Inchworming: a novel motor stereotypy in the BTBR T+ Itpr3tf/J mouse model of autism.

Autism Spectrum Disorder (ASD) is a behaviorally defined neurodevelopmental disorder characterized by decreased reciprocal social interaction, abnormal communication, and repetitive behaviors with restricted interest. As diagnosis is based on clinical criteria, any potentially relevant rodent models of this heterogeneous disorder should ideally recapitulate these diverse behavioral traits. The BTBR T+ Itpr3tf/J (BTBR) mouse is an established animal model of ASD, displaying repetitive behaviors such as increased grooming, as well as cognitive inflexibility. With respect to social interaction an

Altered intrahemispheric structural connectivity in Gilles de la Tourette syndrome.

Gilles de la Tourette syndrome (GTS) is a common developmental neuropsychiatric disorder characterized by tics and frequent psychiatric comorbidities, often causing significant disability. Tic generation has been linked to disturbed networks of brain areas involved in planning, controlling and execution of actions, particularly structural and functional disorders in the striatum and cortico-striato-thalamo-cortical loops. We therefore applied structural diffusion tensor imaging (DTI) to characterize changes in intrahemispheric white matter connectivity in cortico-subcortical circuits engaged i

MeCP2 involvement in the regulation of neuronal alpha-tubulin production.

Rett syndrome (RTT) is a severe neurodevelopmental disorder caused by a dominant mutation in the X-linked methyl CpG binding protein 2 (MeCP2) gene. Neuroanatomically, RTT is characterized by a reduction in dendritic arborization and perikaryal size in the brain. MECP2 binds methylated promoters and facilitates assembly of a multiprotein repressor complex that includes Sin3A and the histone deacetylases HDAC1/HDAC2. MeCP2 has recently been found to be downregulated in autistic spectrum disorders such as Angelman syndrome (AS) and RTT, which share some phenotypic manifestations. We have conduct

Rett Syndrome -- an update.

Rett syndrome is a progressive, usually sporadic and rarely familial, disabling neurodevelopmental disorder with onset in early childhood presenting clinically with mental retardation, behavioral changes, late movement disturbances, loss of speech and hand skills, ataxia, apraxia, irregular breathing with hyperventilation while awake, and frequent seizures. It occurs almost exclusively in females with an estimated prevalence of 1 in 10-22000 births and is considered a manifestation of defective brain maturation caused by dominant mutation of the MeCP2 gene encoding the transcriptional represso

Emerging Personality Pathology in Adolescents With Autism Spectrum Disorder: Clinical Correlates and Assessment Implications.

The link between neurodevelopmental and psychiatric conditions is increasingly recognized in developmental psychiatry, yet the relationship between Autism Spectrum Disorder (ASD) and Personality Disorders (PDs) remains underexplored in adolescence. Dysfunctional personality traits (DPT) may emerge in individuals with ASD, especially when diagnosis is delayed and comorbidities occur. This study assessed 79 adolescents (mean age = 15.15 ± 1.50; 45.6% females) with ASD without intellectual disability using a multimodal protocol (Autism Diagnostic Observation Schedule-Second Edition [ADOS-2], Auti

Exploring offspring behaviour after prenatal COVID-19 vaccination in mice.

Maternal immunisation allows the transfer of protective antibodies to the offspring, reducing the risk of severe infection during early life. While vaccination during pregnancy is clinically recommended, its long-term impact on neurodevelopment remains under investigation. Autism spectrum disorder (ASD) is a neurodevelopmental condition characterised by social and behavioural alterations. Here, we evaluated whether prenatal exposure to the COVID-19 mRNA BNT162b2 vaccine or influenza vaccine affects general and autism-related behavioural outcomes in juvenile (4 weeks) and adult (8 weeks) mouse

Intranasal insulin reduces ADHD-like behaviors and neurodevelopmental deficits following neonatal hypoxia-ischemia in juvenile rats.

Neonatal hypoxia-ischemia (HI) is a leading cause of long-term neurodevelopmental impairment and is increasingly associated with a heightened risk of attention-deficit/hyperactivity disorder (ADHD) and related behavioral abnormalities. Beyond its metabolic role, insulin functions as a neurotrophic and immunomodulatory factor in the developing brain. However, whether early enhancement of central insulin signaling can mitigate the neuroinflammatory and behavioral sequelae of HI remains unclear. Male and female Sprague-Dawley rats were subjected to HI (right common carotid artery ligation followe

GABAB - and GABAA -receptor-mediated regulation of Up and Down states across development.

Slow oscillations, the hallmark of non-REM sleep, and their cellular counterpart, Up and Down states (UDSs), are considered a signature of cortical dynamics that reflect the intrinsic network organization. Although previous studies have explored the role of inhibition in regulating UDSs, little is known about whether this role changes with maturation. This is surprising since both slow oscillations and UDSs exhibit significant age-dependent alterations. To elucidate the developmental impact of GABAB and GABAA receptors on UDS activity, we conducted simultaneous local field potentials and intra

PTBP1: A master regulator of neural development and transdifferentiation-promise and pitfalls.

The polypyrimidine tract-binding protein 1 (PTBP1), a pivotal RNA-binding protein and splicing regulator, is emerging as a master orchestrator of neuronal fate determination, neurodevelopment, and neural function through its central role in alternative splicing. We synthesize current knowledge on PTBP1's multifaceted roles in orchestrating neural tube formation, neuronal migration, and synaptic circuit assembly through precise control of alternative splicing programs. We critically examine the rapidly evolving field of PTBP1-mediated cellular reprogramming, analyzing both supportive evidence a

Single-cell proteomic landscape of the developing human brain.

Profiling protein abundance and dynamics at single-cell resolution in complex human tissues is challenging. Given the discordance between transcript and protein abundance observed in studies of the human cerebral cortex, we developed an optimized workflow that combines label-free single-cell mass spectrometry with precise sample preparation to resolve quantitative proteomes of individual cells from the developing human brain. Our method achieves deep proteomic coverage (~800 proteins per cell) even in small immature prenatal human neurons (diameter ~7-10 μm, ~50 pg protein), capturing major br

Global profiling of protein lactylation in pancreatic ductal adenocarcinoma.

Cancer cells dramatically increase L-lactate production, fueling lysine lactylation (Kla) – a posttranslational modification that rewires protein function and links metabolic flux to gene and pathway regulation. Although emerging studies implicate Kla in diverse diseases, its role in pancreatic ductal adenocarcinoma (PDAC) remains poorly understood. Here, we performed global lactylomic profiling of immunoprecipitated lactylated proteins from PDAC and normal pancreatic cell lines, as well as from patient-derived tumors and adjacent normal tissues. Mass spectrometry revealed distinct Kla signatu

Wdr5 Modulates Synaptic Development and Memory Consolidation in the Hippocampus and Cerebral Cortex.

WD-repeat domain 5 (Wdr5), a core component of the SET1/MLL family of histone methyltransferases, is widely involved in the regulation of development, epigenetic control, transcription, and cell division. Wdr5 has been linked to neurodevelopmental disorders characterized by intellectual disability and developmental delay-yet its regulatory roles in neurons remain poorly understood. Here, we report that postnatal deletion of Wdr5 in excitatory forebrain neurons impairs learning and memory in male Wdr5fl/fl; CaMKIIα-iCre conditional knockout (Wdr5 cKO) mice. We showed that the conditional Wdr5 k

Novel TMEM63A mutation associated with transient hypomyelination of infancy - lessons from a previously negative whole-exome sequencing case: Three case reports.

BACKGROUND: Hypomyelinating leukodystrophies are rare neurodevelopmental disorders characterized by impaired myelin development and early motor delay. Hypomyelinating leukodystrophy type 19 (HLD-19), also termed transient hypomyelination of infancy, is caused by TMEM63A variants. It can clinically resemble other hypomyelinating leukodystrophies but is distinguished by developmental improvement. However, its phenotypic spectrum remains incompletely defined. CASE SUMMARY: We describe 3 related individuals presenting in early infancy with nystagmus, hypotonia, and delayed motor milestones. Brain

Brain Landscape In Situ Crosslinking Mass Spectrometry (BLIS-XL-MS) Enables Global Analysis of Protein Structural Remodeling in the Brain.

Pathological protein conformational remodeling is a key molecular feature of brain disorders. However, conventional approaches to understand the changes in potein conformation are targeted and difficult to scale proteome-wide analysis. Emerging mass spectrometry-based structural proteomic methods broaden coverage but often involve complex workflows and trade- offs between labeling efficiency and preservation of the in vivo molecular state. Thus, quantitative analysis of proteome-wide structural remodeling remains limited. Here we present Brain Landscape In Situ Crosslinking Mass Spectrometry (

Current insights into PHF14: molecular features and functional roles.

Plant Homeodomain Finger Protein 14 (PHF14) is a known epigenetic regulator involved in chromatin-mediated gene regulation. PHF14 belongs to the plant homeodomain (PHD) family, and possesses four PHD zinc-finger domains. The N-terminal PHD1 and PHD2 domain of PHF14 mediate histone recognition through specific PHD1-ZnK-PHD2 (PZP) cassettes and thereby contribute to the regulation of transcriptional activation and gene silencing. PHF14 plays a role in regulating essential cellular processes such as the cell cycle, mesenchymal proliferation, DNA damage response, and B-cell proliferation. Genetic

Molecular mechanism of HUWE1-HAPSTR1-USP7-mediated ubiquitin chain amplification on nuclear proteins.

Rapid protein turnover is essential for cellular stress adaptation. HUWE1 (HECT, UBA, and WWE domain containing 1), a large HECT-type E3 ligase, regulates many short-lived stress-responsive proteins, yet the mechanisms underlying its substrate selectivity remain unclear. Here, we reveal that HUWE1 functions as a ubiquitin chain amplifier that captures pre-ubiquitinated substrates and amplifies the degradation signal by assembling long ubiquitin chains containing K11-K48 branch points, a process regulated by its partners HUWE1-associated protein stress response 1 (HAPSTR1) and USP7 (ubiquitin-s

A stress-dependent postembryonic role for the core CPA factor CFIM-1 in germline integrity.

Post-transcriptional processing of pre-mRNAs by alternative polyadenylation (APA) generates a diversity of transcript isoforms at the 3' untranslated region (3'UTR) that can affect their function and stability. The differential enrichment of transcript isoforms has been implicated in diseases ranging from cancer to neurodevelopmental disorders. However, the postembryonic developmental roles of the core ensemble of cleavage and polyadenylation (CPA) factors that mediate these post-transcriptional changes remain poorly characterized. Here, we report a stress-dependent role for the core CPA facto

Subcortical regional volume vulnerability linked to child neglect without other forms of maltreatment.

Child neglect during critical stages of development has been associated with long-term adverse outcomes, including impairments in social, emotional, and physical health, as well as elevated risks of mental and cardiovascular disorders. This study examined whether alterations in corpus callosum (CC) morphology-essential for interhemispheric communication-are associated with the neural mechanisms underlying psychosocial characteristics in children exposed to neglect. Brain morphology was assessed using T1-weighted magnetic resonance imaging in Japanese children with a history of neglect (neglect
