Manipulation of the somatosensory cortex modulates stimulus-induced repetitive ear movements in a seizure-sensitive strain of gerbil.

Some Mongolian gerbils (Meriones unguiculatus) respond to stimulation by seizures, the pattern of which changes progressively during development. We previously established a seizure-sensitive strain, MGS/Idr, in which all animals exhibit such stimulus-induced seizures. We have now noted that all adults of this strain also show repetitive backward movements of the ears at the ears at the beginning of stimulus-induced seizures, although the incidence varies with the individual. We examined whether the cerebral cortex was involved in these movements and found that electrical stimulation of an are

Perilesional gliosis and seizure outcome: a study based on magnetization transfer magnetic resonance imaging in patients with neurocysticercosis.

We studied the effect of perilesional gliosis on seizure recurrence after stopping antiepileptic drug (AED) therapy in 108 patients with a solitary cysticercal brain cyst. All patients received albendazole therapy in the beginning, and magnetic resonance imaging (MRI) done after 2 seizure-free years showed complete disappearance of the lesion in 67, partial degeneration in 12, and healing by calcification in 29. The gliosis, which was not visible on initial MRI, was observed near the lesion in 22 (20%) patients on T1-weighted magnetization transfer spin-echo (MTSE) MRI. Initial seizure control

Electroencephalographic and histological characteristics of a model of limbic status epilepticus permitting direct control over seizure duration.

Status epilepticus is a neurological emergency associated with substantial morbidity and mortality. Experimental and clinical investigations suggest that prolonged seizure activity is associated with injury to vulnerable neurons. Compounds with neuroprotective properties may minimize such injury. Existing methods of inducing experimental status epilepticus result in seizure activity of variable duration and neuronal injury of variable degree. To minimize such variability, status epilepticus may be stopped with anticonvulsants, but this limits the ability to screen for independent neuroprotecti

ESR spectroscopy for analysis of hippocampal elimination of a nitroxide radical during kainic acid-induced seizure in rats.

To analyze the balance between free radical production and elimination during seizure, techniques were developed to monitor the sequential changes in the level of an exogenous free radical by ESR in the brains of freely moving rats given kainic acid (KA) to induce seizures (n = 6) and control rats given saline (n = 6). The hippocampus of each rat was perfused with a nitroxide radical solution for 120 min by in vivo microdialysis. Then an intraperitoneal injection of KA or the physiological saline was given to each rat, and the perfusate was changed to Ringer's solution. ESR analysis of sequent

Rapid seizure-induced reduction of benzodiazepine and Zn2+ sensitivity of hippocampal dentate granule cell GABAA receptors.

Fast synaptic inhibition in the forebrain is mediated primarily by GABA acting on GABAA receptors (GABARs). GABARs are regulated by numerous positive (barbiturates, benzodiazepines, and neurosteroids) and negative (picrotoxin, bicuculline, and Zn2+) allosteric modulators. The sensitivity of GABARs to GABA and to allosteric modulators changes gradually during normal development, during development of chronic epilepsy, and after prolonged exposure to GABAR agonists. Here we report the development of rapid functional plasticity of GABARs occurring over 45 min of continuous seizures (status epilep

Effect of diaphragmatic respiration with end-tidal CO2 biofeedback on respiration, EEG, and seizure frequency in idiopathic epilepsy.

Breathing rate (RR), end-tidal percent CO2, and EEG were obtained in three groups: psychiatric referral subjects presenting with anxiety, panic phobia, depression and migraine; a group of idiopathic seizure sufferers; and a group of asymptomatic controls. Virtually all the noncontrol subjects were found to show moderate to severe hyperventilation and the accompanying EEG dysrhythmia. The seizure group subjects were taught diaphragmatic respiration with end-tidal percent CO2 biofeedback. The training normalized their respiration and altered their EEGs and seizure frequency.

Anticonvulsant activity of intracerebroventricularly administered glial GABA uptake inhibitors and other GABAmimetics in chemical seizure models.

The antiseizure activities of glial or neuronal GABA uptake inhibitors and GABA agonists were compared following intracerebroventricular administration in 2 acute models of chemoconvulsion in rats. The glia-selective GABA uptake inhibitor, 4,5,6,7-tetrahydroisoxazolo[4,5-c]pyridin-3-ol (THPO), given at doses of 100-750 micrograms, i.c.v., protected against maximal pentylenetetrazol (PTZ) seizures and increased the latency to isonicotinic acid hydrazide (INH) seizures for at least 1 h following central administration. THPO failed to increase PTZ seizure thresholds. In contrast, the more potent

Acute neurogenic pulmonary oedema following generalized tonic clonic seizure. A case report and a review of the literature.

A case of acute neurogenic pulmonary oedema following a generalized tonic clonic seizure is presented. The condition is rare and the exact pathophysiology is unknown. Pulmonary oedema is a serious complication of epileptic seizures, however, and the importance of awareness of this condition and its treatment is emphasized by a high mortality rate in older epileptics and by the demonstration of pulmonary oedema in recent series of young epileptics who died suddenly.

Kindling with the beta-carboline FG7142 suggests separation between changes in seizure threshold and anxiety-related behaviour.

The aim of this paper was to determine whether the prolonged decrease in seizure threshold produced by chemical kindling was accompanied by behavioural changes in tests of anxiety and aggression. The responses of rats in five tests were examined after chronic treatment with the benzodiazepine inverse agonist FG7142. This treatment caused chemical kindling, so that the originally proconvulsant drug caused full seizures. This effect is very long-lasting, and our previous work with mice had suggested that it might be accompanied by an increase in anxiety-related behavior. In the present work no s

Enhancer of seizure: a new genetic locus in Drosophila melanogaster defined by interactions with temperature-sensitive paralytic mutations.

Mutations in the enhancer of seizure (e(sei] locus have been isolated on the basis of their ability to cause temperature-induced paralysis of alleles at the seizure (sei) locus at temperatures at which these mutations ordinarily do not paralyze. This enhancer is specific to the seizure locus and is without effect on other temperature-sensitive paralytic mutants including para, nap, tip-E and shi. This suggests that the enhancer responds specifically to the mechanism of paralysis mediated by the seizure mutations. The e(sei) is a recessive mutation which maps to 39.0 on the left arm of chromoso

Long-term prognosis of convulsive disorders in the first year of life: mental and physical development and seizure persistence.

A follow-up study was made on 304 children (164 boys, 140 girls) with convulsive disorders, excluding occasional convulsions, in the first year of life. All patients except 45 who died were followed until 6 years of age or older. At the final follow-up, the subjects were divided into six groups according to the degree of mental and physical development (groups I-VI). Seizures were regarded as absent if the patient had been seizure free for more than 3 years. At the final follow-up, seizures had ceased in 57.7%, and 43.4% had normal mental and physical development (group I). As to the initial d

Effects of withdrawal from long-term barbital treatment on open-field behaviour and seizure susceptibility of rats.

Rats withdrawn from long-term barbital treatment were tested both for their activity in an open-field and for their sensitivity to convulsant stimuli. Barbital withdrawal induced a significant increase on locomotion and rearing frequency. Drug removal also made the rats more sensitive to convulsions elicited by sound and maximal electroshock. A positive correlation was found between locomotion plus rearing frequency and audiogenic-induced seizures 3, 4 and 5 days after barbital withdrawal. The results show that exploratory behaviour evaluated through the open-field method can be used as a reli

Vitamin D3 prevents epileptic seizures by modulating the ras signalling pathway via Gnb1 and Casr in acute epilepsy mouse models.

OBJECTIVE: This study aimed to investigate the neuroprotective effects of vitamin D3 (cholecalciferol, hereinafter referred to as VitD) in acute epilepsy mouse models by examining its modulation of the calcium-sensing receptor (Casr), the Ras signalling pathway, and the potential mediator role of guanine nucleotide-binding protein subunit beta-1 (Gnb1). METHODS: Acute epilepsy was induced in 135 C57BL/6 J mice via intraperitoneal injection(i.p.) of pentylenetetrazole (PTZ) or kainic acid (KA). Mice were randomly assigned to receive VitD pretreatment, valproate (VPA) emergency treatment, or a

Steroids for Drug-Resistant Seizures and Prolonged Stroke-Like Episode in a Patient With Sturge-Weber Syndrome: A Case Report.

INTRODUCTION: Sturge-Weber syndrome (SWS) brain involvement has been associated with impairments in the blood-brain barrier (BBB) and microglial activation within involved cortical regions. Acute neurological crises, including seizures, stroke-like episodes, and/or significant headaches, are common in these patients. This report describes in detail an adolescent with SWS and acute drug-resistant status epilepticus, headache, and stroke-like episode who improved clinically when treated with high-dose steroids. Review of medical records identified two other patients previously treated with stero

Seizures as the initial presentation of cerebral venous sinus thrombosis in an elderly patient: a case report and literature review.

INTRODUCTION AND IMPORTANCE: Cerebral venous sinus thrombosis (CVST) is a rare cerebrovascular disorder with heterogeneous clinical presentations. Although headache is the most common symptom, atypical presentations without headache may lead to diagnostic challenges, particularly in elderly patients. Seizures are a recognized manifestation of CVST but are occasionally the sole initial presentation. CASE PRESENTATION: We describe a 64-year-old man with recurrent seizures and transient right-sided weakness, without headache. Brain magnetic resonance imaging revealed left frontal encephalomalaci

Prolonged Psychogenic Non-epileptic Seizures Manifested Immediately after Minor Sport-related Head Injury in a Young Judo Athlete:A Case Report.

Psychogenic non-epileptic seizures (PNES) are often associated with a history of mild traumatic brain injury, which may require differentiation with posttraumatic epilepsy, but usually there is a sufficiently long interval between an episode of trauma and their manifestations. Herein we report a case of a young female judo wrestler who experienced PNES immediately after minor head injury sustained during sport contest. It can be speculated that pathophysiological background of PNES in athletes may differ from general population and that disease in such cases may be related to stress of competi

Intramuscular midazolam versus intravenous clonazepam for prehospital treatment of generalized tonic-clonic seizures: A randomized double-blind non-inferiority trial.

BACKGROUND: Seizures are a common neurological emergency associated with significant morbidity and mortality. In the prehospital setting, intravenous (IV) benzodiazepines are commonly used as first-line therapy; however, establishing IV access may delay treatment in actively convulsing patients. Intramuscular (IM) midazolam provides a rapid alternative, but comparative evidence with IV clonazepam remains limited. AIM: To evaluate whether IM midazolam is non-inferior to IV clonazepam for the prehospital treatment of generalized tonic-clonic seizures. METHODS: We conducted a prospective, rando
