Malaria control is challenged by parasite drug resistance and inadequate vaccine protection. Although mosquito gut microbes regulate Plasmodium development, integrated analyses of microbiota-Plasmodium-host interactions are lacking. Innate immunity imbalance is the primary driver of disease pathogenesis. This review proposes a unified "microbiota-barrier-innate immunity" axis hypothesis. Under physiological conditions, gut microbiota modulates host innate immunity via metabolites and maintains barrier integrity. However, Plasmodium infection disrupts this axis, causing microbiota imbalance, me

Neospora caninum is an apicomplexan parasite that causes abortion in cattle, resulting in significant economic losses. There is no commercial treatment for neosporosis, and drug repositioning is a fast strategy to test possible candidates against N. caninum. In this article, we describe the effects of atovaquone, chloroquine, quinine, primaquine and tetracycline on N. caninum proliferation. The IC50 concentrations in N. caninum were compared to the current information based on previous studies for Plasmodium and Toxoplasma gondii, correlating to the described mechanisms of action of each teste

Malaria remains one of the deadliest diseases in the world today. Novel chemoprophylactic and chemotherapeutic antimalarials are needed to support the renewed eradication agenda. We have discovered a novel antimalarial acridone chemotype with dual-stage activity against both liver-stage and blood-stage malaria. Several lead compounds generated from structural optimization of a large library of novel acridones exhibit efficacy in the following systems: (1) picomolar inhibition of in vitro Plasmodium falciparum blood-stage growth against multidrug-resistant parasites; (2) curative efficacy after

The present study aims to develop a method for rapid diagnosis of malaria using loop-mediated isothermal amplification (LAMP) combined with a lateral flow device (LFD). By adding the biotin-labeled and fluorescein amidite-labeled loop primers to the LAMP reaction solution, the end product can be visualized on a LFD. The entire procedure takes approximately 42 minutes to complete, LAMP assay exhibited high sensitivity, as the detection limit was 0.01 pg/μL for all five Plasmodium species. It was demonstrated that all Plasmodium knowlesi (N = 90) and Plasmodium vivax (N = 56) were positively amp

Atovaquone is a hydroxy-naphthoquinone that is used to treat parasitic and fungal infections including Plasmodium falciparum (malaria), Pneumocystis jivorecii (pneumonia) and Toxoplasma gondii (toxoplasmosis). It blocks mitochondrial oxidation of ubiquinol in these organisms by binding to the ubiquinol oxidation site of the cytochrome bc(1) complex. Failure of atovaquone treatment has been linked to the appearance of mutations in the mitochondrially encoded gene for cytochrome b. In order to determine the optimal parameters required for inhibition of respiration in parasites and pathogenic fun

During the course of rodent malaria a marked decrease in the numbers of circulating lymphocytes within the peripheral blood occurred 2-4 days post-infection. Monocytes and polymorphs did not show the same degree of decline. For both avirulent Plasmodium yoelii and lethal Plasmodium berghei infections lymphocyte numbers returned to control levels by day 6-8 post-infection. While these levels were maintained until clearance of P. yoelii infection, a sustained and abnormal increase occurred during P. berghei infection. Early lymphocyte depletion was also observed following Babesia microti and Pla

We report a case of babesiosis in a traveler from India who was diagnosed with malaria on the basis of blood smears. Pan-Plasmodium PCR was positive, though species-specific assays were negative. Reexamination of blood smears and Babesia-specific PCR confirmed babesiosis. We highlight the overlapping clinical and diagnostic features of malaria and babesiosis and the potential cross-reactivity of Plasmodium primers in cases of babesiosis.

A novel compaction domain between the nuclear envelope and the plasma membrane is observed in intraerythrocytic Plasmodium knowlesi and Plasmodium chabaudi. The plasma membrane and the two nuclear membranes stick very closely together thus forming a heptalamina domain. This has an outer diameter of 14.3 +/- 1.1 nm and can be as long as 2/um. Fracture faces in this region appear to be devoid of the typical membrane-intercalating-particles. This compaction domain occurs only in ring-stages and early to mid trophozoites, but never in schizonts.

A case of congenital malaria due to Plasmodium malariae in one of two living Greek twins born in Germany is presented. In the child fever, anaemia and hepatosplenomegaly were first noticed at the age of two months. Plasmodium malariae parasites were demonstrated in the peripheral blood and bone marrow. The sera of mother and infant contained malaria antibodies. However the mother never showed clinical signs of malaria. Diagnosis and therapy of congenital malaria and the possible mode of transmission of this rare disease are discussed.

Glycosylphosphatidylinositols (GPIs) are complex glycolipids present on the surfaces of Plasmodium parasites that may act as toxins during the progression of malaria. GPIs can activate the immune system during infection and induce the formation of anti-GPI antibodies that neutralize their activity. Therefore, an antitoxic vaccine based on GPI glycoconjugates may prevent malaria pathogenesis. To evaluate the role of three key modifications on Plasmodium GPI glycan in the activity of these glycolipids, we synthesized and investigated six structurally distinct GPI fragments from Plasmodium falcip

Despite the relatively high frequency of imported malaria in metropolitan France, the transmission of malaria by transfusion is exceptional. The screening of donations to determine those at risk is performed by an interview, and by the testing of serology for defined groups of donors. However, the exclusion of a candidate 'at risk' as a blood donor, by a pre-donation interview, is not completely mastered and the discrimination by biological examination lacks sensitivity, as much for methodological reasons as for reasons linked to the complex parasitic pathogenic agent (Plasmodium ssp.), as for

Malaria remains a major global health challenge, particularly in sub-Saharan Africa where Anopheles mosquitoes transmit the Plasmodium parasites. Resistance to insecticides remains an obstacle in spite of the efforts to control malaria vector. The interaction between Plasmodium parasites and mosquito vectors, with a focus on the immunity of mosquitoes and approaches to combat malaria, is examined in this review. This review explores the potential of genetic approaches including CRISPR-Cas9, Wolbachia, RNA interference (RNAi), and symbiont-based strategies for the control of malaria vector. The

Malaria is one of the deadliest infectious diseases in the world, annually responsible for over 400,000 deaths. It is caused by parasites of the genus Plasmodium, which undergo remarkable structural changes during their development within different cells across various hosts. An important approach to understand the structural basis of biochemical and physiological processes during Plasmodium infection has been the quantitative measurement of dimensional parameters obtained by different microscopy techniques. In this regard, sample preparation, particularly electron microscopy protocols that re

BACKGROUND: Malaria is an infectious disease caused by parasites of the genus Plasmodium, of which Plasmodium vivax and Plasmodium falciparum are the major species that cause the disease in humans. As there are relatively few alternatives for malaria treatment, it is necessary to search for new chemotherapeutic options. Colombia possesses a great diversity of plants, which are potential sources of new compounds of medical interest. Thus, in this study the antiplasmodial effect of extracts from two species of plants from the families Simaroubaceae and Picramniaceae (Picramnia latifolia and Picr

BACKGROUND: Malaria is one of the most important parasitic infectious diseases for which almost half of the world's population is at risk. Although several diagnostic methods are now available to detect the infection, more sensitive and applicable tests are still required in the field. The loop-mediated isothermal amplification (LAMP) method is a DNA amplification tool in which the DNA amplification can be achieved by incubation at a stable temperature. A malaria detection kit based on this methodology has already been commercialized and is being used in some countries. The kit includes two re

BACKGROUND: A deep understanding of the local epidemiology of malaria is essential for the design and implementation of setting-specific control and elimination efforts. In Côte d'Ivoire, new initiatives are underway to reduce the burden of malaria, which requires high-quality longitudinal data. The epidemiology of malaria was studied in the Taabo health and demographic surveillance system (HDSS) in south-central Côte d'Ivoire and implications for control are discussed.
METHODS: Two cross-sectional surveys were carried out in the rainy season of June/July in 2010 and 2011. Inhabitants of appr

Malaria is a disease caused by parasites of Plasmodium species. It is responsible for around 1-2 million deaths annually, mainly children under the age of 5. It occurs mainly in tropical and subtropical areas. Malaria is caused by five Plasmodium species: P. falciparum, P. malariae, P. vivax, P. knowlesi and P. ovale. Mosquitoes spread the disease by biting humans. The malaria parasite has two stages of development: the human stage and the mosquito stage. The first stage occurs in the human body and is divided into two phases: the liver phase and the blood phase. The invasion of erythrocytes b
