Reanalysis of Published Histological Data Can Help to Characterize Neuronal Death After Spinal Cord Injury.

Neuronal death is a central event in spinal cord injury (SCI) pathophysiology. Despite its importance, we have a fragmentary vision of the process. In our opinion, the research community has accumulated enough information to provide a more detailed, integrated vision of neuronal death after SCI. This work embeds this vision by creating an open repository to store and share data and results from their analysis. We have employed this repository to upload raw images of spinal cord sections from a mouse model of contusive SCI and used this information to compare manual-, threshold-, and neural net

The Vibrational Signature of Alzheimer's Disease: A Computational Approach Based on Sonification, Laser Projection, and Computer Vision Analysis.

Alzheimer's disease (AD) is the most prevalent form of dementia, and accessible biomarkers for early detection remain limited. This study introduces a biomimetic approach in which brain electrical activity is transformed into sound and vibration, emulating natural sensory encoding mechanisms. Resting-state EEG recordings from 36 AD patients and 29 healthy controls were averaged by group, directly sonified, and used to drive a membrane-laser system that projected dynamic vibrational patterns. This transformation mirrors how biological systems convert electrical signals into sensory representati

Decoding COPD-related cognitive impairment: The protective potential of YTHDF1.

Cognitive impairment is a significant complication in individuals with chronic obstructive pulmonary disease (COPD), which presents a substantial challenge to future healthcare resource allocation. However, the underlying therapeutic targets and mechanisms remain inadequately understood. In a study involving mice with COPD, hippocampal damage and deficits in working memory, spatial learning, and memory capabilities were observed following prolonged exposure to cigarette smoke (CS). CS exposure was found to induce apoptosis in hippocampal cells in vivo. Western blot and RT-qPCR analyses demonst

Personnel exposure studies to multi-source diffusion of bioaerosols under biosafety laboratory - Level 2 plus operating conditions.

Biosafety Laboratories are utilized for the handling of hazardous samples and pathogenic agents. However, the accidental bioaerosol dissemination has triggered many laboratory-acquired infections. Thus, this study elucidated the diffusion of leaked bioaerosols in the laboratory. Three operational conditions and three bioaerosol sources were considered. Firstly, bioaerosol experiments were conducted to measure the spatial concentration of bioaerosols. Subsequently, the visualized airflow and real-time distribution of bioaerosol were obtained using Euler-Lagrange model. Furthermore, the infectio

Reducing the Formation of Toxic Byproducts During the Photochemical Release of Epinephrine.

Engineered light-sensitive molecules offer a sophisticated toolkit for the manipulation of biological systems with both spatial and temporal precision. Notably, artificial "caged" compounds can activate specific receptors solely in response to light exposure. However, the uncaging process can lead to the formation of potentially harmful byproducts. For example, the photochemical release of adrenaline (epinephrine) is accompanied by the formation of adrenochrome, which has neuro- and cardiotoxic effects. To investigate this effect in detail, we synthesized and compared two "caged" epinephrine a

De novo assembly of nuclear stress bodies rearranges and enhances NFIL3 to restrain acute inflammatory responses.

The membrane-less nuclear stress bodies (nSBs), with satellite III (SatIII) RNAs as the hallmark, are present in primates upon sensing stresses. We report that SatⅢ DNAs, SatⅢ RNAs, and 30 nSB proteins assemble into well-organized structures shortly after stresses. The activated SatⅢ heterochromatin loci rapidly expand, resulting in reduced spatial distance and enhanced expression of adjacent genes, including the transcription suppressor NFIL3, which is known to dampen proinflammatory cytokine production. Rearranging NFIL3 loci within the nSB territory enhances NFIL3 chromatin accessibility an

Mitigating Nephrotoxicity Risks and Ranking Drug Candidates Using MALDI Mass Spectrometry Imaging: A Case Study.

Drug-induced nephrotoxicity is a major challenge in drug discovery and development, accounting for nearly a quarter of severe adverse effects in current pharmacotherapy. Antimicrobial use may be associated with this problem, with one-third of nephrotoxicity related to these drugs. During the lead optimization stage of our antibacterial programs, nephrotoxicity was observed with renal tubule degeneration and tubular granular casts. To examine the nephrotoxicity mechanisms and triage compounds, matrix-assisted laser desorption/ionization mass spectrometry imaging (MALDI-MSI) was used to investig

Immunological control of skin development: from homeostasis to developmental pathologies.

The skin serves not only as a physical barrier but also as a dynamic immune organ, where immune cells orchestrate tissue morphogenesis, homeostasis, and repair. Recent advances have revealed that immune cells play pivotal roles during skin development, guiding processes such as vascular formation, epidermal stratification, and hair follicle morphogenesis. In this review, we integrate fundamental mechanistic studies based on mouse models with key clinical observations from human diseases to comprehensively elucidate the contributions of critical immune cell populations-including macrophages, La

Langerin+ Dendritic Cells in Cutaneous Fibrosis: The TGF-β1 Signaling Axis.

Cutaneous fibrosis - including hypertrophic scars and keloids - arises when immune, epithelial, and stromal signals fail to re-equilibrate after injury. Langerin+ dendritic cells (DCs) - epidermal Langerhans cells and dermal cDC1 - sit at the center of this process. These DC subsets generate latent transforming growth factor-β1 (TGF-β1) that keratinocyte integrins αvβ6/αvβ8 locally activate, creating an epidermal "cytokine gate" that restrains immunity in homeostasis yet seeds fibrosis when overdriven. Downstream, active TGF-β1 cooperates with mechanosensitive YAP/TAZ to drive fibroblast activ

Semi-Supervised Thyroid Nodule Detection in Ultrasound Videos.

Deep learning techniques have been investigated for the computer-aided diagnosis of thyroid nodules in ultrasound images. However, most existing thyroid nodule detection methods were simply based on static ultrasound images, which cannot well explore spatial and temporal information following the clinical examination process. In this paper, we propose a novel video-based semi-supervised framework for ultrasound thyroid nodule detection. Especially, considering clinical examinations that need to detect thyroid nodules at the ultrasonic probe positions, we first construct an adjacent frame guide

Alzheimer's disease and sleep disorders: A bidirectional relationship.

Alzheimer's disease (AD) is the most prevalent dementia, pathologically featuring abnormal accumulation of amyloid-β (Aβ) and hyperphosphorylated tau, while sleep, divided into rapid eye movement sleep (REM) and nonrapid eye movement sleep (NREM), plays a key role in consolidating social and spatial memory. Emerging evidence has revealed that sleep disorders such as circadian disturbances and disruption of neuronal rhythm activity are considered as both candidate risks and consequence of AD, suggesting a bidirectional relationship between sleep and AD. This review will firstly grasp basic know

Computational modeling of selective attention differentiates subtypes of amnestic mild cognitive impairment.

Individuals with amnestic mild cognitive impairment (aMCI), a prodromal stage of Alzheimer's disease and other dementias, show inhibition deficits in addition to episodic memory. How the latent processes of selective attention (i.e., from perception to motor response) contribute to these inhibition deficits remains unclear. Therefore, the present study examined contributions of selective attention to aMCI-related inhibition deficits using computational modeling of attentional dynamics. Two models of selective attention - the dual-stage two-phase model and the shrinking spotlight model - were f

Mass Spectrometry Advances in Analysis of Glioblastoma.

Some cancers such as glioblastoma (GBM), show minimal response to medical interventions, often only capable of mitigating tumor growth or alleviating symptoms. High metabolic activity in the tumor microenvironment marked by immune responses and hypoxia, is a crucial factor driving tumor progression. The many developments in mass spectrometry (MS) over the last decades have provided a pivotal tool for studying proteins, along with their posttranslational modifications. It is known that the proteomic landscape of GBM comprises a wide range of proteins involved in cell proliferation, survival, mi

Specific and Polyfunctional T Cell Response Against N-Methyl-d-aspartate Receptor in an Autoantibody-Mediated Encephalitis Model.

BACKGROUND: Anti-N-Methyl-d-aspartate receptor (NMDAR) autoimmune encephalitis (NMDAR AE) is an autoimmune disease characterized by severe psychiatric and neurological symptoms. While the pathogenic role of antibodies (Abs) directed against the GluN1 subunit of NMDAR is well described in this disease, the immune mechanisms involved in the generation of the autoimmune B cell response, especially the role of T helper cells, are poorly understood. Previously, we developed a B-cell-mediated mouse model of NMDAR AE by immunization with a GluN1359-378 peptide that drives a series of symptoms that re

Invisible but Insidious Effects of Microplastics.

Increasing evidence on the adverse health impacts of microplastics (MPs) is available, but their associated risks to the well-being of humans and long-term impacts are poorly understood. An indicator of the remote effects of MPs may be their influence on the rate of aging. To assess the effects of MPs on the aging process, we used accelerated senescence OXYS rats that develop a complex of geriatric diseases. We prepared the polyethylene terephthalate MPs (2-6 microns in size) and in OXYS and Wistar (maternal strain) rats assessed the influence of chronic administration of MPs (10 or 100 mg/kg

Neuroinflammatory Biomarkers in Alzheimer's Disease: From Pathophysiology to Clinical Implications.

The identification of neuroinflammation as a critical factor in Alzheimer's disease (AD) has expanded the focus of research beyond amyloid-β and tau pathology. The neuroinflammatory fluid biomarkers GFAP, sTREM2, and YKL-40 have gained attention for their potential in early detection and monitoring of disease progression. Plasma GFAP has demonstrated promise in predicting the conversion from mild cognitive impairment to AD dementia, while sTREM2 highlights microglial activation, although there are conflicting results regarding its dynamics in AD pathogenesis. Advanced imaging techniques, such

Immunologic aspects of asthma: from molecular mechanisms to disease pathophysiology and clinical translation.

In the present review, we focused on recent translational and clinical discoveries in asthma immunology, facilitating phenotyping and stratified or personalized interventions for patients with this condition. The immune processes behind chronic inflammation in asthma exhibit marked heterogeneity, with diverse phenotypes defining discernible features and endotypes illuminating the underlying molecular mechanisms. In particular, two primary endotypes of asthma have been identified: "type 2-high," characterized by increased eosinophil levels in the airways and sputum of patients, and "type 2-low,

Steroid hormone signaling synchronizes cell migration machinery, adhesion and polarity to direct collective movement.

Migratory cells - either individually or in cohesive groups - are critical for spatiotemporally regulated processes such as embryonic development and wound healing. Their dysregulation is the underlying cause of formidable health problems such as congenital abnormalities and metastatic cancers. Border cell behavior during Drosophila oogenesis provides an effective model to study temporally regulated, collective cell migration in vivo. Developmental timing in flies is primarily controlled by the steroid hormone ecdysone, which acts through a well-conserved, nuclear hormone receptor complex. Ecd
