Effects of chronic ethanol exposure on dorsal medial striatal neurons receiving convergent inputs from the orbitofrontal cortex and basolateral amygdala.

Alcohol use disorder is associated with altered function of cortical-amygdala-striatal circuits such as the orbitofrontal cortex (OFC), basolateral amygdala (BLA) and their connections to the dorsal medial striatum (DMS) shown to be involved in goal-directed actions. Using retrobead tracing, we previously reported enhanced excitability of DMS-projecting OFC neurons in mice following 3-to-7-day withdrawal from chronic intermittent ethanol (CIE) exposure. In the same animals, spiking of DMS-projecting BLA neurons was decreased at 3-days post-withdrawal followed by an increase in firing at 7- and

Potential mechanism of impaired perceptual reasoning in children with obstructive sleep apnea syndrome: topological analysis of brain white matter network employing graph theory.

Childhood obstructive sleep apnea syndrome (OSAS) disrupts normal ventilation and sleep structure and affects cognitive functions. However, the neurophysiological mechanisms underlying cognitive impairment are unclear. This study investigates the topological connectivity of white matter networks in children with moderate to severe OSAS and explores the underlying mechanisms of cognitive impairment. We collected clinical data of patients with moderate to severe OSAS (n = 43) and non-OSAS (n = 30). Intelligence testing was conducted using the China Wechsler Intelligence Scale for Children-Fourth

Effect of a single dose of lorazepam on resting state functional connectivity in healthy adults.

Lorazepam is a fast-acting benzodiazepine that is widely used to manage anxiety symptoms through modulation of GABAergic activity. Despite being one of the most prescribed benzodiazepines, the effects of a single dose of lorazepam on brain functional connectivity at rest is not known. In this placebo-controlled, crossover study, twenty healthy adult participants (9 women; 26.2 ± 5.2 years) underwent two resting-state fMRI (rs-fMRI) scans following administration of either lorazepam (2.5 mg) or placebo. Imaging data were analyzed using an individual brain network parcellation approach and diffe

Audio-visual crossmodal connectivity in perceiving distorted speech: A comparison between patients with cleft palate and typical listeners.

This study investigates audio-visual crossmodal connectivity during the perception of distorted speech using task-based functional magnetic resonance imaging (fMRI). A randomized block design was employed, involving 20 patients with cleft palate and 20 typical listeners. Participants underwent perceptual tasks involving both cleft-related glottal stop and typical speech while fMRI scans were conducted. Regional interactions between the auditory and visual cortices were analyzed using dynamic causal modeling (DCM). Individual-level effective connectivity analysis revealed that, during the perce

The ventral hippocampus and nucleus accumbens as neural substrates for cocaine contextual memory reconsolidation.

Addictive drugs engage molecular pathways of associative learning and memory. Drug craving triggered by cues that were once associated with drug intoxication contributes to continued drug-seeking behaviors. Reactivated memories are vulnerable to disruption by interference with the process of reconsolidation, hence targeting reconsolidation could be beneficial in reducing cue-induced drug craving and relapse. Here, we sought to identify the neuronal pathways and neuroplasticity involved in cocaine contextual memory reconsolidation. Mice expressing inhibitory DREADDs in either nucleus accumbens

Extinction of Heroin Seeking Does Not Require the Infralimbic Cortex or Its Projections to the Nucleus Accumbens Shell or Amygdala.

Evidence indicates that the activity of the infralimbic cortex (IL), as well as its projections to the nucleus accumbens shell (NAshell) and amygdala, following an unreinforced lever press is critical for cocaine extinction learning and retention. It is unclear whether the same neural circuitry is involved in extinction encoding for other classes of addictive drugs, including opioids. In this study, we used a behaviour-controlled optogenetic approach in female and male Sprague-Dawley rats to examine the role of the IL and its projections in the extinction of heroin seeking. Rats first underwen

Alterations in static and dynamic functional network connectivity in chronic low back pain: a resting-state network functional connectivity and machine learning study.

Low back pain (LBP) is a prevalent pain condition whose persistence can lead to changes in the brain regions responsible for sensory, cognitive, attentional, and emotional processing. Previous neuroimaging studies have identified various structural and functional abnormalities in patients with LBP; however, how the static and dynamic large-scale functional network connectivity (FNC) of the brain is affected in these patients remains unclear. Forty-one patients with chronic low back pain (cLBP) and 42 healthy controls underwent resting-state functional MRI scanning. The independent component an

Papez circuit remodeling in type 2 diabetes: Enlarged choroid plexus volume partially mediates functional impairments linked to insulin resistance.

AIM: Type 2 diabetes mellitus (T2DM) is a metabolic disorder associated with cognitive decline. This study investigated the interplay between altered effective connectivity (EC) in the Papez circuit, regional atrophy, and choroid plexus volume (CPV) in relation to T2DM-related cognitive deficits. METHODS: Eighty T2DM patients and 75 healthy controls underwent multimodal MRI. Resting-state fMRI data were analyzed using spectral dynamic causal modeling (spDCM) to quantify EC between Papez circuit regions. Structural 3D-T1 images were processed to measure regional volumes and the CPV, which migh

Structural Cerebellar and Lateral Frontoparietal Networks are altered in CUD: An SBM Analysis.

Repetitive drug use results in enduring structural and functional changes in the brain. Addiction research has consistently revealed significant modifications in key brain networks related to reward, habit, salience, executive function, memory and self-regulation. Techniques like Voxel-based Morphometry have highlighted large-scale structural differences in grey matter across distinct groups. Source-based Morphometry (SBM) takes this a step further by incorporating the Independent Component Analysis to detect shared patterns of grey matter variation, all without requiring prior selection of re

Melanin-Concentrating Hormone Projections to the Nucleus Accumbens Enhance the Reward Value of Food Consumption and Do Not Induce Feeding or REM Sleep.

Regulation of food intake and energy balance is critical to survival. Hunger develops as a response to energy deficit and drives food-seeking and consumption. However, motivations to eat are varied in nature and promoted by factors other than energy deficit. When dysregulated, nonhomeostatic drives to consume can contribute to disorders of food intake, adding to the increasing prevalence of restrictive eating disorders and obesity. Melanin-concentrating hormone (MCH) neurons have been implicated in the regulation of feeding behavior, in addition to a number of other fundamental behaviors inclu

Functional connectome-based predictive modeling of suicidal ideation.

Suicide represents an egregious threat to society despite major advancements in medicine, in part due to limited knowledge of the biological mechanisms of suicidal behavior. We apply a connectome predictive modeling machine learning approach to identify a reproducible brain network associated with suicidal ideation in the hopes of demonstrating possible targets for novel anti-suicidal therapeutics. Patients were recruited from an inpatient facility at The Menninger Clinic, in Houston, Texas (N = 261; 181 with active and specific suicidal ideation) and had a current major depressive episode and

Characterizing microstructural patterns within the cortico-striato-thalamo-cortical circuit in Parkinson's disease.

PURPOSE: Parkinson's disease (PD) involves pathological alterations that include cortical impairments at levels of region and network. However, its microstructural abnormalities remain to be further elucidated via an appropriate diffusion neuroimaging approach. This study aimed to comprehensively demonstrate the microstructural patterns of PD as mapped by diffusion kurtosis imaging (DKI). METHODS: The microstructure of grey matter in both the PD group and the matched healthy control group was quantified by a DKI metric (mean kurtosis). The intergroup difference and classification performance

Effects of repetitive transcranial magnetic stimulation on individual variability of resting-state functional connectivity in major depressive disorder.

BACKGROUND: Repetitive transcranial magnetic stimulation (rTMS) is an effective treatment for major depressive disorder (MDD), but substantial heterogeneity in outcomes remains. We examined a potential mechanism of action of rTMS to normalize individual variability in resting-state functional connectivity (rs-fc) before and after a course of treatment. METHODS: Variability in rs-fc was examined in healthy controls (baseline) and individuals with MDD (baseline and after 4-6 weeks of rTMS). Seed-based connectivity was calculated to 4 regions associated with MDD: left dorsolateral prefrontal cor

Altered topology in cortical morphometric similarity network in recurrent major depressive disorder.

BACKGROUND: Recurrent major depressive disorder (RDD) is increasingly understood to be associated with a 'disconnection' within the brain areas. But, the true understanding of cortical connectivities remains challenging. Morphometric similarity network (MSN) with multi-modal magnetic resonance imaging (MRI) could provide more information about cortical micro-architecture changes in individuals with RDD. METHODS: Here, we integrated multi-modal features from T1-weighted imaging, diffusion tensor imaging (DTI), and inhomogeneous magnetization transfer imaging (ihMT) to construct MSN. We used gr

Altered degree centrality and functional connectivity in girls with central precocious puberty.

Girls who suffer from central precocious puberty (CPP) are at risk of experiencing detrimental psychological and behavioural consequences, along with impaired brain development. However, the mechanism by which puberty hormones affect patients with CPP remains unclear. This study aimed to use degree centrality (DC) analysis to explore the impact of premature activation of the hypothalamic-pituitary-gonadal (HPG) axis on brain functional development in girls with CPP. In this cross-sectional study, thirty-four girls (mean ± SD, 7.89 ± 0.81 years) with CPP and 25 age-matched girls without CPP (me

Neural Mechanisms of Resting-State Networks and the Amygdala Underlying the Cognitive and Emotional Effects of Psilocybin.

BACKGROUND: Serotonergic psychedelics, such as psilocybin, alter perceptual and cognitive systems that are functionally integrated with the amygdala. These changes can alter cognition and emotions that are hypothesized to contribute to their therapeutic utility. However, the neural mechanisms of cognitive and subcortical systems altered by psychedelics are not well understood. METHODS: We used resting-state functional magnetic resonance images collected during a randomized, double-blind, placebo-controlled clinical trial of 24 healthy adults under 0.2 mg/kg psilocybin to estimate the directed

Dissociable Roles of the mPFC-to-VTA Pathway in the Control of Impulsive Action and Risk-Related Decision-Making in Roman High- and Low-Avoidance Rats.

BACKGROUND: Impulsive action and risk-related decision-making (RDM) are associated with various psychiatric disorders, including drug abuse. Both behavioral traits have also been linked to reduced frontocortical activity and alterations in dopamine function in the ventral tegmental area (VTA). However, despite direct projections from the medial prefrontal cortex (mPFC) to the VTA, the specific role of the mPFC-to-VTA pathway in controlling impulsive action and RDM remains unexplored. Structural connectivity changes in unilateral hearing loss.

Network connectivity, as mapped by the whole brain connectome, plays a crucial role in regulating auditory function. Auditory deprivation such as unilateral hearing loss might alter structural network connectivity; however, these potential alterations are poorly understood. Thirty-seven acoustic neuroma patients with unilateral hearing loss (19 left-sided and 18 right-sided) and 19 healthy controls underwent diffusion-weighted and T1-weighted imaging to assess edge strength, node strength, and global efficiency of the structural connectome. Edge strength was estimated by pair-wise normalized s
