The nuclear body is a multiprotein complex that may have a role in the regulation of gene transcription. This structure is disrupted in a variety of human disorders including acute promyelocytic leukemia and viral infections, suggesting that alterations in the nuclear body may have an important role in the pathogenesis of these diseases. In this study, we identified a cDNA encoding a leukocyte-specific nuclear body component designated Sp110. The N-terminal portion of Sp110 was homologous to two previously characterized components of the nuclear body (Sp100 and Sp140). The C-terminal region of

We report here the first three-dimensional structure of alpha-latrotoxin, a black widow spider neurotoxin, which forms membrane pores and stimulates secretion in the presence of divalent cations. We discovered that alpha-latrotoxin exists in two oligomeric forms: it is dimeric in EDTA but forms tetramers in the presence of Ca2+ or Mg2+. The dimer and tetramer structures were determined independently at 18 A and 14 A resolution, respectively, using cryo-electron microscopy and angular reconstitution. The alpha-latrotoxin monomer consists of three domains. The N- and C-terminal domains have been

The spontaneous mouse mutant Dominant megacolon (Dom) is a valuable model for the study of human congenital megacolon (Hirschsprung disease). Here we report that the defect in the Dom mouse is caused by mutation of the gene encoding the Sry-related transcription factor Sox10. This assignment is based on (i) colocalization of the Sox10 gene with the Dom mutation on chromosome 15; (ii) altered Sox10 expression in the gut and in neural-crest derived structures of cranial ganglia of Dom mice; (iii) presence of a frameshift in the Sox10 coding region, and (iv) functional inactivation of the resulti

Theoretical methods to delineate antibody inducing epitopes have been employed to predict antigenic determinants on envelope glycoprotein (gpE) of Japanese encephalitis (JE), West Nile (WN) and Dengue (DEN) I-IV viruses. A predicted region on JE virus gpE 74CPTTGEAHNEKRAD87 was synthesized, conjugated to KLH (KLH-peptide) and used in immunization of mice. A mouse monoclonal antibody (MoAb IVB4) reactive to the peptide was also found to react with native JE virus gpE. Characterization of the idiotypic (ID) determinants with the help of polyclonal domain-specific anti-ID antibodies revealed that

Distal radius fractures (DRFs) are among the most common upper extremity fractures encountered in clinical practice. Closed reduction followed by external immobilization remains a fundamental treatment approach; however, the procedure is highly operator-dependent and lacks standardization. This article presents a step-by-step protocol for manual reduction and four-splint immobilization using volar, dorsal, radial, and ulnar splints for acute DRFs. The protocol encompasses patient selection, anesthesia administration, patient positioning, sustained longitudinal traction with countertraction, re

BACKGROUND: With clinical xenotransplantation now underway, optimal patient selection is critical for achieving successful outcomes. Candidates with antibodies targeting Bw4 and Bw6 epitopes on HLA molecules often experience prolonged wait times for allotransplantation because of high calculated panel reactive antibody levels. We investigated whether these antibodies cross-react with class I swine leukocyte antigen (SLA) molecules and could therefore pose a barrier to xenotransplantation.
METHODS: We performed sequence alignment and structural modeling to assess the presence of Bw4 and Bw6 ep

The β-glucosidase from Bacillus thermoamylovorans (Bgl52) has been identified as a thermophilic and acidophilic β-glucosidase, which constitutes the most critical component in the cellulolytic enzyme system for cellulose hydrolysis. Although Bgl52 is a thermophilic enzyme, its poor thermal stability at high temperatures restricts its long-term industrial application under elevated-temperature conditions. In this study, rational design strategies, including two free energy calculation programs (Fireprot and FoldX) and homologous sequence alignment, were integrated to perform single-point and mu

Western flower thrips (WFT), Frankliniella occidentalis (Pergande) (Thysanoptera: Thripidae), is a globally invasive polyphagous pest that relies on olfaction to sense host plants. Odorant-binding proteins (OBPs) in insect sensillar lymph mediate interactions with plant volatiles. However, the physicochemical mechanisms by which OBPs in F. occidentalis bind to host-plant volatiles, the key amino acid residues involved in these interactions and the structural characteristics of the preferred ligands remain unclear. Here, we cloned a novel OBP, FoccOBP8, which belongs to the C-minus OBP subfamil

Mycobacterium tuberculosis (M. tuberculosis) encodes a unique bifunctional AdaA-AlkA enzyme (Rv1317c) that fuses an N-terminal methyltransferase domain with a C-terminal glycosylase-derived domain, an arrangement confined to Actinobacteria and absent in Escherichia coli, where the two functions reside on separate proteins. Multiple sequence alignment across six bacterial species confirmed strict conservation of the four Zn2+-coordinating cysteines in the AdaA domain and the catalytic aspartate in the AlkA domain, while identifying Actinobacteria-specific substitutions (Leu125→Gln329, Tyr222→Va

Sickle cell disease (SCD) is an autosomal recessive hemoglobin disorder that is mainly characterized by the presence of hemoglobin S (HbS; point mutation [Glu6Val] in the beta-globin gene). Under deoxygenated conditions, HbS polymerizes and serves as the primary trigger of oxidative stress in red blood cells (RBCs), promoting polymerization of Band 3, a major membrane scaffold protein that links the lipid bilayer to the spectrin-ankyrin cytoskeletal network. Phosphorylation at key residues within the cytosolic domain of Band 3 induces conformational changes that weaken ankyrin binding and enha

BACKGROUND: Circular RNAs (circRNAs), a conserved class of non-coding RNAs, are hypothesized to play functional roles in diabetic retinopathy (DR), yet their expression landscape and molecular involvement in retinal tissue remain poorly defined.
METHODS: A streptozotocin-induced diabetic mouse model (C57BL/6J, male, n=40) was established, with diabetes confirmed by persistent hyperglycemia (fasting blood glucose >300 mg/dL). Retinal function was assessed by electroretinography (ERG) and vascular pathology by fluorescein fundus angiography (FFA) at 12 weeks post-induction. Retinal circRNA prof

NAD(P)H dehydrogenase [quinone] 1 (NQO1) is a mammalian quinone oxidoreductase, which can react with cell antioxidants and nucleophiles and reduce cell damage. Although NQO1 has been widely studied in mammals, its research in aquatic animals is relatively limited, especially in Nile tilapia (Oreochromis niloticus). In this study, we successfully cloned NQO1 (OnNQO1) of O. niloticus and studied its role in oxidative stress induced by ammonia exposure. The results showed that the open reading frame (ORF) of OnNQO1 was 882 bp, encoding 293 amino acids, and its protein had NAD(P)H dehydrogenase [q

Protein crystallization remains a major bottleneck in X-ray crystallography due to difficulties in achieving favorable molecular arrangements within the crystal lattice. While protein-protein interactions at molecular packing interfaces are crucial for determining crystallization conditions, methods for predicting crystal packing interfaces and systematically exploring crystallization conditions remain limited. In this study, we present MASCL (Molecular Assembly Simulation in Crystal Lattice), a novel approach that integrates AlphaFold with symmetrical docking to simulate crystal packing. To e

Selection pressures often act at the level of protein structure, yet most evolutionary analyses remain confined to linear sequences. Early structure-informed approaches improved interpretation by mapping single-site metrics onto protein structures, and later methods introduced 3D sliding windows to capture spatial neighbourhoods missed by linear analyses. These frameworks, however, are restricted to predefined statistics and narrowly defined 3D window types, limiting the scope of questions that can be addressed. We developed an R package, evo3D, a generalised framework for structure-informed e

PURPOSE: Our study identifies a novel HESX1 variant in two siblings, resulting in isolated growth hormone deficiency (IGHD) associated with empty sella. To the best of our knowledge, this represents the second recognized mutation within the EH1 repressor domain in HESX1. We explore and interpret the potential mechanism, with the aim of guiding pediatricians comprehend this disorder.
METHODS: The clinical data for two Chinese siblings was summarized. Furthermore, multiple sequence alignment and pathogenicity prediction of functional effects through five online algorithms were performed. Additi

The Sox family genes, as a group of transcription factors, are widely expressed in vertebrates and play a critical role in reproduction and development. The present study reported that 26 sox genes were identified from the genome and transcriptome of P. leopardus. The phylogenetic tree construction, chromosome localization, and gene structure analysis were executed to verify the evolutionary relationships, gene duplication, and deletion variations of P. leopardus sox genes in evolution. The sequence alignment revealed the HMG-box domain was highly conserved throughout the Sox gene family. The

BACKGROUND: The optimal sequence of microvascular clamping during free flap transfer is yet to be established. Many surgeons are reluctant to perform temporary declamping and subsequent reclamping during microvascular anastomosis; however, we generally anastomose the artery first and temporarily declamp it before performing venous anastomosis to confirm arterial patency and ensure proper alignment of the flap veins. Herein, we aimed to retrospectively investigate the efficacy and safety of this temporary revascularization method in 126 patients who underwent microvascular head and neck reconst

Candida albicans deploys a variety of mechanisms such as morphological switch and elicitor release to promote virulence. However, the intricate interactions between the fungus and the host remain poorly understood, and a comprehensive inventory of fungal virulence factors has yet to be established. In this study, we identified a C. albicans secretory effector protein Sce1, whose induction and secretion are associated with vagina-simulative conditions and chlamydospore formation. Sequence alignment showed that Sce1 belongs to a Pir family in C. albicans, which is conserved across several fungi
