Glucose dialysate induces mitochondrial DNA damage in peritoneal mesothelial cells.

BACKGROUND: It is known that peritoneal mesothelial cells (PMCs) are denuded in patients undergoing long-term continuous ambulatory peritoneal dialysis (CAPD); the mechanism of damage is not well known. A high quantity of glucose loaded onto PMCs in these patients may generate toxic radicals during the mitochondrial metabolism, leading to mitochondrial DNA damage that accumulates due to the incomplete repair system of this DNA. OBJECTIVE: To study damage to the PMCs of long-term CAPD patients, and to examine whether glucose overload accelerates this damage in vitro. DESIGN: Descriptive clini

Structure, Evolution and Functional Inference on the CDP Gene Family in European Hazelnut.

Food allergy is an IgE-mediated response of the immune system of sensitive subjects to specific proteins. The recent WHO/FAO consultation included hazelnut (Corylus avellana L.) in the list of global priority allergenic ingredients. One of the most common hazelnut allergen groups encompasses the Cupin_1 domain-containing proteins (CDPs). To date, three CDP allergens (Cor a 9, Cor a 11, and Cor a 16) have been deposited into the official WHO/IUIS database. Recently, using a whole-genome approach, the Cor a loci were mapped in four Corylus species. However, the task of identifying novel CDP alle

Delivery architectures for oligonucleotide therapeutics in genetic and rare diseases: GalNAc-based hepatocyte targeting and extrahepatic translation.

Many genetic and rare diseases are molecularly defined but remain difficult to treat because their biology involves intracellular targets, toxic gain-of-function mechanisms, splicing defects, repeat expansions, or chemically undruggable pathways. Although RNA medicines can be designed directly from genetic information, their clinical success depends on delivery architecture rather than sequence design alone. This review examines delivery architectures for oligonucleotide therapeutics, using GalNAc-siRNA as the central clinically validated model. The GalNAc-siRNA platform demonstrates how recep

VSMC-derived 3HAA activates AhR to drive abdominal aortic aneurysm formation via the p62-Nrf2 axis.

BACKGROUND: Abdominal aortic aneurysm (AAA) remains a life-threatening vascular disease without effective pharmacologic therapy. Although 3-hydroxyanthranilic acid (3HAA), a kynurenine-pathway metabolite with dual redox effects, has been implicated in vascular diseases. How 3HAA instigates AAA is poorly defined. METHODS: We combined human AAA tissues, genetically modified mice, pharmacologic kynurenine-pathway inhibition, and mechanistic vascular smooth muscle cell (VSMC) studies to define 3HAA-AhR signaling in aneurysm formation. RESULTS: In angiotensin (Ang) II-infused apoE-/- mice, plasma

Gut microbiota regulation of the IL-6/STAT3 pathway: Linking hematopoiesis to colorectal cancer progression.

BACKGROUND AND AIM: Chronic gut microbiota-driven inflammation fuels colorectal cancer (CRC) progression, but how microbial signals influence systemic immunity and tumor growth remains poorly defined. This review synthesizes evidence on gut microbiota regulation of the interleukin-6 (IL-6)/signal transducer and activator of transcription 3 (STAT3) pathway as the central mechanistic link connecting intestinal dysbiosis, altered hematopoiesis, and CRC progression. Other inflammatory pathways and therapies are discussed only as they intersect with this core axis. METHODS: A structured search of

Peripheral blood immune profiling reveals key signatures in newly diagnosed NK/T cell lymphoma patients.

RATIONALE: Natural killer/T-cell lymphoma (NKTCL) is an aggressive Epstein-Barr virus (EBV)-associated non-Hodgkin lymphoma with a poor prognosis. Recent genomic and transcriptomic studies of tumor tissues have advanced our understanding of NKTCL pathogenesis, but the systemic immune profile at initial diagnosis remains incompletely elucidated. METHODS: In this study, we characterized the immune landscape of peripheral blood mononuclear cells (PBMCs) from 20 newly diagnosed NKTCL patients and 12 healthy donors using single-cell RNA sequencing and confirmed our results through flow cytometry a

Minimally expanded breast cancer tumor-infiltrating-lymphocytes provide guidance for therapeutic selection.

INTRODUCTION: The analysis of tumor-infiltrating lymphocytes often requires techniques that expand their numbers, potentially introducing bias. To address this, we performed a detailed analysis of minimally cultured TILs to evaluate whether this approach better preserves their characteristics. METHODS: The TIL culture method was based solely on tumor tissue with low IL-2 supplementation to minimize artificial alterations. The validity of this approach was confirmed by the correlation between CD3+ T cell percentages in cultures and infiltration patterns observed by immunohistochemistry. Immuno

Single-cell transcriptomic landscape of sciatic nerve after transection injury.

Peripheral nerve injuries, particularly those affecting the sciatic nerve, often result in incomplete functional recovery due to the limited regenerative capacity of adult peripheral nerves. To elucidate the cellular and molecular mechanisms underlying nerve regeneration, we performed single-cell RNA sequencing (scRNA-seq) on rat sciatic nerve tissues at seven time points (Days 0, 1, 3, 5, 7, 10, and 14) following transection injury. Through unsupervised clustering, we identified four major cellular compartments-neurofibroblasts (NFs), glial cells (Glis), immune cells, and vascular cells-and d

Trained Immunity in Bladder ILC3s Enhances Mucosal Defense Against Recurrent Urinary Tract Infections.

Background: Urinary tract infections (UTIs) rank among the most prevalent infectious diseases globally, with recurrent UTIs (rUTIs) posing substantial therapeutic challenges due to the lack of durable protective immunity. While trained immunity augments innate immune responses, its induction and functional significance in bladder-resident group 3 innate lymphoid cells (ILC3s) remain unknown. This study investigates whether ILC3s develop trained immunity following uropathogenic Escherichia coli (UPEC) exposure and how they contribute to mucosal defense against rUTIs. Methods: The ILC3 counts we

Comparative study for evaluation of Ginkgo biloba extract and gold nanoparticle-encapsulated metformin against nephrotoxicity caused by potassium bromate in Sprague Dawley rats.

BACKGROUND: Potassium bromate (KBrO3) is a potent oxidizing agent that can initiate the generation of free radicals through xenobiotic metabolic pathways, according to more research. This chemical disturbs cellular redox balance, causing structural and functional damage to particular tissues and macromolecules. AIM: This study explored the therapeutic impact of a blend of Ginkgo biloba extract and metformin, delivered via gold nanoparticles (AuNPs), in reducing the toxicity of KBrO3 in the kidneys. METHODS: Rats were categorized into eight groups: a control group, a group receiving G. biloba

Fibroblast reprogramming in the dura mater of NTG-induced migraine-related chronic hypersensitivity model drives monocyte infiltration via Angptl1-dependent stromal signaling.

BACKGROUND: Migraine, characterized by recurrent episodes of severe headache, remains mechanistically enigmatic. While traditional theories emphasize trigeminovascular activation, the role of meningeal stromal-immune crosstalk in disease chronicity is poorly understood. METHODS: A migraine-related chronic hypersensitivity model was utilized via intermittent intraperitoneal nitroglycerin (NTG, 10 mg/kg, every other day for 9 days) and peripheral mechanical hypersensitivity was assessed using von Frey filaments. Single-cell RNA sequencing (scRNA-seq) was performed on dura tissues to construct a

Immunosuppressants Rewire the Gut Microbiome-Alloimmune Axis Through Time-Dependent and Tissue-Specific Mechanisms.

BACKGROUND: Lifelong immunosuppressive therapy is required to prevent allograft rejection in organ transplantation. Current immunosuppressants effectively suppress adaptive and innate immune responses, but their broad, antigen-non-specific effects often result in severe off-target complications. It remains a significant unmet medical need in transplant medicine. RESULTS: In this study we investigated immunosuppressant effects of four major immunosuppressant classes, including tacrolimus, prednisone, mycophenolate mofetil (MMF), and fingolimod (FTY), on the gut microbiome, metabolic pathways,

Development and Preclinical Evaluation of a Copper-64-Labeled Antibody Targeting Glycine-Alanine Dipeptides for PET Imaging of C9orf72-Associated Amyotrophic Lateral Sclerosis/Frontotemporal Dementia.

Aggregating poly(glycine-alanine) (poly-GA) is derived from the unconventional translation of the pathogenic intronic hexanucleotide repeat expansion in the C9orf72 gene, which is the most common genetic cause of frontotemporal dementia (FTD) and amyotrophic lateral sclerosis (ALS). Poly-GA accumulates predominantly in neuronal cytoplasmic inclusions unique to C9orf72 ALS/FTD patients. Poly-GA is, therefore, a promising target for PET/CT imaging of FTD/ALS to monitor disease progression and therapeutic interventions. A novel 64Cu-labeled anti-GA antibody (mAb1A12) targeting the poly-GA protein

Low-Dose Interleukin-2 Reverses Traumatic Brain Injury-Induced Cognitive Deficit and Pain in a Murine Model.

OBJECTIVE: Despite the high prevalence, mild traumatic brain injury (mTBI)-induced chronic headache and cognitive deficits are poorly understood and lack effective treatments. Low-dose interleukin-2 (LD-IL-2) treatment soon after mTBI or overexpressing IL-2 in brain astrocytes prior to injury protects mice from developing post-traumatic headache (PTH)-related behaviors and cognitive decline. The present study addresses a clinically relevant knowledge gap: whether LD-IL-2 treatment long after the initial injury is still effective for chronic PTH and cognitive deficits. METHODS: mTBI was induce

Homeodomain-interacting protein kinase maintains neuronal homeostasis during normal Caenorhabditis elegans aging and systemically regulates longevity from serotonergic and GABAergic neurons.

Aging and the age-associated decline of the proteome is determined in part through neuronal control of evolutionarily conserved transcriptional effectors, which safeguard homeostasis under fluctuating metabolic and stress conditions by regulating an expansive proteostatic network. We have discovered the Caenorhabditis elegans homeodomain-interacting protein kinase (HPK-1) acts as a key transcriptional effector to preserve neuronal integrity, function, and proteostasis during aging. Loss of hpk-1 results in drastic dysregulation in expression of neuronal genes, including genes associated with n
