Species-Agnostic Tools for HPAI H5N1 Antibody and Antigen Detection in Dairy Cows, Chickens, and Humans.

High Pathogenicity Avian Influenza (HPAI) A (H5N1) circulates in wild birds, causing outbreaks in domestic poultry, but recently became enzootic in the United States spilling over into various mammals, including cattle. The zoonotic potential of H5N1 clade 2.3.4.4b emphasizes the need for tools for surveillance in mammalian and avian species. We developed two species-agnostic assays using the MESO SCALE DISCOVERY® (MSD) platform to enhance current surveillance methods: a sandwich immunoassay for influenza A nucleoprotein (NP) antigen, and a bridging serology assay for influenza A/H5 specific a

Detection of H5N1 HPAIV Clade 2.3.4.4b Avian Influenza Virus in Backyard Chickens in Costa Rica.

Influenza A virus is a segmented, negative-sense RNA virus. Since the early 2020s, H5 clade 2.3.4.4b viruses have spread widely across Europe, Africa, and Asia, affecting wild birds and poultry. Costa Rica reported its first H5 clade 2.3.4.4b avian influenza virus (AIV) case on 19 January 2023. This study describes an outbreak in backyard chickens and ducks. Initial serum samples collected on 24 January showed three chickens negative for AIV, while one duck tested positive by ELISA and agar gel immunodiffusion (AGID). During a second visit on 27 January, three of four chicken sera collected te

Valorization of Food By-Products into a Zero-Waste Broth: Effects of Yeast Extract on Composition and Sensory Properties.

Food losses and waste occur throughout the food production chain, creating an urgent need for their recovery and valorization into sustainable food products. Sensory acceptance is crucial for consumer interest and market success and can be enhanced using yeast extract. However, strategies that simultaneously valorize meat and vegetable by-products into nutritionally and functionally enriched products while ensuring sensory acceptability have been scarcely investigated. This study examined whether poultry carcasses after mechanical meat separation, combined with root vegetable processing residu

Protective Effect of Synbiotic Supplementation Against Salmonella Typhimurium Infection in Young Broiler Chickens.

Salmonella infection in poultry remains a major concern due to its economic impact and public health risks. Synbiotics have emerged as a natural strategy to improve gut health and resistance to enteric pathogens. This study evaluated the immune response and early protective effects of a synbiotic in broiler chickens using a reduced in vivo model of Salmonella Typhimurium infection. A total of 44 broiler chickens were randomly assigned to four dietary treatments: control (CT), control plus synbiotic (CT + Syn), challenged (ST), and challenged plus synbiotic (ST + Syn). Birds were challenged at

Heat Stress-Derived Plasma Extracellular Vesicles Protect Hepatocytes in Chickens by Suppressing MYD88/NF-κB/MAPK Signaling.

Heat stress is a major systemic challenge in poultry, but the role of circulating extracellular vesicles (EVs) in liver-directed adaptation remains unclear. This study investigated whether plasma-derived EVs from heat-stressed chickens (HS_EV) mediate hepatoprotective responses under thermal stress. EVs were isolated from the plasma of control and heat-stressed chickens and characterized by morphology, size distribution, and marker expression. Their biodistribution in vivo and uptake by primary hepatocytes in vitro were also evaluated. Hepatocyte injury was induced by heat exposure, and the ef

Structural and histological changes in Ascaridia galli exposed in vitro to underutilized anthelmintic plant, Meyna laxiflora Robyns seed extract.

The methanol seed extract of Meyna laxiflora Robyns was evaluated in vitro against the most prevalent poultry nematode, Ascaridia galli, to assess its anthelmintic potential employing the adult worm mortality assay. Worms were subjected to varying concentrations (0.05-100mg/mL) of the extract dissolved in physiological phosphate-buffered saline solution containing 0.1% dimethyl sulphoxide, and incubated at 37 ± 1°C. A parallel assay was performed with the broad-spectrum anthelmintic drug, Albendazole. Exposure to the extract induced complete inactivation and flaccid paralysis, culminating mort

Activation of the ATM-Chk2 DNA damage response pathway by Newcastle disease virus enhances viral replication.

Newcastle disease (ND), caused by virulent strains of the Newcastle disease virus (NDV), is a highly contagious disease that poses significant economic burdens on the global poultry industry. The DNA damage response (DDR) is a critical cellular mechanism that detects and repairs genomic damage to maintain cellular integrity. While viral infections are known to modulate DDR pathways to either inhibit or enhance viral replication, the interaction between NDV and host DDR remains largely underexplored. Here, we demonstrate that NDV infection induces significant DNA damage in DF-1 cells and activa

Nutritional Modulation of Epigenetic Responses to Heat Stress in Farm Animals: Evidence, Limitations and Perspectives.

Rising global temperatures pose a serious threat to the productivity of farmed animals. Prolonged heat exposure impairs growth, reproduction and immunity; suppresses feed intake; and disrupts intestinal barriers, leading to substantial economic losses. Conventional cooling strategies offer limited relief, spurring research into the underlying molecular dysregulation. Epigenetic mechanisms link heat stress (HS) to gene expression. Specific nutrients that modulate these mechanisms could help mitigate HS. This review critically summarizes how methyl donors, short-chain fatty acids (SCFAs), vitami

L-Selenomethylselenocysteine Improves Growth Performance, Antioxidant Status, and Meat Quality in White-Feathered Broilers More Effectively than Selenomethionine.

Selenium (Se) is essential in poultry nutrition for antioxidant defense through selenoproteins. While selenomethionine (Se-Met) is commonly used, L-selenomethylselenocysteine (L-SeMC) may offer more favorable responses for selected measured variables. This study compared Se-Met and L-SeMC effects on growth, antioxidant status, and meat quality in broilers. Six hundred and sixty one-day-old male Cobb 500 broiler chickens were randomly assigned to 11 dietary treatments, with six replicate pens of 10 birds per treatment, comprising a basal control diet and 10 selenium-supplemented diets arranged

Role of O-linked glycosylation modification on internalization and replication of avian leukosis virus subgroup J.

Upon infection, viruses reprogram the host metabolic system to hijack metabolic resources for proliferation. Avian leukosis virus subgroup J (ALV-J), an avian oncogenic virus, poses significant challenges to the poultry industry. ALV-J infection upgrades monosaccharide N-acetylgalactosamine (GalNAc) and galactosyltransferase (core 1 β3-Gal-T) in DF-1 cells, both of which are crucial for O-linked glycosylation. Addition of GalNAc or overexpression of core 1 β3-Gal-T in DF-1 cells can promote ALV-J replication. ALV-J envelope protein (Env) undergoes complex post-translational modifications. Two
