Kaposi Sarcoma Associated with Pemphigus Vulgaris in the Setting of Immunosuppressive Therapy: Case Report and Review of Literature.

INTRODUCTION: Pemphigus vulgaris (PV) is a chronic autoimmune blistering disorder characterized by intraepidermal blisters affecting the skin and mucous membranes. The primary treatment for PV is systemic corticosteroid therapy, which is often prolonged due to the disease's chronic and relapsing course. The interplay between autoimmunity and long-term immunosuppressive treatment can predispose patients to secondary complications, including Kaposi sarcoma (KS), an angioproliferative tumor primarily involving the skin and soft tissues. When KS develops in the context of immunosuppressive therapy

Clear Cell and Histiocytic/Dendritic Cell Sarcomas: Clinical Outcomes, Molecular Features, and Diagnostic Pitfalls.

Background: Dendritic and histiocytic cell sarcoma (DHCS) and clear cell sarcoma (CCS) are ultra-rare soft-tissue sarcomas characterized by diagnostic ambiguity, limited treatment guidelines, and poor outcomes. Their rarity has restricted the development of evidence-based management strategies, leaving clinical decisions reliant on small case series and institutional experience. DHCS typically presents without a unifying molecular driver and is often misclassified without comprehensive immunophenotyping. CCS is defined by EWSR1-ATF1/CREB1 fusions but exhibits low responsiveness to conventional

Testicular histiocytic sarcoma: a case report and literature review of an uncommon histological type of urogenital malignancy.

BACKGROUND: Testicular tumors are relatively uncommon urogenital malignancies, accounting for only 1-2% of all cancers in men, with germ cell tumors representing the predominant histopathological subtype. Histiocytic sarcoma (HS) is a rare and aggressive malignancy that typically arises in extranodal sites, and its standard treatment strategy has yet to be established. Here, we report a rare case of testicular histiocytic sarcoma, which has been sparsely documented in previous studies and for which clinical experience remains limited, thereby providing valuable insights into its diagnostic cha

Recombinant production, biochemical characterization, and in vitro evaluation of an EWS-FLI1-derived peptide for Ewing sarcoma.

Ewing sarcoma is an aggressive malignancy characterized by the EWS-FLI1 fusion oncoprotein, which remains challenging to target because of its intrinsically disordered nature and lack of intrinsic enzymatic activity. EWSPEP, a peptide derived from the EWS-FLI1 fusion junction region, has previously been investigated in chemically synthesized form; however, its recombinant production has not been reported. In this study, recombinant EWSPEP (rEWSPEP) was expressed in Escherichia coli BL21 Star (DE3) using the pET-45b expression system and purified by Ni2+ affinity chromatography, yielding approx

Next-generation sequencing in head and neck sarcoma: a single-centre institutional experience and review of the literature.

Head and neck sarcomas (HNS) are rare, heterogeneous malignancies representing less than 1% of head and neck cancers. Their complex anatomy and overlapping morphologies pose significant challenges for traditional diagnosis. We aimed to evaluate the clinical utility of next-generation sequencing (NGS) within a tertiary referral centre and synthesise these findings with current global molecular standards. We conducted a retrospective review of an original, previously unpublished, cohort of 12 patients with histologically verified HNS treated at University College London Hospital (UCLH) between 2

Microenvironment of Sarcomas Associated and Not Associated with Inoculation Sites in Domestic Cats: The Role of Metalloproteinases and Angiogenic Factors.

Soft tissue sarcomas are malignant neoplasms originating from mesenchymal cells. In cats, there are two groups: inoculation site-associated sarcomas (ISSs) and non-inoculation site-associated sarcomas (NISSs). The aim of this study was to investigate the immunoexpression of metalloproteinases (MMPs) 2 and 9, tissue inhibitors (TIMPs) 1 and 2, and the main molecules related to angiogenesis and vascular count in both sarcoma groups; compare their expression; and evaluate their association with angiogenesis. Both tumor groups expressed all the molecules evaluated. A positive correlation was detec

Next-Generation Sequencing Refines Diagnosis and Expands Precision Medicine Opportunities in Soft Tissue Sarcomas.

Soft tissue sarcomas (STSs) are a heterogeneous group of rare mesenchymal malignancies with overlapping morphological and immunohistochemical features, often making definitive diagnosis challenging. Recent advances in next-generation sequencing (NGS) have enabled the identification of recurrent molecular alterations that contribute to tumor classification, prognostic stratification, and precision oncology approaches. This retrospective study aimed to evaluate the diagnostic and clinical impact of molecular profiling in pediatric soft tissue sarcomas using the Oncomine Childhood Cancer Research

Pioneering women in musculoskeletal pathology: A review highlighting their scientific achievements and remarkable impact in sarcoma care.

Bone and soft tissue pathology - perhaps more than in any other surgical pathology subspecialty - has been profoundly shaped by a cadre of trailblazing female pathologists who have seamlessly integrated morphologic expertise, molecular innovation, and educational leadership. While no single review can capture the full breadth of contributions made by women in this field, this article highlights the remarkable achievements of six such leaders - Drs. Sharon W. Weiss, Adrienne M. Flanagan, Julia A. Bridge, Cristina R. Antonescu, Judith V.M.G. Bovée, and Jeanne M. Meis - whose careers collectively

Distinct T cell functions enable efficient immunoediting and prevent tumor emergence of developing sarcomas.

T cells edit tumors by eliminating neoantigen-expressing tumor cells. Yet, how and when this is achieved remains uncertain. Using a murine sarcoma model with fluorescent neoantigens, we found that tumors developed later and in fewer T cell-sufficient mice (∼53% penetrance) than T cell-deficient mice (∼100%). With T cells, all emergent tumor cells had silenced neoantigens, but neoantigen-negative tumor cells were also present in every T cell-deficient mouse. This suggested silencing was necessary but not sufficient for outgrowth. Genetic removal of neoantigens restored tumor penetrance if imple

Differentiation of uterine fibroids and sarcomas by MRI and serum LDH levels: a multicenter study of the KAMOGAWA study.

OBJECTIVE: In the differential diagnosis between uterine fibroids and uterine sarcomas, real-world magnetic resonance imaging (MRI) diagnostic information is scarce; furthermore, high diagnostic sensitivity is important in clinical practice. We previously developed a diagnostic algorithm to detect uterine sarcoma with high sensitivity using simple MRI images and serum lactate dehydrogenase (LDH) levels. In this multicenter study, we investigated the preoperative diagnosis of sarcoma in the real world and further validated the usefulness of our diagnostic algorithm. METHODS: Of 154 uterine sar

Identification of high-risk age groups for cisplatin-induced nausea and vomiting in young patients with sarcoma.

BACKGROUND: Young age has been reported as a patient-related risk factor for chemotherapy-induced nausea and vomiting (CINV). However, there have been few reports of the status of CINV limited to younger patients. In this study, CINV status after cisplatin administration was investigated in young patients with sarcoma. METHODS: This retrospective, observational study included patients younger than 65 years with sarcoma who received cisplatin-containing chemotherapy at the Department of Orthopaedic Surgery, Cancer Institute Hospital Ariake, Japan, between January 2011 and December 2020. Orthopedic Surgical Management of Soft Tissue Sarcomas in a Referral Center: 5-Year Oncological Outcomes.

INTRODUCTION: Soft tissue sarcomas (STS) are malignant tumors of connective tissue, characterized by a wide heterogeneity in their presentations, resulting in variable therapeutic strategies between reference centers. However, clinical outcomes from an orthopedic and trauma surgery department of university hospitals are poorly documented. We evaluated the 5-year local recurrence rate of patients managed according to our local strategy and operated in our department, which is certified for the management of both soft tissues and bone sarcomas. We also assessed overall survival, metastasis and r

Myeloid sarcoma presenting as intestinal obstruction: A case report of the first presentation of acute myeloid leukemia.

INTRODUCTION AND IMPORTANCE: Myeloid sarcoma (MS), a rare extramedullary tumor composed of immature myeloid cells, most commonly affects the skin, lymph nodes, and gastrointestinal tract. Rarely, it presents in isolation or in pediatric patients. Misdiagnosis is frequent due to its morphological similarity to lymphomas and lack of clinical context. Early identification is crucial as MS often precedes or accompanies acute myeloid leukemia (AML) and requires immediate treatment. CASE PRESENTATION: A two-and-a-half-year-old female presented with fever, oral ulcers, abdominal pain, and bleeding p
