Sonographic findings in tuberculous epididymo-orchitis.

Genitourinary tuberculosis is the most common manifestation of extrapulmonary tuberculosis. The epididymides, the seminal vesicles, and the prostate are the most commonly infected sites. The testes are rarely involved. We describe the unusual sonographic findings in a case of tuberculous epididymo-orchitis that presented with multiple small, hypoechoic nodules within the testis.

Tuberculous orchitis co-existing with tuberculosis of the sternum--case report.

The sternum and testis are infrequent localisations for extra pulmonary tuberculosis. The initial response to a co-existing lesion, as this case depicted is, to implicate a malignant process of some sort. A high index of suspicion is needed to overcome this diagnostic riddle. Biopsy of the lesion and bacteriological work-up are essential ingredients of management.

Gonadal Transcriptomic Atlas Reveals Conserved and Sex-Biased Regulatory Networks in Black-Spotted Croaker (Protonibea diacanthus).

The black-spotted croaker (Protonibea diacanthus) is a commercially important marine fish, in which males possess swimbladders of superior market value. To establish a molecular foundation for all-male breeding, this study presents the first comprehensive gonadal transcriptomic analysis of this species. RNA sequencing of ovary and testis identified 8,251 sex-biased differentially expressed genes. Functional enrichment analyses revealed their significant involvement in key reproductive pathways, including ovarian steroidogenesis and oocyte meiosis. Our analysis validated the sexually dimorphic

Gut Microbial Dysbiosis and Male Reproductive Health: Current Insights and Future Directions.

Male infertility contributes to approximately half of all infertility cases, with a substantial proportion remaining idiopathic. Emerging evidence implicates the gut microbiome as a regulator of male reproductive health through a proposed gut-testis axis. Few studies have shown that gut microbial dysbiosis may impair sperm quality via multiple mechanisms, including disruption of endocrine function (e.g., reduced testosterone production), alterations in microbial-derived metabolites, and impaired testicular energy metabolism. Increased intestinal permeability and systemic inflammation may furth

Loss of TEX30 leads to male subfertility in mice.

Tex30, a member of the "testis-expressed" genes, has been implicated in spermatogenesis due to its high expression in the testes, localized in both the cytoplasmic and soluble nuclear fractions of testicular cells. However, its precise role in germ cell development remains unclear. In this study, we generated Tex30 knockout (KO) mice by utilizing the CRISPR/Cas9-mediated gene KO approach and comprehensively investigated its function in reproduction. The results revealed that the male Tex30 KO mice exhibited increased apoptosis in metaphase I spermatocytes, premature dissociation of XY chromoso

Bdyof is a Y-chromosome-specific gene required for male development in Bactrocera dorsalis.

BACKGROUND: In many organisms, the Y chromosome contains important genes associated with sex determination and male reproductive development. However, there have been few studies of Y-chromosome-specific genes in non-model species due to the incomplete information of Y chromosome genome and difficulty in sequencing. Here, we screened 90 candidate Y-specific sequences in a constructed transcriptome assembly library by using the chromosome quotient method, among which 11 were unreported sequences associated with male reproductive development, including Bactrocera dorsalis Y-specific Oligozoosper

Roles of Cep215/Cdk5rap2 in establishing testicular architecture for mouse male germ cell development.

Cep215/Cdk5rap2 is a centrosome protein crucial for directing microtubule organization during cell division and morphology. Cep215 is a causal gene of autosomal recessive primary microcephaly type 3, characterized by a small brain size and a thin cerebral cortex. Despite previous attempts with Cep215 knockout (KO) mice to elucidate its developmental roles, interpreting their phenotypes remained challenging due to potential interference from alternative variants. Here, we generated KO mice completely lacking the Cep215 gene and investigated its specific contributions to male germ cell developme

New Insights into D-Aspartate Signaling in Testicular Activity.

D-aspartate (D-Asp) is an amino acid found in high concentrations in the testis and pituitary gland. Increasing evidence suggests that D-Asp promotes spermatogenesis by activating testosterone production in the Leydig cells via LH release from the pituitary gland. In vitro studies indicate that D-Asp may also influence steroidogenesis and spermatogenesis through autocrine and paracrine signals. D-Asp enhances StAR and steroidogenic enzyme expressions, facilitating testicular cell proliferation via the GluR/ERK1/2 pathway. Moreover, it supports spermatogenesis by enhancing the mitochondrial fun

DMRT1-mediated regulation of TOX3 modulates expansion of the gonadal steroidogenic cell lineage in the chicken embryo.

During gonadal sex determination, the supporting cell lineage differentiates into Sertoli cells in males and pre-granulosa cells in females. Recently, single cell RNA-seq data have indicated that chicken steroidogenic cells are derived from differentiated supporting cells. This differentiation process is achieved by a sequential upregulation of steroidogenic genes and downregulation of supporting cell markers. The exact mechanism regulating this differentiation process remains unknown. We have identified TOX3 as a previously unreported transcription factor expressed in embryonic Sertoli cells

ATP activation of peritubular cells drives testicular sperm transport.

Spermatogenesis, the complex process of male germ cell proliferation, differentiation, and maturation, is the basis of male fertility. In the seminiferous tubules of the testes, spermatozoa are constantly generated from spermatogonial stem cells through a stereotyped sequence of mitotic and meiotic divisions. The basic physiological principles, however, that control both maturation and luminal transport of the still immotile spermatozoa within the seminiferous tubules remain poorly, if at all, defined. Here, we show that coordinated contractions of smooth muscle-like testicular peritubular cel

Introducing a new scrotal orchiopexy technique: The fat anchor orchiopexy.

Cryptorchidism is the most common congenital anomaly of the male genital tract. Cryptorchidism is associated with potentially serious consequences including reduced fertility, increases risk of cancer, susceptibility to torsion and traumatic injury, as well as future psychological problems related to an abnormal scrotal appearance. The mainstay of orchiopexy is to achieve an adequate tension-free and permanent scrotal fixation of the testis; one that prevents recurrent ascent. A number of suggestions have been made regarding keeping the testis in the scrotal position once it was brought down i

GLI3 resides at the intersection of hedgehog and androgen action to promote male sex differentiation.

Urogenital tract abnormalities are among the most common congenital defects in humans. Male urogenital development requires Hedgehog-GLI signaling and testicular hormones, but how these pathways interact is unclear. We found that Gli3XtJ mutant mice exhibit cryptorchidism and hypospadias due to local effects of GLI3 loss and systemic effects of testicular hormone deficiency. Fetal Leydig cells, the sole source of these hormones in developing testis, were reduced in numbers in Gli3XtJ testes, and their functional identity diminished over time. Androgen supplementation partially rescued testicul

Oxidative Stress and DNA methylation in male rat pups provoked by the transplacental and translactational exposure to bisphenol A.

The epigenetic changes induced by environmental contaminants play important roles in the inheritance of male reproductive dysfunction. The present study investigated DNA methylation changes and some oxidative stress biomarkers induced by bisphenol A (BPA) in male offspring. A total number of 48 female albino rats were administered orally with 50 μg/kg of BPA/day during gestation and/or lactation periods. At postnatal day 60, the samples were collected from the male pups to assess the serum testosterone, malondialdehyde (MDA) level, superoxide dismutase (SOD), glutathione S-transferase, and glu

Arresting of miR-186 and releasing of H19 by DDX43 facilitate tumorigenesis and CML progression.

Cancer-testis (CT) antigens, rarely in normal tissues except testis, are expressed in many tumor types. In recent years, DDX43 has been shown to be expressed in several malignancies. However, the role of DDX43 during tumorigenesis is not well established. In the present study, we explored the function of DDX43 in chronic myeloid leukemia (CML). We found that DDX43 overexpression in CML cell lines enhanced survival and colony formation, inhibited cell apoptosis, promoted tumorigenesis, and CML progression. In contrast, silencing of DDX43 inhibited cell survival and tumorigenesis. Upregulated H1
