Assessing recoveries of branched-chain volatile fatty acids or amino acids in dual-flow continuous cultures limited in rumen-degraded protein without or with supplemental leucine.

Ruminal bacteria can either synthesize branched-chain amino acids (BCAA) from central metabolites or from reductive carboxylation and transamination of the branched-chain volatile fatty acids (BCVFA). There is bidirectional mixing within a metabolic pair (i.e., isoleucine/2-methylbutyrate, leucine/isovalerate, and valine/isobutyrate). The keto acid of valine is the intermediate in its synthesis but also branches off in the leucine synthesis pathway. Therefore, we hypothesized that interconversion of 13C-isobutyrate with its keto acid could label both valine and, to a lesser extent, leucine, es

Physiological Responses in the Hepatopancreas of Litopenaeus vannamei to Carbonate Alkalinity Stress and Subsequent Recovery: Integration of Antioxidant, Immune, and Metabolic Profiles.

High carbonate alkalinity (CA) is a major constraint on shrimp culture in saline-alkaline waters. The hepatopancreas is central to shrimp immunity and metabolic regulation. Litopenaeus vannamei underwent a 7-day exposure period to 5 mmol/L CA, followed by a subsequent 7-day recovery phase after removal of the stressor. The physiological regulatory mechanism of the hepatopancreas during CA stress and recovery was investigated by integrating multiple biological levels including histomorphology, antioxidant and immune indices, energy metabolism, and metabolite profiles. Results showed that CA str

POU4F1 Promotes the Primary Resistance of Melanoma to Anti-PD-1 Therapy by Regulating Glycolysis Through METTL1-Mediated m7G Methylation of PKM2.

Melanoma, a highly malignant tumor originating from melanocytes, has seen a significant increase in global incidence, particularly among the elderly. Anti-PD-1 monoclonal antibodies, which activate the immune system to attack cancer cells by blocking the PD-1/PD-L1 signaling pathway, have improved survival rates but face challenges such as innate resistance. This study enrolls 37 melanoma patients and 7 benign nevus patients, with tissue samples collected for analysis. RT-qPCR and Western blot are used to quantify the expression of the target protein. Flow cytometry is utilized to analyze immu

Label-Free Proteomic Profiling of the dvls2 (CL2006) Caenorhabditis elegans Alzheimer's Disease (AD) Model Reveals Conserved Molecular Signatures Shared With the Human AD Brain.

Alzheimer's disease (AD) is the most common form of dementia, posing significant challenges to cognitive, emotional, social, and financial well-being. The biochemical and molecular pathways associated with AD are complex, making it difficult to study and simulate in patients or through in vitro research. Thus, animal models play a crucial role in investigating the development and progression of AD. One widely used model in neuroscience studies is the free-living nematode Caenorhabditis elegans (C. elegans). The development of transgenic animals has allowed for the construction of the dvls2 (CL

The human gut microbiome and its metabolic pathway dynamics before and during HIV antiretroviral therapy.

The human gut microbiome consists of highly complex microbial populations that play important roles in disease pathogenesis. Changes in the gut microbiome are associated with HIV infection. However, it is unclear whether gut microbiome dysbiosis is causally linked to disease or whether it simply reflects disease-induced changes in the host immune and metabolic systems. This study aims to describe and compare intestinal microbial compositions and derived metabolic pathways in people living with HIV (PL-HIV) from prior to antiretroviral therapy (ART) to 6 and 12 months after ART initiation, comp

The Causal Role of Gut Microbiota and Immune Cell Mediation in Gastroenteropancreatic Neuroendocrine Neoplasms: A Mendelian Randomization Study.

INTRODUCTION: Gastroenteropancreatic neuroendocrine neoplasms (GEP-NENs) are a diverse group of tumors arising from neuroendocrine cells, often characterized by slow growth and subtle symptoms, leading to advanced-stage diagnoses. The gut microbiota (GM) has been implicated in various gastrointestinal malignancies, but its relationship with GEP-NENs remains unclear. METHODS: A two-sample MR analysis was employed using genome-wide association study data to explore causal associations between GM and GEP-NENs. Genetic variants significantly associated with GM traits were selected as instrumental

Embryonic hypoxia alters cardiac gene expression patterns in American alligators, Alligator mississippiensis.

How environmental conditions during embryogenesis shape development, physiology, and phenotype is a key question for understanding the roles of plasticity and environmental factors in determining organismal traits. Answering this question is essential for revealing how early-life environmental variation drives adaptive responses and influences evolutionary processes. Here we examine how hypoxia impacts cardiac gene expression during embryonic development in the American alligator (Alligator mississippiensis). Eggs were incubated in normoxic (21% O2) or hypoxic (10% O2) conditions from 20% to 9

Carbapenem-resistant Enterobacter hormaechei uses mucus metabolism to facilitate gastrointestinal colonization.

The emergence and global spread of carbapenem-resistant Enterobacter cloacae complex species present a pressing public health challenge. Carbapenem-resistant Enterobacter spp. cause a wide variety of infections, including septic shock fatalities in newborns and immunocompromised adults. The intestine may be a major reservoir for these resistant strains, either by facilitating contamination of fomites and transfer to susceptible individuals, or through translocation from the gut to the bloodstream. For this reason, we sought to establish a neonatal mouse model to investigate the mechanisms unde

Delayed Diagnosis of Glutaric Aciduria Type 1: A Case Report.

Newborn screening (NBS) is performed to screen for conditions where early intervention can make a difference in a patient's prognosis. We present the case of a patient with glutaric aciduria type 1 (GA1) that was missed on NBS but was diagnosed through exome sequencing (ES) at eight years of age. We report the case of a patient who was born in Florida in 2011 and underwent routine NBS, which was negative for all tested conditions, including amino acidemias. At 11 months of age, the patient presented with seizures. Upon physical examination, she had hypotonia, ptosis, and macrocephaly. A urinar

Gut microbiome-mediated nutrients alter opportunistic bacterial growth in peritonitis.

Peritonitis is a well-known complication of bowel perforation and abdominal surgery, leading to sepsis and high mortality. Despite its prevalence and severity, the pathogenesis of peritonitis remains incompletely understood, limiting our ability to develop targeted medical therapies. Specifically, little is known about the determinants of the peritoneal nutrient environment for pathogens. The gut microbiome is a well-established source of infectious bacteria in peritonitis, but whether it also modulates levels of nutrients that enable and sustain these infections remains unknown. Using multipl

N 1-methylnicotinamide promotes age-related cochlear damage via the overexpression of SIRT1.

Age-related hearing loss (ARHL) is a complex condition with genetic, aging, and environmental influences. Sirtuins, particularly SIRT1, are NAD-dependent protein deacetylases critical to aging and stress responses. SIRT1 is modulated by nicotinamide N-methyltransferase (NNMT) and its product, N1-methylnicotinamide (MNAM), which influence ARHL progression. While SIRT1 is protective under certain conditions, its overexpression may paradoxically exacerbate hearing loss. This study examines MNAM supplementation's impact on SIRT1 expression and ARHL in low-fat diet (LFD)-fed B6 and CBA mice. Mice w
