetics, a colorimetric method was developed for the detection of 0.05-50.0 μM H2O2, with a detection limit of 0.031 μM. When the catalytic reaction of PB NPs was coupled with the reaction of glucose ox

A localized surface plasmon resonance-based multicolor electrochromic device with electrochemically size-controlled silver nanoparticles.

The first localized surface plasmon resonance (LSPR)-based multicolor electrochromic device with five reversible optical states is demonstrated. In this device, the size of deposited silver nanoparticles is electrochemically controlled by using a voltage-step method in which two different voltages are applied successively. The electrochemically size-controlled silver nanoparticles enable a reversible multiple-color change by a shift of the LSPR band.

Solution processed Al-doped ZnO nanoparticles/TiOx composite for highly efficient inverted organic solar cells.

We investigated the electrical properties of solution processed Al-doped ZnO (AZO) nanoparticles, stabilized by mixing with a TiOx complex. Thin solid films cast from the solution of AZO-TiOx (AZOTi) (Ti/Zn ∼0.4 in the bulk and ∼0.8 on its surface) is processable in inert environment, without a need for either ambient air exposure for hydrolysis or high temperature thermal annealing commonly applied to buffer layers of most metal-oxides. It was found that the electronic structure of AZOTi matches the electronic structure of several electron acceptor and donor materials used in organic electron

Effects of prepubertal exposure to silver nanoparticles on reproductive parameters in adult male Wistar rats.

The incidence of male reproductive pathologies, such as hypospadias, cryptorchidism, testicular cancer, and low sperm production in adulthood, is increasing and may be related to exposure to environmental contaminants. The silver nanoparticles (AgNP) are a new class of chemical compounds commonly used in both medical and nonmedical settings, and they affect development of spermatogonial stem cells in vitro. The aim of this study was to examine the adverse productive toxic effects of AgNPs in male Wistar rats exposed during the prepubertal period and sacrificed at postnatal day (PND) 53 and PND

Modulation of defect-mediated energy transfer from ZnO nanoparticles for the photocatalytic degradation of bilirubin.

In recent years, nanotechnology has gained significant interest for applications in the medical field. In this regard, a utilization of the ZnO nanoparticles for the efficient degradation of bilirubin (BR) through photocatalysis was explored. BR is a water insoluble byproduct of the heme catabolism that can cause jaundice when its excretion is impaired. The photocatalytic degradation of BR activated by ZnO nanoparticles through a non-radiative energy transfer pathway can be influenced by the surface defect-states (mainly the oxygen vacancies) of the catalyst nanoparticles. These were modulated

Metal and silicate particles including nanoparticles are present in electronic cigarette cartomizer fluid and aerosol.

BACKGROUND: Electronic cigarettes (EC) deliver aerosol by heating fluid containing nicotine. Cartomizer EC combine the fluid chamber and heating element in a single unit. Because EC do not burn tobacco, they may be safer than conventional cigarettes. Their use is rapidly increasing worldwide with little prior testing of their aerosol. OBJECTIVES: We tested the hypothesis that EC aerosol contains metals derived from various components in EC. METHODS: Cartomizer contents and aerosols were analyzed using light and electron microscopy, cytotoxicity testing, x-ray microanalysis, particle counting

Sensitive colorimetric detection of Listeria monocytogenes based on isothermal gene amplification and unmodified gold nanoparticles.

Listeria monocytogenes (L. monocytogenes), one of most problematic food-borne bacteria, is mainly transmitted through the food chain and may cause listeriosis. Therefore, the development of rapid and sensitive L. monocytogenes detection technique has become an urgent task. In this study, we proposed a method using hyperbranching rolling circle amplification (HRCA) combined with gold nanoparticle (GNP) based colorimetric strategy to offer an isothermal, highly sensitive and specific assay for the detection of L. monocytogenes. First, a linear padlock probe targeting a specific sequence in the h

Characteristics of magnetic labeling on liver tumors with anti-alpha-fetoprotein-mediated Fe3O4 magnetic nanoparticles.

For preoperative and intraoperative detection of tumor distribution, numerous multimodal contrast agents, such as magnetic nanoparticles (MNPs) with several examination indicators, are currently in development. However, complex materials, configuration, and cost are required for multimodal contrast agents, accompanied by a high possibility of toxicity and low popularity in clinics. Nevertheless, the magnetic labeling of MNPs using bioprobes should be feasible not only in preoperative magnetic resonance imaging (MRI), but also in intraoperative examination based on other magnetic properties. Antimicrobial and anti-inflammatory activity of chitosan-alginate nanoparticles: a targeted therapy for cutaneous pathogens.

Advances in nanotechnology have demonstrated potential application of nanoparticles (NPs) for effective and targeted drug delivery. Here we investigated the antimicrobial and immunological properties and the feasibility of using NPs to deliver antimicrobial agents to treat a cutaneous pathogen. NPs synthesized with chitosan and alginate demonstrated a direct antimicrobial activity in vitro against Propionibacterium acnes, the bacterium linked to the pathogenesis of acne. By electron microscopy (EM) imaging, chitosan-alginate NPs were found to induce the disruption of the P. acnes cell membrane

Flt1 peptide-hyaluronate conjugate micelle-like nanoparticles encapsulating genistein for the treatment of ocular neovascularization.

Flt1 peptide of GNQWFI is an antagonistic peptide for vascular endothelial growth factor receptor 1 (VEGFR1 or Flt1). In this work, Flt1 peptide-hyaluronate (HA) conjugates were successfully synthesized and the resulting micelle-like nanoparticles were exploited to encapsulate genistein, an inhibitor of tyrosine-specific protein kinases, for the treatment of ocular neovascularization. The mean diameter of genistein-loaded Flt1 peptide-HA conjugate micelles was measured to be 172.0±18.7 nm, with a drug-loading efficiency of 40-50%. In vitro release tests of genistein from the genistein-loaded F

Transport of ferrihydrite nanoparticles in saturated porous media: role of ionic strength and flow rate.

The use of nanoscale ferrihydrite particles, which are known to effectively enhance microbial degradation of a wide range of contaminants, represents a promising technology for in situ remediation of contaminated aquifers. Thanks to their small size, ferrihydrite nanoparticles can be dispersed in water and directly injected into the subsurface to create reactive zones where contaminant biodegradation is promoted. Field applications would require a detailed knowledge of ferrihydrite transport mechanisms in the subsurface, but such studies are lacking in the literature. The present study is inte

Immobilization of mercury in field soil and sediment using carboxymethyl cellulose stabilized iron sulfide nanoparticles.

Mercury (Hg) is one of the most pervasive and bio-accumulative metals in the environment. Yet, effective in situ remediation technologies have been lacking. This study investigated the effectiveness of a class of soil-deliverable FeS nanoparticles for in situ immobilization of Hg in two field-contaminated soils from a New Jersey site and one sediment from an Alabama site. The nanoparticles were prepared using sodium carboxymethyl cellulose (CMC) as a stabilizer. Transmission electron microscopy measurements revealed a particle size of 34.3 ± 8.3 nm (standard deviation), whereas dynamic light s
