Metastatic Signet-Ring Cell Carcinoma of the Bladder in Cerebrospinal Fluid.

Primary bladder signet-ring cell carcinoma (SRCC) is extremely rare and associated with an aggressive course. To our knowledge, we describe the first metastatic bladder SRCC identified in cerebrospinal fluid (CSF). A 68-year-old male with 1 year history of primary bladder SRCC with spinal metastasis presented with multiple falls and loss of consciousness. Brain imaging showed high signal in the frontoparietal sulci and superior cerebellum. CSF analysis was significant for increased leukocytes with monocyte predominance while protein and glucose values were within normal range. There was a hype

Visible-light driven biofuel cell based on hierarchically branched titanium dioxide nanorods photoanode for tumor marker detection.

In this work, a novel sensing platform based on visible light driven biofuel cell (BFC) has been facilely designed for sensitive detection of prostate-specific antigen (PSA) with the photo-response bioanode, realizing the dual route energy conversion of light energy and chemical energy to electricity. The hierarchical branched TiO2 nanorods (B-TiO2 NRs) decorated with CdS quantum dots (QDs) act as the substrate to confine glucose dehydrogenase (GDH) for the visible light driven glucose oxidation at the bioanode. Three dimensional flowers like hierarchical carbon/gold nanoparticles/bilirubin ox

Serum and urine tissue kallikrein concentrations in male-to-female transsexuals treated with antiandrogens and estrogens.

BACKGROUND: The expression of human tissue kallikrein genes is regulated by steroid hormones, but most studies have been conducted with cancer cell lines. Our purpose was to examine serum and urinary tissue kallikrein concentration changes in male-to-female transsexuals before and after treatment with antiandrogens and estrogens. METHODS: Thirty-five male-to-female transsexuals receiving cyproterone acetate and estrogens (orally or transdermally) were included in this study. Serum and urine samples were collected before initiation of therapy and 4 and 12 months post therapy. ELISAs were used

Hormonal changes during GnRH analogue therapy in children with central precocious puberty.

Gonadotropin releasing hormone analogues (GnRHa) have been used for treatment of central precocious puberty (CPP) for more than 15 years. They are generally considered safe although data on potential long-term side effects are scarce. However, GnRHa therapy has profound effects on both the hypothalamopituitary-gonadal axis as well as on growth hormone (GH) secretion. Gonadal activity is increased in children with CPP; during GnRHa therapy secretion of gonadal hormones is suppressed as reflected by measurements of LH, FSH, and estradiol/testosterone. More recently, studies of levels of inhibin

Prostate Cancer Incidence and Disease Characteristics in Transgender Women Receiving Gender-affirming Hormone Therapy: A Systematic Review with Meta-analysis.

BACKGROUND AND OBJECTIVE: Transgender women (TGW) retain lifelong prostate cancer (PCa) risk despite gender-affirming hormone therapy (GAHT), yet its association with PCa incidence and severity is poorly synthesised. This study aimed to quantify pooled PCa incidence in TGW on GAHT relative to cisgender men and characterise disease severity. METHODS: This is a systematic review with meta-analysis per Preferred Reporting Items for Systematic Reviews and Meta-Analyses 2020 (International Prospective Register of Systematic Reviews: CRD420261369142). Five databases were searched from inception to

Investigation on Automatic Classification of Bone, Lymph Node, and Local Lesions in CT Images of Prostate Cancer Patients.

Accurate assessment of different types of prostate cancer (PCa) lesions is a crucial part of treatment planning. Following a computed tomography (CT) or a positron emission tomography (PET)/CT imaging scan, a significant amount of time is required from the clinicians to detect lesions and characterize them by their anatomical locations. To facilitate the development of new image analysis applications, we investigated the potential of automated classification of PCa lesions between bone metastases, lymph node metastases, local lesions, and other PCa lesions in CT images via machine learning (ML

Integrated Prognostic Score in Metastatic Castration-resistant Prostate Cancer Treated with Cabazitaxel - A CABASTY Post Hoc Analysis Validated by Two International Prospective Phase 3 Trials.

BACKGROUND AND OBJECTIVE: Optimal therapy after docetaxel and an androgen receptor pathway inhibitor (ARPI) in metastatic castration-resistant prostate cancer (mCRPC) remains debated. We aimed to build and validate the cabascore, a prognostic and predictive score for patients with mCRPC receiving cabazitaxel, to guide treatment strategy. METHODS: In this post hoc analysis of the randomized CABASTY trial, the cabascore was derived from multivariable Cox model hazard ratios for overall survival (OS). The population was stratified into three groups using log-rank optimization for score threshold

Correlation between Serum Testosterone and Tumor Grade in a Black African Population of Men with Prostate Cancer.

BACKGROUND: In the past few decades, it was assumed that high testosterone levels were associated with increased risk of prostate cancer (CAP) while low testosterone was protective. However, more recent studies reported an association between low serum testosterone and high-grade CAP. These studies were largely done in the Western world, thus leaving a knowledge gap as regards such a relationship in Africans. Therefore, the aim of this study was to bridge this gap in knowledge by assessing the nature of the relationship between serum testosterone and tumor grade in a black African population o

Development and validation of a machine learning-based early warning model for bone metastasis in newly diagnosed prostate cancer.

BACKGROUND: Bone metastasis (BM) is common in newly diagnosed prostate cancer (PCa), particularly in patients with advanced disease at presentation. However, the indications for bone scintigraphy remain inconsistent and may lead to unnecessary imaging in low-risk patients. This study aimed to develop and validate a machine learning model for individualized prediction of BM in patients with newly diagnosed PCa. METHODS: We retrospectively collected data from 327 patients with newly diagnosed PCa from two tertiary hospitals. Patients were randomly assigned to a training set (n = 229) and an int

Discovery of 177Lu-PSMA-MN4 as an improved PSMA-targeted radioligand with enhanced antitumor efficacy.

Prostate-specific membrane antigen (PSMA)-targeted radioligand therapy with 177Lu-PSMA-617 improves outcomes in metastatic castration-resistant prostate cancer (mCRPC), yet a substantial subset of patients experiences disease progression, underscoring the need for next-generation agents with improved pharmacological profiles. Guided by the PSMA-617/PSMA co-crystal structure (PDB: 8BOW) and molecular dynamics (MD) simulations, the present study rationally designs seven novel PSMA-617 derivatives (PSMA-MN1-7) to enhance target engagement and pharmacokinetic behavior. Among these, 177Lu-PSMA-MN4

Oncolytic virus OH2 induces PD-L1 upregulation via NF-κB signaling and synergizes with anti-PD-L1 therapy in prostate cancer through a targeted extracellular vesicle delivery system.

BACKGROUND: Prostate cancer (PCa) is a prevalent malignancy with limited treatment options for advanced stages. Oncolytic virotherapy represents a promising immunotherapeutic approach, but its efficacy and underlying mechanisms in PCa, particularly concerning immune checkpoint regulation, remain unclear. METHODS: The antitumor effects of the oncolytic virus oncolytic herpes simplex virus type 2 (OH2) were evaluated in PCa cell lines and mouse models. Transcriptome sequencing, western blot, chromatin immunoprecipitation-sequencing-quantitative PCR, and flow cytometry were employed to investiga

Biochemical Recurrence-Free Survival After Radical Prostatectomy in Patients with High-Risk Prostate Cancer: A Nationwide Study of PSMA PET/CT Versus Conventional Imaging.

Prostate-specific membrane antigen (PSMA) PET/CT offers excellent accuracy in the staging of high-risk prostate cancer (PCa), yet outcomes-based evidence of the clinical benefit remains limited. We aimed to determine whether the rates of biochemical recurrence-free survival (BRFS) are influenced by PSMA-based staging compared with that of conventional imaging among patients undergoing radical prostatectomy (RP), using real-world data and exploiting regional variation in the adoption of PSMA PET/CT across Denmark. Methods: This nationwide cohort study included men with high-risk PCa who underwe
