The Antimicrobial Peptide C14R Is Active Against All Pathogenic Species of the ESKAPE Group.

The global rise in antimicrobial resistance among the ESKAPE pathogens represents a major challenge to public health. Here, we report the broad-spectrum antibacterial activity of the synthetic antimicrobial and pore-forming peptide C14R against all six ESKAPE species. Using a radial diffusion assay and resazurin-based viability testing, C14R exhibited a potent bactericidal effect with minimum inhibitory concentrations (MICs), defined as the lowest concentration of an antimicrobial agent that completely inhibits visible growth of planktonic microorganisms, ranging from 3.4 µg/mL (Enterococcus f

Enhancing the immunogenicity of a recombinant PEDV subunit vaccine: the role of lipoprotein signal peptide.

The spike (S) protein of Porcine Epidemic Diarrhea Virus (PEDV) is a major structural glycoprotein responsible for viral attachment and entry, making it a critical target for the development of subunit vaccines. Within the S1 domain, a well-characterized region spanning residues 499–799, commonly referred to as the core neutralizing epitope (COE) the equivalent of the CO-26 K domain, contains multiple predicted B-cell, T-cell, and virus-neutralizing epitopes, making it highly immunogenic. This study investigates whether the incorporation of a bacterial lipoprotein signal peptide into the COE d

An agent-guided peptide hydrogel bio-stabilizer clamps pericellular viscoelastic drift.

Living tissues continually remodel the extracellular matrix, causing pericellular mechanics to drift over time. Chronic glycation amplifies this drift, destabilizing mechanoregulatory cues and compromising regenerative outcomes. Here we introduce Micro-Mechanical Homeostasis Reset, a material-based strategy that targets cell-to-pericellular viscoelastic homeostasis rather than static bulk stiffness. Using an agent-guided, rule-based enumeration-and-scoring workflow, we design ViscoClamp, an extracellular matrix-interpenetrating peptide hydrogel with affinity for calcium ions and advanced glyca

Glucagon-like Peptide-1 Receptor Agonists: An Alternative for Dysglycemia in Hospitalized Patients?

In the outpatient management of type 2 diabetes mellitus, glucagon-like peptide-1 receptor agonists (GLP-1 RAs) are proven to benefit long-term glycemic control and reduce complications related to chronic inflammation. Emerging clinical data suggest that GLP-1 RAs may serve as an alternative or complement to insulin for glycemic management in hospitalized patients. Preclinical studies indicate that GLP-1 RAs influence the inflammatory response in models of acute inflammation, including ischemia-reperfusion injury, sepsis-induced vascular dysfunction, acute lung injury, and acute kidney injury.

Mitochondrial-derived peptide Humanin protects granulosa cells under oxidative conditions.

In brief: Redox imbalance compromises granulosa cell survival and follicle fate. This study identifies the mitochondrial-derived peptide Humanin (HN) as a cytoprotective factor that protects granulosa cells under oxidative condItions. Abstract: Granulosa cell function is essential for proper ovarian physiology. Redox imbalance compromises granulosa cell survival, thereby impacting follicle fate within the ovary. Humanin (HN), a small mitochondrial-derived peptide, exerts cytoprotective effects in several tissues under pro-oxidant conditions. The present study aimed to evaluate the cytoprotecti

Enhanced Intramuscular mRNA Activity of Peptide-Lipid Nanoparticles with Hydrophobized Head Groups.

A library of xenopeptide-based ionizable lipids (XP-ILs) was evaluated in mRNA lipid nanoparticles (LNPs) to investigate the impact of modifications within the polar oligoamine headgroup. Modifications included hydrophobic or bulky side chains or a reduced number of protonatable amines. The resulting XP-ILs were characterized by their key physicochemical properties (logD, logP, HLB, CMC), which were correlated with LNP transfection efficiency and cytotoxicity. The inverse relationship between computational logP and experimental logD5.5 is consistent with the HLB, indicating a lower tendency fo

Triple helix formation of a collagen model peptide induces cell aggregation formation.

Biofabrication refers to the creation of tissues and organs from biological materials such as cells and proteins, and is often applied in transplantation therapy and drug efficacy evaluation. For such a construct to function as a tissue, it is important for individual cells to interact closely with each other, so tailor-made approaches for the fabrication of cell aggregates to facilitate intercellular interactions are needed. Some researchers are using the DNA chain as a crosslinking site to make cell-cell assemblies, but DNA degradation and the potential risk of an immune response are concern

Dynamic Covalent Peptide-Drug Conjugates Address the Heterogeneity in Alzheimer's Disease Progression.

A growing understanding of the pathophysiological evolution of Alzheimer's disease (AD) underscores the heterogeneity in its progression as a critical factor undermining the success of various candidate interventions and complicating the establishment of effective pharmacotherapeutic regimens. Here, we introduce the development of a hierarchical-responsive therapeutic agent self-assembled from phenylboronate ester-linked Tjernberg's KLVFF peptide-curcumin conjugates (CPKNAs), which is designed to dynamically track the spatiotemporal coordinates of biomarkers associated with AD heterogeneity in

Peptide Substitutes of PEG: Biological Outcomes on Prospective Lipid Nanoparticle Shielding Materials.

Polyethylene glycol (PEG) is a proven component in lipid nanoparticles (LNPs). PEGylated nanoparticles are known to improve LNP circulation half-life and opsonization profiles. However, the drawbacks of PEG are hypersensitivity reactions due to pre-existing PEG antibodies and poor biodegradability. These factors warrant a search for alternatives. Some large peptides have shown potent stealth properties in protein delivery. Here, we demonstrate that repeating units of serine-threonine-glutamic acid-proline (STEP) peptide conjugated to dimyristoylglycerol (DMG-STEP) and incorporated into lipid n

Calcitonin Gene-Related Peptide Signalling in Alcohol Use Disorders: A Scoping Review.

ISSUE: Alcohol use disorders (AUD) represent a major global health burden, and currently available pharmacological treatments show limited and heterogeneous efficacy. Calcitonin gene-related peptide (CGRP) is a pleiotropic neuropeptide involved in stress regulation and neuroimmune signalling, and is an established therapeutic target in migraine. CGRP signalling may also be involved in alcohol-related mechanisms, raising interest in its potential relevance for AUDs. APPROACH: We conducted a scoping review of original studies investigating CGRP and CGRP-related pathways in the context of alcoho

Nature of False Peptide Identifications in Data-Independent Acquisition-Based Proteome Analysis.

Data-independent acquisition (DIA) mass spectrometry is increasingly used in proteomics because it offers a shorter analysis time at higher proteome coverage. Most existing DIA search engines rely heavily on fragmentation spectra and produce identification results, even when precursor ions are not detected. Although the latter is well-known, the errors it can lead to have received little attention. In this work, we studied false identifications in DIA, primarily due to the lack of a corresponding precursor ion envelope. Using a DIA data set with known UPS proteins spiked into Escherichia coli

Label-free quantification of cumulative cytosol-enriched peptide concentrations by mass spectrometry.

The development of label-free cell permeability assays for the detection and quantification of peptide therapeutics remains analytically challenging. Accurate and rapid evaluation of target-agnostic peptides and other beyond-rule-of-five (BRO5) compounds in the pharmaceutical industry would accelerate discovery programs aimed at intracellular targets by enabling earlier prioritization based on intracellular availability. We report a label-free assay designed to quantify intact intracellular peptides, rank-order permeability, and assess intracellular stability. Cumulative peptide levels were qu

Peptide binding at the gasdermin D exosite reveals the structural basis for targeting the site.

Gasdermin D (GSDMD) has been identified as a critical component of the inflammasome, an important intracellular signaling multi-protein complex. Abnormal activation of GSDMD has been linked to a variety of inflammatory diseases, including non-alcoholic steatohepatitis, inflammatory bowel disease and COVID-19, and has been linked to potential pathogenesis of septic shock. While small molecules have been identified to inhibit N-terminal domain (NTD) pore formation and membrane translocation, further clinical application of these molecules is currently limited due to narrow specificity. Here, we

Constrained diffusion-enabled construction of a highly expressed and robust peptide asparaginyl ligase.

Peptide asparaginyl ligases (PALs) hold great promise for peptide macrocyclization and protein bioconjugation, yet their broader application is limited by insufficient thermal stability and low recombinant expression. Here, we report FortiPAL-1, a diffusion-enabled PAL developed through an integrated computational-experimental strategy that couples de novo sequence generation with multi-model-guided mutagenesis and PAL-specific functional refinements. Compared with natural PALs, FortiPAL-1 exhibits markedly enhanced thermal stability (T m = 69 °C versus ∼50 °C for typical PALs) and achieves a

Safety and Efficacy of Approved and Unapproved Peptide Therapies for Musculoskeletal Injuries and Athletic Performance.

Peptides are short chains of amino acids with a unique pharmacological niche between small-molecule drugs and large proteins. Their use in sports medicine is rapidly expanding, driven by patient demand for accelerated injury recovery and performance enhancement. While numerous peptide drugs have undergone a rigorous approval process that evaluates both safety and efficacy, a parallel "gray market" of unapproved compounds has emerged, operating largely outside of regulatory oversight. Our objective is to present the pharmacological mechanisms, safety profiles, and regulatory status of prominent

Antimicrobial peptide chensinin-1b attenuates T2DM progression in atherosclerotic ApoE-/- mice.

AIMS: Epidemiological evidence suggests that atherosclerosis (AS) may precede or coexist with type 2 diabetes mellitus (T2DM); however, whether anti-atherosclerotic interventions can reduce T2DM risk remains unclear. Chensinin-1b (C-1b), an antimicrobial peptide derived from the skin secretions of Rana chensinensis, has previously demonstrated anti-atherosclerotic activity, suggesting a potential therapeutic effect against T2DM in the context of AS. METHODS: In an apolipoprotein E-knockout (ApoE-/-) mice model induced by high-fat diet (HFD), the developed process of pathological changes and i

Molecular arms race classifier for decrypting venom peptide and ion channel interactions.

Animal venoms comprise an astonishing number of peptides, proteins and small molecules. The diversity of venom compounds arises from evolutionary adaptations resulting in both offensive and defensive traits in predators and prey alike. This concept, referred to as "arms race", underpins the specificity and selectivity of venom compounds for certain molecular targets, like ion channels. Ion channels are essential regulators of cellular processes, and their dysfunction or dysregulation facilitates a wide range of diseases. Venom peptides and their derivatives are powerful modulators of ion chann

A Fluorescent Inhibitory Peptide Identifies α6β4 nAChR Distribution in the Rat Urinary Tract.

α6β4 nicotinic acetylcholine receptors (nAChRs) represent potential therapeutic targets for pain management; however, studies on the distribution and function of these receptors have been constrained by the scarcity of pharmacological tools. An analogue of α-conotoxin (α-CTx) LvIC from Conus lividus, [D1G, Δ14Q]LvIC, selectively inhibits heterologous rat α6/α3β4 nAChRs (where α6/α3 denotes the chimeric subunit) and represents a promising pharmacological tool. However, its limited stability may restrict further applications. Rational design based on the [D1G, Δ14Q]LvIC analogue led to the ident
