The endoderm forms the innermost layer of the embryonic gut tube. It gives rise to the epithelial and glandular components of many internal organs, including those of the digestive and respiratory systems. Endodermal organs have essential physiological processes such as nutrient absorption, respiration, detoxification and endocrine regulation. Over the past two decades, work in vertebrate embryos and human pluripotent stem cell models has defined the mechanisms that govern endoderm formation, regionalization and organogenesis. These processes are driven by conserved cell signaling pathways act

Deciphering neuronal circuits is pivotal for deepening our understanding of neuronal functions and advancing treatments for neurological disorders. Conventional neuronal tracers suffer from restrictions such as limited penetration depth, high immunogenicity, and inadequacy for long-term and in vivo imaging. In this context, we introduce an aggregation-induced emission luminogen (AIEgen), MeOTFVP, engineered for enhanced neuronal tracing and imaging. MeOTFVP is strategically designed to target cell membranes by integrating into the phospholipid bilayer through its amphipathy. The donor-acceptor

Exosomes, small extracellular vesicles secreted by various cells, play crucial roles in the pathogenesis and treatment of oral diseases. Recent studies have highlighted their involvement in orthodontics, periodontitis, oral squamous cell carcinoma (OSCC), and hand, foot, and mouth disease (HFMD). Exosomes have a positive effect on the inflammatory environment of the oral cavity, remodeling and regeneration of oral tissues, and offer promising therapeutic options for bone and periodontal tissue restoration. In OSCC tumor-derived exosomes promote cancer progression through cell proliferation, mi

End-stage organ failure can result from various preexisting conditions and occurs in patients of all ages, and organ transplantation remains its only treatment. In recent years, extensive research has been done to explore the possibility of transplanting animal organs into humans, a process referred to as xenotransplantation. Due to their matching organ sizes and other anatomical and physiological similarities with humans, pigs are the preferred organ donor species. Organ rejection due to host immune response and possible interspecies infectious pathogen transmission have been the biggest hurd

The impact of proteins and peptides on the treatment of various conditions including ocular diseases over the past few decades has been advanced by substantial breakthroughs in structural biochemistry, genetic engineering, formulation and delivery approaches. Formulation and delivery of proteins and peptides, such as monoclonal antibodies, aptamers, recombinant proteins and peptides to ocular tissues poses significant challenges owing to their large size, poor permeation and susceptibility to degradation. A wide range of advanced drug delivery systems including polymeric controlled release sys

Using bacteria as therapeutic agents against solid tumors is emerging as an area of great potential in the treatment of cancer. Obligate and facultative anaerobic bacteria have been shown to infiltrate the hypoxic regions of solid tumors, thereby reducing their growth rate or causing regression. However, a major challenge for bacterial therapy of cancer with facultative anaerobes is avoiding damage to normal tissues. Consequently the virulence of bacteria must be adequately attenuated for therapeutic use. By placing an essential gene under a hypoxia conditioned promoter, SalmonellaTyphimurium

Preclinical (in vivo) experiments for efficacy and toxicity are predominantly conducted in mice. Nevertheless, human-derived cells, tissues, and organoids are generally rejected by the immune system of mice. Thus, immunodeficient mice are used as xenograft models to overcome immune rejection in cancer research. Xenograft mouse models are divided into ectopic, orthotopic, and metastatic models based on the transplant sites. Xenograft mouse models are established by transplanting cancer cells, tissues, or organoids derived from patients. These models show diverse success rates and production tim

We developed acellular tissue engineered vessels (ATEV) using small intestine submucosa (SIS) incorporating heparin and a novel protein named H2R5. ATEVs were implanted into the arterial circulation of an ovine animal model, demonstrating high primary patency rates over a period of three months. Implanted grafts were infiltrated by host cells, the majority of which were monocytes/macrophages (MC/MΦ), as demonstrated by scRNA sequencing and immunostaining. They also developed functional endothelial and medial layers that deposited new extracellular matrix leading to matrix remodeling and acquis

The treatment of urethral stricture disease and the prevention of restenosis present considerable challenges in the field of urology. Tissue-engineered materials, particularly bacterial cellulose scaffolds, have emerged as promising solutions due to their abundant sources, excellent mechanical properties, and biocompatibility. However, for attaining superior treatment for patients with USD, further modification of bacterial cellulose is necessary. We have fabricated a dual-network scaffold with enhanced antibacterial properties and cytokines absorption ability through in-situ polymerization of

The biliary tract is a ductal network comprising the intrahepatic (IHBDs) and extrahepatic bile duct (EHBDs). Biliary duct disorders include cholangitis, neoplasms, and injury. However, the underlying mechanisms are not fully understood. With advancements in 3D culture technology, cholangiocyte organoids (COs) derived from primary tissues or induced pluripotent stem cells (iPSCs) can accurately replicate the structural and functional properties of biliary tissues. These organoids have become powerful tools for studying the pathogenesis of biliary diseases, such as cystic fibrosis and primary s

Endometrial tissue lines the inner cavity of the uterus and is under the cyclical control of estrogen and progesterone. It is a tissue that is composed of luminal and glandular epithelium, a stromal compartment, a vascular network, and a complex immune cell population. Mouse models have been a powerful tool to study the endometrium, revealing critical mechanisms that control implantation, placentation, and cancer. The recent development of 3D endometrial organoid cultures presents a state-of-the-art model to dissect the signaling pathways that underlie endometrial biology. Establishing endomet

Urothelium is a specialized multilayer epithelium that lines the urinary tract from the proximal urethra to the kidney. In addition to proliferation and differentiation during development, urothelial injury postnatally triggers a robust regenerative capacity to restore the protective barrier between the urine and tissue. Mounting evidence supports the existence of dedicated progenitor cell populations that give rise to urothelium during development and in response to injury. Understanding the cellular and molecular basis for urothelial patterning and repair will inform tissue regeneration ther

With more and more engineered nanoparticles (NPs) being translated to the clinic, the United States Food and Drug Administration (FDA) has recently issued the latest draft guidance on nanomaterial-containing drug products with an emphasis on understanding their in vivo transport and nano-bio interactions. Following these guidelines, NPs can be designed to target and treat diseases more efficiently than small molecules, have minimum accumulation in normal tissues, and induce minimum toxicity. In this Minireview, we integrate this guidance with our ten-year studies on developing renal clearable

Fabry's disease is a rare X-linked inborn error of glycosphingolipid metabolism characterised by an abnormal lipid storage due to a defect of lysozomal alphagalactosidase. The consequence is a storage of glycosphingolipides in all tissues. This storage in vessels's endothelial cells is responsible, in males, for severe ischemic lesions leading to progressive kidney failure, cardiac and cerebral dysfunctions. Similarly, it involves ocular tissues, mainly the cornea, the conjunctiva and the lens. The corneal storage, known as cornea verticillata, is a clinical marker easy to recognize by slit la

Respiratory diseases pose a severe threat to global health, with notable limitations in current diagnosis and treatment, such as insufficient sensitivity of diagnostic tools and a lack of effective targeted therapies. Due to their highly efficient information transmission capabilities and excellent safety profile, exosomes carrying non-coding RNA, particularly microRNA (miRNA), are increasingly attracting attention. Compared with free miRNAs, exosomes can protect miRNAs from nuclease degradation, prolong their circulation time in the body, thereby improving the stability and bioavailability of

Pulmonary artery reconstruction is frequently required for congenital heart disease repair, yet current graft options remain limited by the scarcity of autologous conduits and the poor durability of synthetic grafts, which often fail due to thrombosis, stenosis, and calcification. Challenges are exacerbated in the hypoxic, low-pressure pulmonary circulation. Because a functional endothelium is critical for preventing thrombosis and neointimal hyperplasia, we engineered ready-to-use endothelialized tissue-engineered vascular grafts (TEVGs) using a stepwise shear-preconditioning regimen that mim

Glaucoma is among the leading causes of blindness worldwide that is characterized by irreversible damage to the retinal ganglion cell axons in the lamina cribrosa (LC) region of the optic nerve head (ONH), most often associated with elevated intraocular pressure (IOP). The LC is a porous, connective tissue structure that provides mechanical support to the axons as they exit the eye and the biomechanics of the LC microstructure likely play a crucial role in protecting the axons passing through it. There is a limited knowledge of the IOP-driven biomechanics of the LC microstructure, primarily du

BACKGROUND AND OBJECTIVE: Macrophages are being investigated as adoptive cell therapies for solid tumors because they can migrate into tissues, phagocytose tumor cells, present antigen, secrete cytokines, and interact with stromal and other myeloid cells. However, their plasticity is also a liability. Once transferred into solid tumors, macrophages are exposed to hypoxia, metabolic stress, suppressive cytokines, and tumor-derived signals that may reduce phagocytosis, dampen inflammatory activation, and redirect them toward tumor-associated macrophage (TAM)-like, tumor-supportive phenotypes. Translating preclinical research findings to clinical applications presents challenges due to the metabolic and anatomical disparities between animal models and humans. Although organ-on-a-chip models replicate the organization of human cells, they lack the complexity of the microenvironment and anatomical fidelity. Developing alternative research models with greater physiological and anatomical relevance to humans holds the potential to enhance translational success and reduce reliance on animal experimentation. In this study, we have showcased the technical details and conditions to maintain

Over the past few years, biomaterial-based periodontal tissue engineering has gained popularity. An ideal biomaterial for treating periodontal defects is expected to stimulate periodontal-derived cells, allowing them to contribute most efficiently to tissue reconstruction. The present study focuses on evaluating the in vitro behavior of human periodontal ligament-derived stromal cells (hPDL-MSCs) when cultured on gelatin/Polycaprolactone prototype (GPP) and volume-stable collagen matrix (VSCM). Cells were cultured onto the GPP, VSCM, or tissue culture plate (TCP) for 3, 7, and 14 days. Cell mo
