We report a 79-year-old woman with T-cell prolymphocytic leukemia (T-PLL) who was successfully treated with fludarabine monophosphate. She was admitted to our hospital because of dyspnea on effort. On admission, anemia and hepatosplenomegaly were apparent but lymphadenopathy was absent. Peripheral blood examination showed anemia and leukocytosis with 29.5% abnormal lymphocytes. To assess the involvement of the immune system in Parkinson's disease we studied the phenotype of circulating lymphocytes in 30 untreated and 34 treated patients. We found a numeric decrease in helper T cells (higher in CD4(+)CD45RA(+) than in CD4(+)CD29(+)) and B cells, and a rise in activated, CD4(+)CD25(+) lymphocytes that was correlated with lymphocyte depletion. Coccidia antigen-specific T lymphocyte proliferation induced by Eimeria acervulina was measured in chickens congenic at the major histocompatibility complex and in two unrelated lines. A panel of monoclonal antibodies specific for TcR V gene families was used to study TcR V region expression in 28 cases of malignant and reactive T-cell expansions including four cases of mixed cellularity Hodgkin's disease (HD) and five reactive cases. Gamma delta (γδ) T cells exhibit unique antiviral, antibacterial, and antitumor properties. Advancing the fundamental understanding of γδ T cell immunobiology and unlocking their full therapeutic potential requires detailed immunophenotypic profiling alongside optimized strategies for ex vivo activation and expansion. In this review, we synthesize current knowledge of γδ T cell immunobiology, emphasizing (i) comprehensive phenotypic characterization using flow cytometry and (ii) emerging approaches for their therapeutic implementation, with a focus on activation and ex vivo expansion methods.
