Interventional Effects of Edible Bird's Nest or Sialic Acids on Sepsis-Induced Immunosuppression in Mice.

Edible bird's nest (EBN) and its key component, sialic acid (SA), were investigated for their potential to counteract sepsis-induced immunosuppression. In a murine "two-hit" sepsis model, EBN and SA significantly enhanced survival, ameliorated multi-organ damage, and reduced bacterial dissemination. Crucially, they reversed immunosuppression by restoring the TNF-α-secreting capacity of immune cells. Proteomic and KEGG analyses identified T cell proliferation as a central mechanism, which was validated by CFSE assay. Furthermore, we demonstrated that this enhanced T cell proliferation is mediat

Predictors of death in diabetes-associated Enterobacteriaceae sepsis: A Mendelian randomization and machine learning study.

Diabetes mellitus is a common comorbidity that may increase susceptibility to severe infections and worse outcomes. We aimed to identify clinical predictors of in-hospital mortality in diabetic patients with Gram-negative Enterobacteriaceae sepsis using a data-driven feature-selection and machine learning (ML) approach, and to examine the causal relationship between diabetes and sepsis using Mendelian randomization (MR). We performed a retrospective cohort study of patients admitted to The Second Affiliated Hospital of Harbin Medical University between January 2018 and August 2025 with diabete

Changes in plasma inflammatory injury factors and their clinical significance in pediatric bacterial pneumonia and sepsis.

Bacterial pneumonia and sepsis are common severe infectious diseases in pediatrics. The overlapping early clinical manifestations of these two conditions often lead to misdiagnosis. Establishing a precise early differential diagnostic method is crucial for improving patient outcomes. This retrospective cohort study enrolled 126 children with bacterial pneumonia, 116 children with sepsis, and 65 healthy children as controls. Plasma procalcitonin (PCT), C-reactive protein (CRP), and peripheral blood leukocyte subset indicators were measured, and their differential diagnostic efficacy was analyze

The Role of Routine Surveillance Cultures in Optimising Sepsis Management in High-Risk Patient Groups.

BACKGROUND: Sepsis remains a leading cause of morbidity and mortality, particularly when caused by multidrug-resistant organisms (MDROs). Early identification of colonising or infecting pathogens may inform initial antimicrobial selection. Surveillance cultures, providing microbiological data prior to infection onset, could guide timely and targeted therapy. This retrospective study analysed routine surveillance culture results from patients with bloodstream infection (BSI) episodes, assessing pathogen prevalence, resistance phenotypes, and concordance with specimen type in haemato-oncology (H

Longitudinal and adjusted prognostic assessment of calprotectin in Gram-negative sepsis with acute respiratory failure.

Sepsis caused by Gram-negative bloodstream infection is characterized by dynamic host responses, but the longitudinal and incremental prognostic value of calprotectin remains uncertain. We conducted a multicenter retrospective observational study of 82 adults with Gram-negative bloodstream infection, sepsis, and acute respiratory failure. Serum calprotectin, routine biomarkers, cytokines, and growth factors were measured at diagnosis before antibiotic therapy (T0), day 3 (T1), and day 7 (T2), and compared according to 28-day survival. Sixty-four patients survived, and 18 died. Calprotectin dec

The burden of neonatal sepsis and its risk factors in Africa. a systematic review and meta-analysis.

BACKGROUND: Neonatal sepsis is a significant cause of newborn mortality in low- and middle-income countries (LMICs). Together, infections, complications of preterm birth, and intrapartum-related conditions contribute to nearly 90% of all neonatal deaths. Africa experiences high rates of neonatal deaths due to sepsis, with insufficient prevention efforts. Understanding the burden of neonatal sepsis is essential to reducing these deaths in the region. This study aims to estimate the pooled magnitude of neonatal sepsis and identify its associated risk factors in Africa. METHOD: For this study, w

A mouse model of sepsis-associated DIC induced by Kappa-carrageenan and Lipopolysaccharides: Establishment and characteristics.

BACKGROUND: No animal models fully replicate the pathogenesis and clinical features of sepsis-associated disseminated intravascular coagulation (DIC), which hinders mechanistic understanding and treatment development. Kappa-carrageenan (KCG) and lipopolysaccharides (LPS) induce thrombosis and systemic inflammation in mice, respectively. The combination of LPS and KCG provides a promising method for establishing a mouse model of sepsis-associated DIC. OBJECTIVE: This study aimed to establish a standardized mouse model of sepsis-associated DIC using KCG and LPS. METHODS: Kunming (KM) mice were

The role of lncRNAs in sepsis-induced acute lung injury: Molecular mechanisms and therapeutic potential.

Sepsis, a life-threatening syndrome, results from a dysregulated immune and hemostatic response, contributing to acute lung injury (ALI) and its progression into acute respiratory distress syndrome (ARDS). The development of septic ALI is complex, involving excessive inflammatory mediator production that damages endothelial and epithelial cells, leading to vascular leakage, edema, and vasodilation-key factors in ALI pathogenesis. Long noncoding RNAs (lncRNAs), over 200 nucleotides in length, play critical roles in various biological processes, including sepsis regulation. They exhibit both pro

A Quinoxaline Derivative as a New Therapeutic Agent for Sepsis through Suppression of TLR4 Signaling Pathways.

Sepsis is a severe systemic inflammatory syndrome and one of the leading causes of global morbidity and mortality. Preclinical studies have identified several quinoxaline-based compounds with anti-inflammatory properties, but their effects in sepsis have not been investigated. This study aimed to identify a quinoxaline derivative with anti-inflammatory properties in sepsis. Examining the inflammatory response of primary mouse macrophages to Lipopolysaccharides (LPS) revealed that 2-methoxy-N-(3-quinoxalin-2-ylphenyl)benzamide (2-MQB) is a promising molecule. It suppressed the production of sev

Design of a quinoxalinone-based AIE probe for the detection of ROS in in vitro and in vivo sepsis models.

Sepsis is one of the major causes of long-term mortality; the identification of potential biomarkers and developing specific and sensitive imaging and detection methods are crucial for timely diagnosis and progression evaluation. Reactive oxygen species (ROS) may serve as a potential detection and imaging marker for sepsis. Herein, we designed and synthesized a near-infrared quinoxalone framework-based aggregation-induced emission probe (QuinoNS NPs). We evaluated the selectivity, cytotoxicity, and detection and imaging ability in an in vitro LPS induced inflammatory model and an in vivo sepsi

Identification and Verification of Necroptosis-Related Genes in Patients With Sepsis by Bioinformatic Analysis and Molecular Experiments.

Although necroptosis is an emerging mechanism of multiple organ dysfunction in sepsis, data on the mechanistic link between necroptosis and sepsis are scarce. Bioinformatic analysis was performed to compare the gene profiles between the sepsis (n = 133) and healthy control (n = 12) groups and identify necroptosis-related differentially expressed genes (DEGs). The identified necroptosis-related DEGs were verified by three-step molecular experiments: (1) quantitative real-time PCR and enzyme-linked immunosorbent assay; (2) cell culture, transfection and Western blotting; and (3) cytokine array w
